Childhood mitochondrial myopathy with ophthalmoplegia.

A 14-year-old boy with mitochondrial myopathy is described, and the findings on muscle biopsy shown. He presented with mild weakness, and severe exercise intolerance; examination showed ptosis, external ophthalmoplegia and severe muscle wasting. There was a possible family history of a similar disorder. Metabolic study demonstrated severe lactic acidosis on exercise. Oxygen consumption was measured and found abnormally high at rest and on exercise. Biochemical study of extracted muscle mitochondria showed decreased respiratory rates with NAD-linked substrates. These and other results suggest t

Presenile dementia with motor neuron disease -an additional case report-.

A 51-year-old male has suffered the gradual onset of difficulty with memory, concentration and cognition at age 50. He has progressively developed severe dementia accompanied by muscle wasting and fasciculation prominent in the upper arms and shoulders and the bulbar muscles, EMG and muscle biopsy revealed denervation patterns and a CT-scan showed moderate cerebral atrophy. Cases of presenile dementia with motor neuron disease reported in Japan have been reviewed and the possibility of a new clinico-pathologic entity is discussed.

[Clinicoelectroneuromyographic characteristics of neuralgic amyotrophy].

Clinical and genealogical examinations of 155 patients with neural amyotrophies were carried out. Use was made of general and local electromyography, determinations of the speed of impulse transmission along the efferent fibres of the peripheral nerves, and examinations of the H-reflex. The examinations revealed a great diversity of the clinical forms of the neural amyotrophies, each forms having a definite clinical picture, type of hereditary transmission, and characteristic electrophysiological changes. The electroneuromyographic examinations showed that in cases of Charcot-Marie-Tooth's cla

Studies on asterognosis and amyotrophy of the hand in brainstem syndromes. Relation to the symptomatology of tumours at the spinocranial junction.

Fifteen patients with symptoms of an ischaemic lesion in the vertebrobasilar territory were investigated with respect to the presence of asterognosis and amyotrophy of the hand muscles. Stereognostic disturbances were noted in 4 patients with a medullary lesion and in two patients with brainstem syndromes of less defined level. The findings in these cases suggested a lesion of the medial lemniscus. Amyotrophy of the hand muscles was not seen in these patients. The absence of association of astereognosis and amyotrophy of the hand muscles in syndromes due to lower brainstem lesions supports the

[Ringed fibres in skeletal muscles--a contribution to the statement of these structures in unselected obductions (author's transl)].

26 muscles with one or more ringed fibres, corresponding to 2,83%, have been found in 939 muscles out of 313 unselected obductions. Taking into consideration that in the M. rectus femoris alone. 21 ringed fibres were found, raises the quota to 6.7%. After desintegration into primary diseases, the cardiovascular illnesses have been accompanied with ringed fibres in 20.6%, in comparison to the other disease groups which have been rarely or not at all affected. Apart from one case probably traumatic ringed fibres formation, individuals above 49 years old are exclusively striken, at which by far t

Polydeoxyribonucleotide attenuates dexamethasone-induced skeletal muscle atrophy by suppressing oxidative stress and regulating muscle protein turnover.

Glucocorticoid-induced muscle atrophy is characterized by excessive oxidative stress and disrupted muscle protein homeostasis. Polydeoxyribonucleotide (PDRN) exerts tissue-protective and regenerative effects; however, its therapeutic potential in muscle atrophy remains unclear. This study investigated the protective effects of PDRN against dexamethasone (DEX)-induced muscle atrophy in vitro and in vivo. Differentiated C2C12 myotubes were treated with PDRN in the presence or absence of DEX (100 µM). PDRN showed no cytotoxicity, and concentrations of 10, 25, and 50 µg/mL were selected for subseq

Medicinal plants in the management of muscle loss and sarcopenia: A narrative review of preclinical and clinical evidence from eight species.

BACKGROUND: Muscle loss, whether due to aging (sarcopenia), chronic disease, or disuse, is a major contributor to frailty, reduced mobility, and diminished quality of life. With limited pharmacological options available, there is growing interest in natural therapies that support muscle preservation. This narrative review investigates the potential of eight medicinal plants, Cannabis sativa, Chrysanthemum morifolium, Crocus sativus, Eriobotrya japonica, Glycine max, Olea europaea, Salvia rosmarinus, and Sesamum indicum, in promoting muscle health and mitigating sarcopenia and atrophy. These sp

Mechanosensitive Piezo1/Osteocalcin/Irisin Axis Protects Against Disuse-Induced Muscle Atrophy.

Disuse-induced muscle atrophy remains a therapeutic challenge due to its complex etiology. Osteocalcin (OCN) is a bone-derived hormone with metabolic functions, while its role in muscle atrophy remains poorly understood. Here, we identified a mechanosensitive Piezo1/osteocalcin/Irisin axis linking bone mechanotransduction to muscle homeostasis. We showed that bilateral hindlimb immobilization (IMM) markedly reduced circulating undercarboxylated OCN. OCN deficiency exacerbated IMM-induced muscle atrophy, whereas exogenous OCN attenuated muscle atrophy and promoted recovery. Mechanistically, Pie

Muscle atrophy after ACL reconstruction involves molecular mechanisms beyond unloading.

Anterior cruciate ligament reconstruction (ACLR) leads to profound muscle atrophy and weakness that remain resistant to rehabilitation. Although early recovery typically involves a brief period of limb unloading, the degree to which disuse alone accounts for muscle pathology after ACLR remains unclear. Here, we leveraged publicly available RNA-seq datasets of muscle biopsies from vastus lateralis obtained 7 days after ACLR or 10 days after unilateral lower limb suspension (ULLS), each with matched control limbs, to directly compare disuse-driven and ACLR-specific early transcriptional response

Aqueous extract of Atractylodes macrocephala Koidz. improves dexamethasone-induced skeletal muscle atrophy in mice by enhancing mitochondrial biological function.

PURPOSE: The study aims to investigate the therapeutic effects of the aqueous extract of Atractylodes macrocephala Koidz. (AEA) on dexamethasone (Dex) -induced sarcopenia in mice and to explore its possible mechanisms of action. METHODS: This study utilized bioinformatics analysis to explore the primary pathogenic mechanisms of age-related sarcopenia and Dex-induced muscle atrophy. In animal experiments, a mouse model of muscle atrophy was established using Dex, and different doses of AEA were administered for treatment. The therapeutic effects of AEA were evaluated through tests of motor abi

The role of microRNAs in dexamethasone-induced skeletal muscle atrophy.

Muscle atrophy is characterized by a decrease in muscle mass, strength, and activity. Recently, it was determined that microRNAs (miRNAs) can regulate muscle atrophy and that dexamethasone (Dex), an allergy and autoimmune disorder treatment that can induce muscle atrophy. Therefore, this study was designed to identify miRNAs expressed in Dex-induced muscle atrophy in mice using small RNA sequencing. A total of 820 miRNAs were identified, with 58 miRNAs expressed explicitly in atrophic muscles. Dex-induced muscle atrophy miRNAs clustered separately from the differential miRNAs in aging, disuse,
