CD4+ T Cell Interstitial Migration Controlled by Fibronectin in the Inflamed Skin.

The extracellular matrix (ECM) is extensively remodeled during inflammation providing essential guidance cues for immune cell migration and signals for cell activation and survival. There is increasing interest in the therapeutic targeting of ECM to mitigate chronic inflammatory diseases and enhance access to the tumor microenvironment. T cells utilize the ECM as a scaffold for interstitial migration, dependent on T cell expression of matrix-binding integrins αVβ1/αVβ3 and tissue display of the respective RGD-containing ligands. The specific ECM components that control T cell migration are unc…

Urinary proteome of dogs with kidney injury during babesiosis.

BACKGROUND: Acute kidney injury is the most frequent complication of babesiosis in dogs and may provide a natural model for identifying earl … g hormone receptors, inflammatory responses mediated by chemokine and cytokine signalling pathways, integrins, interleukins, and TGF-β (transforming growth factor β) pathways. Those pathways were linked by interleukin-13, bone morphogenetic protein 7, α2(1) collagen, and tyrosine protein kinase Fer, which are potential biomarkers of damage during babesiosis …

On the Cellular and Molecular Mechanisms of Drug-Induced Gingival Overgrowth.

INTRODUCTION: Gingival overgrowth has been linked to multiple factors such as adverse drug effects, inflammation, neoplastic processes, and … n and glycosaminoglycans. Drug-induced gingival overgrowth confirms the key role of collagenase and integrins, membrane receptors present in the fibroblasts, due to their involvement in the catabolism of collagen. The three drug categories implicated: calcineuron inhibitors (immunosuppressant drugs), calcium channel blocking agents and anticonvulsant drugs …

Nonhistone Protein Acetylation Reprograms the ECM-Receptor Interaction Pathway: Multiomics Profiling of Cartilage Degeneration in Rat Models of Osteoarthritis.

Nonhistone acetylation is a key epigenetic mechanism in osteoarthritis (OA), but its specific role in cartilage degeneration remains unclear. Here, we integrated transcriptomics, proteomics, and acetyl-proteomics to systematically analyze molecular changes in rat OA cartilage. Multiomics coenrichment identified the ECM-receptor interaction pathway as the core target, reprogrammed by increased acetylation of Fn1, Tnc, and Itgb1. Using 113 machine-learning strategies, we identified 11 core diagnostic genes, with Wisp3 showing the highest efficacy validated in independent data sets. In vitro expe

Identification of proliferating CD103+ CD8+ tissue-resident memory-like T-cells in villitis of unknown etiology in human placenta.

Villitis of unknown etiology (VUE) is a destructive inflammatory lesion of unknown cause in the human placenta. It may be caused by semiallogeneic rejection of fetal tissue by the mother. CD8+ tissue-resident memory T (TRM) cells reside in the peripheral tissues and have an important role in the local defense against reinfection. These cells also contribute to immunopathogenesis under some circumstances and cause local tissue damage and autoimmune disease. In this study, we examine the contribution of CD103+ CD8+ TRM-like cells to the development of VUE. The study included 23 cases of human pl

High expression of ITGB3 ameliorates asthma by inhibiting epithelial-mesenchymal transformation through suppressing the activation of NF-kB pathway.

Integrin β3 (ITGB3) has been identified as an asthma-associated gene; however, its molecular mechanisms remain poorly understood. Epithelial-mesenchymal transition (EMT) is a critical driver of airway remodeling in asthma, which underpins disease progression. This study aimed to elucidate the role of ITGB3 in asthma pathogenesis by investigating its regulation of EMT. Asthma models were established in vivo using C57BL/6 mice and in vitro with A549 cells, both exposed to house dust mite (HDM) extract. The effects of HDM and ITGB3 modulation on cellular viability, apoptosis, and inflammatory cyt

The CTSZ-TRA2A-IL32 axis defines a targetable macrophage-dependent pathway in metastatic prostate cancer.

BACKGROUND: Emerging evidence underscores the pivotal role of M2-polarized tumor-associated macrophages (M2-TAMs) in orchestrating immunosuppressive tumor microenvironments that fuel metastatic dissemination in prostate cancer (PCa), yet the fundamental mechanisms governing M2-TAM trafficking in lethal PCa progression remain poorly understood. METHODS: Multi-cohort transcriptomic analyses were performed to identify metastasis-associated genes, with CTSZ prioritized as a key cathepsin linked to prostate cancer progression. Circulating tumor and bone metastatic mouse models were employed to inv

ITGB4 is a prognostic biomarker and correlated with lung adenocarcinoma brain metastasis.

BACKGROUND: The aim of this study is to explore the prognostic value and immune signature of ITGB4 expression in lung adenocarcinoma (LUAD) brain metastasis. METHODS: We comprehensively screened genes associated with LUAD brain metastasis by integrating datasets from the GEO database and TMT-based quantitative proteomics profiles. Univariable survival and Multivariate Cox analysis was used to compare several clinical characteristics with survival, and a risk model was constructed. The biological functions were explored via GO and KEGG analysis. Gene set enrichment analysis (GSEA) was performe

PEX3 promotes regenerative repair after myocardial injury in mice through facilitating plasma membrane localization of ITGB3.

The peroxisome is a versatile organelle that performs diverse metabolic functions. PEX3, a critical regulator of the peroxisome, participates in various biological processes associated with the peroxisome. Whether PEX3 is involved in peroxisome-related redox homeostasis and myocardial regenerative repair remains elusive. We investigate that cardiomyocyte-specific PEX3 knockout (Pex3-KO) results in an imbalance of redox homeostasis and disrupts the endogenous proliferation/development at different times and spatial locations. Using Pex3-KO mice and myocardium-targeted intervention approaches, t

Anti-human platelet antigen-5b antibodies and fetal and neonatal alloimmune thrombocytopenia; incidental association or cause and effect?

Most cases of fetal and neonatal thrombocytopenia (FNAIT) are caused by maternal anti-human platelet antigen-1a antibodies (anti-HPA-1a). Anti-HPA-5b antibodies are the second most common antibodies in suspected FNAIT cases. Given the high prevalence of anti-HPA-5b antibodies in pregnant women delivering healthy newborns, the association with FNAIT may be coincidental. This review of the literature related to FNAIT using the MEDLINE database was conducted according to PRISMA guidelines. A retrospective analysis of a single-centre cohort of 817 suspected FNAIT cases was conducted. The pooled pr

Marginating transitional B cells modulate neutrophils in the lung during inflammation and pneumonia.

Pulmonary innate immunity is required for host defense; however, excessive neutrophil inflammation can cause life-threatening acute lung injury. B lymphocytes can be regulatory, yet little is known about peripheral transitional IgM+ B cells in terms of regulatory properties. Using single-cell RNA sequencing, we discovered eight IgM+ B cell subsets with unique gene regulatory networks in the lung circulation dominated by transitional type 1 B and type 2 B (T2B) cells. Lung intravital confocal microscopy revealed that T2B cells marginate in the pulmonary capillaries via CD49e and require CXCL13
