Magnesium Disorders: Core Curriculum 2024.

Magnesium is ubiquitous in nature. It sits at the origin of the food chain, occupying the center of chlorophyl in plants. In humans, magnesium is critical to diverse molecular and catalytic processes, including energy transfer and maintenance of the genome. Despite its abundance, hypomagnesemia is common and often goes undiagnosed. This is in spite of epidemiologic data linking low magnesium with chronic diseases including diabetes mellitus. Clinically significant hypermagnesemia is encountered less frequently, but the presentation may be dramatic. Advances in molecular biology and the elucida

Fatty acid metabolization and insulin regulation prevent liver injury from lipid accumulation in Himalayan marmots.

Fat storage and weight gain are dominant traits for hibernating mammals. However, excessive fat accumulation may cause liver damage. Here, we explore the lipid accumulation and metabolic processes of the Himalayan marmot (Marmota himalayana), a hibernating rodent species. We find that the unsaturated fatty acid (UFA) content in food was consistent with a large increase in the body mass of Himalayan marmots. Metagenomic analysis shows that Firmicutes Bacterium CAG:110 plays a synergistic role by synthesizing UFAs, which is demonstrated by fecal transplantation experiments, indicating that the g

A novel electrochemical sensor based on dual-functional MMIP-CuMOFs for both target recognition and signal reporting and its application for sensing bisphenol A in milk.

In this work, novel magnetic molecularly imprinted CuMOFs (MMIP-CuMOFs) were synthesized and applied to construct an electrochemical bisphenol A sensor. The constructed sensor used an electrode modified with reduced graphene oxide (RGO/GCE) as the sensing platform to improve its stability and sensitivity. The Fe3O4 nanoparticles in magnetic MOFs simplified the preparation process. Moreover, the combination of CuMOFs and molecular imprinting methodology was beneficial for improving the detection specificity, and the electroactive copper hexacyanoferrate generated by the reaction of Cu2+ in CuMO

An Overview on Different L-Thyroxine (l-T4) Formulations and Factors Potentially Influencing the Treatment of Congenital Hypothyroidism During the First 3 Years of Life.

Congenital hypothyroidism (CH) is a relatively frequent congenital endocrine disorder, caused by defective production of thyroid hormones (THs) at birth. Because THs are essential for the development of normal neuronal networks, CH is also a common preventable cause of irreversible intellectual disability (ID) in children. Prolonged hypothyroidism, particularly during the THs-dependent processes of brain development in the first years of life, due to delays in diagnosis, inadequate timing and dosing of levothyroxine (l-thyroxine or l-T4), the non-compliance of families, incorrect follow-up and

Metabolic Profiling of the Hypothalamus of Mice during Short-Term Food Deprivation.

Nutrient availability and utilization in hypothalamic cells are directly associated with the regulation of whole-body energy homeostasis. Thus, establishing metabolic profiling in the hypothalamus in response to metabolic shift is valuable to better understand the underlying mechanism of appetite regulation. In the present study, we evaluate the alteration of lipophilic and hydrophilic metabolites in both the hypothalamus and serum of fasted mice. Fasted mice displayed an elevated ketone body and decreased lactate levels in the hypothalamus. In support of the metabolite data, we further confir

Addressing social and racial justice in public policy for healthy living.

Long-standing health disparities stemming from the historical, inequitable distribution of wealth, power, and privilege in the United States exist across almost every health indicator and outcome. There is a need for equitable policy, systems, and environment changes that are rooted in an understanding of the historical arc of structural racism across obesity prevention and treatment, ending tobacco and nicotine addiction and increasing access to healthy, affordable foods and physical activity opportunities and infrastructure. This paper explores the influence of structural inequities on the p

Glucagon and other proglucagon-derived peptides in the pathogenesis of obesity.

Because of differential processing of the hormone precursor, proglucagon, numerous peptide products are released from the pancreatic alpha cells and the intestinal L-cells in which the (pro)glucagon gene is expressed. Of particular interest in relation to obesity are glucagon from the pancreas and oxyntomodulin and GLP-1 from the gut, all of which inhibit food intake, but the other products are also briefly discussed, because knowledge about these is required for selection and evaluation of the methods for measurement of the hormones. The distal intestinal L-cells also secrete the appetite-inh

Antioxidant and anti-ageing effects of enzymatic polysaccharide from Pleurotus eryngii residue.

The wide industrial cultivation of Pleurotus eryngii (P. eryngii) has resulted in the massive production of mushroom residues (MR) with low-efficiency utilization. In the present study, the P. eryngii enzymatic residue polysaccharide (PERP) was obtained from the P. eryngii residues. The characterization analysis showed that PERP was polysaccharides comprised of five kinds of monosaccharides with molecular weight of 2.05 × 103 Da. PERP also showed rough surface and appeared as spherical structure dispersed in aqueous solution. The animal experiment analysis demonstrated that PERP exhibited pote

Is LRP2 Involved in Leptin Transport over the Blood-Brain Barrier and Development of Obesity?

The mechanisms underlying the transport of leptin into the brain are still largely unclear. While the leptin receptor has been implicated in the transport process, recent evidence has suggested an additional role of LRP2 (megalin). To evaluate the function of LRP2 for leptin transport across the blood-brain barrier (BBB), we developed a novel leptin-luciferase fusion protein (pLG), which stimulated leptin signaling and was transported in an in vitro BBB model based on porcine endothelial cells. The LRP inhibitor RAP did not affect leptin transport, arguing against a role of LRP2. In line with

Analysis of geometrical isomers of dietary carotenoids.

Carotenoids are versatile isoprenoids of great importance in food science and nutrition. These compounds are natural colorants and key to combat vitamin A deficiency, a major global nutritional problem. In addition, carotenoids have health-promoting biological actions that can contribute to reduce the risk of serious diseases, including cancer, cardiovascular disease, skin, bone and eye conditions and metabolic disorders. Beneficial roles of carotenoids in cognition and fetal development are also attracting increased attention. The presence of double bonds cause carotenoids to be prone to geom

Shaping the Innate Immune Response by Dietary Glucans: Any Role in the Control of Cancer?

β-glucans represent a heterogeneous group of naturally occurring and biologically active polysaccharides found in many kinds of edible mushrooms, baker's yeast, cereals and seaweeds, whose health-promoting effects have been known since ancient times. These compounds can be taken orally as food supplements or as part of daily diets, and are safe to use, nonimmunogenic and well tolerated. A main feature of β-glucans is their capacity to function as biological response modifiers, exerting regulatory effects on inflammation and shaping the effector functions of different innate and adaptive immuni

Rationally designed imidazole derivative as colorimetric and fluorometric sensor for selective, qualitative and quantitative cyanide ion detection in real time samples.

A new imidazole derivative of 1,2-diaminoanthraquinone and fluorene-2-carboxaldehyde was designed as a sensor B2 to selectively detect the cyanide (CN-) ion through colorimetric and/or fluorometric methods. The photochemical characterizations of sensor B2 were tested using absorption and emission spectral studies in CH3CN-H2O (8:2) semi-aqueous medium. An excited state proton transfer process (ESIPT) was proved by theoretical and spectral studies. The colorimetric and fluorescence detection limit of CN- ion was found to be 5.3 × 10-6 M and 4.11 × 10-8 M, respectively. 1H NMR titration, electro

Pathophysiology of Drug-Induced Hypomagnesaemia.

Magnesium (Mg2+) is the second most abundant intracellular and fourth extracellular cation found in the body and is involved in a wide range of functions in the human cell and human physiology. Its role in most of the enzyme processes (ATP-ases)-stabilisation of nucleic acids (DNA, RNA), regulation of calcium and potassium ion channels, proliferation, glucose metabolism and apoptosis-make it one of the most important cations in the cell. Three pathogenetic mechanisms are mainly implicated in the development of hypomagnesaemia: reduced food intake, decreased intestinal absorption and increased

Characterization of thermo-oxidative behavior of ethylcellulose oleogels.

The thermo-oxidative behavior crucial to the applicability of ethylcellulose (EC) oleogels is characterized. Not only did we take into account the composition of the gel network in relation to textural attributes, but also the dynamic chemical changes occurred during formation, heating, and holding of the gels. EC oleogel oxidative stability showed that at 6.0% EC100 concentration in the oleogels the movement of liquid oil trapped in the gel network was hindered by its high viscosity and stable gel network, thus retarding oxidation. Processing temperature ≤ 120 °C for <2 h was recommended when

Daratumumab in multiple myeloma.

The development of effective monoclonal antibodies for the treatment of myeloma has been a long journey of clinical and drug development. Identification of the right target antigen was a critical part of the process. CD38 as a target has been considered for some time, but clinically, daratumumab, a CD38 monoclonal antibody, was the first to be tested, and it has delivered the best clinical responses as a single agent to date. Its proven safety and efficacy in combination with other antimyeloma agents have led to several US Food and Drug Administration approvals for treating myeloma. Furthermor

International Commission on Trichinellosis: Recommendations on pre-harvest control of Trichinella in food animals.

Transmission of Trichinella to domestic livestock, notably pigs, is limited to certain risk factors including feeding of raw meat-containing waste products or animal carcasses and exposure to infected rodents and wildlife. Prevention of infection in food animals is dependent on eliminating risk of exposure to these potential sources of Trichinella. By implementing conditions of controlled management, pig herds can be considered to pose a negligible risk for Trichinella, and groups of herds which follow these same conditions can be considered to be a compartment with negligible risk for Trichin

The Gordon Memorial Lecture: novel approaches to controlling bacterial infections.

1. There is huge emphasis in veterinary and agricultural science in understanding the basics of processes and exploiting them for benefits to the economy and human and animal welfare. It is always valuable to be able to step back from existing or favourite hypotheses and paradigms to look at an area of work or problem and see whether a different approach might be productive particularly by drawing parallels with other sometimes unrelated problems. 2. This approach has been used to explore (i) the use of live, attenuated Salmonella vaccines to generate a new form of competitive exclusion, (ii)

Identification of Antioxidant Peptides in Enzymatic Hydrolysates of Carp (Cyprinus Carpio) Skin Gelatin.

The protein by-products from carp (Cyprinus carpio) are normally discarded as industrial waste during fish processing. The objective of this study was to identify and characterise the peptides with a potential antioxidant activity that are released from carp skin proteins during hydrolysis by the Protamex enzyme mixture. This study shows that a hydrolysate of carp skin gelatin and its reversed-phase chromatography fractions have strong in vitro antioxidant properties. Among these fractions, the alanine-tyrosine (Ala-Tyr) dipeptide was identified as the major compound with high antioxidant pote
