The Adaptive Roles of Active Transposable Elements in Insect Hosts.

Active transposable elements (TEs) are capable of generating new insertions in genomes and have historically been viewed as genomic parasites due to their largely detrimental or neutral effects. However, emerging evidence suggests that these elements also play a crucial role in driving adaptive evolution in insects. This mini-review synthesizes recent findings on how active TEs contribute to insect adaptation through various mechanisms, including regulation of gene expression, structural variation, and epigenetic effects. Notable examples of adaptation driven by active TEs include their roles

Identification, Synthesis, Behavioral Activity, and Field Assessment of Components of the Female-Produced Sex Pheromone of Helopeltis theivora (Hemiptera: Miridae), a Devastating Pest of Tea.

Helopeltis theivora is a major pest of tea, cocoa and cashew, and increasing insecticide resistance highlights the need for sustainable alternatives such as pheromones. Previous studies failed to identify female pheromone components attractive to males in the field. Re-examination of female extracts by gas chromatography with electroantennographic detection (GC-EAD) revealed a single active compound, identified by GC-MS as octyl 3-(butanoyloxy)butanoate (III). Enantioselective analysis showed females produce both (3R)-and (3S)-enantiomers. Two related but EAD-inactive compounds, hexyl (3R)-3-(

Integrated Venom Gland Transcriptomic and Venom Proteomic Analyses of the Digger Wasp Cerceris japonica.

Digger wasps are solitary apoid wasps that excavate ground nests and use venom to paralyze insect prey. Recent phylogenetic analyses suggest that digger wasps belong to an ancient lineage of aculeate Hymenoptera and share an evolutionary ancestry with other venomous hymenopterans, including bees and ants. Although the venoms of ants, bees, and social wasps have been extensively studied, those of digger wasps remain poorly characterized in terms of their composition, molecular diversity, and biological activity. In this study, we investigated the venom of the digger wasp Cerceris japonica using

Emamectin benzoate seed coating provides systemic protection of maize against fall armyworm (Spodoptera frugiperda).

The fall armyworm, Spodoptera frugiperda, is a major invasive pest causing severe damage to maize. This study evaluated the efficacy of emamectin benzoate (EB) as a seed-coating insecticide and examined its systemic movement within maize plants. EB coatings did not affect seed germination or seedling vigor. Larval mortality was significantly higher in EB-treated plants, with PRIME GREEN G106 (PG)+EB at 0.5-1.0% producing 69-72% mortality at 20 days after germination. Leaf injury in treated plants remained below 25%, as compared with more than 40% in controls. Synchrotron FTIR (SR-FTIR) and pri

Further development of cinnamodial analogues as mosquito (Aedes aegypti) toxicants for vector control.

Aedes aegypti mosquitoes are vectors of deadly arboviruses such as dengue, yellow fever, chikungunya, and Zika which kill millions worldwide. Prior research by our group discovered a drimane-type sesquiterpene cinnamodial (CDIAL, 1), which was isolated from endemic and medicinal Malagasy plants, that showed promising mosquitocidal activities against A. aegypti. Subsequent synthetic medicinal chemistry efforts to develop more stable and potent mosquitocides derived from CDIAL led to the discovery of novel active compounds with drimane or quinone chemotypes. Here we use medicinal chemistry to fu

Gamma-selinene synthase catalyzes the first step of dihydroagarofuran sesquiterpene alkaloid biosynthesis in Tripterygium.

Tripterygium wilfordii and T. hypoglaucum, which belong to family Celastraceae, are perennial vine shrubs with medicinal uses in treating rheumatoid arthritis, cancer, and male contraception. Among the bioactive ingredients, the macrocyclic dihydroagarofuran sesquiterpene alkaloids (DASAs), also serving as chemotaxonomic markers of Celastraceae, are well-known for cytotoxicity and insecticidal properties. Despite intensive phytochemical investigations over the last half-century, the DASAs biosynthetic pathway remains unsolved. Here, we mined multi-omics data of Tripterygium species and identif

Molecular mechanism of miR-203a targeting Runx2 to regulate thiram induced-chondrocyte development.

Thiram is a kind of organic compound, which is commonly used for sterilization, insecticidal and deodorization in daily life. Its toxicology has been broadly studied. Recently, more and more microRNAs have been shown to participate in the regulation of cartilage development. However, the potential mechanism by which microRNA regulates chondrocyte growth is still unclear. Our experiments have demonstrated that thiram can hamper chondrocytes development and cause a significant increase in miR-203a content in vitro and in vivo trials. miR-203a mimic significantly decrease in mRNA and protein expr

Crystal structure and Hirshfeld-surface analysis of an etoxazole metabolite designated R13.

The etoxazole metabolite R13, systematic name 4-(4-tert-butyl-2-eth-oxy-phen-yl)-2-(2,6-di-fluoro-phen-yl)oxazole (C21H21F2NO2), results from the oxidation of etoxazole, a chitin synthesis inhibitor belonging to the oxazoline class, widely used as an insecticide/acaricide since 1998. The structure of R13 features a central oxazole ring with attached 2,6-di-fluoro-phenyl and 4-t-butyl-2-eth-oxy-phenyl moieties. The overall conformation gives dihedral angles between these rings and the oxazole of 24.91 (5)° (with di-fluoro-phen-yl) and 15.30 (6)° (with t-butyl-eth-oxy-phen-yl), indicating an ove

Adsorption-desorption mechanisms and migration behavior of fluchlordiniliprole in four different soils under varied conditions.

Utilizing infrared spectroscopy coupled with batch equilibrium methods, the adsorption and desorption characteristics of the novel Insecticide fluchlordiniliprole were assessed in four different soil types. It was found that fluchlordiniliprole's adsorption and desorption in these soils were consistent with the Freundlich isotherm, exhibiting adsorption capacities (KF-ads) ranging from 8.436 to 36.269. Temperature fluctuations, encompassing both high and low extremes, impaired the ability of soil to adsorb fluchlordiniliprole. In addition, adsorption dynamics were modulated by several other fa

The effects of Spinosad on zebrafish larvae and THP-1 cells: Associated with immune cell damage and NF-kappa B signaling pathway activation.

Spinosad is a highly effective macrolide insecticide with a wide range of applications. However, few studies have been reported on the effects of Spinosad on immune cells. The immune system is an important line of defense in the human body and plays an important role in maintaining the normal functioning of the organism. Meanwhile, macrophages, neutrophils and Thymic T cells are an important component of the immune system. We studied the immunotoxicity of Spinosad using zebrafish and THP-1 cells. In vivo, Spinosad (0-20 μM) did not cause developmental toxicity in zebrafish, but induced damage

Genus Zanthoxylum as Sources of Drugs for Treatment of Tropical Parasitic Diseases.

The tropical parasitic infections account for more than 2 billion infections and cause substantial morbidity and mortality, and account for several million deaths every year. Majorly parasitic infections in humans and animals are caused by protozoa and helminths. Chronic infections in the host can cause retardation, impairment of cognitive skills, development in young children and weaken the immune system. The burden is felt to a greater extent in developing countries due to poverty, inaccessibility to medicines and resistance observed to drugs. Thus, human health continues to be severely harm

Tanning Beds, Rue, and Major Burns: An Alarming Association.

Skin contact with phytotoxic chemicals present in Ruta graveolens L. and UV light exposure might cause a nonallergic contact dermatitis known as phytophotodermatitis. We present a major burns case report associated with the use of rue (an herb used in Colombia for cultural beliefs) before entering a tanning bed. The patient presented superficial wounds that evolved to deeper ones in a phytophotodermatitis reaction that acted as a chemical burn. The patient presented an estimated percentage of total burn surface area of 85% with second-degree burns. It took two and a half months to apply multip

Characterisation of thymol effects on RDL receptors from the bee parasite Varroa destructor.

A major contributor to bee colony decline is infestation with its most devastating pest, the mite Varroa destructor. To control these mites, thymol is often used, although how it achieves this is not understood. One well-documented action of thymol is to modulate GABA-activated ion channels, which includes insect RDL receptors, a known insecticidal target. Here we have cloned two Varroa RDL subunits, one of which is similar to the canonical RDL subunit, while the other has some differences in M4, and, to a lesser extent, M2 and its binding site loops. Expression of this unusual RDL receptor in

Pregnancy Interventions to Improve Birth Outcomes: What Are the Effects on Maternal Outcomes? A Scoping Review.

Objectives: Interventions in pregnancy are commonly evaluated for their effects on birth outcomes because maternal infection and poor nutrition are the primary contributors to adverse pregnancy outcomes, especially in low- and middle-income countries (LMICs). However, the extent to which such interventions directly impact maternal health and nutrition has not been succinctly characterized. Methods: We conducted a scoping review of systematic reviews and meta-analyses of 27 pregnancy interventions to summarize the evidence of impact on maternal outcomes. Results: Overall, these were reported in
