Proteomic Signatures and an Injury-Stress Endotype in Myositis-Associated Interstitial Lung Disease.

RATIONALE: Idiopathic inflammatory myopathy-associated interstitial lung disease (IIM-ILD) is a major cause of morbidity and mortality. We tested whether quantitative myositis-specific autoantibodies and proteomic profiling capture biological heterogeneity and prognosis beyond categorical serology. METHODS: Myositis-specific autoantibodies were quantified using the luciferase immunoprecipitation systems assay, and 184 serum proteins were measured in 226 IIM patients; 199 with higher-ILD-risk autoantibodies (Jo-1/MDA5/PL-7/PL-12/EJ), 27 with lower- ILD-risk autoantibodies (Mi-2/NXP2/TIF1γ) and

Male Breast Cancer: Bridging the Gaps of Neglect-A Global Call for Equitable Care and Inclusion.

BACKGROUND AND AIMS: Male breast cancer (MBC) is a rare but increasingly recognized malignancy that remains underrepresented in research, policy, and clinical practice. Persistent misconceptions that breast cancer is exclusively female disease contribute to delayed presentation, advanced-stage diagnosis, and poorer outcomes in men. This article examines global disparities affecting awareness, diagnosis, treatment, research inclusion, and psychosocial care, and proposes strategic actions to advance equitable and inclusive oncology. METHODS: This article is based on a targeted review of peer-re

AKAP12 as a signal-compartmentalizing Hub across diseases: Context-dependent functions and translational potential.

A-kinase anchoring protein 12 (AKAP12; Gravin/SSeCKS) is a multidomain scaffold that coordinates spatially restricted signaling rather than acting as a single linear effector. By organizing protein kinase A (PKA), protein kinase C (PKC), phosphodiesterases (PDEs), proto-oncogenic non-receptor tyrosine kinase (Src)-related modules, and cytoskeleton- or adhesion-associated proteins within local signaling microdomains, AKAP12 regulates kinase output, PDE-dependent cyclic adenosine monophosphate (cAMP) gradients, cytoskeletal remodeling, cell migration, barrier integrity, inflammatory activation,

HMGB1-mediated neuroinflammation: molecular mechanisms and emerging therapeutic approaches.

High mobility group box 1 (HMGB1) has emerged as a central inflammatory mediator linking cellular stress and tissue injury to sustained neuroinflammation in the central nervous system. Although originally characterized as a nuclear chromatin-binding protein, HMGB1 acquires potent pro-inflammatory activity following cytoplasmic translocation and extracellular release, where it functions as a damage-associated molecular pattern. The inflammatory actions of HMGB1 are regulated by its redox state and post-translational modifications, which determine receptor engagement and downstream signaling. Ex

The Childhood Cancer and Leukemia International Consortium (CLIC): Expanding global collaboration in pediatric cancer etiology research.

Childhood cancers are rare, but incidence has risen modestly in countries with robust registration, partly reflecting improved diagnosis. In high-income countries, cancer is the leading cause of disease-related death in children. Marked inequities in incidence, survival, and research capacity underscore the need for large-scale collaboration to identify environmental, genetic, and contextual determinants of risk. The Childhood Cancer and Leukemia International Consortium (CLIC) was established in 2007 to study the etiology of childhood leukemia and later expanded in 2019 to include other child

A Mini-Review on the Symptoms and Pathogenesis of Psoriasis and Recent Nanotechnological Developments in its Treatment Strategies.

Psoriasis is a chronic disease that is common and incurable. In psoriasis, skin cells proliferate more quickly than normal cells, suggesting a possible immune system connection. The topical treatment of psoriasis is best when applied in combination with anti-inflammatory medications; however, the lack of an appropriate delivery method limits the drugs' ability to be delivered. Due to several problems, including adverse reactions and problems with penetration, the current oral and topical treatments for psoriasis fall short of meeting the need for an optimal drug delivery method. None of them,

A review of recent studies on CRISPR/Cas9-mediated genome editing in a variety of muscle-related genetic disorders.

The human body is capable of mutating a single gene to produce a wide range of debilitating disorders. Genomic editing for disease prevention via phenotypic reversal was a significant challenge prior to the development of clustered regulatory interspaced short tandem repeats (CRISPR) and CRISPR-associated protein (Cas) systems. Gene therapy-editing a patient's DNA to correct a particular mutation-and treating human diseases that have not responded to conventional medicine are two areas where CRISPR/Cas9 technology shows the most promise as a therapeutic tool. This powerful instrument has shown

N-oxide-based and trioxolane-based fluorescent probes for monitoring labile iron and labile heme: an update.

Iron (Fe) and heme are essential for numerous biological processes, but their dysregulation contributes to cancer, neurodegeneration, and ferroptosis. This review summarizes the development and applications of N-oxide chemistry-based fluorescent probes and 1,2,4-trioxolane-based probes for selective monitoring of labile Fe(II) and heme in living systems over the past decade. We describe our discovery that Fe(II) selectively deoxygenates tertiary amine N-oxides, enabling conversion of non-fluorescent N-oxide compounds into highly fluorescent tertiary amines. This approach has yielded a comprehe

KIF4A in disease pathogenesis and therapeutics: from molecular mechanisms to clinical translation.

Kinesin family member 4 A (KIF4A) is a multifunctional motor protein essential for chromosome condensation, spindle dynamics, and cytokinesis. Beyond its classical mitotic functions, emerging evidence positions KIF4A as a central regulator of tumorigenesis, therapy resistance, metabolic reprogramming, and immune modulation across diverse cancer types. However, no comprehensive review has integrated its molecular mechanisms with its roles in both oncological and non-oncological diseases, nor clarified its context-dependent behavior, including paradoxical tumor-suppressive effects in cervical ca

Unraveling Cross-Cellular Communication in Cannabis sativa Sex Determination: A Network Ontology Transcript Annotation (Nota) Analysis.

Cannabis sativa L. (marijuana, hemp) is a dioecious angiosperm species currently evolving sex chromosomes. Genetic mechanisms, primarily controlled by an XY chromosome system, appear to dictate sex expression in C. sativa. However, sexual expression is also governed by the interplay of hormone regulatory gene networks, influenced by both genetic and environmental factors. Within the species, some populations exhibit dioecy, monoecy, or a gradient of both. Dioecious individuals produce exclusively male or female flowers, while monoecious plants bear both male and female flowers. However, throug
