Genetically linked site-specificity of disuse osteoporosis.

UNLABELLED: The genetic influence on bone loss in response to mechanical unloading was investigated within diaphyseal and distal femoral regions in three genetically distinct strains of mice. One mouse strain failed to lose bone after removal of function, whereas osteopenia was evident in multiple regions of the remaining two strains but in different areas of the bone. INTRODUCTION: It is well recognized that susceptibility to osteoporosis is, in large measure, determined by the genome, but whether this influence is systemic or site-specific is not yet known. Here, the extent to which genetic

Genomic characterization of bla KPC transmission via Tn4401a and Tn4401l transposons across Enterobacteriaceae and ESKAPE in Ecuador.

Carbapenem resistance in Enterobacteriaceae poses a major threat to public health because of the limited therapeutic options. The dissemination of the bla KPC gene, which encodes KPC carbapenemases, typically occurs through Tn4401 transposons and plasmids that facilitate its transmission. In this study, we analysed 802 genomes of ESKAPE pathogens and other Enterobacteriaceae from Ecuador, including both clinical and non-clinical isolates, to determine the prevalence and genetic context of bla KPC-2 and bla KPC-3 and their associated mobile genetic elements. Whole-genome analysis revealed sever

Plasma metabolic signature of cardiovascular-kidney-metabolic syndrome and incident stroke: A nationwide prospective cohort study.

Cardiovascular-kidney-metabolic (CKM) syndrome has increasingly affected the middle-aged and elderly population. The relationship between CKM risk factors and stroke at metabolomic level and underlying genetic alterations remains unknown. Elastic net regression was utilized to establish a metabolic signature indicative of CKM risk factors in 108,305 participants in UK Biobank. Multivariable COX proportional hazard models were utilized to determine the association between CKM plasma metabolic signature and incident stroke. Causal relationships between metabolites and stroke were explored throug

Germline mutations and somatic mosaicism in steatotic liver diseases and related liver carcinogenesis.

Steatotic liver diseases, encompassing metabolic dysfunction-associated steatotic liver disease and alcohol-related liver disease, affect nearly one-third of the global population and are a leading cause of cirrhosis and hepatocellular carcinoma. The substantial interindividual variation in disease progression indicates important genetic contributions beyond environmental factors. Genome-wide association studies have identified multiple common variants predominantly affecting lipid metabolism, with PNPLA3 I148M representing the strongest genetic determinant across the disease spectrum. The sha

Sex-specific gut microbiota and metabolite signatures in Parkinson's disease: implications for personalized therapeutics.

Parkinson's disease (PD) is characterized by dopaminergic neuron loss and α-synuclein aggregation in the substantia nigra pars compacta (SNpc). It is a multifactorial disorder with motor and non-motor manifestations and growing evidence suggests that gastrointestinal dysfunction may precede motor onset. Sex differences influence PD risk, onset and clinical features, with men exhibiting higher prevalence and earlier onset, driven by hormonal, genetic, and metabolic factors. The gut microbiota communicates bidirectionally with the central nervous system (CNS) via the gut-brain axis, modulating n

Obesity and employment: Causal evidence from Genetic Instruments in the UK Biobank.

This study explores the causal impact of obesity on employment using data from the UK Biobank, a comprehensive dataset combining genetic, health, and socioeconomic information. We apply a Mendelian Randomisation (MR) approach, using a genetic risk score for a higher body mass index (BMI) as an instrumental variable. This method leverages genetic predispositions determined at conception, enabling us to isolate the exogenous variation in obesity while addressing key endogeneity concerns, such as omitted variable bias and reverse causation. To account for the heterogeneity in obesity's impact acr

Genetic Evolution and Molecular Characterization of PRRSV GP5 in Germany.

Porcine reproductive and respiratory syndrome (PRRS) has been prevalent in Germany for over 30 years, posing a significant threat to the local swine industry. There are limited analyses to offer German PRRSV ORF5 genetic and evolutionary characteristics and GP5 structural and functional features. In this study, a total of 518 sequences of the PRRSV GP5 gene were obtained from the GenBank database, encompassing 102 sequences from Germany to investigate the genetic relationships and GP5 structural and functional features. Similarly, phylogenetic and recombination analyses were used to identify g

Lung squamous cell carcinoma with germline EGFR P848L mutation: a case report.

BACKGROUND: Germline epidermal growth factor receptor (EGFR) mutations have been increasingly recognized as genetic risk factors for non-small cell lung cancer (NSCLC), with T790M, V843I, and R776H constituting the mostly reported variants. The germline EGFR p848L mutation (c.2543C>T, p.Pro848Leu), initially identified by de Gunst et al as a rare germline polymorphism suppressor, presents unsolved clinical challenge due to its ambiguous association with familial NSCLC risk and lack of established therapeutic guidelines. CASE PRESENTATION: We presented a case of a 57-year-old male with a famil

Revisiting the W-Index and Waardenburg Syndrome: A Retrospective Review of Waardenburg Syndrome Diagnoses at a Single Site Hearing Loss Clinic and the Sensitivity, Specificity, and Genotype-Phenotype Correlations of an Elevated W-Index.

Waardenburg syndrome (WS) is a genetically heterogenous condition characterized by variable clinical features including congenital sensorineural hearing loss, pigmentation differences, dysmorphic features including dystopia canthorum, and other manifestations. Pathogenic variants in WS genes, including PAX3, MITF, SOX10, EDNRB, EDN3, and KITLG, account for approximately 3% of congenital hearing loss cases. The genetic heterogeneity, variable expressivity, and different subtypes of WS have made diagnosis challenging. Recent literature suggests that W-index measurements evaluating for dystopia c

Sex chromosome identification and genome curation from a single individual with SCINKD.

In most animal species, the sex-determining pathway is typically initiated by the presence/absence of a primary genetic cue at a critical point during development. This primary genetic cue is often located on a single locus-referred to as sex chromosomes-and can be limited to females (in a ZZ/ZW system) or males (in an XX/XY system). One trademark of sex chromosomes is a restriction or cessation of recombination surrounding the sex-limited region (to prevent its inheritance in the homogametic sex). This may lead to-through a variety of mechanisms-higher amounts of genetic divergence within thi

Podocyte mPGES-2 Determines Renal Aging and Contributes to Senile Osteoporosis.

Renal aging shortens healthspan and propagates organ dysfunction beyond the kidney, yet its molecular drivers remain incompletely defined. Here we identify microsomal prostaglandin E synthase-2 (mPGES-2) as a critical regulator of renal aging and its skeletal consequence. Genetic ablation of Ptges2 improved health indices in aged mice, prolonged median survival, and markedly alleviated glomerulosclerosis, podocyte injury, and renal senescence. Single-cell transcriptomic analysis, together with podocyte- and tubule-specific knockout models, showed that podocyte mPGES-2, rather than tubular mPGE

Non-Isolated Dandy-Walker Malformation: Exome Sequencing Efficacy and Phenotypic Expansions.

Dandy-Walker malformation (DWM) is a rare congenital abnormality of the posterior fossa and the cerebellum and has an incidence of 1 in 10 000 to 30 000 births. Although DWM can present in isolation, it is often associated with other central nervous system (CNS) abnormalities or extra-CNS anomalies (DWM+). A molecular cause is not identified in the majority of individuals with DWM+. This is due, in part, to uncertainty regarding optimal testing strategies and an incomplete understanding of the genetic causes of DWM+. In this study, we analyzed clinical exome sequencing (cES) data from 91 indiv

Genetic predisposition to elevated total immunoglobulin E levels defines a distinct adult-onset-predominant asthma phenotype.

Asthma heterogeneity remains a major barrier in precision medicine. Although elevated total serum immunoglobulin E (IgE) is a hallmark of asthma, even in nonatopic patients, its causal role in asthma pathogenesis is debated. We hypothesized that genetic predisposition to increased IgE defines a distinct asthma endotype. A genome-wide association study of total serum IgE in 1,287 non-asthmatic Japanese adults was used to construct IgE polygenic risk scores (IgE_PRS). Applying IgE_PRS to 745 patients with asthma, we performed cluster analysis incorporating age at onset, total IgE levels, IgE_PRS

Clinical utility of lipoprotein(a) and the ApoB/ApoA-I ratio in the risk assessment of acute coronary syndrome: from single biomarkers to integrated risk stratification.

Acute coronary syndrome (ACS) is a critical clinical emergency resulting from the rupture or erosion of coronary atherosclerotic plaques and subsequent thrombosis. Its pathogenesis is driven by a complex interplay of dysregulated lipid metabolism, inflammatory responses, thrombotic processes, and genetic susceptibility. Although traditional lipid parameters are widely utilized in ACS risk assessment, they remain limited in their ability to account for residual cardiovascular risk, inherited lipid abnormalities, and the overall atherosclerotic burden. Lipoprotein(a) [Lp(a)], an independent, gen

Paleogenomic sex inference of mammoth remains sheds light on the anthropogenic nature of bone accumulations.

Whether large accumulations of woolly mammoth (Mammuthus primigenius) bones reflect natural mortality or deliberate human resource exploitation has long been debated, with major implications for understanding Late Pleistocene human-megafaunal interactions.1,2,3,4,5 Here, we use ancient DNA to investigate site-formation hypotheses by comparing genetic sex ratios from mammoth remains recovered in putative anthropogenic bone accumulations and from geographically dispersed, non-anthropogenic contexts. We studied genome-wide data from 521 woolly mammoths-including 100 mammoths from bone accumulatio

Human genetic ancestry, Mycobacterium tuberculosis diversity, and tuberculosis disease severity in Dar es Salaam, Tanzania.

Infectious diseases have affected humanity for millennia and are among the strongest selective forces. Tuberculosis (TB) is an ancient disease, caused by the human-adapted members of the Mycobacterium tuberculosis complex (MTBC). The outcome of TB infection and disease is highly variable, and co-evolution between human populations and MTBC strains may account for some of this variability. Particular human genetic ancestries have been associated with higher susceptibility to TB, but sociodemographic aspects of the disease can confound such associations. Here, we studied 1000 TB patients from Da

A scoping review of the cost-effectiveness of precision treatment in chronic lymphocytic leukemia.

Chronic lymphocytic leukemia (CLL) is a common, incurable leukemia. Precision treatment for CLL uses genetic testing to align therapeutic selection with patient characteristics. Insurers are uneven in their reimbursement of precision CLL treatment, partly due to uncertain evidence of cost-effectiveness. This review surveys the current cost-effectiveness evidence for precision CLL treatment and identifies areas for future research. We conducted a scoping review of economic evaluations of precision CLL treatments indexed in PubMed, Embase, and Web of Science and published by October 2024. Eight
