Increased efficacy of PARP inhibitors against cisplatin-sensitive and -resistant ovarian cancer cells mediated via ATR and ATM inhibition.

PARP inhibitors (PARPi) are approved for the treatment of platinum-based therapy-responsive ovarian cancer. However, this severely restricts their therapeutic potential, since there is only limited knowledge on the efficacy of PARPi in platinum drug-resistant ovarian cancer cells. Here, we studied three approved PARPi, niraparib, olaparib, and rucaparib in three ovarian cancer cell lines and their cisplatin-resistant sublines. Complex response profiles demonstrated that cisplatin resistance was not consistently associated with cross-resistance to PARPi. The combination of PARPi with inhibitors

Promoter hypermethylation of SOX11 promotes the progression of cervical cancer in vitro and in vivo.

The development of cervical cancer (CC) is a multi‑gene, multi‑step carcinogenic process that involves complex genetic and epigenetic mechanisms. SRY‑related HMG‑box gene 11 (SOX11) is a member of the SOX family of transcription factors with an emerging crucial role in the development of various tumor types. To elucidate the function of SOX11 in cervical carcinogenesis, the expression level of SOX11 during the development of human CC was analyzed by immunohistochemistry and western blot analysis. Additionally, the methylation status of the SOX11 was examined using bisulfite sequencing and meth

Reduction of hematopoietic cell-specific tyrosine phosphatase SHP-1 gene expression in natural killer cell lymphoma and various types of lymphomas/leukemias : combination analysis with cDNA expression array and tissue microarray.

To investigate the lymphomagenesis of NK/T lymphoma, we comprehensively and systematically analyzed the expression pattern of the human NK/T cell line (NK-YS) genome by cDNA expression array and tissue microarray. We detected significant changes in the gene expression of NK-YS cell line: an increase in 18 and a decrease in 20 genes compared to normal NK cells or peripheral blood mononuclear cells. Among these genes, we found a strong decrease in hematopoietic cell specific protein-tyrosine-phosphatase SH-PTP1 (SHP1) mRNA by cDNA expression array and reverse transcriptase-polymerase chain react

AP2M1 is a prognostic marker associated with cell cycle arrest and the tumor immune microenvironment in acute myeloid leukemia.

Acute myeloid leukemia (AML) is a heterogeneous clonal disease of hematopoietic progenitor cells and the most common malignant myeloid disease in adults. Although significant progress has been made in treatment, the outlook remains bleak, and new therapeutic targets need to be sought. AP-2 complex subunit mu (AP2M1) is a core component of the clathrin-mediated endocytic machinery, AP2M1 plays a critical role in cancer progression. However, its function in acute myeloid leukemia (AML) progression remains unclear. Our study reveals that AP2M1 is highly expressed in AML and is associated with poo

Integrative single-cell and bulk RNA-seq analysis identifies glycosyltransferases-related signature in triple negative breast cancer.

Triple-negative breast cancer (TNBC) represents a formidable subtype with a grim prognosis. This study aims to pinpoint the molecular targets of Glycosyltransferases (GTs) in TNBC, with the goal of improving prognostic accuracy and boosting the effectiveness of immune therapies. Using publicly available datasets, we combined differentially expressed and correlated genes based on AUCell scores from single-cell sequencing. These were then subjected to enrichment analysis and utilized for constructing a risk model. A total of 780 genes were identified as being closely associated with GTs. Using 1

Unleashing the potential role of tumor-associated NK cells as a novel immunotherapeutic target in triple-negative breast cancer.

Breast cancer is the most prevalent malignancy among women worldwide, with triple-negative breast cancer (TNBC) comprising 10–20% of cases. Although immunotherapy has recently emerged as a promising treatment, its efficacy remains limited in immunodeficient subtypes such as TNBC. Tumor-associated NK cells (TaNKs), an immunologically exhausted NK cell subset, have not yet been characterized in TNBC. Elucidating the TNBC tumor microenvironment through TaNKs may therefore provide novel diagnostic and therapeutic insights. Here, we employed single-cell transcriptomics to characterize TaNKs, which

Machine learning identifies the role of SMAD6 in the prognosis and drug susceptibility in bladder cancer.

BACKGROUND: Bladder cancer (BCa) is among the most prevalent malignant tumors affecting the urinary system. Due to its highly recurrent nature, standard treatments such as surgery often fail to significantly improve patient prognosis. Our research aims to predict prognosis and identify precise therapeutic targets for novel treatment interventions. METHODS: We collected and screened genes related to the TGF-β signaling pathway and performed unsupervised clustering analysis on TCGA-BLCA samples based on these genes. Our analysis revealed two novel subtypes of bladder cancer with completely diff

PI3K/AKT inhibitor BEZ-235 targets CCND2 and induces G1 arrest in breast implant-associated anaplastic large cell lymphoma.

Breast implant-associated anaplastic large cell lymphoma (BIA-ALCL) is a mature, CD30-positive T-cell lymphoma lacking expression of the anaplastic lymphoma kinase (ALK). In contrast to ALK-positive ALCL, BIA-ALCL cells express cyclin D2 (CCND2) which controls cyclin dependent kinases 4 and 6 (CDK4/6). DNA methylation and expression analyses performed with cell lines and primary cells suggest that the expression of CCND2 in BIA-ALCL cell lines conforms to the physiological status of differentiated T-cells, and that it is not the consequence of genomic alterations as observed in other hematopoi

The impact of C-MYC gene expression on gastric cancer cell.

The upregulation or mutation of C-MYC has been observed in gastric, colon, breast, and lung tumors and in Burkitt's lymphoma. However, little is known about the role C-MYC plays in gastric adenocarcinoma. In the present study, we intended to investigate the influence of C-MYC on the growth, proliferation, apoptosis, invasion, and cell cycle of the gastric cancer cell line SGC7901 and the gastric cell line HFE145. C-MYC cDNA was subcloned into a constitutive vector PCDNA3.1 followed by transfection in normal gastric cell line HFE145 by using liposome. Then stable transfectants were selected and

Autologous multiantigen-targeted T cell therapy for pancreatic cancer: a phase 1/2 trial.

T cell therapy has proven challenging for pancreatic ductal adenocarcinoma (PDAC), partly due to heterogeneous expression of tumor-associated antigens (TAAs). To address tumor heterogeneity and mitigate immune evasion, an ex vivo expanded, polyclonal, T helper 1 cell-polarized T cell product targeting five TAAs-PRAME, SSX2, MAGEA4, Survivin and NY-ESO-1-was developed. These antigens were chosen based on their tumor specificity, oncogenicity, immunogenicity and level of expression. In a phase 1/2 trial, this autologous nonengineered T cell product was administered (1 × 107 cells m-2 per infusio

Tumor diagnosis by intermediate filament typing: a novel tool for surgical pathology.

In most cell types intermediate or 10-mm filaments (IF) are a major cytoskeletal organization and, thus, directly or indirectly influence the structural appearance of the cytoplasm. In line with the cell type-specific expression patterns of different IF proteins in normal animal and human tissue, IF typing distinguishes the major tumor groups, as documented by results with several hundred human tumors classified by conventional histologic methods. Carcinomas are characterized by cytokeratins, sarcomas of muscle cells by desmin, nonmuscle sarcomas by vimentin, and gliomas by glial fibrillary ac

Asbestos mineral fibre exposure significantly affects volatile organic compound profiles of mesothelial cell lines in vitro.

Malignant mesothelioma (MM) is a rare cancer caused by exposure to asbestos, this condition continues to represent a significant diagnostic and therapeutic challenge. Due to the long disease latency, non-invasive diagnostic modalities such as breath analysis may enhance MM early detection by identifying the disease specific pattern of volatile organic compounds (VOCs) in exhaled breath. In this study, solid phase microextraction (SPME) and gas chromatography-mass spectrometry (GC-MS) was used to extract VOCs from the headspace of two mesothelioma cell lines: MSTO-211 H (biphasic mesothelioma)

Study on the Mechanism of Acanthopanax senticosus Extract Intervening in the Regulation of Parkinson's Disease by BV2 Microglial Cell-Derived Exosomes.

Parkinson's disease (PD) is the second most prevalent neurodegenerative disease. With the intensification of population aging, its incidence rate continues to rise. Microglia play a significant role in regulating neuronal functions. As carriers of intercellular communication, exosomes can mediate intercellular information transmission and signal regulation. This study aimed to explore the molecular mechanism by which Acanthopanax senticosus extract (AS) interferes with the PD process by regulating BV2 microglial cells exosome communication. A PD-related inflammation model was established by in

MEK Inhibition Synergizes with TYK2 Inhibitors in NF1-Associated Malignant Peripheral Nerve Sheath Tumors.

PURPOSE: Malignant peripheral nerve sheath tumors (MPNST) are aggressive sarcomas with limited treatment options and poor survival rates. About half of MPNST cases are associated with the neurofibromatosis type 1 (NF1) cancer predisposition syndrome. Overexpression of TYK2 occurs in the majority of MPNST, implicating TYK2 as a therapeutic target. EXPERIMENTAL DESIGN: The effects of pharmacologic TYK2 inhibition on MPNST cell proliferation and survival were examined using IncuCyte live cell assays in vitro, and downstream actions were analyzed using RNA-sequencing (RNA-seq), qPCR arrays, and v
