Interleukin-2 (IL-2) in flounder (Paralichthys olivaceus): Molecular cloning, characterization and bioactivity analysis.

Interleukin-2 (IL-2) is mainly produced by CD4+ T helper lymphocytes, which is an important immunomodulatory cytokine that primarily promotes activation, proliferation and differentiation of T cells. In the present study, flounder (Paralichthys olivaceus) interleukin 2 homologue (poIL-2) was identified for the first time, and its expression patterns were characterized in healthy, virus- or bacteria-infected flounder. The full-length cDNA sequences of poIL-2 was 989 bp with an open reading frame of 423 bp coding a polypeptide of 140 amino acids (aa). The deduced aa sequences shared low similari

A Salmonella type III secretion effector interacts with the mammalian serine/threonine protein kinase PKN1.

Essential to salmonellae pathogenesis is an export device called the type III secretion system (TTSS), which mediates the transfer of bacterial effector proteins from the bacterial cell into the host cell cytoplasm. Once inside the host cell, these effectors are then capable of altering a variety of host cellular functions in order to promote bacterial survival and colonization. SspH1 is a Salmonella enterica serovar Typhimurium TTSS effector that localizes to the mammalian nucleus and down-modulates production of proinflammatory cytokines by inhibiting nuclear factor (NF)-kappaB-dependent gen

D-alanine aminotransferase (Dat) promotes Staphylococcus aureus colonization fitness on human nasal respiratory epithelium.

Nasal colonization by Staphylococcus aureus is an established risk factor for invasive infection, yet bacterial determinants promoting fitness on human nasal mucosa remain incompletely defined. To identify genes required for early colonization of human nasal respiratory epithelium, we colonized human nasal epithelial organoids differentiated at air-liquid interface (HNO-ALI) with a high-density transposon (Tn) library of the methicillin-resistant USA300 strain LAC. TnSeq analysis identified 165 genes that met our threshold for candidate colonization fitness factors. Among these, genes involved

Factors Contributing to the Evolution of mecA-Mediated β-lactam Resistance in Staphylococci: Update and New Insights From Whole Genome Sequencing (WGS).

The understanding of the mechanisms of antibiotic resistance development are fundamental to alert and preview beforehand, the large scale dissemination of resistance to antibiotics, enabling the design of strategies to prevent its spread. The mecA-mediated methicillin resistance conferring resistance to broad-spectrum β-lactams is globally spread in staphylococci including hospitals, farms and community environments, turning ineffective the most widely used and efficient class of antibiotics to treat staphylococcal infections. The use of whole genome sequencing (WGS) technologies at a bacteria

Massively Increased Caries Susceptibility in an Irf6 Cleft Lip/Palate Model.

Patients with cleft lip/palate (CLP) have been reported, in some studies, to exhibit an increased prevalence of caries, although the underlying cause for this increase is unknown. In genetically defined mouse models, studies of postnatal sequelae associated with CLP have been hampered by neonatal lethality. Using a conditional targeting approach, we ablated the major CLP gene Irf6 only in the late embryonic oral epithelium ( Irf6 cKO), bypassing the role of the gene in lip and palate morphogenesis and thus ensuring survival to adulthood. We report that Irf6 cKO mice present with 1) dysplastic

X-linked agammaglobulinemia. A clinical and molecular analysis.

X-linked agammaglobulinemia (XLA), characterized by a profound deficiency of B lymphocytes due to an arrest in B lymphocyte development, is caused by mutations in the gene encoding Btk (Bruton tyrosine kinase). The BTK gene has been cloned and the genomic organization determined. BTK codes for 19 exons and is expressed in all hematopoietic cell lineages but is selectively down-regulated in T lymphocytes and plasma cells. The different Btk domains include PH, TH, SH3, SH2, and the kinase (SH1) domains. Btk, a cytoplasmic protein tyrosine kinase, is involved in cell signaling, although the preci

Molecular features and expression characteristics of a novel tumour necrosis factor-α paralog from snubnose pompano (Trachinotus blochii).

Tumour necrosis factor alpha (TNF-α) is a pivotal cytokine in vertebrate immunity. Considering the increasing scientific interest for TNF-α in piscine biology, the paper detailed the characteristics of a novel tnf-α gene from a high-value mariculture species, Trachinotus blochii. The tbtnf-α complementary DNA (cDNA) of 1385 bp encoded an open reading frame of 762 bp, 3' untranslated region (3' UTR) of 484 bp and 5' UTR of 139 bp. The deduced Tbtnf-α1 showed the highest sequence identity to that of Seriola dumerili (∼83%). The comparative phylogenetics identified the protein as the TNF-α paralo

Role of IL-36γ/IL-36R Signaling in Corneal Innate Defense Against Candida albicans Keratitis.

PURPOSE: Interleukin (IL)-36 cytokines have been shown to play either beneficial or detrimental roles in the infection of mucosal tissues in a pathogen-dependent manner, but their involvement in fungal keratitis remains elusive. We herein investigated their expression and function in mediating corneal innate immunity against Candida albicans infection. METHODS: Gene expression in mouse corneas with or without C. albicans infection was determined by regular RT- and real-time (q)-PCR, Western blot analysis, ELISA or proteome profile assay. The severity of C. albicans keratitis was assessed usin

An orthogonal mammalian gene circuit for selective sensing of taurine-conjugated bile acids.

Beyond their traditional role as dietary lipid emulsifiers, bile acids (BAs) function as pleiotropic signaling molecules that regulate metabolism, inflammation, and diverse disease processes. Taurine-conjugated bile acids (Tau-BAs) and the glycine-to-taurine conjugation ratio (G/T ratio) have emerged as sensitive biomarkers of multiple pathological conditions, including chronic liver diseases, metabolic disorders, and neurodegenerative diseases. However, existing bile acid-responsive sensors and synthetic circuits largely rely on endogenous receptors or signaling pathways, thereby lacking true

Submersion Treatment of Chimeric MrN-VLPs Encapsulating Therapeutic Double-Stranded RNA Effectively Rescues Prawn Viral Infection.

One of the most successful applications to protect aquatic animals from viral infection is the use of RNA interference (RNAi) in the form of double-stranded RNA (dsRNA). However, its administration into animals is usually via injection route (laboratory scale) or mixing feed pellets with bacterial extracts (containing therapeutic compounds); the outcome of which remains to be improved. In this study, we encapsulated purified dsRNA into a virus-like particle (VLP) and delivered it into the prawn tissues by a submersion route using interiorly modified Macrobrachium rosenbergii nodavirus derived

Critical Involvement of the Thioredoxin Reductase Gene (trxB) in Salmonella Gallinarum-Induced Systemic Infection in Chickens.

Salmonella enterica serovar Gallinarum biovar Gallinarum (SG) causes fowl typhoid, a notifiable infectious disease in poultry. However, the pathogenic mechanism of SG-induced systemic infection in chickens remains unclear. Thioredoxin reductase (TrxB) is a redox protein crucial for regulating various enzyme activities in Salmonella serovar, but the role in SG-induced chicken systemic infection has yet to be determined. Here, we constructed a mutant SG strain lacking the trxB gene (trxB::Cm) and used chicken embryo inoculation and chicken oral infection to investigate the role of trxB gene in t

Characterization of Global Gene Expression, Regulation of Metal Ions, and Infection Outcomes in Immune-Competent 129S6 Mouse Macrophages.

Nutritional immunity involves cellular and physiological responses to invading pathogens, such as limiting iron, increasing exposure to bactericidal copper, and altering zinc to restrict the growth of pathogens. Here, we examine infection of bone marrow-derived macrophages from 129S6/SvEvTac mice by Salmonella enterica serovar Typhimurium. The 129S6/SvEvTac mice possess a functional Slc11a1 (Nramp-1), a phagosomal transporter of divalent cations that plays an important role in modulating metal availability to the pathogen. We carried out global RNA sequencing upon treatment with live or heat-k

Transmission of two viruses that cause Barley Yellow Dwarf is controlled by different loci in the aphid, Schizaphis graminum.

Clonal populations of the aphid, Schizaphis graminum, have been separated into biotypes based on host preference and their ability to overcome resistance genes in wheat. Recently, several biotypes were found to differ in their ability to transmit one or more of the viruses that cause barley yellow dwarf disease in grain crops, and vector competence was linked to host preference. The genetics of host preference has been studied in S. graminum, but how this may relate to the transmission of plant viruses is unknown. Sexual morphs of a vector and nonvector S. graminum genotype were induced from p

The Broad-Spectrum Antimicrobial Potential of [Mn(CO)4(S2CNMe(CH2CO2H))], a Water-Soluble CO-Releasing Molecule (CORM-401): Intracellular Accumulation, Transcriptomic and Statistical Analyses, and Membrane Polarization.

AIMS: Carbon monoxide (CO)-releasing molecules (CORMs) are candidates for animal and antimicrobial therapeutics. We aimed to probe the antimicrobial potential of a novel manganese CORM. RESULTS: [Mn(CO)4S2CNMe(CH2CO2H)], CORM-401, inhibits growth of Escherichia coli and several antibiotic-resistant clinical pathogens. CORM-401 releases CO that binds oxidases in vivo, but is an ineffective respiratory inhibitor. Extensive CORM accumulation (assayed as intracellular manganese) accompanies antimicrobial activity. CORM-401 stimulates respiration, polarizes the cytoplasmic membrane in an uncoupler

Cross-kingdom RNA interference in the dysbiosis-mediated network of vocal fold fibrosis: a pathogenic hypothesis and molecular intervention targets.

Vocal fold fibrosis is the principal pathological basis of intractable voice disorders. Previous etiological models have attributed vocal fold scarring solely to mechanical trauma and chemical irritation, whereas the local mucosal microbiota of the vocal folds and its cross-kingdom regulation of host gene expression have long received little attention. Integrating interdisciplinary advances, this review proposes a cross-kingdom pathogenic hypothesis: phonotrauma induces laryngeal dysbiosis and disassembly of the vocal fold epithelial tight junctions, and opportunistic pathogens under stress re

Ferulic acid promotes wound healing through ACOD1-dependent redox balance and tissue regeneration.

BACKGROUND: Acute infected wounds remain a major clinical challenge owing to persistent inflammation, oxidative stress, and impaired angiogenesis. While natural phenolic compounds, like ferulic acid (FA), have antioxidant properties, their role in coordinating wound repair across immune and vascular cells remains unclear. PURPOSE: This study aims to elucidate the multi-modal mechanisms by which ferulic acid coordinates wound repair, focusing on its effects on redox regulation, immune modulation, and angiogenic activation in both cellular and murine models. METHODS: A combined in vitro and in

Mechanisms of Mitochondrial Toxicity and Cytotoxicity Caused by Pseudomonas aeruginosa Pyocyanin in Human Nasal Epithelial Cells.

BACKGROUND: Pseudomonas aeruginosa is an opportunistic pathogen in cystic fibrosis-related chronic rhinosinusitis (CF-CRS) that produces phenazine metabolites pyocyanin and 1-hydroxyphenazine (1-HP), which may have detrimental effects on mitochondria, reactive oxygen species (ROS), Ca2+ signaling, and apoptosis. However, prior studies utilized lung cancer cells or dissociated animal cells. We sought to better define human nasal epithelial responses to phenazines, including the role of Ca2+. METHODS: Live cell imaging was used to measure Ca2+ and mitochondrial function in RPMI2650 nasal carcin

In vitro neurotoxicity comparison: Colistimethate induces higher toxicity than colistin via formaldehyde accumulation.

OBJECTIVES: Colistin as a last-line of defence for treating multidrug-resistant Gram-negative bacterial pathogens is most commonly administered as its inactive prodrug, colistimethate (CMS), for its reduced nephorotoxity. This study addresses the critical gap in understanding neurotoxicity differences between colistin and CMS. Specifically, we aimed to compare the cytotoxicity of CMS and colistin in human neural SH-SY5Y cells and identify the key mediators of CMS-induced neurotoxicity and their underlying mechanisms. METHODS: We used a multi-experimental approach to link phenotypic toxicity t

Deficiency of intestinal alkaline phosphatase affects behavior and microglia activity in mice.

The gut microbiota plays crucial roles in the development and functions of the central nervous system (CNS) as well as in modulation of neurobehavior in heath and disease. The gut brush border enzyme intestinal alkaline phosphatase (IAP) is an important positive regulator of gut microbial homeostasis. In mice, IAP is encoded by Akp3 gene, which is specifically expressed in the duodenum of the small intestine. IAP deficiency alters gut bacterial composition and gut barrier function. Decreased IAP activity has been observed in aging, gut inflammatory diseases, and metabolic disorders. We hypothe

Disruption of MenT2 toxin impairs the growth of Mycobacterium tuberculosis in guinea pigs.

Toxin-antitoxin (TA) systems are abundantly present in the genomes of various bacterial pathogens. TA systems have been implicated in either plasmid maintenance or protection against phage infection, stress adaptation or disease pathogenesis. The genome of Mycobacterium tuberculosis encodes for more than 90 TA systems and 4 of these belong to the type IV subfamily (MenAT family). The toxins and antitoxins belonging to type IV TA systems share sequence homology with the AbiEii family of nucleotidyl transferases and the AbiEi family of putative transcriptional regulators, respectively. Here, we
