Rise-and-fall dynamics reveal a molecular and cellular vulnerability axis in prion-like α-synuclein propagation.

The spread of misfolded proteins through neuronal circuits is a defining feature of neurodegenerative disease, yet the dynamics underlying this process remain poorly understood. Most studies rely on sparsely sampled datasets that capture spatial patterns of pathology but not their temporal evolution. Here, we analyze longitudinal histopathology measurements of α-synuclein pathology across hundreds of brain regions in a mouse model of Parkinson's disease. Network-based dynamical modeling shows that regional pathology does not simply accumulate but instead follows rise-and-fall trajectories acro

[The blood ecosystem: from concept to practice].

The introduction of the blood ecosystem concept marks a paradigm shift in hematology from traditional cell- or organ-centered analyses toward a framework focused on cross-organ interactions characterized by systemic integration, connectivity, openness, and dynamic regulation. This framework emphasizes blood as a dynamically coupled network that integrates whole-body homeostasis, rather than a collection of isolated components. Enabled by advances in single-cell multi-omics, spatial profiling, and artificial intelligence-based analytical approaches, the blood ecosystem can now be interrogated w

Genomic Epidemiology of Salmonella enterica Serovar Hadar Strain Linked to Poultry-Associated Salmonellosis Outbreaks, United States.

Nontyphoidal Salmonella enterica serovar Hadar causes poultry-associated salmonellosis outbreaks in the United States. One persisting strain of Salmonella Hadar caused 5 multistate outbreaks resulting in ≈2,000 human cases during 2020-2023. The Centers for Disease Control and Prevention designated the strain as reoccurring, emerging, or persisting (REP), related within 26 core-genome allele differences. That REP strain has caused human illnesses by consumption of commercial poultry food products or contact with backyard poultry. To investigate the REP strain's evolution and identify possible m

Peptides in the Diagnosis and Treatment of Pancreatic Cancer and Other Pancreatic Diseases from Basic Research to Clinical Translation.

Peptides have emerged as pivotal agents in biomedicine due to their exceptional physicochemical properties, including high binding affinity, minimal immunogenicity, and precise target specificity. This review critically examines recent advancements in peptide-based strategies for pancreatic disorders, encompassing inflammatory conditions (e.g. pancreatitis), metabolic dysfunctions (e.g. diabetes), and notably, pancreatic cancer. We delineate the evolution of peptide therapeutics, emphasizing rational drug design approaches such as backbone cyclization and N-methylation to enhance metabolic sta

Early-Onset Oral Tongue Squamous Cell Carcinoma in the Absence of Traditional Risk Factors: A Case Report with Whole-Exome Sequencing Analysis.

Oral squamous cell carcinoma (OSCC) typically develops in individuals with established risk factors such as tobacco and alcohol use, yet an increasing number of cases occur in young non-smoking, non-drinking (NSND) patients. We report a case of oral tongue OSCC in a 33-year-old woman who is a never-smoker and never-drinker without identifiable environmental or local risk factors. The patient underwent surgical treatment followed by adjuvant radiotherapy and remains disease-free 15 months after therapy. Whole-exome sequencing (WES) revealed a pathogenic truncating TP53 mutation together with ad

Acanthamoeba ATP synthase structure reveals the TCA cycle is tethered to OXPHOS.

Oxidative phosphorylation (OXPHOS) is a key metabolic process that couples redox energy to ATP production. While some core OXPHOS complex subunits are found across all domains of life, many have diverged or expanded across evolution-as seen in the protozoan pathogen Acanthamoeba castellanii. By integrating cryo-electron microscopy of unenriched mitochondrial lysate with mass spectrometry proteomics, we resolved the structures of endogenous mitochondrial ATP synthase (complex V), Hsp60, and respiratory complex III from Acanthamoeba. We capture Acanthamoeba ATP synthase in an IF1-inhibited state

Beyond the Solid Solution: Ordered Enantiomerically Unbalanced Packing in Surface-Confined Tetrahelicene Monolayers.

The two-dimensional crystallization and on-surface chemistry of chiral polycyclic aromatic hydrocarbons provide access to surface-confined symmetry breaking and novel covalently bonded nanostructures. Here, we investigate the self-assembly of racemic 2,3-dicarbonitrile tetrahelicene on Ag(111). Using low-temperature scanning tunneling microscopy in combination with density functional theory calculations, we resolve the evolution of molecular organization as a function of coverage. After room-temperature deposition at submonolayer coverage, the tetrahelicene moiety spontaneously resolves into e

Convergence in Coevolved Systems: A Two-Axis Filter for Biomimetic Transferability.

Biomimetics often treats convergent evolution as the strongest sign that a biological solution is general. That inference is safest when the constraint does not counter-adapt. Hosts do. In coevolved host-interface systems, recurrence alone cannot tell us whether a solution is translatable. Biomimetic transferability depends first on two axes: conservation of the host target and versatility of the attacking lineage. A conserved target behaves, for translational purposes, like a biochemical constraint, a broad-host parasite has already tested its mechanism across biological variation. The window

Topological Integration of Slide-Ring and Entangled Networks.

Topological regulation of polymer networks has emerged as an effective strategy for enhancing the mechanical performance of elastomeric materials. Among various topological architectures, slide-ring networks and entangled networks have attracted considerable attention, owing to their unique dynamic mobility and spatial interlocked effects, respectively. However, these two topological motifs have so far been explored separately, and their synergistic integration within a single elastomer network remains a significant challenge. Herein, we construct a synergistic topological elastomeric network

Programming Iron Nuclearity and Coordination for Hydride-Mediated Nitroarene Reduction.

Multielectron catalytic reactions intrinsically rely on cooperative metal centers, yet constructing few-atom cluster catalysts (FACCs) with controlled nuclearity, metal-metal connectivity, and electronic structure remains a fundamental challenge. Here, we show that atomically defined iron-oxo clusters can serve as structure-encoded precursors to generate supported Fe ensembles on N-doped carbon (NC), enabling systematic construction of active sites spanning isolated atoms to size-defined multinuclear clusters. During ligand removal and interfacial reconstruction, the molecular structural infor

Tracking the volatile fingerprint of crayfish by-product shrimp sauce during a rapid fermentation: A synergistic strategy using flavoromics and molecular docking.

Crayfish heads were rapidly converted into shrimp sauce via enzymatic pretreatment and fermentation within 30 days. The evolution of volatile organic compounds (VOCs) during shrimp sauce fermentation was analyzed. Electronic nose analysis showed the flavors changed over time, with distinct VOCs between the fermented product and the enzymolysis solution. GC-MS identified 174 VOCs, including aldehydes, aromatics, and alcohols like 1-octen-3-ol, furfural, etc. Gas chromatography-ion mobility spectrometry (GC-IMS) detected 63 VOCs, indicating obvious differences during fermentation. 1-Octen-3-ol,

The evolutionary impact of Robertsonian fusions: linking genomic architecture, recombination and reproduction.

Determining the genomic basis of speciation remains a central challenge in evolutionary biology, including the role of large-scale structural genome reorganizations, such as inversions and chromosomal fusions. While extensive theoretical and empirical work has shown that inversions can facilitate adaptation and speciation by suppressing recombination and maintaining co-adapted gene complexes, the evolutionary role of chromosomal fusions remains comparatively underexplored. This gap contrasts with the widespread occurrence of chromosomal fusions across eukaryotes. Here, we synthesize current kn

Dynamic strength of iron under pressure-temperature conditions of Earth's inner core.

Iron (Fe) is a primary constituent of terrestrial planetary cores, yet its rheological properties under extreme conditions remain uncertain. Here we present direct measurements of Fe strength at 310-430 GPa pressures and 3700-5800 K temperatures, obtained using Rayleigh-Taylor (RT) instability experiments at the National Ignition Facility. Single-crystal α-Fe samples with [001] and [111] orientations are shock-ramp compressed past the α-ε transition along paths approaching Earth's inner core conditions. We find that ε-Fe derived from [001] α-Fe is consistently stronger (11-20 GPa) than that fr

Heat stress induces the formation of unreduced male gametes by targeting translation in Arabidopsis meiocytes.

Heat stress promotes the formation of unreduced (2n) male gametes through meiotic restitution, a driving force of evolutionary polyploidization. Here we report that the molecular mechanism underlying heat tolerance of the meiotic division program in Arabidopsis thaliana relies on sustained protein translation of cell cycle genes. By leveraging natural variation in the Arabidopsis population, we identified heat-sensitive and heat-tolerant alleles of TARDY ASYNCHRONOUS MEIOSIS/CYCLIN A1;2 (TAM). We show that TAM associates with specialized biomolecular condensates in meiotic cells under high tem

Unraveling the modulating mechanism of Maillard reaction gradients on structural properties and emulsifying stabilization of Prunus mume melanoidins.

Fruit-derived melanoidins remain poorly characterized despite the ubiquity of dietary Maillard reaction products. This study investigates how processing intensity affects the structural evolution and functional properties of Prunus mume (Wumei) melanoidins (MWMs). Three fractions were prepared via hot air drying (MDW), conventional smoking (MSW), and deep smoking (MDSW), and benchmarked against coffee melanoidins (MC). Their composition, structure, thermal behavior, wettability, bioactivities, polyphenol release, and emulsion stability were evaluated. Increasing thermal intensity raised molecu

Enhancing the Robustness of Nitrilase from Betaproteobacteria bacterium by Multistrategy Synergistic Evolution.

Nicotinic acid is vital in pharmaceuticals, feed, food, and cosmetics. Nitrilase can catalyze the conversion of 3-cyanopyridine to nicotinic acid, but its industrial application is limited by poor thermostability and substrate tolerance. In this study, nitrilase from Betaproteobacteria bacterium was engineered via multistrategy synergistic evolution, including C-terminal loop truncation, consensus mutation, loop engineering and protein surface engineering. Through iterative saturation mutagenesis, we obtained mutant 4 M (L194F/A201Q/S208L/M180Q-Δ47). Its activity reached 12.78 U/mL. After 1 h

Acute deep vein thrombosis: who still needs hospital admission?

Hospital admission for acute deep vein thrombosis (DVT) was historically considered mandatory, reflecting the therapeutic limitations and safety concerns of previous decades. Over time, advances in anticoagulant therapy, diagnostic strategies, and healthcare organization have profoundly changed this paradigm. Low-molecular-weight heparins and, more recently, direct oral anticoagulants (DOACs) have enabled effective and safe outpatient treatment for a large proportion of patients. Nonetheless, hospitalization remains common in clinical practice, often driven by habit, uncertainty, or organizati

Comprehensive Modeling of Heat and Diffusional Drug Transfer in Photothermal-Responsive Microneedles Applied to Skin.

Photothermal-responsive microneedles (PRMNs) are promising tools for controlled, on-demand transdermal delivery of low-permeability drugs like Methotrexate. While effective, their underlying heat and drug transfer mechanisms, particularly the role of Soret effect-induced thermodiffusion, remain unclear. This study develops a theoretical framework that integrates heat transfer induced by light irradiation with molecular transport governed by both Fick's diffusion and the Soret effect while accounting for skin heterogeneity and temperature-dependent diffusion. Using a geometric PRMN-skin model,
