RATIONALE: Pairing casino-inspired audiovisual cues with reward delivery significantly increases risky decision-making in laboratory-based gambling tasks across rats and humans. The neurocognitive mechanisms underlying this effect are unclear. Data from the rat gambling task (rGT) suggests win-paired cues render choice insensitive to reinforcer devaluation, indicating habitual control of decision-making. Histone deacetylase enzymes (HDACs) negatively mediate gene expression. Inhibiting this process can facilitate the formation of long-term memories and accelerate habit formation.

BACKGROUND AND OBJECTIVE: There is a huge amount of rare diseases, many of which have associated important disabilities. It is paramount to know in advance the evolution of the disease in order to limit and prevent the appearance of disabilities and to prepare the patient to manage the future difficulties. Rare disease associations are making an effort to manually collect this information, but it is a long process. A lot of information about the consequences of rare diseases is published in scientific papers, and our goal is to automatically extract disabilities associated with diseases from t

The immune system can suppress tumour development both by eliminating malignant cells and by preventing the outgrowth and spread of cancer cells that resist eradication1. Clinical and experimental data suggest that the latter mode of control-termed cancer-immune equilibrium1-can be maintained for prolonged periods of time, possibly up to several decades2-4. Although cancers most frequently originate in epithelial layers, the nature and spatiotemporal dynamics of immune responses that maintain cancer-immune equilibrium in these tissue compartments remain unclear. Here, using a mouse model of tr

The rising burden of neurodegenerative diseases has exposed critical gaps in the development of diagnostic and therapeutic tools, particularly regarding their limited sensitivity, poor spatiotemporal resolution, and lack of treatment specificity. These shortcomings highlight the need for technologies that enable early disease detection, real-time monitoring of regenerative processes, and precise therapeutic interventions. This review focuses on recent advances in photonic biosensing and optogenetics, examining how these technologies are reshaping neural regeneration research and assessing thei

BACKGROUND: Human papillomavirus (HPV), particularly high-risk types such as HPV16, is associated with several malignancies, including cervical cancer. Existing therapeutic vaccines targeting HPV oncoproteins E6 and E7 show limited immunogenicity and high production costs. Self-amplifying mRNA (saRNA) vaccines offer a promising alternative by enabling robust antigen expression at low doses. This study evaluated the immunogenicity and antitumor efficacy of a novel saRNA vaccine, JJ-saRNA-HPV01, targeting HPV16 E6/E7 in a preclinical mouse model.
METHODS: JJ-saRNA-HPV01, encoding an HPV16 E6-li

Emerging studies find arteriolar dysfunction in offspring with in utero electronic-cigarette (Ecig) exposure, but the long-term effects on offspring's cerebrovascular and neurocognitive health are poorly understood. Ecigs provides a unique opportunity to directly evaluate the contributions of inhaled nicotine from the vehicle e-liquid, which was not possible with cigarettes. Moreover, many Ecigs have variable power settings, which can alter aerosol toxicity. We hypothesize that maternal vaping at different wattages will have variable effects on offspring cerebrovascular function and would be i

Aging and Alzheimer's disease are associated with chronic elevations in neuronal calcium influx via L-type calcium channels. The hippocampus, a primary memory encoding structure in the brain, is more vulnerable to calcium dysregulation in Alzheimer's disease. Recent research has suggested a link between L-type calcium channels and tau hyperphosphorylation. However, the precise mechanism of L-type calcium channel-mediated tau toxicity is not understood. In this study, we seeded a human tau pseudophosphorylated at 14 amino acid sites in rat hippocampal cornu ammonis 1 region to mimic soluble pre

Obesity has dramatically increased over the last 30 years and reaches according to World Health Organization dimensions of a global epidemic. The obesity-associated chronic low-level inflammation contributes to severe comorbidities and directly affects many immune cells leading to immune dysfunction and increased susceptibility to infections. Thus, prophylaxis against vaccine-preventable diseases is crucial, yet the responsiveness to several vaccines is unclear under obesity. In order to assess the responsiveness to tick-borne encephalitis (TBE) vaccine, we revaccinated 37 obese individuals an

Impaired excitatory synaptic function and synapse loss are early hallmarks of Alzheimer's disease (AD). There is strong biochemical, genetic, physiological, and anatomical evidence that the accumulation of soluble amyloid beta (Aβ) oligomers in the brain leads to AD-related synapse dysfunction and cognitive impairment. Long-term potentiation (LTP), a key form of synaptic plasticity for learning and memory, is disrupted in several mouse models harboring familial early-onset AD-linked mutations that lead to Aβ accumulation, and even acute applications of Aβ oligomers block LTP within minutes and

BACKGROUND: Multiple myeloma (MM) remains an incurable disease despite significant advancements in treatment strategies. Chimeric antigen receptor (CAR) T cell therapies targeting B-cell maturation antigen have demonstrated clinical promise; however, their effectiveness is often limited by disease relapse, partly due to CAR T cell exhaustion. B7-homolog 3 (B7-H3), an immune checkpoint molecule that is overexpressed in MM and may suppress T cell function, represents a potential alternative target to improve CAR T cell efficacy and enhance disease control.
METHODS: We engineered and characteriz

Statins are highly effective and widely prescribed cholesterol lowering drugs. However, statins cross the blood-brain barrier and decrease neural cholesterol in animal models, raising concern that long-term statin use may impact cholesterol-dependent structures and functions in the brain. Cholesterol is a fundamental component of cell membranes and experimentally decreasing membrane cholesterol has been shown to alter cell morphology in vitro. In addition, brain regions that undergo adult neurogenesis rely on local brain cholesterol for the manufacture of new neuronal membranes. Thus neurogene

INTRODUCTION: Joint injuries, a major type of human musculoskeletal disorder, are often accompanied by soft tissue damage, and restoring ligament integrity is a key strategy for reconstructing joint function. However, existing artificial ligaments face a critical challenge: reconciling robust biomechanical performance with intrinsic self-healing capability, especially under cyclic loading and accidental overload conditions. Conventional materials like polyethylene terephthalate (PET) and polytetrafluoroethylene (PTFE) struggle with long-term durability, while emerging self-healing designs are

The ability of Epstein-Barr virus (EBV) to switch between latent and lytic infection is key to its long-term persistence, yet the molecular mechanisms behind this switch remain unclear. To investigate transcriptional events during the latent-to-lytic switch, we utilized Precision nuclear Run On followed by deep Sequencing (PRO-Seq) to map cellular RNA polymerase (Pol) activity to single-nucleotide resolution on the host and EBV genome in three different models of EBV latency and reactivation. In latently infected Mutu-I Burkitt lymphoma (BL) cells, Pol activity was enriched at the Qp promoter,
