Genome-wide linkage analyses of extended Utah pedigrees identifies loci that influence recurrent, early-onset major depression and anxiety disorders.

Major depressive disorder (MDD) is a common, clinically heterogeneous disorder often found comorbid with other disorders. We studied recurrent, early-onset MDD (MDD-RE) and anxiety disorders in combination to define powerful phenotypes for genetic study. We used 87 large, extended Utah pedigrees to investigate linkage to 3 phenotypes: "MDD-RE;" "MDD-RE or anxiety;" and "MDD-RE and anxiety;" where in the latter definition the disorders must appear comorbid within an individual. Pedigrees ranged in size from 2 to 6 generations and contained 3 to 42 individuals affected with MDD or anxiety (718 t…

Adaptation of the structured clinical interview for DSM-IV disorders for assessing depression in women during pregnancy and post-partum across countries and cultures.

BACKGROUND: To date, no study has used standardised diagnostic assessment procedures to determine whether rates of perinatal depression vary across cultures. AIMS: To adapt the Structured Clinical Interview for DSM-IV Disorders (SCID) for assessing depression and other non-psychotic psychiatric illness perinatally and to pilot the instrument in different centres and cultures. METHOD: Assessments using the adapted SCID and the Edinburgh Postnatal Depression Scale were conducted during the third trimester of pregnancy and at 6 months postpartum with 296 women from ten sites in eight countries.

The impact of self-stigma on empowerment in major depressive disorder: The mediating role of self-esteem and the moderating effects of socioeconomic and cultural context in an international multi-site study.

BACKGROUND: Major Depressive Disorder (MDD) is a prevalent mental health condition often accompanied by self-stigma, negatively impacting self-esteem and empowerment. This study examines the mediating role of self-esteem in the relationship between self-stigma and empowerment and explores the moderating influence of socioeconomic and cultural contexts in an international sample. METHODS: This cross-sectional study, part of the ASPEN/INDIGO-Depression project, included participants with a diagnosis of MDD from 34 countries. Self-stigma, self-esteem, and empowerment were assessed using, respect…

Associations between COVID pandemic-related post-traumatic stress disorder symptoms and self-care behaviors, fear of hypoglycemia, and depressive symptoms among Chinese adults with type 2 diabetes in the post-pandemic era.

BACKGROUND: The COVID-19 pandemic significantly impacted individuals with type 2 diabetes (T2D), especially in China, where stringent public health measures disrupted healthcare access and heightened psychological stress. The long-term associations of pandemic-related post-traumatic stress disorder (PTSD) symptoms with self-care behaviors, fear of hypoglycemia (FoH), and depressive symptoms in T2D patients remain underexplored. This study examines these associations in Chinese adults with T2D in the post-pandemic period. METHODS: We conducted a cross-sectional study with 242 adults with T2D a

Systematic Analysis of the Mechanism of Polygoni Multiflori Caulis in Improving Depressive Disorder in Mice via Network Pharmacology Combined with Ultra-High Performance Liquid Chromatography Coupled with Quadrupole Exactive Orbitrap Mass Spectrometer.

BACKGROUND AND OBJECTIVE: Depressive disorder (DD) is a common chronic and highly disabling disease. Polygoni Multiflori Caulis (PMC), a traditional Chinese medicine, has been listed in the 2020 edition of the Chinese Pharmacopoeia. Here, the antidepressant effects and mechanisms of PMC were explored for the first time. METHODS: We observed the safety of PMC at a 10-fold clinically equivalent dose. Depressed mice were induced by chronic unpredictable mild stress (CUMS) and were used to evaluate the antidepressant effects of PMC via the sucrose preference test and the tail suspension test. The…

Determining Whether a Definitive Causal Relationship Exists Between Aripiprazole and Tardive Dyskinesia and/or Dystonia in Patients With Major Depressive Disorder, Part 2: Preclinical and Early Phase Human Proof of Concept Studies.

This series of columns has 3 main goals: (1) to explain class warnings as used by the United States Food and Drug Administration, (2) to increase awareness of movement disorders that may occur in patients treated with antipsychotic medications, and (3) to understand why clinicians should refrain from immediately assuming a diagnosis of tardive dyskinesia/dystonia (TD) in patients treated with antipsychotics. The first column in this series began with the case of a 76-year-old man with major depressive disorder who developed orofacial dyskinesias while being treated with aripiprazole as an anti…

Do regional brain volumes and major depressive disorder share genetic architecture? A study of Generation Scotland (n=19 762), UK Biobank (n=24 048) and the English Longitudinal Study of Ageing (n=5766).

Major depressive disorder (MDD) is a heritable and highly debilitating condition. It is commonly associated with subcortical volumetric abnormalities, the most replicated of these being reduced hippocampal volume. Using the most recent published data from Enhancing Neuroimaging Genetics through Meta-analysis (ENIGMA) consortium's genome-wide association study of regional brain volume, we sought to test whether there is shared genetic architecture between seven subcortical brain volumes and intracranial volume (ICV) and MDD. We explored this using linkage disequilibrium score regression, polyge…

Examining the effects of two dietary intervention approaches on disordered eating-related thoughts and behaviors in adults with moderate to moderately severe depressive symptoms.

BACKGROUND: Ultra-processed foods comprise a majority of the American diet and have been linked to increased risk for depression. In response, minimally processed dietary interventions are increasingly being evaluated as tools to improve mental health, including within the growing "Food Is Medicine" movement. However, because these approaches emphasize reducing ultra-processed foods, they raise concerns that dietary restriction may inadvertently increase disordered eating-related outcomes. This paper reports an exploratory analysis of data from a randomized pilot trial that tested two minimall

Interrelationships of postpartum depression and childbirth-related post-traumatic stress disorder with birth memory characteristics among postpartum women: A network perspective.

BACKGROUND: Despite prior research linking birth memory characteristics to postpartum depression (PPD) and childbirth-related post-traumatic stress disorder (CB-PTSD), the intricate pathways are not fully understood. Using network analysis, this study shifts the focus from overall diagnoses to specific symptoms, aiming to uncover the network structure connecting PPD and CB-PTSD symptoms with birth memory characteristics. METHODS: Primiparous women aged 18 to 49 years who had a vaginal delivery and had a child aged 0 to 12 months were recruited from three tertiary level public hospitals in Chi

Esketamine in treatment-resistant depression with and without comorbid borderline personality disorder: A real-world longitudinal study of suicidal ideation and self-harm.

BACKGROUND: Suicidal ideation and deliberate self-harm are frequent and persistent in treatment-resistant depression (TRD), particularly when comorbid borderline personality disorder (BPD) is present. Real-world evidence on antisuicidal outcomes during intranasal esketamine in this population remains limited. METHODS: In this multicentre prospective observational study, 90 outpatients with TRD received intranasal esketamine and were followed for 6 months across five timepoints (baseline, 2 weeks, 1 month, 3 months, 6 months). Forty-five participants had comorbid BPD and 45 had no personality

Contribution of depressive symptom load on prediction of transition to bipolar disorders: Results from the prospective-longitudinal multi-method Early-BipoLife study.

BACKGROUND: Depressive episodes are associated with a higher risk for the onset of bipolar disorder (BD). This study investigates the contribution of specific depressive symptoms using a multi-method approach in persons at-risk for bipolar disorders. METHODS: In the Early-BipoLife study, N = 1083 participants at risk for BD were examined over two years (baseline, 6, 12, 18, and 24 months). Out of 1083 participants, N = 57 (5.3%) transitioned to BD and/or were prescribed with a mood stabilizing medication (lithium, lamotrigine, valproic acid, carbamazepine, quetiapine). At baseline, N = 880 pa
