Co-ordinate increase in the expression of type I and type III collagen genes in progressive systemic sclerosis fibroblasts.

Progressive systemic sclerosis (PSS), is a connective tissue disease characterized by excessive accumulation of collagen in the skin and various internal organs which is due, at least in part, to increased collagen production by PSS fibroblasts. In order to examine the molecular mechanisms responsible for this abnormality, we compared the kinetics of collagen biosynthesis, the intracellular degradation of collagen and the expression of Types I and III procollagen genes between normal and PSS dermal fibroblasts in culture. Two age- and sex-matched normal and PSS dermal fibroblast cell lines wer…

Heparan sulfate proteoglycans from mouse mammary epithelial cells. Cell surface proteoglycan as a receptor for interstitial collagens.

A heparan sulfate-rich proteoglycan is on the surface of NMuMG mouse mammary epithelial cells apparently intercalated into their plasma membranes. Mild treatment of the cells with trypsin releases the GAG-bearing region (ectodomain) of this molecule as a discrete proteoglycan which is readily purified. At physiological pH and ionic strength, the ectodomain binds collagen types I, III, and V but not types II, IV, or denatured type I. The proteoglycan binds to a single class of high affinity saturable sites on type I collagen fibrils, sites which are selective for heparin-like glycosaminoglycans…

Basement membrane laminin and type IV collagen in various benign and malignant adnexal tumors of the skin: an immunohistochemical study.

Thirty benign and seven malignant adnexal tumors of the skin and one lymph node metastasis were stained for laminin and type IV collagen with rabbit antibodies against the human basement membrane (BM) proteins using the immunoperoxidase technique. Fifteen of the benign sweat gland, sebaceous gland, and hair follicle tumors showed a continuous and distinct BM around the tumor aggregates. The cylindromas and eccrine spiradenomas seemed to produce excessive amounts of BM material, part of which was seen as amorphic patches within the tumor cell clusters, whereas the trichofolliculomas, trichoepit…

Cancer in collagen disease. A statistical analysis by reviewing the Annual of Pathological Autopsy Cases (Nippon Boken Shuho) in Japan.

Cases of systemic lupus erythematosus, systemic sclerosis, periarteritis nodosa, and dermato- and polymyositis autopsied in Japan during a recent, 7-year period (1972-1978) were studied to assess the incidence of cancer as a complication. In comparison with the other three groups, cases of dermato- and polymyositis (DP) showed quite a high incidence of malignancy: 53 of 99 for males (53.5%) and 36 of 139 for females (25.9%). In patients older than 50 years of age cancer was recognized in the autopsies of 65.7% of males and 38.2% of females with DP. The stomach was both the primary site and had

Methods in laboratory investigation. Monoclonal antibodies to type IV collagen: probes for the study of structure and function of basement membranes.

Type IV collagen is one of the main constituents of basement membranes, yet it is unknown whether the structural framework at different sites is assembled from one unique type of molecule or whether different type IV collagen molecules exist. To study the composition, chemical identity, and organization of this protein in different organs we have prepared monoclonal antibodies to a type IV collagen preparation from human placenta. Swiss Webster mice were hyperimmunized, and splenic cells were fused with the three different myeloma cell lines SP2/0, NS1, and U1. Type IV collagen-specific hybrid…

Platelet interaction with collagen fibrils in flowing blood. II. Impaired adhesion-aggregation in bleeding disorders. A comparison with subendothelium.

Anticoagulated whole blood from patients and control subjects was circulated through an annular perfusion chamber in which the fibrillar collagen of alpha chymotrypsin-digested subendothelium and intact subendothelium were exposed. The blood flow conditions corresponded to those in arteries (830 sec-1 wall shear rate). Platelet surface interaction was measured morphometrically. Decreased adhesion to fibrillar collagen associated with normal spreading and normal adhesion-induced formation of platelet thrombi was found with blood of patients with von Willebrand's disease and the Bernard Soulier …

Gel electrophoretic studies on proteoglycans and collagen of abnormal human growth cartilage: proteoglycan abnormalities in pseudoachondroplasia and in Kniest's disease.

The microchemical study of growth cartilage biopsies may improve the classification and the genetic advice of some types of growth disturban … eral cases were difficult to assess. In all cases except mucolipidosis III and Kniest's disease the collagen was extracted by using limited cleavage and solubilization with pepsin, purified and analyzed by polyacrylamide gel electrophoresis. A type of collagen with a single alpha band was found.

A Case of Autosomal Dominant Alport Syndrome with Normal Type IV Collagen α5 Staining Diagnosed by a Genetic Analysis.

A 30-year-old woman with persistent hematuria, mild proteinuria, and a family history of end-stage kidney disease was initially diagnosed with thin basement membrane nephropathy based on renal biopsy findings of a nearly normal glomerular basement membrane with only focal thinning, no lamellation, and normal type IV collagen α5 staining. However, genetic testing identified a heterozygous COL4A4 missense variant (p.Gly527Cys), thus confirming a diagnosis of autosomal dominant Alport syndrome. After starting angiotensin receptor blocker therapy, the patient's kidney function remained stable. Thi…

Effect of Membrane Perforations With and Without Collagen Membrane Coverage in Vertical Ridge Augmentation: An Experimental Pilot In Vivo Study.

OBJECTIVES: Given the proposed bioactive role of collagen membranes, we aimed to evaluate whether the perforation size of titanium-reinforced dPTFE membranes, with or without an additional collagen membrane, influences outcomes of vertical alveolar ridge augmentation (VARA). MATERIALS AND METHODS: In 12 beagle dogs (24 sites), standardized bilateral chronic mandibular ridge defects (8 × 30 mm) were created and healed for 12 weeks. Sites were randomized (1:1:1) to VARA using porcine xenograft covered with either: (i) non-perforated titanium-reinforced dPTFE (NON-PERF), (ii) perforated titanium…

Tissue-specific differences in ECM organization and collagen extractability under estrogen deficiency: A comparative study of skin and tendon.

Estrogen deficiency is known to affect connective tissues, yet its impact on collagen organization beyond bone remains insufficiently understood. In this study, we investigated tissue-specific alterations in type I collagen in rat skin and tendon following ovariectomy, with and without alendronate treatment. Native tissues were analyzed in parallel with corresponding crude collagen isolates to distinguish between collagen-related properties and extracellular matrix (ECM)-level contributions. Estrogen deficiency was associated with tissue-dependent changes, with skin exhibiting increased collag…

Supraspinatus Tendon Thickness Is Associated With Collagen Disorganization in Symptomatic and Asymptomatic Shoulders of Individuals With Unilateral Rotator Cuff Tendinopathy.

Rotator cuff (RC) tendinopathy is a common source of shoulder pain and is frequently linked to structural changes in the supraspinatus tendon of greater thickness and disrupted collagen organization. This study aimed to characterize differences in tendon thickness and collagen organization between individuals with unilateral symptomatic RC tendinopathy, their contralateral asymptomatic shoulder, and asymptomatic controls, and to examine the relationship between thickness (macro-morphology) and collagen organization (micro-morphology). This is a cross-sectional observational study of 64 partici…

Labeling of Collagen in Human Musculoskeletal Connective Tissues with Stable Isotope Tracers and Preparation of Amino Acids for Mass Spectrometry.

Collagen-rich musculoskeletal connective tissues are in constant turnover, reflecting a balance between synthesis and breakdown, which is crucial for tissue remodeling and regeneration. Stable isotopes, particularly 15N-proline and 2H2O, offer a precise method for tracing collagen turnover. This chapter details the use of these isotopes to study collagen dynamics in response to various interventions. The protocol includes the administration of stable isotope tracers, tissue sampling, collagen isolation, and derivatization of amino acids for analysis by mass spectrometry.

Potential Supportive Effects of AP Collagen Peptides on Skin Hydration in a Mouse Model of Oxazolone-Induced Atopic Dermatitis.

Atopic dermatitis (AD) is a chronic and relapsing inflammatory skin disorder characterized by impaired skin barrier function and Th2-mediated immune responses. Collagen tripeptide (CTP), an amino acid-rich dietary supplement with low antigenicity, has been reported to improve skin hydration and barrier function. However, its therapeutic effects in AD, particularly under conditions of barrier dysfunction, have not yet been fully established. In this study, we evaluated the potential effects of AP collagen peptide (APCP), which contains CTP, on skin hydration and inflammatory responses, in an ox…

Electrochemical Characterization of a Molecularly Imprinted Polymer Sensor for the Selective Recognition of Type II Collagen in Joint Degeneration Monitoring.

Type II collagen is a primary fibrillar component of articular cartilage, and its early degradation is a key biomarker of joint-degenerative disorders such as osteoarthritis, rheumatoid arthritis, gout, etc. Reliable detection at low concentrations remains challenging due to limited assay accessibility, complex analytical procedures, and nonspecific responses in multicomponent biological matrices. This research reports the development of a Molecularly Imprinted Polymer (MIP)-based electrochemical sensor engineered for the selective recognition of type II collagen. A series of monomer formulati…

Efficacy and safety of hydrolyzed collagen supplementation on skin health outcomes: a systematic literature review of randomized controlled trials.

Oral hydrolyzed collagen (HC) supplementation is widely recommended for improving age-related skin changes. However, clinical evidence remains inconsistent. This systematic literature review (SLR) synthesizes the findings from randomized controlled trials (RCTs) evaluating the efficacy, safety, and quality of life (QoL) outcomes of HC supplementation in adults. We systematically searched Embase, MEDLINE, and MEDLINE In-Process from inception through April 1, 2023, to identify RCTs assessing oral HC versus placebo or active comparators. Outcomes of interest included skin hydration, elasticity, …
