Strand-seq is a single-cell sequencing method that preserves directionality of DNA template strands, enabling chromosome-length haplotyping, structural variant discovery, and sister chromatid exchange mapping. The conventional, microliter-scale protocol is reliable but consumable-intensive, requiring five bead-based DNA cleanups and individual per-cell processing across a 96-well plate. A nanoliter-scale, one-pot (OP) adaptation nearly eliminates bead cleanups and plastic consumables in sub-microliter volumes on bespoke nanoarrays, but requires an acoustic dispenser, custom array hardware, and

Structural variants (SVs) may increase SOX3 expression in the gonads and have been observed in individuals with ovotesticular differences in sex development (OT-DSD) and XX testicular differences in sex development (T-DSD). Most of the SVs found in OT-DSD individuals are whole-gene duplications, and to date, only one SV affecting SOX3 expression by a positional effect has been described. We report an individual raised as a female with SRY-negative OT-DSD. Karyotype analysis showed a pericentric inversion in one of the X chromosomes - 46,X, inv(X)(p22;q27). The breakpoints and fusion were mappe

Escalating pressures of global climate change necessitate developing agricultural systems and crop varieties with enhanced resilience. Polyploidy, the state of possessing multiple complete sets of chromosomes arising from whole genome duplication (WGD), is a major evolutionary force in plants, often conferring novel genetic and regulatory capacities that facilitate adaptation. Allotetraploid cotton (Gossypium spp.), which formed through the merger of distinct A and D subgenomes approximately 1 to 1.6 Mya, is an exemplary model for elucidating polyploid genome evolution and molecular mechanisms

Holoprosencephaly results from incomplete separation of the cerebral hemispheres. Cyclopia is a facial manifestation of Holoprosencephaly, characterized by a midline single orbit and proboscis. Prenatal diagnosis is done by ultrasonography and can be supplemented by MRI. This case uniquely presents ultrasound and gross morphology correlation of cyclopia in a resource limited setting, and a correlation between facial features, intra-cranial anatomy, and lifespan. We report a 39-year-old gravida 3 para 2 woman with a history of alcohol consumption who presented for her first antenatal checkup at

Replication stress, defined as impaired DNA replication leading to genomic instability, is a central feature of tumour development and progression. In cancer cells, it arises from oncogene activation, transcription-replication conflicts, altered nucleotide metabolism and DNA secondary structures that hinder DNA synthesis. These pressures compromise accurate genome duplication and promote chromosomal instability and mutagenesis. Recent advances have clarified the molecular mechanisms that stabilize, remodel and restart stressed replication forks, and highlighted the essential role of the ataxia

Nodule inception (NIN)-like proteins (NLPs) are plant-specific transcription factors regulating nutrient absorption, growth, and stress tolerance; however, their roles in stress responses remain largely uncharacterized. Cultivated strawberry (Fragaria × ananassa 'Camarosa') serves as an ideal model for dissecting the evolution and function of NLP genes. In this study, 37 FaNLP genes were identified genome-wide. Phylogenetic analysis classified them into three subfamilies, which are evenly distributed across seven chromosomes. Divergent exon-intron structures and conserved motif compositions su

Sea snake venom systems provide exceptional models for studying marine adaptation and are rich sources of bioactive molecules for drug discovery. To comprehensively characterize the venom diversity of the medicinal sea snake Hydrophis cyanocinctus, this study employed an integrated multi-omics strategy. We performed deep proteomic profiling of its venom and venom gland using both data-independent acquisition (DIA) and tandem mass tag (TMT) techniques, underpinned by a previously established chromosome-level genome and PacBio full-length transcriptomics. Our analysis revealed a distinctive prot

Activation of the alternative lengthening of telomeres (ALT) pathway accounts for cellular immortalization in 75% of pediatric osteosarcoma. ALT does not rely on a single enzyme but instead, catalyzes telomere elongation via homologous recombination. There has been steady progress in defining the mechanisms that regulate the ALT pathway. However, the spectrum of genetic mutations that underlie activation of ALT remains unclear. Osteosarcomas, like many cancers, frequently harbor inactivating mutations in the tumor suppressor gene TP53. However, instead of single nucleotide variants that lead t

The CHRNA7 gene, located on chromosome 15q13.3, encodes the α7 nicotinic acetylcholine receptor (α7 nAChR) subunit and lies within a genomic region characterized by high recombination rates and associations with multiple neuropsychiatric disorders. Within this region, the human specific gene CHRFAM7A arose through partial duplication, rearrangement, and fusion between CHRNA7 and FAM7A. The direct CHRFAM7A allele has been shown to negatively regulate α7 nAChR function. The inverted allele (CHRFAM7AΔ2bp), harboring a two-base pair deletion in exon 6, is linked to schizophrenia, bipolar disorder,

Sarcopenia is a common age-associated condition characterized by the progressive loss of skeletal muscle mass, strength, and physical functionality. While large-scale genome-wide association studies (GWAS) have previously addressed isolated traits of sarcopenia, the multifactorial genetic architecture underlying this condition remains largely undefined. To characterize the common genetic basis of sarcopenia-related traits, genomic structural equation modeling (Genomic-SEM) was implemented. Multiple post-GWAS analytic approaches were integrated to pinpoint susceptibility loci. These analyses en

Congenital adrenal hyperplasia (CAH) is an autosomal recessive disorder, commonly caused by variants in CYP21A2 (chr6p21.33), which encodes the 21-hydroxylase enzyme. Genetic diagnosis is challenging due to the high homology between CYP21A2 and its nearby pseudogene CYP21A1P. The current gold standard, PCR-based Sanger sequencing combined with multiplex ligation-dependent probe amplification (MLPA), is labor-intensive, costly, and amenable to PCR bias. Furthermore, it is not reliable in detecting complex structural variants, and it provides no information on whether variants are located on the

46,XY disorders of sexual development (46,XY DSD) are conditions characterized by deviations from typical male sex development, which encompasses a broad spectrum of clinical features such as hypospadias, decreased sperm production, dysgenetic testes, bifid scrotum, and the presence of a mature uterus and fallopian tubes. The WWOX gene, which has been implicated in various cancers, is also thought to play a role in sex development, although its involvement in 46,XY DSD remains poorly understood. This study aimed to investigate the WWOX gene in a patient presenting with hypospadias. A 7-month-o

Chimeric antigen receptor (CAR) T-cell therapy has transformed the treatment of haematological malignancies, but conventional viral transduction results in semi-random genomic integration, contributing to heterogeneous CAR expression and potential insertional effects. Site-specific genome engineering offers an alternative by directing CAR insertion to defined genomic loci, allowing greater control over transgene expression and cellular function. This narrative review evaluates advances in site-specific CAR T-cell engineering, focusing on integration mechanisms, donor platforms, genomic loci, s

Lissencephaly is a migrational disorder that results in abnormal gyration and cortical lamination. Type 1 lissencephaly is characterized by absent or reduced number of gyri giving the brain a smooth appearance, while type 2 lissencephaly (cobblestone lissencephaly) is described as over-migration of neurons or neuronal precursors beyond the glia-pial limitans giving rise to a cobblestone appearance of the cerebral hemispheres. Both types of lissencephaly are typically thought of as congenital anomalies secondary to genetic defects while cases of lissencephaly due to acquired injury is rare. The

Klebsiella aerogenes is an opportunistic pathogen increasingly associated with healthcare-associated infections and severe respiratory complications. However, it's in vivo adaptation during host colonization remains poorly understood. We characterized three clonal K. aerogenes ST93 isolates sequentially recovered on days 4, 14, and 19 of hospitalization from a patient with fatal viral pneumonia and concurrent abdominal sepsis. While initially identified generically as Klebsiella spp. by automated clinical systems, Whole-Genome Sequencing and Average Nucleotide Identity confirmed their identity

Ex vivo genome editing of hematopoietic stem and progenitor cells (HSPCs) holds significant therapeutic potential but remains constrained by genotoxic risks associated with nuclease-induced DNA double-strand breaks, DNA donor template delivery and sensing, and proliferation-induced stress during ex vivo manipulation. These processes can lead to chromosomal instability, large on-target deletions, donor mis-integration, off-target events, and impaired long-term stem cell function, raising safety concerns for clinical translation. Here, we evaluate the impact of transient p38 MAPK inhibition on g

AT-hook motif nuclear-localized (AHL) genes play critical roles in chromatin remodeling and gene transcription regulation, profoundly influencing plant growth, development, and stress responses. While AHL genes have been extensively characterized in multiple plant species, their biological functions in pepper (Capsicum annuum L.) remain largely uncharacterized. In this study, we identified 45 CaAHL genes in the pepper genome through bioinformatics approaches. Comprehensive analyses were conducted to examine their chromosomal distribution, phylogenetic relationships, and the structural and func

This study investigated milk protein (MP)-based edible films incorporated with clove and camphor essential oils (EOs) as antimicrobial agents against dental pathogens. Gas chromatography-mass spectrophotometry revealed clove oil contained 72.2 % eugenol while camphor oil contained 69.51 % citronellal. The optimum formulation as in Run 12 (2.5 % camphor oil + 3.0 % clove oil, 24 h contact time) achieved 100 % growth reduction against Streptococcus mutans, Aggregatibacter actinomycetemcomitans, and Porphyromonas gingivalis using Box-Behnken design. Clove oil films exhibited superior inhibition z

BACKGROUND: Iron (Fe) is indispensable to plants, playing a significant role in life activities such as respiration, chlorophyll biosynthesis, and photosynthetic electron transport. The decrease in pH caused by iron deficiency is related to the activation of H+-ATPase in the root plasma membrane, although the reaction mechanism of this enzyme is not well understood in apples. The H+-ATPase (HA) gene family has been extensively studied in Arabidopsis but is rarely reported in other species.
RESULTS: In this study, 14 HA genes were identified from the apple genome database through whole genome

The Paraoxonase (PON) gene family consists of three paralogues (PON1, PON2 and PON3) that are tandemly located on chromosome 7. In this review paper, the structure and function of the encoded proteins is summarized. In addition, an overview is given on the generated animal models. Finally, their involvement in the pathogenesis of different diseases is discussed, starting from an extended screening of the literature using PUBMED and Web of Science. PON1 and PON3 are mainly expressed in the liver and released into the bloodstream, bound to high-density lipoprotein. PON2 is expressed in various t
