Ammonium-nitrogen addition at the seedling stage does not reduce grain cadmium concentration in two common wheat (Triticum aestivum L.) cultivars.

High cadmium (Cd) concentration in common wheat (Triticum aestivum L.) grains poses potential health risks. Several management strategies have been used to reduce grain Cd concentration. However, limited information is available on the use of ammonium-nitrogen (NH4+-N) as a strategy to manage Cd concentration in wheat grains. In this study, NH4+-N addition at the seedling stage unchanged the grain Cd concentration in the high-Cd accumulator, Zhoumai 18 (ZM18), but dramatically increased that in the low-Cd accumulator, Yunmai 51 (YM51). Further analysis revealed that the effects of NH4+-N addit

Vmp1, Vps13D, and Marf/Mfn2 function in a conserved pathway to regulate mitochondria and ER contact in development and disease.

Mutations in Vps13D cause defects in autophagy, clearance of mitochondria, and human movement disorders. Here, we discover that Vps13D functions in a pathway downstream of Vmp1 and upstream of Marf/Mfn2. Like vps13d, vmp1 mutant cells exhibit defects in autophagy, mitochondrial size, and clearance. Through the relationship between vmp1 and vps13d, we reveal a novel role for Vps13D in the regulation of mitochondria and endoplasmic reticulum (ER) contact. Significantly, the function of Vps13D in mitochondria and ER contact is conserved between fly and human cells, including fibroblasts derived f…

Cellular Stress in the Pathogenesis of Muscular Disorders-From Cause to Consequence.

Cellular stress has been considered a relevant pathogenetic factor in a variety of human diseases. Due to its primary functions by means of contractility, metabolism, and protein synthesis, the muscle cell is faced with continuous changes of cellular homeostasis that require rapid and coordinated adaptive mechanisms. Hence, a prone susceptibility to cellular stress in muscle is immanent. However, studies focusing on the cellular stress response in muscular disorders are limited. While in recent years there have been emerging indications regarding a relevant role of cellular stress in the patho

Melatonin protects mouse testes from palmitic acid-induced lipotoxicity by attenuating oxidative stress and DNA damage in a SIRT1-dependent manner.

Palmitic acid (PA), the main component of dietary saturated fat, has been known to increase in patients with obesity, and PA-induced lipotox … C-Caspase12, and CHOP in type B spermatogonial stem cells. Changes in the expression of endoplasmic reticulum (ER) stress markers (p-IRE1, p-PERK, ATF4) and intracellular Ca2+ levels showed that melatonin relieved PA-induced ER stress. Mechanistically, melatonin stimulated the expression and nuclear translocation of SIRT1 through its receptors and prevented …

The microscopic anatomy of endothelial cells in human atherosclerosis: Focus on ER and mitochondria.

Once regarded merely as a bland lipid storage disease consequence of aging, atherosclerosis is currently considered a slow and continuous in … herosclerosis. Over the years, there has been evidence highlighting the central role of endoplasmic reticulum (ER) in the maintenance of endothelial function and perturbations in ER biology have been proposed to adversely affect a diverse range of endothelial functions. Of particular interest is the evidence that under certain pathophysiological circumstanc …

ER stress-related molecules induced by Hantaan virus infection in differentiated THP-1 cells.

Endoplasmic reticulum stress (ER stress) can be induced by virus infection. In this part, we explored whether Hantaan virus (HTNV) infection could induce ER stress in differentiated THP-1 (dTHP-1) cells. It showed that the mRNA and protein levels of ER stress-related 78 kDa glucose-regulated protein (GRP78, HSPA5) and mRNA levels of X box-binding protein 1 (XBP-1), activating transcription factor 6(ATF6) and PKR-like ER kinase (PERK) after HTNV infection, were significantly higher than that in uninfected control group. However, the mRNA levels of C/EBP homologous protein (CHOP), glucose-regula…

ER-to-Golgi trafficking of procollagen in the absence of large carriers.

Secretion and assembly of collagen are fundamental to the function of the extracellular matrix. Defects in the assembly of a collagen matrix lead to pathologies including fibrosis and osteogenesis imperfecta. Owing to the size of fibril-forming procollagen molecules it is assumed that they are transported from the endoplasmic reticulum to the Golgi in specialized large COPII-dependent carriers. Here, analyzing endogenous procollagen and a new engineered GFP-tagged form, we show that transport to the Golgi occurs in the absence of large (>350 nm) carriers. Large GFP-positive structures were obs…

The cellular stress response in hepatitis C virus infection: A balancing act to promote viral persistence and host cell survival.

Oxidative- and endoplasmic reticulum (ER)-stress are common events during hepatitis C virus (HCV) infection and both regulate cell survival and determine clinical outcome. In response to intrinsic and extrinsic cellular stress, different adaptive mechanisms have evolved in hepatocytes to restore cellular homeostasis like the anti-oxidant response, the unfolded protein response (UPR) and the integrated stress response (ISR). In this review, we focus on the cellular stress response in the context of acute and chronic HCV infection. The mechanisms of induction and modulation of oxidative- and ER-…

Modulation of chewing behavior and reticular pH in nonlactating cows challenged with concentrate-rich diets supplemented with phytogenic compounds and autolyzed yeast.

Feeding of concentrate-rich diets impairs chewing behavior and leads to rumen acidosis in cattle. Because of their modulatory effects on ruminal fermentation, phytogenic compounds (PHY) and autolyzed yeast derivatives (AY) may alleviate the negative consequences of high-concentrate diets. Therefore, this research investigated if chewing behavior and the reticular pH dynamics are modulated by AY and PHY supplementation during repeated concentrate-rich challenges used to simulate intermittent rumen acidotic insults. Eight rumen-cannulated, dry, and nonpregnant Holstein cows were assigned to an i

Gross Measurements of the Digestive Tract and Visceral Organs of Addax Antelope (Addax nasomaculatus) Following a Concentrate or Forage Feeding Regime.

Differences in macroscopic measurements of the gastrointestinal tract have been hypothesized to correlate with the browser-grazer continuum in the natural diet of ruminants. However, to what extent these characteristics represent species-specific traits, or respond to the actually ingested diet, remains to be investigated. Twelve surplus addax antelope (Addax nasomaculatus) were divided into two groups and fed, for 3 months, either their usual diet, consisting of a concentrate feed with a limited amount of hay, or a diet of unlimited hay only. After culling, macroscopic measurements were compa

ER stress and basement membrane defects combine to cause glomerular and tubular renal disease resulting from Col4a1 mutations in mice.

Collagen IV is a major component of basement membranes, and mutations in COL4A1, which encodes collagen IV alpha chain 1, cause a multisystemic disease encompassing cerebrovascular, eye and kidney defects. However, COL4A1 renal disease remains poorly characterized and its pathomolecular mechanisms are unknown. We show that Col4a1 mutations in mice cause hypotension and renal disease, including proteinuria and defects in Bowman's capsule and the glomerular basement membrane, indicating a role for Col4a1 in glomerular filtration. Impaired sodium reabsorption in the loop of Henle and distal nephr

The activity of RTA2, a downstream effector of the calcineurin pathway, is required during tunicamycin-induced ER stress response in Candida albicans.

In this study, we demonstrate a novel function of a downstream effector molecule of the calcineurin pathway, RTA2 (Resistance To Aminocholesterol), in ER stress response. The deletion of RTA2 increases susceptibility to the ER stressor tunicamycin and morpholine-like drug, 7-aminocholesterol. Additionally, the expression of RTA2 is also transcriptionally induced by ergosterol biosynthesis inhibitors and cell-wall-damaging agents. As tunicamycin induces the unfolded protein response pathway (UPR) via the transcription factor, HAC1, we monitored the expression of a subset of HAC1-dependent UPR t

Enhanced synthesis of saturated phospholipids is associated with ER stress and lipotoxicity in palmitate treated hepatic cells.

High levels of saturated FAs (SFAs) are acutely toxic to a variety of cell types, including hepatocytes, and have been associated with diseases such as type 2 diabetes and nonalcoholic fatty liver disease. SFA accumulation has been previously shown to degrade endoplasmic reticulum (ER) function leading to other manifestations of the lipoapoptotic cascade. We hypothesized that dysfunctional phospholipid (PL) metabolism is an initiating factor in this ER stress response. Treatment of either primary hepatocytes or H4IIEC3 cells with the SFA palmitate resulted in dramatic dilation of the ER membra…

The unfolded protein response (UPR) pathway in Cryptococcus.

Unique and evolutionarily conserved signaling pathways allow an organism to sense, respond to, and adapt to internal and external environmental cues at its biological niche. In eukaryotic cells, the unfolded protein response (UPR) pathway regulates endoplasmic reticulum (ER) homeostasis upon exposure to environmental changes causing ER stress. The UPR pathway of Cryptococcus neoformans, an opportunistic fungal pathogen, which causes life-threatening meningoencephalitis in immunocompromised individuals, consists of the evolutionarily conserved Ire1 kinase, a unique bZIP transcription factor, Hx…

Expression of the hyperphosphorylated tau attenuates ER stress-induced apoptosis with upregulation of unfolded protein response.

The neural dysfunction in Alzheimer's disease (AD) could arise from endoplasmic reticulum (ER) stress and deficits of the unfolded protein response (UPR). To explore whether tau hyperphosphorylation, a hallmark of AD brain pathologies, plays a role in ER stress-induced alterations of cell viability, we established cell lines with stable expression of human tau (HEK293/tau) or the vector (HEK293/vec) and treated the cells with thapsigargin (TG), an ER stress inducer. We observed that the HEK293/tau cells were more resistant than the HEK293/vec cells to the TG-induced apoptosis, importantly, a t…

[Viability period of the rabbit skeletal muscle in the Ca(2+)-induced Ca2+ release (CICR) test].

BACKGROUND: In Japan, malignant hyperthermia susceptibility (MHS) is diagnosed by detecting an enhanced rate of CICR from the sarcoplasmic reticulum in skinned skeletal muscle fibers. We usually consider the CICR test should be completed within 48 hours of muscle excision. We measured the viability period of the CICR test in the following four treatment groups. METHODS: Muscle bundles prepared from a rabbit were dissected and stored in muscle relaxant solution at -30 degrees C, 4 degrees C, 20 degrees C, and 37 degrees C. The CICR rate was measured in a part of the biopsied muscle using chemi…

A class of dynamin-like GTPases involved in the generation of the tubular ER network.

The endoplasmic reticulum (ER) consists of tubules that are shaped by the reticulons and DP1/Yop1p, but how the tubules form an interconnected network is unknown. Here, we show that mammalian atlastins, which are dynamin-like, integral membrane GTPases, interact with the tubule-shaping proteins. The atlastins localize to the tubular ER and are required for proper network formation in vivo and in vitro. Depletion of the atlastins or overexpression of dominant-negative forms inhibits tubule interconnections. The Sey1p GTPase in S. cerevisiae is likely a functional ortholog of the atlastins; it s…
