Hyperglycaemia in pregnant rats causes sex-related vascular dysfunction in adult offspring: role of cyclooxygenase-2.

What is the central question of this study? Hyperglycaemia during pregnancy induces vascular dysfunction and hypertension in male offspring. Given that female offspring from other fetal programming models are protected from the effects of fetal insult, the present study investigated whether there are sex differences in blood pressure and vascular function in hyperglycaemia-programmed offspring. What is the main finding and its importance? We demonstrated that hyperglycaemia in pregnant rats induced vascular dysfunction and hypertension only in male offspring. We found sex differences in oxidat

A disparate trace element metabolism in zebu (Bos indicus) and crossbred (Bos indicus × Bos taurus) cattle in response to a copper-deficient diet.

Copper deficiency is a commonly diagnosed problem in cattle around the globe. In Jimma, Ethiopia, 8 zebu (Bos indicus) and 8 zebu ×: Holstein Friesian cross (Bos taurus ×: Bos indicus) heifers were used in an 11-wk study to investigate breed type differences and effects of Cu deficiency on concentrations of trace elements in plasma and edible tissues as well as mRNA expression of Cu-related genes. Heifers were fed a grass diet (6.4 ± 0.2 [SEM] mg Cu/kg DM) supplemented with 1 mg Mo/kg DM in wk 1 to 4 and 2 mg Mo/kg DM in wk 5 to 11, with blood samples collected every 2 wk and tissue collection

A new horizon into the pathobiology, etiology and treatment of migraine.

Sexual dimorphism in the prevalence of migraine (70% women 30% men) suggests the involvement of reproductive hormones in a women's life. Excessive estrogen during menstruation directly stimulate estrogen receptor alpha thickly populated in trigeminal ganglia and periaqueductal gray which manifest as menstrual migraine. In contrast increased progesterone during pregnancy evokes progesterone receptors A/B, which coexist with ERs, providing complete remission from migraine episodes. Moreover, estrogen also increases nociception through extracellularly signal-regulated kinase (ERK) stimulation and

Nitric oxide dynamics and endothelial dysfunction in type II model of genetic diabetes.

Although diabetes is a major risk factor for vascular diseases, e.g., hypertension and atherosclerosis, mechanisms that underlie the "risky" aspects of diabetes remain obscure. The current study is intended to examine the notion that diabetic endothelial dysfunction stems from a heightened state of oxidative stress induced by an imbalance between vascular production and scavenging of reactive oxygen/nitrogen species. Goto-Kakizaki (GK) rats were used as a genetic animal model for non-obese type II diabetes. Nitric oxide (NO) bioavailability and O2- generation in aortic tissues of GK rats were

Spermidine protects mitochondria against amyloid-β oligomers.

BACKGROUND: Alzheimer's disease (AD) is a neurodegenerative condition characterized by amyloid-β (Aβ) plaques in the brain. In early pathological conditions, Aβ oligomers localize in mitochondria to promote disease progression by causing mitochondrial dysfunction. OBJECTIVE: Establish a robust AD pathological in vitro model and assess the therapeutic effect of spermidine (SPD) against extracellular Aβ oligomers. METHODS: In this study, high-resolution respirometry, flow cytometry, and immunoblotting techniques were used to characterize the negative effects of Aβ oligomer localization in mito

Metal-organic framework nanozymes as next-generation biosensing platforms: from catalytic design to point-of-care detection in real-world samples.

Metal-organic frameworks (MOFs) have emerged as a transformative class of nanozymes, offering unprecedented tunability in catalytic activity, structural porosity, and surface functionality for biosensing applications. This review traces the evolution of MOF nanozymes from their foundational design principles to their integration into advanced point-of-care (POC) diagnostic devices. It further examines the structural and mechanistic basis of enzyme-mimicking behavior in MOFs-encompassing peroxidase-, oxidase-, catalase-, and superoxide dismutase-like activities-alongside signal transduction str

A Small-Molecule Platform Demonstrates Light-Activated Synergy Between Cuproptosis and Photodynamic Tumor Therapy.

Cuproptosis offers a strategy to overcome chemotherapy resistance; however, it lacks tumor selectivity. Although glutathione (GSH) is highly expressed in tumors and serves as a conventional trigger for selective cuproptosis, it paradoxically inhibits cuproptosis by binding copper ions. Although recent nanoplatforms combined photodynamic therapy (PDT) and cuproptosis to address these challenges, their complexity raises safety concerns and obscures mechanisms. Herein, a copper(II) complex (NC), synthesized by linking 8-hydroxyquinoline to Nile Blue, represents the first small-molecule platform t

Photodynamic activity of a mitochondria-localized G-quadruplex-responsive fluorescent ligand: Cellular effects and photo-enhanced cytotoxicity.

Mitochondrial localization concentrates photosensitizing molecules near redox-sensitive organelles. In this study, we investigated SPN, a mitochondria-targeted G-quadruplex (G4)-responsive ligand. Photochemical characterization confirmed that SPN generates robust light-triggered ROS and exhibits photooxidative activity in vitro. In cancer cell models under dark conditions, SPN treatment was associated with cell-cycle perturbation, loss of mitochondrial membrane potential, altered mitochondrial morphology, elevated mitochondrial superoxide-sensitive fluorescence, and accumulation of MDC-positiv

Cold plasma-induced oxidation of tannic acid enhances bactericidal activity via quinone-driven singlet oxygen generation.

This study investigates the potential of cold plasma to induce a pro-oxidant transition in tannic acid for antimicrobial applications. Cold plasma treatment of aqueous tannic acid produced plasma-activated tannic acid solution (PAT) with distinct oxidative structural modifications. Browning kinetics and spectroscopic analyses (FTIR and XPS) indicated the conversion of tannic acid into reactive quinone structures without degradation into gallic acid. This oxidative transformation triggered the generation of reactive oxygen species (ROS), including superoxide anion, hydrogen peroxide, and single

Nicotinamide counteracts Rotenone-induced mitochondrial and neuronal dysfunction in a translational early-life model.

Mitochondrial function is essential for key neurodevelopmental processes, including cellular proliferation, differentiation, migration, synaptogenesis, and synaptic pruning. Previous research from our group demonstrated that neonatal administration of Rotenone (Rot), a mitochondrial complex I inhibitor, led to mitochondrial dysfunction and schizophrenia-like behavioral phenotypes. In this study, we aimed to identify possible cellular pathways disrupted by Rot exposure that may underlie these behavioral alterations. Thus, primary cortical neurons were treated with 1.325 nM Rot for 24 h, a conce

Azelaic Acid delays aging and prolongs lifespan via resisting oxidation stress mediated by IIS pathway coupling hsf-1 in C. elegans.

Azelaic acid (AzA), an antioxidant and anti-inflammatory compound from cereals used in food preservatives and cosmetics, was investigated for anti-aging effects in Caenorhabditis elegans. AzA improved locomotive activities and muscle morphology during aging, extended lifespan under both normal and oxidative stress conditions, and reduced paralysis in neurodegenerative models. It decreased reactive oxygen species levels and enhanced superoxide dismutase (SOD) activity and SOD-3 protein expression. Quantitative real-time PCR (qRT-PCR) analysis revealed that AzA differentially regulated oxidative

Multienzyme-mimetic nanozym es for food safety detection: Cascade catalytic design, activity regulation, and sensing applications.

Ultrasensitive detection of trace-level hazardous substances (pesticide residues, heavy metals, bacteria, veterinary drugs, food additives, organic pollutants) is critical for food safety. Conventional methods are costly, complex, or insufficiently sensitive in complex matrices. Multienzyme-mimetic nanozymes, integrating peroxidase, oxidase, catalase, superoxide dismutase, and laccase activities, enable self-cascading signal amplification, overcoming the limited versatility of single-activity nanozymes. This review discusses their rational design and activity modulation, emphasizing how intern

Effect of the insecticide cypermethrin on behaviour and oxidative stress in the spider Loxosceles laeta.

Loxoscelism, caused by bites from Loxosceles spiders, is a global health concern. Control of these spiders with insecticides remains poorly understood and requires assessment at both biochemical and behavioural levels. We evaluated the ability of Loxosceles laeta to detect cypermethrin-treated surfaces and examined the effects of exposure on enzymatic activity, oxidative stress, biomolecule oxidation and locomotor behaviour. Spiders preferred untreated surfaces and increased movement speed on treated ones, indicating a repellent effect. Acetylcholinesterase activity remained unchanged, whereas

Visible-light-promoted catalyst-free benzylic C-H oxidation using molecular oxygen as a green oxidant.

Herein, we report a catalyst- and additive-free strategy for the visible-light-promoted carbonylative oxidation of benzylic C-H bonds employing molecular oxygen as the sole oxidant. This operationally simple protocol enables the conversion of a broad range of aromatic alkanes into valuable ketones, including complex drug derivatives and intermediates under mild conditions using ethyl acetate as a green solvent. Mechanistic studies reveal that the reaction proceeds via a substrate-oxygen charge-transfer (CT) complex upon photoexcitation, generating reactive oxygen species (singlet oxygen and su

Photo-responsive Ce-hemin nanozyme-based portable colorimetric sensor for tetracyclines assay.

A cerium ion-modified hemin polymer (Ce-hemin) was synthesized via a facile route, exhibiting photo-responsive oxidase-like activity and excellent stability. The Ce doping effectively prevented hemin aggregation, facilitated electron transfer, enhanced photocatalytic activity, and created a high adsorption capacity for tetracyclines (TCs). Taking advantage of the photo-sensitivity of TCs, a "turn-on" colorimetric sensing platform based on Ce-hemin nanozymes and smartphone-readout was constructed for visual analysis of TCs. This sensor offers a wide detection range and a low limit of detection

Photoinduced electron transfer in a bilirubin-oxygen complex: the triplet-reactant computational challenge.

Although bilirubin photochemistry is central to neonatal jaundice phototherapy, the mechanism of bilirubin photooxidation remains unclear. Here, we use a comprehensive computational approach to investigate whether this mechanism might be initiated by photoinduced electron transfer (PET) to molecular oxygen (O2), generating superoxide (O2˙-). For this purpose, we employed a simplified bilirubin model compound-tetramethyldipyrrinone (TMD). We use a combination of multireference, coupled cluster methods, and density functional theory techniques to assess the feasibility of the TMD-O2 complex form

Covalent Modulation of Protein Misfolding and Aggregation Processes in the Context of Neurodegenerative Diseases.

Misfolded protein aggregates represent major histopathological hallmarks of neurodegenerative diseases, differing in the structural components and brain regions affected. Furthermore, the formed assemblies act as key players in developing and fostering neurotoxic processes, with distinct mechanisms depending on the stage of the amyloid cascade. Particularly, the oligomer intermediates are now considered as the main drivers of neurotoxicity, thus requiring an early antiaggregant therapeutic intervention to achieve a significant neuroprotective efficacy. Among different strategies, direct intera
