Evaluation of volumetric DBS for lead analysis using ICP-MS/MS: addressing matrix effects and hematocrit variability.

BACKGROUND: Lead testing in dried blood spots (DBS) offer a convenient alternative to venous blood draw; however, its adoption has been limited by variable extraction efficiency, matrix effects, and hematocrit-dependent recovery. To mitigate these challenges, we explored the utility of volumetric DBS (vDBS) cards, which are known to deposit a fixed volume of blood that gets analyzed. MATERIALS AND METHODS: Samples (quality control, proficiency testing, lead spiked, and patient samples that were positive for lead) were pipetted onto the backs of vDBS cards. Extracts were analyzed using Inducti…

Combined physiological, transcriptomic, and metabolomic analyses reveal the response mechanisms of Phragmites australis to lead (Pb) stress.

Phragmites australis (P. australis) has potential for wetland phytoremediation through nutrient and heavy metal accumulation; however, the molecular mechanisms underlying its tolerance to lead (Pb) remain poorly understood. This study conducted a pot experiment to investigate physiological, transcriptomic, and metabolomic responses of P. australis to a gradient of Pb concentrations (300, 400, and 500 mg/kg) over three exposure durations (50, 75, and 100 d). The results showed that Pb stress inhibited biomass accumulation in a concentration-dependent manner. The antioxidant system responded dyn…

Lead(II) Imides: A New Family of Photoluminescent Heavy Main-Group Compounds With Oxidatively Induced Nitrene Reactivity.

A new family of low-valent lead(II) imide dimers, [R1,R2ArNPb]2 (1-4) (R1,R2ArNH2 = 2,6-((4-R1-C6H4)2CH)2-4-R2-C6H2NH2), features a rhombic Pb2N2 core stabilized by short intramolecular Pb⋯Pb contacts and bulky ligands. These complexes exhibit red/near-infrared solid-state phosphorescence (λmax ≈ 730-790 nm) at 77 K, characterized by sub-microsecond lifetimes and significant ligand-to-metal charge-transfer (LMCT) character. Computational analysis reveals that standard density functional theory (DFT) struggles to capture the energetics of this heavy-element emission, establishing these systems …

Clay minerals alleviate lead contamination induced by soil speciation and oat translocation in neutral and saline soils.

Lead (Pb) contamination in agricultural soils poses a critical environmental challenge, necessitating the development of effective remediation strategies. Clay minerals are widely employed in soil remediation owing to their natural abundance and low cost. Nevertheless, their effectiveness is highly dependent on soil pH conditions. This study evaluated the efficacy of three clay minerals (palygorskite (Pa), sepiolite (Sep), bentonite (Be)) in remediating Pb-contaminated soils under neutral and saline conditions. The addition of clay minerals reduced total Pb by 12-26% in neutral soils, compared…

Effects of hydrogen sulfide donors on inflammatory and oxidative factors in the hippocampus of lead-poisoned rats.

Lead is one of the main environment pollutants, which has strong inhibitory effects on the functioning of the CNS. Exposure to high amounts of lead, even shortly, stimulates glial cells and activates inflammatory pathways in brain. Lead can also increase the amount of oxidative stress. In this study, the effect of hydrogen sulfide (H2S) donors on lead-induced neurotoxicity was investigated. The protective effects of H2S have been proven in various neurological diseases. Lead and H2S were administered for 2 weeks. The animals' cognitive performance, exploratory behavior, and anxiety were evalua…

Concentration-dependent alleviation of lead toxicity in rapeseed by elevated CO2 is associated with rhizosphere microbiome remodeling.

Interactive effects of elevated CO2 (eCO2) and heavy metal pollution on plant-microbe systems are critical for predicting ecosystem responses under future climate scenarios. We investigated how eCO2 modulates lead (Pb) toxicity in rapeseed (Brassica napus) and its rhizosphere microbiome using a two-factor experiment with three CO2 concentrations (400, 550, and 700 ppm) and three Pb levels (0, 70, and 400 mg·kg-1). The alleviating effect of eCO2 on Pb toxicity was strongly concentration-dependent. Moderate eCO2 (550 ppm) significantly increased biomass, photosynthetic rate, nutrient uptake, and…

Integrated Phenotypic and Transcriptomic Profiling Positions ONC212 as a Lead Imipridone in Androgen-Independent Prostate Cancer Models.

Prostate cancer (PCa) remains lethal at advanced stages, partly due to stem-like subpopulations known as prostate cancer stem cells (PCSCs) … potential and mass in a context-dependent manner. Collectively, these findings position ONC212 as a leading imipridone candidate in AIPC models, combining potent inhibition of tumor and stem-like cell functions with a coherent stress-response signature that supports further translational evaluation.

Study on the reproductive toxicity of co-exposure to lead and microplastics at environmental concentrations in zebrafish.

This study aimed to assess the reproductive toxicity of co-exposure to environmental concentrations of lead (Pb) and polyethylene microplastics (PE-MPs) in zebrafish, providing a theoretical basis for evaluating the ecotoxicity and reproductive health risk of combined Pb and PE-MPs exposure at environmental concentrations. Wild-type AB strain zebrafish were divided into four groups: blank control (Ctrl), Pb exposure (Pb), PE-MPs exposure (PE-MPs), and Pb+PE-MPs co-exposure (Pb+PE-MPs). Each group contained 30 adult fish (1:1 sex ratio) and was exposed for 30 days. Results indicated that co-exp…

Halide-Triggered Oxidative Etching as a Key Step in the Formation of Lead-Free Double Perovskite Nanocrystals.

The colloidal synthesis of Ag-based lead-free double perovskite nanocrystals (NCs) involves Ag+-reduction mediated pathways, however, the role of Ag redox chemistry in lattice formation remains poorly understood. Here, we elucidate the crystallization-driven redox processes that govern the formation of Cs2AgBiBr6 NCs. An alternative synthesis has been proposed by carefully studying the effect of reaction temperature behind the in-situ Ag-reduction aided from Ag-oleate complex decomposition at >175 °C, followed by its oxidation mediated by the addition of exclusively used halide precursor, oley…

Unipolar diathermy applied via the terminal pin to release an entrapped LBBAP lead helix: a case report.

BACKGROUND: Left bundle branch area pacing (LBBAP) is increasingly being adopted as a physiological solution to both bradycardia and resynchronization indications. Deep septal positioning of both stylet-less and stylet driven leads can be complicated by unexpected entrapment in myocardial or fibrotic tissue, making advancement or removal of the lead challenging. CASE SUMMARY: A patient undergoing LBBAP implantation demonstrated suboptimal electrical parameters after initial lead advancement. Both helical retraction and counter clockwise rotation was unsuccessful in displacing the lead for a s…

Many roads lead to autophagy: the connection between sulfur metabolism and autophagy during metal stress in plants.

During plant development and in response to stress conditions, autophagy contributes to the intracellular degradation of cellular components and subsequent nutrient recycling. As this process is highly connected to the nutrient status of the plant, autophagy also contributes to the mobilisation of sulfur from source to sink tissues as well as the maintenance of primary sulfate assimilation. In turn, sulfur signals regulate autophagy, with sulfide (an intermediate of primary sulfate assimilation) exerting a repressive effect and sulfur deficiency having a stimulatory effect. In addition to a su

Halogen Substitution as a "Chirality Editor" for Amplifying Spin-Selective Transport in Lead-Free Metal-Halide Hybrids.

Lead-free metal-halide hybrids hold promise as flexible semiconductors for X-ray detection, but their application is constrained by inefficient carrier transport. Here, we introduce halogen substitution on chiral molecular cations as a "chirality editor" to modulate the chiral electrostatic environment and thereby amplify spin-selective transport in bismuth-based hybrids. A series of nine (S/rac/R-XPEA)4Bi2I10 compounds, where X = F, Cl, or Br and XPEA = 1-(4-halophenyl)ethylamine, is systematically investigated, revealing that substitution from F to Br modulates molecular dipoles and local st…

Environmentally friendly vortex-assisted liquid-phase microextraction using a deep eutectic solvent for lead determination in seafood.

This work presents a simple, environmentally friendly vortex-assisted liquid-phase microextraction (VA-LPME) method using a hydrophobic deep eutectic solvent composed of caprylic acid and menthol (molar ratio 1 : 0.1) for the preconcentration and determination of Pb in seafood samples by flame atomic absorption spectrometry (F AAS). The main parameters affecting extraction performance were optimized, including DES type and volume, solution pH, and the use of diluents. The optimized conditions were: sample volume: 10.0 mL; extractant volume: 100 µL; pH: 7.5; rich-phase diluent: HNO3-methanol (1

Sex Differences in Oxidative Stress and Depression-Like Behavior Following Chronic Lead Exposure from Pregnancy to Adulthood.

Lead (Pb) is a neurotoxic heavy metal that crosses the placental barrier and may affect fetal development. However, the effects of chronic developmental Pb exposure on oxidative stress, stress-related endocrine responses, and depression-like behavior, particularly regarding sex differences, remain unclear. This study evaluated the effects of chronic Pb exposure from gestation to adulthood on redox balance, corticosterone responses, and depression-like behavior in male and female mice. Pregnant C57BL/6J mice received 250 ppm lead acetate in drinking water from gestational day 0 until offspring …

Repeated epicardial lead dislodgement in a child with trisomy 21 and complete atrioventricular block: A case report.

UNLABELLED: Lead dislodgement is a rare complication of epicardial leads, which are generally chosen for pacemaker implantation in pediatric patients. Here, we report the case of a 12-year-old girl with trisomy 21 and a complete atrioventricular septal defect. The patient required epicardial pacemaker implantation; however, the epicardial left ventricular lead unexpectedly dislodged twice. This report discusses the potential causes and outlines our management strategy. LEARNING OBJECTIVE: Age, body size, growth-related changes, lifestyle, and durability complicate pacemaker implantation in ch…

Missense substitutions in the BTB domain of ZBTB24 can lead to protein instability and cause ICF2 syndrome.

ZBTB24 is a member of a protein family containing a Broad-Complex, Tramtrack, and Bric a Brac (BTB) domain, which functions in protein-protein interactions. ZBTB24, a transcription factor, binds its DNA targets through its C-terminal zinc finger (ZF) domain. Biallelic ZBTB24 pathogenic variants lead to the rare autosomal recessive Immunodeficiency, Centromeric instability and Facial anomalies type 2 (ICF2) syndrome. The majority of ICF2 patients carry biallelic loss-of-function variants in ZBTB24. The remaining patients harbor missense variants in the ZF domain that compromise the ability of Z…

A novel low-profile self-expanding epidural lead array system that is fully collapsible, deployable, and retrievable.

OBJECTIVE: Introduced in 1970s, Spinal Cord Stimulator (SCS) devices have played a crucial role in managing a wide range of complex and refractory chronic pain, particularly back/leg pain as well as neuropathic pain. Currently, 2 primary types of leads, cylindrical and paddle leads, are prevalent in pain management. While both effectively alleviate pain, cylindrical leads, due to their small size, are susceptible to movement and migration as well as a smaller surface area for coverage, leading to device displacement and failure to provide pain relief. On the other hand, paddle leads offer a la…
