New methodology of influential point detection in regression model building for the prediction of metabolic clearance rate of glucose.

Identifying outliers and high-leverage points is a fundamental step in the least-squares regression model building process. The examination of data quality involves the detection of influential points, outliers and high-leverages, which cause many problems in regression analysis. On the basis of a statistical analysis of the residuals (classical, normalized, standardized, jackknife, predicted and recursive) and diagonal elements of a projection matrix, diagnostic plots for influential points indication are formed. The identification of outliers and high leverage points are combined with graphs

Management parameters affecting the outcome of Japanese encephalitis.

The aim of this hospital-based, prospective interventional study was to identify the influence of various therapeutic measures on survival in children with Japanese encephalitis. Fluid restriction, avoidance of routine sedation and higher doses of mannitol were the basis of protocol A (used in the first season). Unrestricted fluid supplementation, guided by central venous pressure monitoring, along with routine sedation and a lower dose of mannitol (only when needed) was used in managing those children with a Glasgow Coma Score (GCS) less than or equal to 8 in the second season. In those child

Analysis and modeling of violations crossing behavior of elderly pedestrians at signalized intersections.

OBJECTIVE: As the population ages, traffic violations by elderly pedestrians at signalized intersections and related issues have become increasingly prominent. Given that signalized intersections are areas where traffic conflicts are highly concentrated, their safety and operational conditions have drawn significant attention. An in-depth examination of the different types of traffic violations committed by older pedestrians and the variations in their causes will help provide a clear understanding of the characteristics of older pedestrians' crossing behavior. Therefore, the study aims to dev

The role of extracranial carotid duplex in predicting functional outcome in first time lacunae strokes.

BACKGROUND: Stroke is the third largest cause of death both worldwide and in Taiwan. Among the various stroke subtypes, lacunae strokes account for 20 to 30% of the total stroke population. Through vigorous risk control factors, the effective prevention and the long-term functional outcome remains are yet to be investigated. Carotid duplex is a non-invasive neuroimaging modality that is routinely applied to stroke patients. In the current research, we correlate baseline carotid duplex examination parameters with functional outcome assessment in an attempt to ascertain important variables for e

Estimating Optimal Site-Specific Individualized Treatment Rules With Limited Data Sharing.

Estimation of optimal individualized treatment rules (ITRs) tailored to patient characteristics is a crucial component of precision medicine. When such problems arise in multicenter randomized trials with known treatment assignment probabilities, the optimal ITR may vary across sites because of site-level heterogeneity, and directly pooling data across centers can lead to biased ITR estimation. Meanwhile, privacy constraints often prevent investigators from accessing individual-level data from all centers. We therefore propose Heterogeneous Distributed Learning (HD-learning) to estimate optima

A one-parameter poisson-emrem model for count data: Theory and applications to cytogenetic and COVID-19 studies.

This paper proposes the Poisson-Emrem (PEm) distribution, a new one-parameter discrete probability model obtained by compounding the Poisson and Emrem distributions. The model preserves the mathematical tractability of the Poisson law while offering greater flexibility for analyzing count data in health and biological sciences. A theoretical study establishes its principal properties, showing that increases in the parameter lead to systematic reductions in the mean, variance, and interquartile range, with a corresponding increase in skewness. Additional features such as moments, dispersion ind

Bayesian Additive Regression Trees for Modeling Multiple Exposures With Measurement Error.

We develop a Bayesian Additive Regression Trees with Measurement Error (BART-ME) model for flexibly estimating exposure-response functions when multiple covariates are measured with classical error. Unlike existing approaches, BART-ME accommodates nonlinearities, interactions, correlated exposures, and correlated measurement errors, while exploiting replicate measurements to estimate the error variance-covariance structure. Posterior inference is obtained via a Metropolis-within-Gibbs algorithm, yielding estimates of exposure-response functions, variable importance, and measurement reliability

Assessing treatment efficacy for interval-censored endpoints using multistate semi-Markov models fit to multiple data streams.

We introduce a computationally efficient and general approach for utilizing multiple, possibly interval-censored, data streams to study complex biomedical endpoints using multistate semi-Markov models. Our motivating application is the REGEN-2069 trial, which investigated the protective efficacy (PE) of the monoclonal antibody combination REGEN-COV against SARS-CoV-2 when administered prophylactically to individuals in households at high risk of secondary transmission. Using data on symptom onset, episodic RT-qPCR sampling, and serological testing, we estimate the PE of REGEN-COV for asymptoma

Multicentre validation and clinical interpretation of an explainable gradient-boosting model for dental-implant survival/failure prediction.

OBJECTIVE: To evaluate, on unseen multicentre data, the discrimination, calibration, clinical utility, and transparency of an interpretable gradient-boosting model for implant survival/failure prediction, and to report centre-wise transportability with an internal-external (leave-one-centre-out) validation. MATERIALS AND METHODS: We analysed 910 unique implants from three centres (A, B, C) and created a stratified 80/20 hold-out test set (N=182). Within the training split, we performed randomized hyper-parameter search (five-fold cross-validation; macro-F1 scoring) and tuned a single decision

Augmenting Treatment Arms With External Data Through Propensity-Score Weighted Power Priors: An Application in Expanded Access.

The incorporation of real-world data to supplement the analysis of trials and improve decision-making has spurred the development of statistical techniques to account for introduced confounding. Recently, "hybrid" methods have been developed through which measured confounding is first attenuated via propensity scores and unmeasured confounding is addressed through (Bayesian) dynamic borrowing. Most efforts to date have focused on augmenting control arms with historical controls. Here we consider augmenting treatment arms through "expanded access", which is a pathway of nontrial access to inves

Two-stage randomized clinical trials with a right-censored endpoint: Comparison of frequentist and Bayesian adaptive designs.

Adaptive randomized clinical trials are of major interest when dealing with a time-to-event outcome in a prolonged observation window. No consensus exists either to define stopping boundaries or to combine p $$ p $$ values or test statistics in the terminal analysis in the case of a frequentist design and sample size adaptation. In a one-sided setting, we compared three frequentist approaches using stopping boundaries relying on α $$ \alpha $$ -spending functions and a Bayesian monitoring setting with boundaries based on the posterior distribution of the log-hazard ratio. All designs compr

Shotgun DNA sequencing for human identification: Dynamic SNP selection and likelihood ratio calculations accounting for errors.

Shotgun sequencing is a DNA analysis method that potentially determines the nucleotide sequence of every DNA fragment in a sample, unlike PCR-based genotyping methods that is widely used in forensic genetics and targets predefined short tandem repeats (STRs) or predefined single nucleotide polymorphisms (SNPs). Shotgun DNA sequencing is particularly useful for highly degraded low-quality DNA samples, such as ancient samples or those from crime scenes. Here, we developed a statistical model for human identification using shotgun sequencing data and developed formulas for calculating the evident

Sequential monitoring of time-to-event safety endpoints in clinical trials.

Background/aimsSafety monitoring is a crucial requirement for Phase II and III clinical trials. To protect patients from toxicity risk, stopping rules may be implemented that will halt the study if an unexpectedly high number of events occur. These rules are constructed using statistical procedures that typically treat the toxicity data as binary occurrences. Because the exact dates of toxicities are often available, a strategy that handles these as time-to-event data may offer higher power and require less calendar time to identify excess risk. This work investigates several statistical metho
