PURPOSE: Rod spherules are the site of the first synaptic contact in the retina's rod pathway, linking rods to horizontal and bipolar cells. Rod spherules have been described and characterized through electron micrograph (EM) and other studies, but their morphological diversity related to retinal circuitry and their intracellular structures have not been quantified. Most rod spherules are connected to their soma by an axon, but spherules of rods on the surface of the Mus musculus outer plexiform layer often lack an axon and have a spherule structure that is morphologically distinct from rod sp…

BACKGROUND: At least in normal-pressure glaucoma a vascular genesis with hypoperfusion and regulation impairment is discussed. This may lead to malnutrition of retinal ganglion cells and apoptosis. The retinal microvasculature has a small functional reserve. In addition, the retinal microvessels lack the autonomic nerves that are normally found in other tissues. Thus, no systemic influences reach the retinal capillaries apart from circulating hormones or transmitters. Blockers of carbonic anhydrase (CA) may modulate regional blood flow by mediating changes in extra- and intracellular pH. Howev…

Optical coherence tomography is a crucial imaging technique for the detection and analysis of retinal diseases. Precise classification of optical coherence tomography images helps ophthalmologists and healthcare providers design personalized treatment plans in clinical practice. In this paper, we focus on optical coherence tomography image classification for seven types of retinal diseases and normal retina. Although existing deep neural networks could be applied to optical coherence tomography images for the classification, the features were extracted within same hyperspace, causing "feature …

PURPOSE: To investigate a new method for controlling the direction of subretinal hematoma migration during tissue plasminogen activator (tPA) treatment by selecting the tPA injection site.
METHODS: Retrospective, interventional case series. An artificial retinal detachment was created by subretinal injection of tPA into the healthy lower retina of 18 patients, temporal to the fovea. The posterior pole of the detached retina was continuous with the subretinal hematoma, allowing tPA to access the lesion. After tPA infusion, the vitreous cavity was filled with SF 6 gas, and the surgery was concl…

Retinoic acid (RA) is a key morphogen in human retinal development, activating transcriptional programs that drive retinal progenitor differentiation and photoreceptor development, ensuring proper spatial organisation within the neural retina, essential for vision. Despite its well-established role in retinal patterning, the concentration-dependent effects of RA on human retinal cell fate specification and the regional definition of the primate macula and peripheral retina remain poorly understood. Here, we show that temporal and dosage-dependant modulation of RA during human retinal organoid …

Retinal ex vivo systems have been widely used to investigate diverse physiological and pathological processes in the retina. Although retinal ex vivo systems are generally considered to approximate in vivo environment of the retina, careful evaluation of how ex vivo conditions affect cellular function is necessary for accurate interpretation of such data. Here we analyzed the responses of retinal Müller glia to photoreceptor injury in vivo and ex vivo to assess the effects of ex vivo culture on the phenotypes of Müller glia in pathological contexts. A rat model of photoreceptor injury was gene…

Synaptic failure predicts cognitive decline in Alzheimer's disease (AD), yet its impact and molecular drivers in the human retina remain unclear. Leveraging the retina as an accessible central nervous system (CNS) proxy, we integrated spatially resolved histopathology of retinal cross-sections with ultrastructural, proteomic, and biochemical profiling across independent postmortem cohorts spanning normal cognition, mild cognitive impairment due to AD (MCI), and AD dementia. We uncover early, progressive degeneration of excitatory glutamatergic synapses, evidenced by losses of presynaptic vesic…

Mutations in the ciliary gene INPP5E, encoding inositol polyphosphate-5-phosphatase E (INPP5E), can cause retinal degeneration as part of the ciliopathy Joubert syndrome or non-syndromic retinitis pigmentosa (RP). INPP5E regulates the membrane makeup of the primary cilium; however, its function in the specialized sensory photoreceptor cells of the human retina remain unclear. Here, we utilize control and CRISPR/Cas9-generated INPP5E knockout (INPP5ED477N/D477N) human induced pluripotent stem cells (iPSCs) to generate retinal organoids (ROs). Through proteomic and immunofluorescence analysis, w…

N6-methyladenosine (m6A) modification is the most prevalent posttranscriptional epigenetic modification in mammalian mRNAs, and it has been implicated in the regulation of nervous system development by modulating mRNA metabolism. VIRMA is the largest core subunit of the m6A methyltransferase complex and is essential for the assembly and stability of the m6A methyltransferase complex. In the retina, m6A methylation modification is widely distributed in various cellular layers and is essential for retinal homeostasis. Here, we demonstrate that VIRMA-mediated m6A modification is essential for ret…

KEY MESSAGES: WHAT IS KNOWN : • The retina, one of the most energy-demanding neural tissues in the human body, predominantly relies on glycolysis for energy production even under aerobic conditions. Lactate is the primary metabolic byproduct of glycolysis, and its functional scope significantly surpasses that of a mere metabolic waste product. • Lactate exerts extensive influence over diverse biological processes and cellular metabolic pathways. Notably, lactate can serve as a precursor for a novel post-translational modification of proteins, known as lactylation. Emerging research has increas…

Vision impairment following ischemic stroke is a prevalent complication that significantly compromises patients' quality of life. Inflammatory responses critically contribute to retinal dysfunction in this condition. Retinal myeloid cells contributed to the retinal inflammatory response, which presented heterogeneity after retinal injury. In this study, we employed the classical middle cerebral artery occlusion (MCAO) mouse model to simulate ischemic stroke. We demonstrated that stroke-induced retinal damage manifests as diminished photoreceptor responses and increased retinal cell apoptosis b…

The morphogenesis and cellular interactions in developing retina are incompletely characterized. The full understanding needs a precise mapping of the gene expression with a single-cell spatial resolution. Here, we present a spatial transcriptomic (ST) resource for the developing human retina at six developmental stages. Combining the spatial and single-cell transcriptomic data enables characterization of the cell-type-specific expression profiles at distinct anatomical regions at each developmental stage, highlighting the spatiotemporal dynamics of cellular composition during retinal developm…

Astrocytes throughout the central nervous system mediate a variety of functions to support proper tissue physiology, including the regulation of blood flow and providing metabolic support to neurons. There is also growing appreciation for their role in directly modulating neuronal excitability and information transfer. Recently, we reported that astrocytes in the retina exhibit an array of neuronal-associated microstructural motifs whose structure and placement suggest roles in monitoring neuronal electrical activity or direct modulation of excitability. In this study, we record whole-cell pat…

Early and accurate diagnosis of retinal diseases is crucial, as many of these conditions can be prevented or managed effectively with timely treatment. Optical Coherence Tomography (OCT) is a widely used medical imaging technique that provides high-resolution visualization of the retina, allowing the detection of abnormalities. Current state-of-the-art methods predominantly rely on extracting either local or global features using Convolutional Neural Networks or Vision Transformers from OCT images. Moreover, few existing ensemble methods primarily focus on concatenation and decision-level feat…

Microglia, the parenchymal macrophage of the central nervous system, serve crucial remodeling functions throughout development. Microglia are transcriptionally heterogenous, suggesting that distinct microglial states confer discrete roles. Currently, little is known about how dynamic these states are, the cues that promote them, or how they impact microglial function. In the developing retina, we previously found a significant proportion of microglia express CD11c (Integrin αX, Itgax, subunit of complement receptor 4) which has also been reported in other developmental and disease contexts. He…

Astigmatic eyes are commonly believed to focus on distant targets with a circle of least confusion on the retina when the spherical equivalent (SE) is 0. We tested this hypothesis in young adults by measuring ocular focus under induced astigmatism. Twenty healthy volunteers (20.2 ± 0.8 years) first received full refractive correction. We then applied three regular astigmatic states (1, 2, and 3 D; axis 180°), each with an SE of 0 D. Over-refraction was recorded for 3 s at 5 Hz using an open-field autorefractor. The objective SEs were significantly myopic under all conditions (- 0.33 ± 0.09 D, …

Efforts in vision restoration have been focused on a condition called Retinitis Pigmentosa, where photoreceptors in the retina degenerate while the rest of the visual pathway remain mostly intact. Retinal implants that directly stimulate retinal ganglion cells have shown promising but limited results in patients so far. Apart from technical limitations, cross-modal plasticity of visual areas might contribute to this problem. We therefore investigated if the primary visual cortex (V1) of the rd10 mouse model for retinal degeneration became more sensitive to auditory or tactile sensory inputs. A…

Adaptation is a universal aspect of neural systems that changes circuit computations to match prevailing inputs. These changes facilitate efficient encoding of sensory inputs while avoiding saturation. Conventional artificial neural networks (ANNs) have limited adaptive capabilities, hindering their ability to reliably predict neural output under dynamic input conditions. Can embedding neural adaptive mechanisms in ANNs improve their performance? To answer this question, we develop a new deep learning model of the retina that incorporates the biophysics of photoreceptor adaptation at the front…
