Piglet diarrhea is a swine disease responsible for serious economic impacts in the pig industry. Clostridium perfringens beta2 toxin (CPB2), which is a major toxin of C. perfringens type C, may cause intestinal diseases in many domestic animals. N6-methyladenosine (m6A) RNA methylation plays critical roles in many immune and inflammatory diseases in livestock and other animals. However, the role of m6A methylation in porcine intestinal epithelial (IPEC-J2) cells exposed to CPB2 has not been studied. To address this issue, we treated IPEC-J2 cells with CPB2 toxin and then quantified methylation

Dorsal root ganglia (DRG) neurons regenerate spontaneously after traumatic or surgical injury. Long noncoding RNAs (lncRNAs) are involved in various biological regulation processes. Conditions of lncRNAs in DRG neuron injury deserve to be further investigated. Transcriptomic analysis was performed by high-throughput Illumina HiSeq2500 sequencing to profile the differential genes in L4-L6 DRGs following rat sciatic nerve tying. A total of 1,228 genes were up-regulated and 1,415 down-regulated. By comparing to rat lncRNA database, 86 known and 26 novel lncRNA genes were found to be differential.

Immunotherapy with checkpoint inhibitors has revolutionized cancer therapy and enabled considerable therapeutic advances for treating various diseases, including lung cancer. However, the molecular determinants of variable patient outcomes remain unclear. Using long-read RNA sequencing, we mapped the landscape of alternative RNA splicing in human primary lung adenocarcinomas. We identified more than 149,000 full-length messenger RNA (mRNA) isoforms, more than half of which were unannotated and many that occurred in immune-related genes, particularly within the type I interferon response pathwa

Emerging evidence implicates the Stuxnet (Stx) protein in human disease, extending beyond its known role in proteasome-independent degradation. Exploring this further, our investigation into stx downstream targets in Drosophila reveals that loss of the U snoRNA host gene 5 (Uhg5) gene disrupts sleep. This sleep phenotype is linked to inefficient translation of mitochondrial genes, as Uhg5 produces small nucleolar RNAs (snoRNAs) that directly regulate mitochondrial transcripts. Using GoldCLIP technology, we discover that Stx interacts with both Uhg5 and parkin mRNAs. parkin is a key regulator o

Dopaminergic (DA) neurons are highly susceptible to endoplasmic reticulum (ER) burden and redox imbalance, which drive their degeneration and contribute to Parkinson's disease (PD) pathogenesis. Previous work established METTL14-mediated N6-methyladenosine (m6A) modification as critical for dopaminergic (DA) neuron survival. Here, we delineate the underlying mechanism by which m6A dysregulation triggers neurodegeneration through the post-transcriptional modulation of key target genes. Using Mettl14 conditional knockout mice, we identified the ER calcium channel ATP2A3-a key calcium homeostasis

The advent of cystic fibrosis transmembrane conductance regulator modulators has revolutionized cystic fibrosis (CF) care by restoring epithelial ion transport and improving airway physiology in people with CF. These therapies reduce infection burden, especially from Pseudomonas aeruginosa, through enhanced mucociliary clearance and immune responses. Yet, P. aeruginosa often persists, raising concerns about microbial adaptation. Emerging evidence suggests that modulator-altered airways impose novel selective pressures, favouring mutations in genes linked to biofilm formation, iron metabolism a

Medulloblastoma (MB) is the most common malignant tumor of the cerebellum in children. The Sonic Hedgehog (SHH) subgroup of MB (SHH-MB) is driven by aberrant activation of the SHH pathway; however, mutations in genes related to this pathway are relatively rare, posing challenges for therapeutic development. Glypican-6 (GPC6), a heparan sulfate proteoglycan, is highly expressed in SHH-MB. In this study, we demonstrate the synchronous expression of GPC6 with GLI family zinc finger 1 (GLI1) in both the developing cerebellum and medulloblastoma. GPC6 promotes cell proliferation, migration, and inv

Platelets, derived from megakaryocytes (MKs), are crucial for blood clotting. Identifying genes that regulate MK development and platelet production could advance treatments for blood disorders. We found that KxDL motif-containing 1 (Kxd1) knockout (KO) mice exhibited doubled platelet counts without impairment of individual platelet function. Kxd1-KO mice showed enhanced MK progenitor differentiation and rapid polyploid MK formation in vivo and in vitro. Mechanistically, KXD1 deficiency increased TSPAN14 levels by disrupting its endolysosomal trafficking, thus activating the ADAM10-Notch axis

Cold stress is a major environmental constraint limiting crop productivity and quality. Here, we identify a transcription factor MdGeBP3 in apple, which responds to cold stress at both transcriptional and post-translational levels. Functional analysis revealed that overexpression of MdGeBP3 significantly enhances cold tolerance in both apple calli and the model plant Arabidopsis, and the transcript levels of cold-responsive genes were induced in MdGeBP3 overexpression plants. Furthermore, MdGeBP3 was found to participate in anthocyanin biosynthesis. Overexpression of MdGeBP3 in Arabidopsis and

Lysosomes and peroxisomes are essential for cellular homeostasis, yet how their activities are coordinated remains poorly understood. Here, we identify peroxisome-derived ether lipids as key regulators of lysosomal function. A genome-wide CRISPR/Cas9 screen in LYSET-deficient mucolipidosis V cells revealed that disruption of ether lipid synthesis genes or peroxins markedly reduces lysosome accumulation and restores degradative capacity. Genetic or pharmacological inhibition of ether lipid synthesis enhanced lysosomal exocytosis and promoted the clearance of undigested material independently of

The stimulator of interferon genes (STING) is a pivotal regulator of type I interferon (IFN) responses. Although the IFN system is confined to vertebrates, STING is present across metazoans and in some unicellular eukaryotes, suggesting involvement in distinct functions prior to vertebrate divergence. Here we explore the conservation of STING-mediated regulation of polyunsaturated fatty acid (PUFA) metabolism. We find that STING homologs from vertebrates, invertebrates, and unicellular eukaryotes interact with fatty acid desaturase 2 (FADS2), the rate-limiting enzyme in PUFA metabolism, and in

Transcription Factor EB (TFEB) is widely recognized as a key transcription factor regulating lysosomal biogenesis and autophagy. Although the TFEB gene is highly expressed in the testes, the mechanism by which it affects male fertility remains unclear. Here, we report that spermatogonia-specific deletion of TFEB in mice results in a multifaceted reproductive phenotype, including impaired fertility, compromised sperm motility, and attenuated androgen production. Immunofluorescence results showed a significant decrease in the expression levels of TNP1, a marker for spermiogenesis. Under electron

The role of micro-nanoplastics (MNPs) in the elevated risk of papillary thyroid carcinoma (PTC) observed in patients with Hashimoto's thyroiditis (HT) is unclear. We employed an integrated computational and experimental approach to investigate this. By intersecting predicted MNP targets with HT/PTC transcriptomic data, we identified 34 key genes enriched in immune and cancer pathways. Machine learning pinpointed the epigenetic regulator EZH2 as the core driver. Molecular docking and dynamics simulations confirmed stable binding between MNP monomers and EZH2. Intervention with MNPs in vitro HT

Pneumococcal meningitis is the most common form of bacterial meningitis, with high case fatality rates. Bacterial genetics influence disease severity, but the underlying mechanisms remain poorly understood. Here, we evaluated top variants from a genome-wide association study (GWAS) linking Streptococcus pneumoniae genetic variation to disease outcome. Fifteen S. pneumoniae knockout mutants were tested for virulence in a zebrafish embryo meningitis model; six showed reduced virulence compared to wild-type, but four were excluded following competitive index and growth assays. Loss of SPV_1044 (l

Plant growth regulators (PGRs) are extensively applied in cherry cultivation to enhance fruit quality. While sodium 4-chlorophenoxyacetate (4-CPANa) is a registered plant growth regulator for cherries, its specific effects on fruit quality and flavor are poorly understood. Herein, the effects of 4-CPANa on both the flavor enhancement and the undesirable grassy aroma formation in 'Meizao' cherry, as well as the underlying mechanisms, were investigated using integrated metabolomic and transcriptomic analyses. The results indicated that while 4-CPANa improved the taste quality of cherry fruit by

Tumor cells often evade immune pressure via metabolic reprogramming, yet the key metabolic regulators orchestrating this process remain incompletely defined. Here, using in vivo metabolic CRISPR screening under distinct immune pressures, we identified tumor cell-intrinsic solute carrier family 1 member 5 (SLC1A5) as a metabolic node that sustains an immunosuppressive tumor microenvironment. SLC1A5-mediated glutamine metabolism in tumor cells modulated CD8 T cell infiltration and effector function, reshaping tumor responses to immune checkpoint blockade therapy. Glucose deprivation up-regulated

Lymph nodes are uniquely organized niches for immune interactions. Fibroblastic reticular cells (FRCs) facilitate immune cell communication and regulate immune function by producing growth factors, chemokines, and inflammatory cues. Stromal expression of the glycoprotein podoplanin (PDPN) is required for lymph node development, but the requirement for PDPN signaling in FRCs in adult lymph nodes has not been directly tested. Using a conditional in vivo deletion model PDGFRαmGFPΔPDPN, single-cell RNA sequencing (scRNA-seq) reveals that PDPN deletion increases inflammatory signaling in lymph node

Lineage plasticity is a hallmark of pancreatic ductal adenocarcinoma (PDAC) and contributes to tumor heterogeneity and therapeutic resistance. Here, we identify KLF5 as a dynamic master regulator of epithelial lineage identity in PDAC, with dichotomous roles in promoting either classical or basal-like transcriptional programs. Through unbiased proteomic and genetic screens, we uncover the AAA+ ATPases RUVBL1 and RUVBL2 as essential coactivators of KLF5 across both lineage states. We demonstrate that ATP hydrolysis by RUVBL1/2 is required for the stable interaction with an intrinsically disorde

BACKGROUND: Gastric cancer (GC) remains a leading cause of cancer-related mortality globally, with current immunotherapies exhibiting limited therapeutic efficacy owing to the immunosuppressive tumour microenvironment (TME).
MAIN BODY: Lactate, a hallmark metabolic by-product of tumour glycolysis, also functions as a critical metabolic driver. By promoting TME acidification, immunosuppression, and epigenetic reprogramming, lactate emerges as a pivotal regulator in reshaping GC immune evasion and therapeutic resistance, while also strongly correlating with chemotherapy resistance. Furthermore,
