BpCYCD3;1 regulates organ growth by activating the cell cycle module in white birch.

Cyclin D proteins are key regulators of cell cycle progression and plant growth. Here, we identified BpCYCD3;1, a CYCD3 family member in birch, and investigated its function. Phylogenetic and sequence analyses showed that BpCYCD3;1 is evolutionarily conserved and contains canonical cell cycle-related domains. Promoter and expression analyses suggested responsiveness to environmental and hormonal signals, with preferential expression in leaves. BpCYCD3;1 overexpression promoted leaf expansion and biomass accumulation, whereas CRISPR/Cas9 knockout reduced vegetative growth. Cytological analyses

Computational model of flower pattern evolution predicts spontaneous emergence of boundary cell types across the petal epidermis.

Petal patterns play an important role in the reproductive success of flowering plants by attracting pollinators and protecting against environmental factors. Some transcription factors (TFs) involved in petal epidermal cell differentiation have been identified, but little is known about the upstream processes that pre-pattern the petal surface to establish their expression domains. Here, we have developed a computational model of petal pattern evolution to investigate this pre-patterning phase, selecting for gene regulatory network (GRNs) that divide the petal into proximal and distal domains

An Argonaute protein traffics from nematode to mouse and is a vaccine against parasitic nematodes.

Argonautes are ancient proteins with well-characterised functions in cell-autonomous gene regulation and genome defence, but less clear roles in non-cell-autonomous processes. Extracellular Argonautes have been reported across plants, animals and protozoa, yet their biochemical and functional properties remain elusive. Here, we demonstrate that an extracellular Argonaute (exWAGO) released by the rodent-infective nematode Heligmosomoides bakeri is detectable inside mouse cells during the natural infection. We show that exWAGO is released from H. bakeri in both vesicular and non-vesicular forms

Hydrogen sulfide donor sodium hydrosulfide-induced heat tolerance in tobacco (Nicotiana tabacum L) suspension cultured cells and involvement of Ca(2+) and calmodulin.

Hydrogen sulfide (H(2)S) is considered as a new emerging cell signal in higher plants. Hydrogen sulfide donor, sodium hydrosulfide, pretreatment significantly increased survival percentage of tobacco suspension cultured cells under heat stress and regrowth ability after heat stress, and alleviated decrease in vitality of cells, increase in electrolyte leakage and accumulation of malondialdehyde (MDA). In addition, sodium hydrosulfide-induced heat tolerance was markedly strengthened by application of exogenous Ca(2+) and its ionophore A23187, respectively, while this heat tolerance was weakened

AaORA, a trichome-specific AP2/ERF transcription factor of Artemisia annua, is a positive regulator in the artemisinin biosynthetic pathway and in disease resistance to Botrytis cinerea.

· Six transcription factors of APETALA2/ethylene-response factor (AP2/ERF) family were cloned and analyzed in Artemisia annua. Real-time quantitative polymerase chain reaction (RT-Q-PCR) showed that AaORA exhibited similar expression patterns to those of amorpha-4,11-diene synthase gene (ADS), cytochrome P450-dependent hydroxylase gene (CYP71AV1) and double bond reductase 2 gene (DBR2) in different tissues of A. annua. · AaORA is a trichome-specific transcription factor, which is expressed in both glandular secretory trichomes (GSTs) and nonglandular T-shaped trichomes (TSTs) of A. annua. Characterization of the defect in a variant of HL-60 promyelocytic leukemia cells with reduced transferrin receptor expression.

The mechanism by which a clone of HL-60 human promyelocytic leukemia cells designated Tf-Gel-1 expresses reduced levels of the transferrin receptor (TfR) was investigated. Tf-Gel-1 was developed by continuous exposure of HL-60 cells to human iron-saturated transferrin covalently linked to the plant toxin gelonin (Tf-Gel); this variant was five- to sixfold more resistant to Tf-Gel than parental HL-60 cells. The amount of cell surface, as well as of solubilized, TfR and the cycling pools of TfR in Tf-Gel-1 cells, as measured by the binding of [125I]Tf, were all decreased to 20-30% of the levels

Structural elucidation of anti-inflammatory withanolides from Physalis minima.

The investigation of the whole plants of Physalis minima led to the isolation of four novel withanolides, designated phyminiolides F-I, together with six known analogues. The structures of 1-4 were elucidated through comprehensive analysis of their spectroscopic data, including HRESIMS, NMR and ECD, while those of 5-10 were identified by comparing their spectroscopic data with the reported literature values. Biologically, compounds 1, 3-4, and 8-10 exhibited moderate anti-inflammatory activities by inhibiting nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated murine RAW 264.7

Disseminated Bisifusarium infection following toxic epidermal necrolysis in a child with B-cell acute lymphoblastic leukemia.

Fusarium is a polyphyletic genus of plant pathogens, members of which can cause opportunistic human infections with varying superficial and systemic presentations, including disseminated infections which typically occur in immunocompromised patients and have a poor prognosis. Treatment is challenging due to intrinsic resistance to many antifungal agents, and antifungal susceptibility testing is therefore essential. Early suspicion, isolation of the organism, and prompt initiation of management are crucial to improving survival. We present a case of disseminated Bisifusarium infection following

NtPDR3, an iron-deficiency inducible ABC transporter in Nicotiana tabacum.

In plants, the ABC transporter PDR (pleiotropic drug resistance) subfamily is composed of approximately 15 genes, few of which have been analyzed. We have identified NtPDR3, a Nicotiana tabacum PDR gene belonging to a cluster for which no functional data was previously available. NtPDR3 was found to be induced in suspension cells treated with methyl jasmonate, salicylic acid, 1-naphthalene acetic acid, or cembrene, a macrocyclic diterpene. In agreement with the identification of a putative iron deficiency element in the NtPDR3 transcription promoter region, we found that iron deficiency in the

Agmatine signaling: odds and threads.

Agmatine is a metabolite of L-arginine. It is formed by the decarboxylation of L-arginine via arginine decarboxylase in bacteria, plants and mammals. It is becoming clear that it has multiple physiological functions as a potential transmitter. Agmatine binds to alpha2-adrenoceptors and to imidazoline binding sites. It blocks NMDA receptors and other ligand-gated cation channels. It also inhibits nitric oxide synthase, induces release of peptide hormones and antizyme and plays a role during cell proliferation by interacting with the generation and transport of polyamines. Although the precise f

The highly conserved defender against the death 1 (DAD1) gene maps to human chromosome 14q11-q12 and mouse chromosome 14 and has plant and nematode homologs.

We have cloned the cDNA encoding the mouse DAD1 (defender against apoptotic cell death) protein. While showing an expected high homology with the previously cloned human and Xenopus DAD1-encoding cDNAs, this sequence has striking homology to partial cDNA sequences reported from O. sativa (rice) and C. elegans (nematode), suggesting the existence of plant and invertebrate homologs of this highly conserved gene. The human and mouse DAD1 genes map to chromosome 14q11-q12 and chromosome 14, respectively. This mapping data supports and extends the previously reported similarities between human chro

Up-regulation of a magnesium transporter gene OsMGT1 is required for conferring aluminum tolerance in rice.

Magnesium (Mg)-mediated alleviation of aluminum (Al) toxicity has been observed in a number of plant species, but the mechanisms underlying the alleviation are still poorly understood. When a putative rice (Oryza sativa) Mg transporter gene, Oryza sativa MAGNESIUM TRANSPORTER1 (OsMGT1), was knocked out, the tolerance to Al, but not to cadmium and lanthanum, was decreased. However, this inhibition could be rescued by addition of 10 μm Mg, but not by the same concentration of barium or strontium. OsMGT1 was expressed in both the roots and shoots in the absence of Al, but the expression only in t

Integrative molecular mechanisms of lodging resistance in rice (Oryza sativa L.) culms: from DELLA-OsGSK2-BZR1 signaling to cell wall engineering and QTL-based breeding.

Rice lodging, permanent culm displacement from the vertical position, reduces grain yield and quality, potentially causing up to ~80% yield loss. Lodging resistance is a multifactorial trait shaped by stem morphology, internal anatomy, and cell wall chemistry. A central unresolved question in the field concerns the relative contribution of lignin versus cellulose to culm mechanical strength, a debate that has significant implications for targeted breeding and cell wall engineering. This review offers a comprehensive integration of two key questions: (1) What are the current molecular regulator

An Improved Immunochromatographic Strip Based on Plant-Derived E2 for Detection of Antibodies against Classical Swine Fever Virus.

Vaccination is an effective method to control the spread of classical swine fever virus (CSFV), which is a major cause of economic losses to the swine industry. Although serological detection assays are commonly used to assess immune status, current methods for monitoring of antibodies (Abs) are time-consuming, expensive, and require cell culture and virus manipulation. To address these problems, the E2 protein of CSFV was expressed in transgenic rice seeds as a labeled antigen for the development of an immunochromatographic test strip (ICTS) for rapid, precise, and cost-effective detection of

An oral vaccine based on U-Omp19 induces protection against B. abortus mucosal challenge by inducing an adaptive IL-17 immune response in mice.

As Brucella infections occur mainly through mucosal surfaces, the development of mucosal administered vaccines could be radical for the control of brucellosis. In this work we evaluated the potential of Brucella abortus 19 kDa outer membrane protein (U-Omp19) as an edible subunit vaccine against brucellosis. We investigated the protective immune response elicited against oral B. abortus infection after vaccination of mice with leaves from transgenic plants expressing U-Omp19; or with plant-made or E. coli-made purified U-Omp19. All tested U-Omp19 formulations induced protection against Brucell

Cytological and transcriptomic responses of WI-38 human lung fibroblasts to plant-exposed metal nanoparticle residues.

Metal-based nanoparticles are increasingly encountered in human exposure scenarios, yet the relationship between residue-level exposure and cellular hazard remains unclear. In this study, iron (Fe), copper (Cu), and silver (Ag) nanoparticles were applied to chilli pepper plants to estimate plant-associated metal nanoparticle residues in the edible fruits. Subsequently, exposure-relevant concentration ranges were defined and examined in normal human WI-38 fibroblasts using integrated cytological and transcriptomic approaches. Metal accumulation was higher in vegetative tissues than fruits, with

P450 Fusion Protein Expressed in E. coli for Regioselective Hydroxylation of Flavonoids.

Plant cytochrome P450 monooxygenases (CYPs) are valuable biocatalysts for the regioselective hydroxylation of aromatic compounds. However, their expression in bacterial hosts is hampered by poor solubility, membrane anchoring and the requirement for redox partners. In this work, we report the design and characterization of modular expression systems that enable the functional production of SbCYP82D1.1 from Scutellaria baicalensis (SbF6H) in Escherichia coli. Both independent expression and synthetic fusion systems were evaluated by combining a CYP with a compatible reductase (ATR2_tr from Arab

Environmental Preconditioning Shapes the Expression and Post-Formulation Stability of Plant Growth-Promoting Traits in Native Actinobacteria.

The functional expression of plant growth-promoting (PGP) traits in soil actinobacteria is conditioned by abiotic factors, yet the combined effects of pH and temperature on their metabolic profiles and the stability of these profiles after encapsulated formulation and post-processing stress remain insufficiently characterized. This study aimed to evaluate the physiological plasticity of native actinobacteria and the expression of plant growth-promoting (PGP) traits under different pH and temperature conditions, as well as their stability after encapsulation, dehydration, and exposure to UV irr
