Severe mutilating recessive dystrophic epidermolysis bullosa presents with extensive blistering, scarring, and pseudosyndactylies. The skin of most affected individuals lacks normal anchoring fibrils and contains no, or drastically reduced amounts of, collagen VII, the major fibril component. To detect and analyse Onchocerca volvulus mRNAs possessing a spliced leader (SL) sequence, a SL cDNA library was constructed from O. volvulus RNA using a PCR method. Three of the sixty cDNAs analysed displayed significant homology to a known sequence. These cDNAs encode a hsp70, a collagen-like protein and a 60S ribosomal protein. The remaining cDNAs analysed either encoded proteins with no apparent homology to known sequences or they appeared to be generated from RNAs with an internal SL and did not possess an open reading frame.

Osteogenesis imperfecta (OI) is a heritable disorder of connective tissue associated with fractures, osteopenia, and short stature. A prospective trial compared spray thrombin to a bovine collagen haemostatic agent on skin graft donor sites. Twenty donor sites had half of the area covered with the collagen sheet and the other half sprayed with thrombin. Chronic ulcers are well-known complication of diabetes mellitus, often compounded by infection. Healing of lesions is problematic in some cases using conventional treatment. We have treated a group of six hospitalized diabetic patients with chronic ulcer by a combined regimen consisting of metabolic control, parenteral antibiotics, growth factor and porcine graft-young collagenous wettable membrane (YCWM) treatment. Autosomal dominant inheritance of a mild form of osteogenesis imperfecta (osteogenesis imperfecta type I) with different phenotypic expression was found in a family. Processing was markedly increased by growing cells for 2-4 days postconfluence and then adding ascorbate to the medium for 2 days prior to labeling with [3H] proline. Electron microscopic findings of the skin biopsy of a boy with spondyloepiphyseal dysplasia tarda showed several novel features. The basement membrane of the epidermis showed focal irregular thickening and reduplication. The collagen fibres showed foci of fibrous long spacing collagen and marked variation in fibre diameter. Taken together these abnormalities have not been reported in any other condition.
