A ShbHLH34-ShPUR Module Activates ShTPS1-mediated β-caryophyllene Biosynthesis for Insect Resistance in Sugarcane.

Terpenoids represent one of the largest and most structurally diverse classes of plant secondary metabolites, playing essential roles in defense against herbivorous insects. Despite their importance, the mechanisms regulating terpenoid biosynthesis remain largely unexplored. Here, in sugarcane, we identified and characterized a terpene synthase gene from Saccharum spp. Hybrids, ShTPS1, which is strongly induced by feeding from fall armyworm Spodoptera frugiperda and sugarcane stem borer Chilo infuscatellus. Enzymatic assays in vitro revealed that ShTPS1 catalyzes the biosynthesis of volatile t

Photodynamic therapy improves sensitivity to temozolomide in glioblastoma cells via inhibiting autophagy.

OBJECTIVE: Chemotherapy resistance from temozolomide (TMZ) continues to be a major cause of glioblastoma (GBM) recurrence. Emerging evidence indicates that cytoprotective autophagy constitutes a key resistance mechanism to TMZ. Photodynamic therapy (PDT) has been shown to modulate the sensitivity of glioblastoma cells (GCs) to TMZ through multiple pathways. This study aims to explore how PDT regulates the sensitivity of GCs to TMZ chemotherapy by inhibiting autophagy formation. METHODS: The vitality and proliferation ability of U251 cells were evaluated under different treatments using cell

Ferulic Acid Increases Temozolomide Sensitivity in Glioblastoma Cells, Causing DNA Damage and Inhibiting Cell Proliferation.

AIM: To investigate the antitumor effects of ferulic acid (FA) on glioblastoma multiforme (GBM) cells both alone and in combination with temozolomide (TMZ), and also to determine the potential of this synergy for treatment processes. MATERIAL AND METHODS: Human glioblastoma U87-MG cells were used in this study. To evaluate the potential constructive interaction between temozolomide (TMZ) and ferulic acid (FA), a sequential treatment protocol was applied in which cells were first treated with TMZ (20, 40, and 80 μM) for 48 hours, followed by FA (1000 μM and 1500 μM) for an additional 24 hours.

Non-surgical periodontal therapy and systemic inflammatory biomarkers in patients with periodontitis and carotid plaque burden: a pilot study.

INTRODUCTION: Systemic inflammation is a key contributor to the pathophysiology of carotid plaque burden (CpB). Increasing evidence supports a link between periodontitis and systemic conditions, including endothelial dysfunction, and CpB. This study aimed to explore inflammatory mediators involved in cardiovascular disease that may represent shared mechanistic pathways between periodontitis and carotid plaque burden, and to determine whether non-surgical periodontal therapy modulates systemic inflammatory activity. METHODS: A pilot study was conducted on subjects presenting with both periodon

Systemic inflammation index is a predictive and prognostic factor in patients with liposarcoma or leiomyosarcoma treated with trabectedin.

Trabectedin is a chemotherapeutic agent that has shown activity in the treatment of patients with soft tissue sarcomas after failure of anthracycline-based therapy and the best results were recorded in the treatment of L-sarcomas. There is a need for prognostic and predictive factors that will help clinicians with therapy selection. In this study we aim to analyze treatment results, prognostic and predictive factors in patients treated with trabectedin in routine clinical practice. We retrospectively analyzed the medical records of patients who started treatment with trabectedin between 04/200

Oral Cyclophosphamide for Patients With Metastatic Castration-Resistant Prostate Cancer in a Scenario of Limited Health Care Resources.

PURPOSE: Prior noncontemporary studies showed that oral cyclophosphamide is an active treatment of metastatic castration-resistant prostate cancer (mCRPC). However, cyclophosphamide is currently underutilized in routine clinical practice given the lack of survival benefit and the emergence of more effective treatments. METHODS: We retrospectively reviewed our institutional database to identify patients with mCRPC treated with cyclophosphamide. Prostate-specific antigen decrease ≥50% from baseline (PSA50) response was determined as the proportion of patients achieving a prostate-specific antig

The potential ameliorative effect of curcumin nanoemulsion on busulfan-induced testicular toxicity in adult rat model.

Many factors contribute to infertility. Among these are chemotherapeutic drugs as they interfere with cell division and spermatogenesis. Busulfan (BU) is one of these drugs that is applied in the treatment of many malignancies. Latest studies have focused on the protective impact of medicinal plant products, such as curcumin (CU), for ameliorating tissue damage through their antioxidant properties. Nanoparticles are now used to improve drug delivery and efficacy. Our study was aimed at assessing the potential therapeutic effect of curcumin nanoemulsion (CU-NE) on busulfan-induced testicular to

Pearls & Oy-sters: Acute Obstructive Hydrocephalus Co-Occurrence With Vigabatrin-Associated MRI Changes in an Infant With LIS1-Related Classical Lissencephaly.

We present a case of a 4-month-old infant with classical type I lissencephaly treated with vigabatrin for hypsarrhythmia. The patient subsequently developed hypotonia and respiratory failure. Imaging revealed vigabatrin-associated brain abnormalities on MRI (VABAM), with acute obstructive hydrocephalus requiring emergent extraventricular drainage. VABAM involve a predictable pattern of restricted diffusion, are generally clinically silent, and are reversible on cessation of the medication. This case presents a rare, life-threatening complication in association with VABAM. In this report, we re

MicroRNA-143 overexpression enhances the chemosensitivity of A172 glioblastoma cells to carmustine.

As an aggressive malignancy, glioblastoma multiforme (GBM) is the most common type of brain tumor. The existing treatments have shown limited achievement in increasing the overall survival of patients. Therefore, identifying the key molecules involved in GBM will provide new potential therapeutic targets. Carmustine is an alkylating agent used as a supplementary therapeutic option for GBM. However, the extensive use of carmustine has been limited by uncertainty about its efficacy. MicroRNAs (miRNAs) are essential in post-transcriptional gene regulation. Many aberrantly expressed miRNAs have be

Long-term remission with temozolomide for AIDS-related primary central nervous system lymphoma.

Primary Central Nervous System Lymphoma (PCNSL) is an aggressive brain tumour with a median survival rarely exceeding 3 months without treatment when seen in association with advanced HIV. High dose methotrexate (HD-MTX) in association with combination antiretroviral therapy (cART) is the recommended chemotherapy. However, HD-MTX may be not feasible due to poor performance status and concerns about toxicity. The 2023 guidelines from the European Association of Neuro-Oncology recommend that for people living with HIV (PLWH) presenting with PCNSL who have morbidities and/or poor functional statu

Cyclophosphamide exposure factors in family caregivers for pediatric cancer patients.

The exposure of family caregivers to anticancer drugs for pediatric patients with malignancy is a potential health risk that needs to be minimized. We monitored the amount of cyclophosphamide (CPM) that had adhered to the undershirts of patients and the personal protective equipment (PPE) of family caregivers as well as the caregivers' urine levels of CPM within the first three days after the first and second courses of high-dose CPM therapy. Liquid chromatography/mass spectrometry (LC/MS/MS) detected >0.03 ng/ml of CPM in 26% (23/88) of urine samples from 8 of 11 (72.7%) patients' family care

Aptamer-functionalized triptolide with release controllability as a promising targeted therapy against triple-negative breast cancer.

Targeted delivery and precise release of toxins is a prospective strategy for the treatment of triple-negative breast cancer (TNBC), yet the flexibility to incorporate both properties simultaneously remains tremendously challenging in the X-drug conjugate fields. As critical components in conjugates, linkers could flourish in achieving optimal functionalities. Here, we pioneered a pH-hypersensitive tumor-targeting aptamer AS1411-triptolide conjugate (AS-TP) to achieve smart release of the toxin and targeted therapy against TNBC. The multifunctional acetal ester linker in the AS-TP site-specifi

Magnolol and Temozolomide exhibit a synergistic anti-glioma activity through MGMT inhibition.

Temozolomide (TMZ) is the leading chemotherapeutic agent used for glioma therapy due to its good oral absorption and blood-brain barrier permeability. However, its anti-glioma efficacy may be limited due to its adverse effects and resistance development. O6-Methylguanine-DNA-methyltransferase (MGMT), an enzyme associated with TMZ resistance, is activated via the NF-κB pathway, which is found to be upregulated in glioma. TMZ also upregulates NF-κB signaling like many other alkylating agents. Magnolol (MGN), a natural anti-cancer agent, has been reported to inhibit NF-κB signaling in multiple my
