Unique molecular signature in mucolipidosis type IV microglia.

BACKGROUND: Lysosomal storage diseases (LSD) are a large family of inherited disorders characterized by abnormal endolysosomal accumulation of cellular material due to catabolic enzyme and transporter deficiencies. Depending on the affected metabolic pathway, LSD manifest with somatic or central nervous system (CNS) signs and symptoms. Neuroinflammation is a hallmark feature of LSD with CNS involvement such as mucolipidosis type IV, but not of others like Fabry disease. METHODS: We investigated the properties of microglia from LSD with and without major CNS involvement in 2-month-old mucolipi

Selective Persistence of Sensorimotor Mismatch Signals in Visual Cortex of Behaving Alzheimer's Disease Mice.

Neurodegenerative processes in Alzheimer's disease (AD) affect the structure and function of neurons [1-4], resulting in altered neuronal activity patterns comprising neuronal hypo- and hyperactivity [5, 6] and causing the disruption of long-range projections [7, 8]. Impaired information processing between functionally connected brain areas is evident in defective visuomotor integration, an early sign of the disease [9-11]. The cellular and neuronal circuit mechanisms underlying this disruption of information processing in AD, however, remain elusive. Recent studies in mice suggest that visuom

The expanding syndrome of amyotrophic lateral sclerosis: a clinical and molecular odyssey.

Recent advances in understanding amyotrophic lateral sclerosis (ALS) have delivered new questions. Disappointingly, the initial enthusiasm for transgenic mouse models of the disease has not been followed by rapid advances in therapy or prevention. Monogenic models may have inadvertently masked the true complexity of the human disease. ALS has evolved into a multisystem disorder, involving a final common pathway accessible via multiple upstream aetiological tributaries. Nonetheless, there is a common clinical core to ALS, as clear today as it was to Charcot and others. We stress the continuing

Synaptic alterations in the rTg4510 mouse model of tauopathy.

Synapse loss, rather than the hallmark amyloid-β (Aβ) plaques or tau-filled neurofibrillary tangles (NFT), is considered the most predictive pathological feature associated with cognitive status in the Alzheimer's disease (AD) brain. The role of Aβ in synapse loss is well established, but despite data linking tau to synaptic function, the role of tau in synapse loss remains largely undetermined. Here we test the hypothesis that human mutant P301L tau overexpression in a mouse model (rTg4510) will lead to age-dependent synaptic loss and dysfunction. Using array tomography and two methods of qua

[Molecular pathogenesis of motor neuron diseases].

Spinal and bulbar muscular atrophy (SBMA) and amyotrophic lateral sclerosis (ALS) are representative motor neuron diseases in which selective neuronal degeneration occurs. In this paper, some molecular aspects are discussed related to the pathogenesis of the neuronal degeneration. SBMA is a an X-linked neurodegenerative disease caused by the expansion of a CAG repeat in the first exon of the androgen receptor (AR) gene. To date, eight CAG repeat diseases have been identified, including spinal and bulbar muscular atrophy (SBMA), Huntington's disease (HD), dentatorubralpallidoluysian atrophy (DR

Chronic Exposure to Hypoxia Inhibits Myelinogenesis and Causes Motor Coordination Deficits in Adult Mice.

Exposure to chronic hypoxia is considered to be a risk factor for deficits in brain function in adults, but the underlying mechanisms remain largely unknown. Since active myelinogenesis persists in the adult central nervous system, here we aimed to investigate the impact of chronic hypoxia on myelination and the related functional consequences in adult mice. Using a transgenic approach to label newly-generated myelin sheaths (NG2-CreERTM; Tau-mGFP), we found that myelinogenesis was highly active in most brain regions, such as the motor cortex and corpus callosum. After exposure to hypoxia (10%

Off-Target Expression of Cre-Dependent Adeno-Associated Viruses in Wild-Type C57BL/6J Mice.

Adeno-associated viruses (AAVs) are a commonly used tool in neuroscience to efficiently label, trace, and/or manipulate neuronal populations. Highly specific targeting can be achieved through recombinase-dependent AAVs in combination with transgenic rodent lines that express Cre-recombinase in specific cell types. Visualization of viral expression is typically achieved through fluorescent reporter proteins (e.g., GFP or mCherry) packaged within the AAV genome. Although nonamplified fluorescence is usually sufficient to observe viral expression, immunohistochemical amplification of the fluoresc

Deciphering MECP2-associated disorders: disrupted circuits and the hope for repair.

MECP2 is a critical gene for neural development, mutations or duplication of which led to severe neurodevelopmental disorders, such as Rett syndrome (RTT) and autism spectrum disorders (ASD). Extensive works during the past decade yield ample insights into the molecular and cellular functions of MeCP2 in neural development. Furthermore, genetic manipulations in Mecp2 mouse models strongly suggested that deficiency in synaptic plasticity and various behaviors of Mecp2 null or transgenic mice could be rescued in adulthood. Further studies elucidating neural circuits responsible for symptoms in M

Prions in skeletal muscles of deer with chronic wasting disease.

The emergence of chronic wasting disease (CWD) in deer and elk in an increasingly wide geographic area, as well as the interspecies transmission of bovine spongiform encephalopathy to humans in the form of variant Creutzfeldt Jakob disease, have raised concerns about the zoonotic potential of CWD. Because meat consumption is the most likely means of exposure, it is important to determine whether skeletal muscle of diseased cervids contains prion infectivity. Here bioassays in transgenic mice expressing cervid prion protein revealed the presence of infectious prions in skeletal muscles of CWD-i

Chemical synthesis of poliovirus cDNA: generation of infectious virus in the absence of natural template.

Full-length poliovirus complementary DNA (cDNA) was synthesized by assembling oligonucleotides of plus and minus strand polarity. The synthetic poliovirus cDNA was transcribed by RNA polymerase into viral RNA, which translated and replicated in a cell-free extract, resulting in the de novo synthesis of infectious poliovirus. Experiments in tissue culture using neutralizing antibodies and CD155 receptor-specific antibodies and neurovirulence tests in CD155 transgenic mice confirmed that the synthetic virus had biochemical and pathogenic characteristics of poliovirus. Our results show that it is

A transgenic model for listeriosis: role of internalin in crossing the intestinal barrier.

Listeria monocytogenes is responsible for severe food-borne infections, but the mechanisms by which bacteria cross the intestinal barrier are unknown. Listeria monocytogenes expresses a surface protein, internalin, that interacts with a host receptor, E-cadherin, to promote entry into human epithelial cells. Murine E-cadherin, in contrast to guinea pig E-cadherin, does not interact with internalin, excluding the mouse as a model for addressing internalin function in vivo. In guinea pigs and transgenic mice expressing human E-cadherin, internalin was found to mediate invasion of enterocytes and

Molecular basis of osteogenesis imperfecta and related disorders of bone.

Recent work has demonstrated that more than 90% of patients with osteogenesis imperfecta (OI) have mutations in the gene for either the pro alpha 1 (I) chain or the gene for the pro alpha 2 (I) chain of type I procollagen. Three molecular mechanisms have explained the devastating effects of these mutations. In addition, the data provide several general conclusions about mutations in collagen genes. Studies in cell cultures with antisense oligonucleotides and studies in transgenic mice with an antisense gene raised the possibility that several strategies eventually may be developed to treat OI

Ext1-targeted gene therapy ameliorates tauopathy by inhibiting heparan sulfate-dependent tau propagation.

The propagation of pathological tau protein in neuronal cells is a critical driver of neurodegeneration in tauopathies, including Alzheimer's disease. Heparan sulfate (HS) critically regulates pathological tau seeding and cellular uptake processes closely linked to the development of tau pathology in tauopathies, identifying HS as a key potential therapeutic target. Here, we focused on targeting Exostosin-1 (Ext1), an essential enzyme for HS biosynthesis, in the tauP301S transgenic (PS19) tauopathy mouse model. Conditional knockdown of neuronal Ext1 in PS19 mice by delivering an adeno-associat

Immunogenicity and protective efficacy of MERS CoV, NL140422, and HKU4 spike protein nanoparticle vaccines.

While vaccines against Middle East respiratory syndrome coronavirus (MERS-CoV) have had substantial preclinical and clinical development, few vaccine candidates target other related viruses with potential for human spillover within the Merbecovirus subgenus, like NL140422 and HKU4. We designed nanoparticle vaccines displaying the spike ectodomains of MERS-CoV, NL140422, and HKU4 and evaluated their immunogenicity and protective efficacy in mice. All vaccines elicited high IgG antibody titers against the corresponding vaccine spike protein with moderate cross-binding to mis-matched spike protei

In vitro and in vivo inhibition of amyloid β aggregation by a Ru(II)-naphthalene diimide complex.

Amyloid deposits of amyloid-β (Aβ) and hyperphosphorylated tau are pathological hallmarks of Alzheimer's disease (AD), which accounts for most dementia cases worldwide. This study investigates the effect of the complex [Ru(phen)2(pNDIp)]2+ (RuNDI; phen = 1,10-phenanthroline, pNDIp = N,N'-di(1,10-phenanthroline)-1,4,5,8-naphthalenetetracarboxylic diimide) on Aβ aggregation in vitro and in vivo. In vitro, RuNDI markedly attenuated Aβ42 aggregation, as shown by nephelometry, circular dichroism, and transmission electron microscopy, by suppressing β-sheet formation and promoting amorphous assembli

Stochastic misfolding drives the emergence of distinct α-synuclein strains.

α-Synuclein conformational strains provide a potential explanation for the clinical and pathological differences among synucleinopathies such as Parkinson's disease and multiple system atrophy. However, how distinct α-synuclein strains arise remains unknown. Here, we observed conformational heterogeneity between individual preparations of α-synuclein pre-formed fibrils (PFFs) generated by polymerizing wild-type or A53T-mutant human α-synuclein under identical conditions. Moreover, we found that α-synuclein aggregates formed spontaneously in the brains of a transgenic synucleinopathy mouse mode

Targeting peptidyl-arginine deiminase 4 suppresses SARS-CoV-2 replication and modulates the inflammatory response.

SARS-CoV-2, the causative agent of COVID-19, remains a global concern due to gaps in understanding its pathogenesis. Peptidyl-arginine deiminases (PADs) are emerging as key regulators of viral replication and inflammation. PADs catalyze the conversion of arginine into citrulline, a modification linked to autoimmune diseases. Here, we show that PAD-mediated citrullination plays a crucial role in SARS-CoV-2 infection. Using human cell lines and in K18-hACE2 transgenic mice infected with different SARS-CoV-2 strains, we demonstrate that viral replication is associated with increased PAD4 expressi
