Endocannabinoid degradation inhibitors ameliorate neuronal and synaptic alterations following traumatic brain injury.

Our previous work showed that lateral fluid percussion injury to the sensorimotor cortex (SMC) of anesthetized rats increased neuronal synaptic hyperexcitability in layer 5 (L5) neurons in ex vivo brain slices 10 days postinjury. Furthermore, endocannabinoid (EC) degradation inhibition via intraperitoneal JZL184 injection 30 min postinjury attenuated synaptic hyperexcitability. This study tested the hypothesis that traumatic brain injury (TBI) induces synaptic and intrinsic neuronal alterations of L5 SMC pyramidal neurons and that these alterations are significantly attenuated by in vivo post-

Formaldehyde induces ferroptosis in hippocampal neuronal cells by upregulation of the Warburg effect.

The mechanisms underlying formaldehyde (FA)-induced neurotoxicity have not yet been fully clarified. Ferroptosis is a novel regulatory cell death and the Warburg effect is involved in regulating neural function. In this study, we investigated whether FA-induced neurotoxicity is implicated in neuronal ferroptosis and determined whether the Warburg effect mediates FA-induced neuronal ferroptosis. We found that FA (0.1, 0.5 and 1.0 mM, 6 h) induced cell death in HT22 cells (a cell line of mouse hippocampal neuron), as evidenced by a decrease in cell viability and an increase in cell mortality; en

Axonal sprouting in the dorsal cochlear nucleus affects gap‑prepulse inhibition following noise exposure.

One of the primary theories of the pathogenesis of tinnitus involves maladaptive auditory‑somatosensory plasticity in the dorsal cochlear nucleus (DCN), which is assumed to be due to axonal sprouting. Although a disrupted balance between auditory and somatosensory inputs may occur following hearing damage and may induce tinnitus, examination of this phenomenon employed a model of hearing damage that does not account for the causal relationship between these changes and tinnitus. The present study aimed to investigate changes in auditory‑somatosensory innervation and the role that axonal sprout

Nutritional status-dependent endocannabinoid signalling regulates the integration of rat visceral information.

KEY POINTS: Vagal sensory inputs transmit information from the viscera to brainstem neurones located in the nucleus tractus solitarii to set physiological parameters. These excitatory synapses exhibit a CB1 endocannabinoid-induced long-term depression (LTD) triggered by vagal fibre stimulation. We investigated the impact of nutritional status on long-term changes in this long-term synaptic plasticity. Food deprivation prevents LTD induction by disrupting CB1 receptor signalling. Short-term refeeding restores the capacity of vagal synapses to express LTD. Ghrelin and cholecystokinin, respective

The selective 5-HT6 receptor antagonist SLV has putative cognitive- and social interaction enhancing properties in rodent models of cognitive impairment.

In the present study, our aim was to investigate whether the novel highly selective 5-hydroxytryptamine6 (5-HT6) receptor antagonist SLV can ameliorate impairments in cognition and social interaction with potential relevance for both schizophrenia and Alzheimer's disease (AD). SLV sub-chronically - treated Wistar rats reared in isolation showed significantly enhanced prepulse inhibition (PPI) and object recognition performance when compared to vehicle - treated rats. In the isolated rats, also a significant reduction in expression of hippocampal neural cell adhesion molecule polysialylation (N

Early Odorant Exposure Increases the Number of Mitral and Tufted Cells Associated with a Single Glomerulus.

UNLABELLED: The highly specific organization of the olfactory bulb (OB) is well known, but the impact of early odorant experience on its circuit structure is unclear. Olfactory sensory neurons (OSNs) project axons from the olfactory epithelium to the OB, where they form spherical neuropil structures called glomeruli. These glomeruli and the postsynaptic targets of OSNs, including mitral and tufted cells (M/TCs) and juxtaglomerular cells, form glomerular modules, which represent the basic odor-coding units of the OB. Here, we labeled M/TCs within a single glomerular module of the mouse OB and s

Ineffective esophageal motility and the vagus: current challenges and future prospects.

Ineffective esophageal motility (IEM) is characterized by low to very low amplitude propulsive contractions in the distal esophagus, hence primarily affecting the smooth muscle part of the esophagus. IEM is often found in patients with dysphagia or heartburn and is commonly associated with gastroesophageal reflux disease. IEM is assumed to be associated with ineffective bolus transport; however, this can be verified using impedance measurements or evaluation of a barium coated marshmallow swallow. Furthermore, water swallows may not assess accurately the motor capabilities of the esophagus, si

Brain Lipotoxicity of Phytanic Acid and Very Long-chain Fatty Acids. Harmful Cellular/Mitochondrial Activities in Refsum Disease and X-Linked Adrenoleukodystrophy.

It is increasingly understood that in the aging brain, especially in the case of patients suffering from neurodegenerative diseases, some fatty acids at pathologically high concentrations exert detrimental activities. To study such activities, we here analyze genetic diseases, which are due to compromised metabolism of specific fatty acids, either the branched-chain phytanic acid or very long-chain fatty acids (VLCFAs). Micromolar concentrations of phytanic acid or of VLCFAs disturb the integrity of neural cells by impairing Ca(2+) homeostasis, enhancing oxidative stress or de-energizing mitoc

Tonic Local Brain Blood Flow Control by Astrocytes Independent of Phasic Neurovascular Coupling.

According to the current model of neurovascular coupling, blood flow is controlled regionally through phasic changes in the activity of neurons and astrocytes that signal to alter arteriole diameter. Absent in this model, however, is how brain blood flow is tonically regulated independent of regional changes in activity. This is important because a large fraction of brain blood flow is required to maintain basal metabolic needs. Using two-photon fluorescence imaging combined with patch-clamp in acute rat brain slices of sensory-motor cortex, we demonstrate that reducing resting Ca(2+) in astro

Dopamine-derived salsolinol derivatives as endogenous monoamine oxidase inhibitors: occurrence, metabolism and function in human brains.

Salsolinol, 1-methyl-6,7-dihydroxy-1,2,3,4-tetrahydroisoquinoline, is an endogenous catechol isoquinoline detected in humans by M. Sandler. In human brain, a series of catechol isoquinolines were identified as the condensation products of dopamine or other monoamines with aldehydes or keto-acids. Recently selective occurrence of the (R)enantiomers of salsolinol derivatives was confirmed in human brain, and they are synthesized by enzymes in situ, but not by the non-enzymatic Pictet-Spengler reaction. A (R)salsolinol synthase catalyzes the enantio-specific synthesis of (R)salsolinol from dopami

Transmission through the dorsal spinocerebellar and spinoreticular tracts: wakefulness versus thiopental anesthesia.

BACKGROUND: Most of what is known regarding the actions of injectable barbiturate anesthetics on the activity of lumbar sensory neurons arises from experiments performed in acute animal preparations that are exposed to invasive surgery and neural depression caused by coadministered inhalational anesthetics. Other parameters such as cortical synchronization and motor ouflow are typically not monitored, and, therefore, anesthetic actions on multiple cellular systems have not been quantitatively compared. METHODS: The activities of antidromically identified dorsal spinocerebellar and spinoreticu

A spermine-coupled cholesterol metabolite from the shark with potent appetite suppressant and antidiabetic properties.

OBJECTIVE: We describe the pharmacological properties of a novel spermine-cholesterol adduct, MSI 1436 (3beta-N-1(spermine)-7alpha, 24R-dihydroxy-5alpha-cholestane 24-sulfate), which causes reversible suppression of food and fluid intake in mammals resulting in profound weight loss, not associated with other signs or symptoms of illness, and which exhibits antidiabetic properties in genetically obese mice. METHODS: Wild-type rodents and strains with genetic obesity were studied. Effects on food and fluid intake, body weight and composition were examined along with pharmacological and toxicolo

Transient receptor potential melastatin 3-dependent disruption of hypothalamic paraventricular nucleus-medial prefrontal cortex serotonergic signaling promotes depressive-like behavior in ovariectomized rats.

Menopause is a natural biological process in women driven by estrogen deficiency and is associated with multiple postmenopausal physiological and behavioral changes. Notably, menopause increases the risk of depression. However, the precise neural mechanisms by which estrogen deficiency contributes to depressive phenotypes remain poorly understood. Estrogen deficiency has been reported to activate transient receptor potential melastatin 3 (TRPM3) channels and promote neuroinflammation by inducing microglial activation and upregulating proinflammatory cytokines. Although the paraventricular nucl

Ventromedial hypothalamus IL-6 signaling mediates the excessive aggressive behavior induced by late-gestational sleep deprivation in male mice.

BACKGROUND: Aggressive behavior is an evolutionarily conserved trait that supports resource acquisition and survival; however, excessive or dysregulated aggression is closely associated with neuropsychiatric disorders and social dysfunction. Maternal sleep deprivation (SD) during late gestation is a common yet underexplored environmental stressor that may disrupt offspring neurodevelopment through immune-mediated pathways. The present study investigated whether SD during late pregnancy alters aggressive behavior in offspring during early adulthood, explored the underlying neuroimmune mechanism

Neddylation as a molecular integrator of metabolic dysfunction and endothelial dysfunction in type 2 diabetes.

Type 2 diabetes mellitus (T2D) is a major global health challenge associated with a high burden of cardiovascular and microvascular complications. Endothelial dysfunction is a hallmark of T2D and represents a critical link between metabolic abnormalities and vascular disease. Chronic hyperglycemia, insulin resistance, obesity-associated inflammation, oxidative stress, and mitochondrial dysfunction disrupt endothelial homeostasis, resulting in impaired nitric oxide bioavailability, vascular inflammation, endothelial senescence, barrier dysfunction, and defective vascular repair. However, the mo

Multi-Omics Analysis Identifies TTYH3 and MPG as Candidate Osteoporosis-Associated Biomarkers in Osteoblasts and Characterizes Spatial Heterogeneity of SPP1 in the Osteoporotic Bone Microenvironment.

Background: Osteoporosis (OP) arises from dysregulated bone metabolism driven by genetic and epigenetic factors. Osteoblasts are central to bone formation, and their functional heterogeneity-shaped by genetic background and receptor expression profiles-may critically influence OP susceptibility. This study aimed to identify osteoblast-specific genes robustly associated with OP and elucidate their underlying pathogenic mechanisms. Methods: We analyzed single-cell RNA sequencing (scRNA-seq), bulk RNA sequencing (bulk RNA-seq), and spatial transcriptomics (ST) datasets of osteoblasts. Differentia

Spinal cord transverse section hinders Wallerian degeneration and neoangiogenesis after peripheral nerve transection: Involvement of Schwann cells' miR-134-5p.

Patients with spinal cord injury (SCI) frequently develop chronic wounds in paralyzed limbs, underscoring the importance of neural regulation and peripheral nerve integrity for tissue repair; however, it remains unclear how concomitant SCI alters the distal responses to peripheral nerve injury (PNI) and the associated microRNA (miRNA) profile. Sprague-Dawley rats underwent sciatic nerve transection alone (PNI group) or complete spinal cord transection at L1-L2 combined with sciatic transection (SCI+PNI group), and distal sciatic nerve stumps were harvested at 0, 3, and 7 days for gross observa

An H₂O₂/Acidic microenvironment-responsive theranostic nanoplatform promotes spinal cord injury repair via MRI-guided synergistic anti-ferroptosis.

The secondary injury cascade following spinal cord injury (SCI) involves complex pathological processes, with the interplay between ferroptosis and oxidative stress serving as a key impediment to neural repair. Ferrostatin-1 (Fer-1), an inhibitor of ferroptosis, faces limitations in clinical translation due to its low delivery efficiency and poor targeting in vivo. To address this, we constructed a theranostic nanoplatform based on mesoporous polydopamine (M-PDA), designated M-PDA@MnO₂@Fer-1, aiming to achieve targeted drug delivery and visual monitoring of the treatment process. The MnO₂ shel

Peak alpha frequency and spectral exponent in postoperative delirium: a high-density EEG cohort study.

BACKGROUND: Present concepts around the neurophysiology of delirium suggest impairments in excitatory/inhibitory balance, leading to EEG slowing, altered synaptic activity and integration of information. We aim to clarify the mechanisms involved, hypothesising that, in postoperative delirium, the EEG peak alpha frequency (PAF; decreased peak frequency indicating slowing in alpha frequency) would slow and the spectral exponent (SE; a higher slope indicating an increased neural inhibitory tone) would be steeper compared with a baseline collected before surgery. The primary aim was to determine t
