Purification and culture of central nervous system pericytes.

Pericytes are found on the abluminal surface of endothelial tubes. The function of these cells is still being elucidated, but they have been … eural cell types. We have developed an immunopanning protocol to specifically purify pericytes from capillaries in the rodent optic nerve. This method relies on a combination of negative and positive selection criteria and allows prospective, acute isolation of pericytes. Use of this method will facilitate studies of pericyte cell biology and function and per …

Evaluation of enzyme-based tear glucose electrochemical sensors over a wide range of blood glucose concentrations.

Miniature enzyme-based amperometric and coulometric glucose sensors were fabricated and applied to measure tear glucose concentrations in anesthetized rabbits. Without perturbing the eyeball, 3 µL of tear fluid was sampled from the marginal conjunctiva under the lower eyelid of anesthetized rabbits at various time points via a microliter glass capillary tube, and the miniature sensors were then inserted into the volume of collected tear fluids within the capillaries for detection. Intravenous bolus doses of insulin were administrated to the rabbits to lower the elevated blood glucose concentra…

Retinitis Pigmentosa Associated with Vasoproliferative Tumors and Coats-like Fundus.

PURPOSE: To report two cases of retinitis pigmentosa (RP) associated with vasoproliferative tumors (VPTs) and Coats-like fundus. CASE REPORTS: Two patients with RP presented with recent loss of vision due to combined VPTs and Coats-like retinal vascular alterations. One patient had two VPTs with adjacent capillary nonperfusion, telangiectasia and aneurysmal vascular changes in one eye. The other patient had prominent VPT with Coats-like retinal vascular alterations in both eyes. These lesions received treatment resulting in improved vision in both patients. CONCLUSION: Although rare, VPTs an

A 7-year-old with pulmonary hypertension.

The authors discuss the case of a 7-year-old female who presented with exertional cyanosis and was found to have pulmonary arterial hypertension. Despite normal left-sided heart function, the patient developed pulmonary oedema in response to pulmonary vasodilator therapy, increasing suspicion for pathology in the pulmonary capillaries and veins. Lung biopsy confirmed a diagnosis of pulmonary capillary haemangiomatosis (PCH), a rare cause of pulmonary hypertension in both children and adults. The diagnosis requires lung biopsy and is often made postmortem. She was treated with interferon α-2a a…

Encephalomalacia with Enterococcus durans infection in the brain stem and cerebral hemisphere in chicks in Japan.

Fourteen chicks, 7-12 days old, that died or had neurological signs (depression, paralysis, torticollis, incoordination, and rolling) were investigated pathologically and microbiologically. Variably sized foci of malacia were present within the parenchyma of the brain stem (cerebral peduncle, optic lobe, and medulla oblongata) and the cerebral hemisphere. Capillary thrombosis with congestion and hemorrhage was frequently observed within the malacia lesions. Gram-positive cocci were recognized in the blood capillary of malacia lesions. Bacteriologically, Enterococcus durans was isolated from th

The human testicular artery and the pampiniform plexus--where is the connection?

The aim of the study was the microscopic evaluation of the human spermatic cord vessels, with special attention to the connection between the testicular artery and the veins of the pampiniform plexus. We used the corrosive cast method to visualise the angioarchitecture of the spermatic cord. Casts were evaluated using a scanning electron microscope. We observed that there is a narrow space (previously filled with the testicular artery wall), between the casts of the testicular artery and the veins of the pampiniform plexus. This area contains a capillary vessel net, which connects the testicul

Experimental study on brain oxygenation in relation to tissue water redistribution and brain oedema.

The aim of the present experimental research was to study brain oxygenation parameters in relation to tissue water movement and brain cortex oedema caused by focal brain ischemia. It has been demonstrated that local osmotic dehydration of the parietal brain cortex, mercury compounds (aquaporin inhibitors) and brain cortex oedema resulting from focal brain ischemia all influence extra-capillary oxygen transport lowering tissue respiration rates and oxygen transfer coefficients. The changes of brain oxygenation parameters in case of cortex ischemic oedema are reflected in the gas composition (ox

[Chronic edema, monoclonal dysglobulinemia and profuse telangiectasia: a distinct entity?].

INTRODUCTION: Monoclonal gammopathy and edema are features encountered during several diseases, especially systemic capillary leak syndrome. The diagnoses of POEMS syndrome, edematous systemic scleroderma and a fortuitous association may be also discussed. We report the cases of two patients which did not fulfill the criteria for such diagnoses. CASE REPORT: Although the 2 cases share some discrepancies, they have also similar and particular features: association of chronic edema, monoclonal gammopathy and profuse and acquired telangiectasias. DISCUSSION: The meaning of these cases remains t

Anionic sites of charge barrier in the guinea pig crista ampullaris.

In present studies we obtained anionic sites in the epithelial and capillary basement membranes in the dark cell area of the crista ampullaris in the guinea pig. The immersion method with cationic tracer polyethyleneimine (PEI) was applied. Electronmicroscopically, the arrangement of PEI particles was observed as two strata along the basement membrane. The number of particles could be counted and compared in each portion. The control test with protamine sulfate showed that the number of PEI particles decreased in both the epithelium and capillaries of the dark cell area. In the experiment usin…

Hemangioma of the ciliary body: a case report and review of the literature.

This paper documents a case of ciliary body hemangioma discovered in the eye of a 72-year-old white man during routine ocular examination. The correct differential diagnosis was not made clinically despite the use of A and B scan ultrasonography and also a fluorescein-angiographic study. This rare tumor was removed by a sector cyclectomy after apparent growth, onset of anterior uveitis and demonstration of fluorescein dye leakage. Histopathologic examination revealed that the tumor consisted of capillaries and large vascular loculi containing numerous erythrocytes. The histopathologic diagnosi…

Pathophysiology of hydrops fetalis.

Hydrops fetalis occurs when the rate of interstitial fluid production by capillary ultrafiltration exceeds the rate of interstitial fluid return to the circulation via lymphatic vessels. Developmental differences in the microcirculation and lymphatic system of the fetus, as compared with mature subjects, renders the fetus susceptible to interstitial fluid accumulation. These differences include greater capillary permeability, more compliant interstitial compartment, and greater influence of venous pressures on lymphatic return. The balance between interstitial fluid production and removal is m

The microvasculature of human infant oral mucosa using vascular corrosion casts and india ink injection. II. Palate and lip.

The microvasculature of human hard and soft palate and lip originating from four infant males and six females, aged 6 months to 2 years was studied by scanning electron microscopy of vascular corrosion casts and light microscopy of India ink injected specimens. The capillary loops of the hard palate mucosa and vermilion border of the lips were found to be tall, numerous and consisted of primary, secondary and tertiary loops. Those of the soft palatal and labial mucosa were short, few in number and demonstrated a simple hair-pin shape originating directly from the subpapillary vascular network.

Regulation of vascular permeability by macrophage-derived chemotactic factor produced in Japanese encephalitis.

The vascular effects of Japanese encephalitis virus (JEV)-stimulated splenic macrophage-derived neutrophil chemotactic factor (MDF) were evaluated in mice. Intraperitoneal injection of MDF in mice resulted in a rapid increase in capillary permeability in a dose-dependent manner as assessed by leakage of intravenously injected radiolabelled albumin ([125I]-albumin) or Evans blue dye. Intradermal inoculation of MDF in rabbits caused [51Cr]-labelled neutrophil emigration and accumulation into injected sites. Peak plasma leakage and neutrophil infiltration were observed at 1 h following MDF inocul

Pericyte physiology.

Endothelial cells are the primary physical barrier between blood and tissue in microvessels. The other capillary and post-capillary venule wall cell is the pericyte. The literature on the biology of endothelium is appreciable but less is known about pericytes. Pericytes are morphologically, biochemically, and physiologically heterogeneous. Some of pericyte functional characteristics observed in vivo and in vitro are that they: regulate endothelial proliferation and differentiation; contract in manners that either exacerbate or stem endothelial cell junctional inflammatory leakage; function as

Case report: nephrotic syndrome associated with a total hydatidiform mole.

The authors describe a 51-year-old Japanese woman who developed nephrotic syndrome in association with a total hydatidiform mole. The nephrotic syndrome remitted completely following hysterectomy. A renal biopsy performed before the operation showed diffuse mesangial cell proliferation of a moderate degree, and thickened capillary walls with focal and segmental subendothelial deposits, as well as circumferential mesangial interposition. Occasional foci of the mesangiolysis were also observed. Immunofluorescence microscopy revealed granular staining of IgM along the glomerular capillary walls i

Cochlear microcirculation in young and old gerbils.

Human postmortem studies as well as epidemiologic investigations have implicated the peripheral vascular system as one of the etiologic fact … vascularis). The decrease of blood flow in older animals was clearly not related to loss of strial capillaries with aging. The decreased flow to the lateral portion must arise from either decreased perfusion pressure or from increased vascular resistance. This decreased blood flow in the aged animals supports a vascular theory of presbycusis.

Influence of antihypertensive therapy on development and progression of diabetic glomerulopathy.

Capillary hypertension is suggested to be the underlying cause of microvascular disease affecting the kidney, the retina, and other organs and tissues in diabetic patients and animals. Hyperglycemia causes an expansion of extracellular volume, which induces a vasodilatory response. Hemodynamic adaptation to vasodilation leads to an increase in intracapillary hydraulic pressure, which subsequently causes vascular damage. In experimental animals, restoration of capillary pressure to normal levels by ingestion of a low-protein diet or administration of an angiotensin I-converting enzyme inhibitor

Local flow velocities in the cat carotid body tissue.

With the aid of a new three-dimensional mathematical model local flow velocities in the specific carotid body tissue of the cat measured by hydrogen clearances were calculated to have a mean value of 0.006 cm/s at a perfusion pressure of 50 mm Hg, 0.014 cm/s at a perfusion pressure of 120 mm Hg, and 0.018 cm/s at a perfusion pressure of 170 mm Hg. These results indicate that the carotid body specific tissue possesses a distinct flow heterogeneity with normal capillary flow velocities and a high shunt flow. During hypoxia, the smallest decrease in tissue PO2 was significantly correlated with th

Fetal microcirculation of abnormal human placenta. II. Scanning electron microscopy of placental vascular casts from fetus with severe erythroblastosis fetalis.

The latex injection-corrosion cast technique coupled with scanning electron microscopy was applied to the study of severe Rh sensitization of the placenta. The main observed ultrastructural changes in fetal vasculature in this type of placenta were very similar to those of the small for gestational age placenta; large-sized vessels exhibited less branching than those of the normal placenta, and the capillary network showed numerous vascular buds and "H"-shaped anastomoses. In addition, in Rh-sensitized placentas, the capillary network was remarkably sparse, and the cast also revealed smooth-su
