Aminoglycosides in gynecologic infections.

Aminoglycosides have always had a major role in the treatment of gynecologic infections. This therapeutic emphasis has been based upon the bactericidal activity of the aminoglycosides against gram-negative aerobes. This group of organisms is important to the gynecologist because of their frequent isolation from soft tissue, urinary sites, and the bloodstream in women with nosocomial or community-acquired pelvic infections. In the past decade, there has been increasing clinical awareness of the multi-bacterial nature of these soft tissue pelvic infections. A major therapeutic change directed ag

GROMTools: scalable individual-level GReX imputation for mega-biobank-scale cohorts.

MOTIVATION: The computational burden of individual-level genetically regulated gene expression (GReX) imputation has risen sharply with the growth of human mega-biobanks and the rapid expansion of transcriptomic imputation models across tissues and single-cell hierarchies. Existing tools were not designed for this setting and require complex, memory-intensive workflows that are poorly matched to shared and cloud-based compute environments, where runtime, memory, and I/O directly determine cost and throughput. GROMTools is an open-source C ++ engine with an R interface that exploits sparse pred

Paired Single-Cell Profiling Reveals Glutathione-Dependent Immune Evasion in Breast Cancer Liver Metastasis.

Breast cancer liver metastasis (BCLM) represents a major clinical challenge. Characterizing the differences between primary and metastatic tumors could help identify therapeutic targets for preventing and treating metastasis. Through single-cell transcriptomic sequencing of matched primary tumors and BCLMs from 13 treatment-naïve patients, we uncovered a fundamental paradox in metastatic progression: BCLMs exhibited a highly immune-infiltrated microenvironment characterized by cytotoxic T cell expansion, inflammatory macrophage polarization, and depletion of regulatory T cells, yet they succes

A shape-morphing bilayer hydrogel microrobot featuring prolonged retention for targeted cell delivery.

Soft robots have attracted extensive attention in biomedical applications due to their adaptability, mechanical compliance, and conformability to biological tissues. The integration of soft materials such as hydrogels with functional components enables robots to respond to environmental stimuli and perform targeted tasks, including cargo transport and release. However, many soft systems lack effective retention mechanisms, limiting their efficiency in sustained delivery applications such as drug or cell therapy. In this study, we present a reversibly deformable microrobot for targeted delivery

Design principles of the cytotoxic CD8+ T cell response.

Cytotoxic T lymphocytes eliminate infected or malignant cells, safeguarding surrounding tissues. Although experimental and systems-immunology studies have cataloged many molecular and cellular actors involved in an immune response, the design principles governing how the speed and magnitude of T cell responses emerge from cellular decision-making remain elusive. Here, we recast the T cell response as a feedback-controlled program, wherein the rates of activation, proliferation, differentiation, and death are regulated through antigenic, pro- and anti-inflammatory cues. By exploring a broad cla

Soluble PD-L1-secreting umbilical cord-derived mesenchymal stromal cells transiently attenuate established xenogeneic T cell activation in vivo.

Immune overactivation damages tissues in T cell-mediated disorders, and cell-based therapies are being developed to suppress T cell responses. Mesenchymal stromal cells (MSCs) modulate immunity; however, their suppressive effects depend on inflammatory cytokine priming. Umbilical cord-derived MSCs are attractive for non-invasive collection and expansion, but their effects diminish after intravenous infusion. PD-1/PD-L1 signaling lowers T cell activation; therefore, soluble PD-L1 (sPD-L1)-secreting MSCs may provide contact-independent checkpoint engagement and we tested whether sPD-L1-MSCs can

Comparative Genomics of the Tea XTH Gene Family Reveals the Involvement of CsXTH27 in Internode Elongation.

Xyloglucan endotransglucosylase/hydrolases (XTHs) are key cell-wall-remodeling enzymes that mediate xyloglucan modification, cell expansion, and organ elongation, yet the composition and variation in the XTH gene family across tea accessions remain unclear. In this study, we systematically identified and comparatively analyzed the XTH gene family across the genomes of nine tea accessions, yielding 477 putative XTH genes. Phylogenetic, conserved-motif, protein-domain, and conserved catalytic-motif analyses indicated that the major structural features of tea XTH proteins are broadly conserved, w

Mechanical stress-induced STAT3/YAP signaling in fibroblasts programs immunosuppressive lymph node remodeling in lung squamous carcinoma.

Lung squamous cell carcinoma (LUSC) frequently metastasizes to lymph nodes, yet how tumor-intrinsic biomechanical cues influence nodal immune environments remains unclear. Here, we identify solid stress, generated during tumor expansion, as a key driver of pre-metastatic niche formation in tumor-draining lymph nodes (TDLNs). Using murine models with defined solid stress levels, we show that high-stress tumors induce early TDLN remodeling characterized by fibroblast activation and accumulation of immunosuppressive S100a8⁺ myeloid cells. Single-cell and spatial transcriptomics reveal that Col1a1

Proteomic profiling of Ascaridia galli reveals sex- and stage-specific protein signatures.

Ascaridia galli is one of the most prevalent intestinal nematodes of domesticated chickens and the primary cause of avian ascaridiasis, a globally important disease in poultry production. Infection rates often reach 70-90 % in free-range systems due to its direct life cycle and efficient environmental transmission. Despite its impact, the molecular mechanisms governing A. galli host-parasite interactions remain poorly understood. A. galli belongs to Clade III nematodes, which include several species of major veterinary importance. Comparative genomic studies Clade III-nematodes have revealed e

METTL16 promotes PD-L1 expression and mediates immunosuppression in B-cell lymphomas via m6A methylation.

OBJECTIVES: Immune Checkpoint Inhibitors (ICIs) have changed cancer treatment. Altered expression of methyltransferase-like 16 (METTL16), one of the initial m6A "writers", is associated with malignant progression. METTL16 is a methyltransferase that may influence the treatment response of B-cell lymphoma (BCL). METHODS: Overexpression and knockdown strategies were used to regulate METTL16 expression in BCL cells, and then qPCR and western blot were employed to measure the levels of METTL16 and programmed death ligand 1 (PD-L1). A BCL-T cell co-culture system was employed to study immune evasi

What if each voxel were measured with a different diffusion protocol?

Expansion of diffusion MRI (dMRI) both into the realm of strong gradients, and into accessible imaging with portable low-field devices, brings about the challenge of gradient nonlinearities. Spatial variations of the diffusion gradients make diffusion weightings and directions non-uniform across the field of view, and deform perfect shells in the q-space designed for isotropic directional coverage. Such imperfections hinder parameter estimation: Anisotropic shells hamper the deconvolution of fiber orientation distribution function (fODF), while brute-force retraining of a nonlinear regressor f

Combining T cell receptor sequencing and transcriptomics to characterize tissue-resident T cells from human gut biopsies.

Gastrointestinal T cells (GI-T) play a critical role in mucosal immunity, balancing tolerance and pathogen defence. T cells recognize antigens via T cell receptors (TCRs). Next-generation sequencing (NGS) is utilized to analyse TCR repertoires in contexts such as health, haematological diseases, autoimmunity, and inflammation. While some studies have explored T cell involvement in GI conditions, the integration of different techniques and examination of diverse tissues remain underdeveloped. In our "proof of concept" study, for the first time, we combined flow cytometry, TCR sequencing and tra

Sending the Signal to Bone: How Tumor-Derived EVs Orchestrate Pre-Metastatic Niche Formation and Skeletal Colonization.

Bone is a preferred site for disseminated tumor cells, yet the molecular mechanisms that prepare the skeletal microenvironment for metastatic colonization are only beginning to be understood. At the heart of this process are extracellular vesicles (EVs), nano-sized, lipid-encapsulated particles secreted by cancer cells and stromal components. This review consolidates current findings that position EVs as key architects of the bone-metastatic niche. We detail the biogenesis of EVs and their organotropic distribution, focusing on how integrin patterns and bone-specific ligands guide vesicle homi

Cardiothoracic Wound Management in Pediatrics: A Scoping Review.

BACKGROUND: Congenital heart defects represent the most prominent cause of mortality in a child's first year of life. Management of cardiothoracic wounds in pediatric populations is complex, attributed to the fragility of soft tissue, with healing progressing differently in adults. Understanding the breadth of wound care management is essential in establishing its applicability to clinical practice, especially in the prevention of complications. OBJECTIVE: The aim of this study was to expansively explore preoperative, intraoperative, and postoperative cardiothoracic wound care in pediatrics.

PD1+ T Regulatory Cells Are Not Sufficient to Protect from Gestational Hypertension.

Tolerance to foetal tissues in pregnancy depends on the match between mother and child. CD4+Foxp3+ regulatory T cells (Tregs), which are involved in peripheral tolerance, may facilitate this effect. Previous findings have indicated that the number of missing KIR ligands (MSLs) between mother and child correlates with the risk of gestational hypertension (GH) and preeclampsia (PE). This study tested whether Tregs are involved in the pathogenesis of gestational disorders. In total, 57 pregnant women participated, including 39 with hypertensive disorders of pregnancy and 18 healthy controls. Treg

Loss of GNB2L1 promotes expansion of CD74+ dendritic cells and contributes to development of diabetic foot ulcers.

Refractory diabetic foot ulcers (DFUs) are characterized by low-grade chronic inflammation both locally and systemically. GNB2L1 has been reported to be associated with wound healing in diabetic rats, but the specific mechanism by which it acts has not been explored. scRNA-seq data from foot skin samples of three diabetic patients without DFU (non-DFU) and four diabetic patients with non-healing DFU (DFU) were downloaded from the GSE165816 dataset and analyzed. Twenty-five different cell clusters and fourteen cell types were identified, and the dendritic cell (DC) cluster was identified as a m
