Intermolecular Complementation between Two Varicella-Zoster Virus pORF30 Terminase Domains Essential for DNA Encapsidation.

UNLABELLED: The herpesviral terminase complex is part of the intricate machinery that delivers a single viral genome into empty preformed capsids (encapsidation). The varicella-zoster virus (VZV) terminase components (pORF25, pORF30, and pORF45/42) have not been studied as extensively as those of herpes simplex virus 1 and human cytomegalovirus (HCMV). In this study, VZV bacterial artificial chromosomes (BACs) were generated with small (Δ30S), medium (Δ30M), and large (Δ30L) ORF30 internal deletions. In addition, we isolated recombinant viruses with specific alanine substitutions in the putati

First report of coexistence of blaKPC-2 and blaNDM-1 in carbapenem-resistant clinical isolates of Klebsiella aerogenes in Brazil.

Klebsiella aerogenes is an important opportunistic pathogen with the potential to develop resistance against last-line antibiotics, such as carbapenems, limiting the treatment options. Here, we investigated the antibiotic resistance profiles of 10 K. aerogenes strains isolated from patient samples in the intensive-care unit of a Brazilian tertiary hospital using conventional PCR and a comprehensive genomic characterization of a specific K. aerogenes strain (CRK317) carrying both the blaKPC-2 and blaNDM-1 genes simultaneously. All isolates were completely resistant to β-lactam antibiotics, incl

Cardiovascular malformations and complications in Turner syndrome.

BACKGROUND: Turner syndrome (gonadal dysgenesis with sex chromosome abnormalities) is recognized to be a disorder in which cardiovascular malformations are common. The prevalence and natural history of these findings, the risk for aortic dissection, and the occurrence of cardiovascular disease have all been the subject of debate, as have been the American Academy of Pediatrics recommendations for cardiac screening of patients with Turner syndrome. OBJECTIVE: To evaluate a large population of patients both cross-sectionally and longitudinally to determine the prevalence of cardiovascular malfo

[Increased AFP in maternal serum as an indication for invasive diagnosis].

The association of increased maternal serum alpha-fetoprotein levels (MS-AFP) with certain morphologic anomalies of the fetus is fully established. These anomalies are abdominal wall defects (e.g. omphalocele, gastroschisis, complete eventration), neural tube defects (anencephalus, spina bifida, encepalocele) and other malformations (e.g. coccygeal teratoma). The present study compares MS-AFP levels with amniotic fluid alpha-fetoprotein (AF-AFP) and acetylcholinesterase activity, the results of ultrasound, genetic and morphologic examination. Between April 1992 and November 1995 60 patients we

Prenatal Diagnosis and Individualized Perinatal Care in Apparently Nonmosaic Trisomy 9.

BACKGROUND: Nonmosaic Trisomy 9 is a rare chromosomal abnormality, accounting for approximately 2.2%-2.7% of spontaneous abortions caused by autosomal trisomies, and is characterized by a heterogeneous multisystem phenotype and an extremely poor prognosis. Prenatal imaging may demonstrate severe multisystem abnormalities; however, these findings are not specific to Trisomy 9, making cytogenetic testing essential for definitive diagnosis. We report a case of prenatally diagnosed apparently nonmosaic Trisomy 9 in which diagnosis enabled individualized perinatal care. CASE: A 44-year-old multipa

Telomere variant sequences encode the genetic blueprint for allele-specific telomere length.

The ends of human chromosomes are capped by specialized nucleoprotein structures, termed telomeres, which are essential for genome stability. Recent advances in long-read sequencing have enabled allele-specific telomere length measurements at nucleotide resolution, uncovering extreme heterogeneity in telomere length between alleles. The progressive telomere shortening over time makes steady-state telomere length in human somatic cells a promising biomarker for age-associated diseases. However, the mechanisms underlying allele-specific telomere maintenance and its stability remain poorly unders

Genome-wide analysis of the HSF gene family in lily and functional characterization of LdHSFC1 under heat stress.

Genome-wide analyses reveal extensive HSF expansion in lily, and functional assays identify LdHSFC1 as a heat-induced transcriptional repressor that reduces thermotolerance by directly suppressing LdHSFA2. Heat shock transcription factors (HSFs) are key regulators of plant thermotolerance, but their genome-wide characteristics and functions in lily are poorly characterized. We identified 48, 62, and 114 HSF genes in Lilium davidii var. unicolor, L. sargentiae, and L. regale, respectively. Comparative analyses of phylogeny, gene structure, conserved motifs, chromosomal distribution, and synteny

Sucrose synthase gene family analysis in loquat reveals EjSUS4 as a key regulator of sugar accumulation.

INTRODUCTION: Soluble sugar composition critically determines the commercial quality of loquat (Eriobotrya japonica), a prominent Rosaceae fruit. Sucrose synthase (SUS) plays a central role in carbohydrate partitioning and fruit sink strength, yet the SUS gene family in loquat remains uncharacterized. METHODS: Seven EjSUS genes were identified from the loquat genome and analyzed via phylogenetics, chromosomal mapping, synteny, and structural characterization. Spatiotemporal expression profiling was performed during fruit development in two cultivars ('YingShuang' and 'ZheHong 16'). Soluble su

Characterization of GmNPF genes prioritizes candidate genes for soybean yield and protein improvement.

Nitrate uptake and translocation are crucial for plant growth and development, with Nitrate Transporter 1/Peptide Transporter family (NRTs) being critical in mediating nitrogen supply. Soybean (Glycine max) has been a major source of plant-based protein globally, and it requires high amounts of nitrogen to support the plant's development and protein accumulation in seeds. However, genes accounting for nitrogen uptake and usage have yet to be systematically identified. This study focuses on the characterization of Nitrate Transporter 1/Peptide Transporter family (NPF) genes belonging to the NRT

Direct visualization of MCM helicase activation and replisome coupling in situ.

Deciphering the spatial organization of molecular machines that copy the genome remains a fundamental challenge in biology. Essential for eukaryotic DNA replication, Mini-Chromosome Maintenance (MCM2-7) helicases are loaded during G1 as double hexamers (DHs) to license replication origins. Upon activation in S phase, each DH is thought to split into two single hexamers (SHs) that form the active CMG helicases and travel bidirectionally. However, the field has long been divided: biochemical and structural studies define CMG helicases as autonomous, independent motors, while genomic and cellular

Copy Number Variant Detection by Exome/Genome Sequencing Versus Chromosomal Microarray: A Comparative Study of Over 9,000 Clinical Cases.

PURPOSE: Copy number variants (CNVs) are implicated in many health conditions. Chromosomal microarray (CMA) has traditionally been the first-tier test for CNV detection. However, exome and genome sequencing (ES/GS) can identify CNVs alongside other variant types. This study compared CNV detection using CMA versus ES/GS in a large clinical cohort. METHODS: CNV calls from CMA and ES/GS were analyzed in a diverse cohort of over 9,000 individuals tested in a high-throughput clinical laboratory. Concordance between platforms was evaluated, with discordant findings reviewed to determine their natur

An update on the characterization of immunoglobulin loci in Ambystoma mexicanum.

BACKGROUND: We previously reported the genomic characterization of immunoglobulin loci in Ambystoma mexicanum of the laboratory d/d white strain, where the IGH locus gene orientation was incompatible with VDJ recombination, suggesting scaffold orientation errors. A novel 29.1 Gbp A. mexicanum genome derived from an F1 cross between A. mexicanum and A. tigrinum (UKY_AmexF1_1) has recently been released, containing only 220 unmapped scaffolds. Here, we present an updated description of the immunoglobulin loci based on this improved genome assembly. METHODS: Using our previously annotated AmbMex

A dominant mutation in tomato DNA POLYMERASE DELTA 1 causes geminivirus DNA replication catastrophe.

Geminiviruses pose a severe threat to grain and vegetable crops worldwide, often resulting in significant economic losses. In cultivated tomato (Solanum lycopersicum), Ty resistance alleles have been introduced from wild tomato relatives, providing partial to strong resistance to geminivirus infections. The Ty-6 resistance locus from Solanum chilense was previously mapped to chromosome 10. It was recently shown to contain a mutant allele of the DNA POLYMERASE DELTA 1 (POLD1) gene that provides resistance to Tomato yellow leaf curl virus (TYLCV) infections. However, the resistance mechanism rem

Genome-wide characterization of the Bursaphelenchus xylophilus UGT gene family and its potential roles in detoxification and host interaction.

INTRODUCTION: Pine wilt disease caused by Bursaphelenchus xylophilus constitutes a severe threat to global pine forests. The UDP-glycosyltransferase (UGT) gene family is involved in xenobiotic detoxification, environmental adaptation, and other biological processes, but systematic identification of this family in B. xylophilus has not yet been performed. This study aimed to comprehensively identify UGT family members in B. xylophilus and clarify their molecular roles in xenobiotic detoxification and host adaptation regulation. METHODS: Based on genome-wide data, we identified 47 UGT genes in

The hexosamine biosynthetic pathway alters the cytoskeleton to modulate cell proliferation and migration in aggressive prostate cancer.

Castration-resistant prostate cancer (CRPC) progresses despite androgen deprivation therapy, as cancer cells adapt to grow without testosterone, becoming more aggressive and prone to metastasis. CRPC biology complicates the development of effective therapies, posing challenges for patient care. Recent gene-expression and metabolomics studies highlight the Hexosamine Biosynthetic Pathway (HBP) as a critical player, with key components like GNPNAT1 (Glucosamine-phosphate N-acetyltransferase 1) being downregulated in CRPC. GNPNAT1 knockdown in pre-clinical models has been shown to increase growth

MutMap Reveals a Structural Deletion at the Chalcone Synthase Locus Controlling Black Seed Coat in a Gamma-Irradiated Vietnamese Soybean Mutant.

Black soybean is an important functional crop valued for its high anthocyanin content and associated health benefits, attracting increasing interest in plant breeding and mutation-based approaches to enhance its nutritional and agronomic traits. Here, we investigated a DT26 black seed-coat mutant (DT26BS) derived from the Vietnamese cultivar DT26 following gamma irradiation. Genetic analysis of F2 populations indicated that the phenotype is controlled by a single recessive mutation, with additional epistatic interactions observed in other genetic backgrounds. MutMap analysis based on whole-gen

Analysis of DNA double-strand breaks induced by DNA damaging agents in trophoblast organoids.

DNA double-strand break (DSB) is one of the most genotoxic lesions, and unrepaired DSBs can lead to chromosomal aberrations and eventually cause carcinogenesis, ageing and dysfunctions of organs. Therefore, studies of DNA repairs and DNA damage responses (DDRs) corresponding to DSBs has been gathered intense interest. Meanwhile, understanding of the clinical symptoms how dysfunction and accelerated ageing of various organs caused by the effects of DNA damages has been less accomplished. To address this, we have developed a new method that we could investigate DSB formation in trophoblast organ
