MeCP2 Lactylation Protects against Ischemic Brain Injury by Transcriptionally Regulating Neuronal Apoptosis.

Lactate plays diverse roles in brain pathophysiology, including ischemic stroke. Here, the role of lysine lactylation, an epigenetic modification of lactate, in cerebral ischemia is investigated. Using a mouse model of transient middle cerebral artery occlusion, increased brain lactate levels and global protein lactylation are observed. Proteomics analysis reveals significant lactylation of non-histone proteins in the ischemic penumbra. Lactylation of MeCP2, a transcriptional regulator, is identified as a protective mechanism against stroke-induced neuronal death. Inhibition of MeCP2 lactylati

Unraveling an enhancer-silencer regulatory element showing epistatic interaction with a variant that escaped genome-wide association studies.

Regulation of gene expression has recently been complicated by identifying Epromoters, a subset of promoters with enhancer function. Here, we uncovered a dual cis-regulatory element, "ESpromoter," exhibiting both enhancer and silencer function as a regulator of the nearby genes ATP2B4 and LAX1 in single human T cells. Through an integrative approach, we pinpointed functional rs11240391, a severe malaria-risk variant that escapes detection in genome-wide association studies, challenging conventional strategies for identifying causal variants. CRISPR-modified cells demonstrated the regulatory ef

Dermal T cell immunity and key regulatory signaling pathways: Implications in immune-mediated alopecia and hair regeneration.

Mammalian hair follicles undergo periodic regeneration, with recent research highlighting the immunological niche as a critical regulator of stem cell activity and hair follicle regeneration. Chemotactic signals from hair follicles attract macrophages and T cells, which, in turn, control the resting and differentiation of epithelial stem cells in both healthy and damaged conditions. T cells play a pivotal role in hair follicle regeneration, contributing to injury-induced hair neogenesis and physiologic hair cycling. However, disruption of this interaction can lead to clinically significant imm

The ADCY1-mediated cAMP signaling pathway mediates functional effects of montelukast treatment in brain organoids.

Montelukast (MTK) is a drug widely used for treating allergic rhinitis and asthma. However, severe neuropsychiatric adverse events related to MTK have been reported, with limited understanding of the underlying mechanisms. Here we leveraged human forebrain organoids (hFOs) and showed that MTK exposure in hFOs downregulated the expression of genes associated with multiple neuronal functions and neuropsychiatric disorders. The following integrative analysis highlighted adenylate cyclase 1 (ADCY1), a main regulator of the cAMP signaling pathway, as a hub gene mediating the functional effects of M

Ursolic Acid Activates Mitochondrial Unfolded Protein Response to Enhance Innate Immunity via Transcription Factor ATFS-1/ATF5.

Ursolic acid (UA), a pentacyclic triterpenoid found in various plants, exhibits diverse biological activities. The mitochondrial unfolded protein response (UPRmt) acts as a pivotal regulator in orchestrating innate immune responses. However, it remains unclear whether UA modulates innate immunity, and the molecular mechanisms involved have yet to be elucidated. In this study, we established that UA enhances the survival of Caenorhabditis elegans against Pseudomonas aeruginosa PA14 infection by promoting intestinal bacterial clearance, independent of inhibitory effects on bacterial growth or ev

Maternal thyroid hormones affect zebrafish embryo development.

Thyroid hormones (THs), including thyroxine (T4) and triiodothyronine (T3), regulate vertebrate development, influencing critical processes during embryogenesis and larval stages. However, their maternal transfer and developmental roles in fish remain poorly understood. This study investigates maternal THs' impact on zebrafish development, focusing on the somatotropic axis, a key regulator of growth and metabolism. Female zebrafish were fed diets with 12.5 or 25 μg T4/g body weight/day for five days, while controls received vehicle-only food. Plasma TH levels were measured, and females were br

Elucidating the chromatin-driven transcription regulatory networks response to Streptococcus agalactiae infection under low temperature in Nile tilapia.

Low temperature impairs immune function in ectothermic vertebrates, but the underlying regulatory mechanisms remain unclear. Here, we show that hypothermia significantly decreases bacterial load and increases mortality in Nile tilapia (Oreochromis niloticus) following Streptococcus agalactiae infection. Integrative RNA-seq and ATAC-seq profiling of head kidney tissue revealed widespread suppression of immune gene expression under cold stress, including ifrd2, isg15, cxcr1, ccr2, ccr9a, and socs1b. Footprinting analysis of 259 transcription factor binding elements in accessible chromatin enable

The Regulation of Catecholamine Biosynthesis by the Gas Transmitters Carbon Monoxide and Hydrogen Sulfide.

The gas transmitters nitric oxide (NO), carbon monoxide (CO), and hydrogen sulfide (H2S) play important roles in physiological regulation, including adrenal function. Among them, only NO has been directly implicated in controlling catecholamine biosynthesis. This study examined whether CO and H2S exert similar effects by treating PC12 cells with a CO donor (CORM-2) or an H2S donor (NaHS), with or without glucocorticoid stimulation. Gene expression of tyrosine hydroxylase (Th), dopamine β-hydroxylase (Dbh), and phenylethanolamine N-methyltransferase (Pnmt) was assessed by RT-qPCR, and catechola

Diazepam exerts immunosuppressive effects on macrophages isolated from the head kidney of catfish (Silurus asotus).

Diazepam (DZ), a common benzodiazepine, is increasingly detected in aquatic environments, yet its immunological effects in fish remain poorly understood. We investigated the impact of DZ at concentrations ranging from 1 to 10 mg/L on head kidney-derived macrophages of Amur catfish (Silurus asotus). Cell viability was unaffected, but DZ induced dose-dependent macrophage aggregation and significantly reduced phagocytic activity. Transcriptomic profiling revealed 1053 differentially expressed genes, with suppression of immune-related pathways, including PI3K-Akt, MAPK, phagosome, and toll-like re

ZNF865 (BLST) Regulates Back Pain via Cell Senescence and DNA Damage Mechanisms.

Senescence has been shown to contribute to the progression of aging related diseases, yet its regulation in the intervertebral disc (IVD) remains poorly understood. Recently, ZNF865 (BLST) was identified as a previously uncharacterized zinc finger protein, that regulates a wide array of genes related to protein processing, cell senescence and DNA damage repair. Here, we show that ZNF865 expression decreases with age and pathology in human and mouse IVD samples. Depletion of ZNF865 induces senescence, SASP expression and DNA damage in both human and rat healthy NP cells. Conversely, restoration

Discovery of isoindoline-2(1H)-carboxamide STING inhibitors as anti-inflammatory agents.

STING (stimulator of interferon genes) is an endoplasmic reticulum-resident membrane-spanning protein that is widely expressed in mammalian cells and functions as a central regulator for the innate immunity. Aberrant activation of the STING axis due to loss-of-function or gain-of-function mutation leads to various autoimmune and autoinflammatory disorders such as Aicardi-Goutières syndrome, systemic lupus erythematosus, and STING-associated vasculopathy with onset in infancy. Here we report the design, synthesis, and structure-activity relationship (SAR) of the isoindoline-2(1H)-carboxamide ST

Circulatory age-associated B cells: Their distinct transcriptomic characteristics and clinical significance in drug-naïve patients with rheumatoid arthritis.

Age-associated B cells (ABCs) have been implicated in the pathogenesis of autoimmune diseases. However, the global gene expression and clinical significance of circulatory ABCs in rheumatoid arthritis (RA) remain poorly understood. Here, single-cell RNA sequencing identified nine B cell subsets in peripheral blood of RA patients, including ABCs. Increased phagocytosis and antigen presentation were functionally enriched by the genes expressed differentially in ABCs. Network analysis and in vitro experiments demonstrated SYK as a key regulator defining the myeloid-like phenotypes in ABCs. Flow c

TLX3 regulates CGN progenitor proliferation during cerebellum development and its dysfunction can lead to autism.

Tlx3, a master regulator of the fate specification of excitatory neurons, is primarily known to function in post-mitotic cells. Although we have previously identified TLX3 expression in the proliferating granule neuron progenitors (GNPs) of cerebellum, its primary role is unknown. Here, we demonstrate that the dysfunction of Tlx3 from the GNPs significantly reduced its proliferation through regulating anti-proliferative genes. Consequently, the altered generation of GNPs resulted in cerebellar hypoplasia, patterning defects, granule neuron-Purkinje ratio imbalance, and aberrant synaptic connec

Amelioration of nonalcoholic fatty liver disease by inhibiting the deubiquitylating enzyme RPN11.

Nonalcoholic fatty liver disease (NAFLD), including its more severe manifestation nonalcoholic steatohepatitis (NASH), is a global public health challenge. Here, we explore the role of deubiquitinating enzyme RPN11 in NAFLD and NASH. Hepatocyte-specific RPN11 knockout mice are protected from diet-induced liver steatosis, insulin resistance, and steatohepatitis. Mechanistically, RPN11 deubiquitinates and stabilizes METTL3 to enhance the m6A modification and expression of acyl-coenzyme A (CoA) synthetase short-chain family member 3 (ACSS3), which generates propionyl-CoA to upregulate lipid metab

Molecular mechanisms of the obesity associated with Bardet-Biedl syndrome: An update.

Obesity is a prominent feature of Bardet-Biedl syndrome (BBS), which represents a major and growing public health problem. More than half of BBS patients carry mutations in one of eight genes that encode subunits of a protein complex known as the BBSome, which has emerged as a key regulator of energy and glucose homeostasis. However, the mechanisms underlying obesity in BBS are complex. Numerous studies have identified a high prevalence of insulin resistance and metabolic syndrome among individuals with BBS. However, the exact mechanisms are not fully understood. This review summarized evidenc

RhoA downregulation in the murine intestinal epithelium results in chronic Wnt activation and increased tumorigenesis.

Rho GTPases are molecular switches regulating multiple cellular processes. To investigate the role of RhoA in normal intestinal physiology, we used a conditional mouse model overexpressing a dominant negative RhoA mutant (RhoAT19N) in the intestinal epithelium. Although RhoA inhibition did not cause an overt phenotype, increased levels of nuclear β-catenin were observed in the small intestinal epithelium of RhoAT19N mice, and the overexpression of multiple Wnt target genes revealed a chronic activation of Wnt signaling. Elevated Wnt signaling in RhoAT19N mice and intestinal organoids did not a

Rapid expansion and international spread of M1UK in the post-pandemic UK upsurge of Streptococcus pyogenes.

The UK observed a marked increase in scarlet fever and invasive group A streptococcal infection in 2022 with severe outcomes in children and similar trends worldwide. Here we report lineage M1UK to be the dominant source of invasive infections in this upsurge. Compared with ancestral M1global strains, invasive M1UK strains exhibit reduced genomic diversity and fewer mutations in two-component regulator genes covRS. The emergence of M1UK is dated to 2008. Following a bottleneck coinciding with the COVID-19 pandemic, three emergent M1UK clades underwent rapid nationwide expansion, despite lack o
