Genetic counseling and carrier screening in candidates for gamete donation at a Portuguese center.

OBJECTIVE: Genetic counseling and carrier screening are part of the gamete donation process by healthy individuals. We aim to review the findings of genetic counseling and carrier screening of a cohort of candidates at our public gametes bank. METHODS: Thirty-four male and 64 female candidates had genetic counseling with a medical geneticist before donation. Of these, one female candidate voluntarily dropped-out. Thirty-four males and 63 females performed karyotype and screening for the more common pathogenic variants for CFTR-related cystic fibrosis and spinal muscular atrophy (SMN1) in the

Sleep patterns, genetic susceptibility, and risk of new-onset gout: The UK Biobank prospective cohort study.

OBJECTIVE: The association between sleep behaviors and gout risk remains uncertain. We aimed to evaluate the relationship of sleep patterns based on a combination of five major sleep behaviors with the risk of new-onset gout, and to explore whether genetic risks of gout may modify this association in the general population. METHODS: 403,630 participants without gout at baseline in UK Biobank were included. A healthy sleep score was created by combining five major sleep behaviors, including chronotype, sleep duration, insomnia, snoring, and daytime sleepiness. Genetic risk score for gout was c

Gene Variants at Loci Related to Blood Pressure Account for Variation in Response to Antihypertensive Drugs Between Black and White Individuals.

Selection of antihypertensive treatment according to self-defined ethnicity is recommended by some guidelines but might be better guided by individual genotype rather than ethnicity or race. We compared the extent to which variation in blood pressure response across different ethnicities may be explained by genetic factors: genetically defined ancestry and gene variants at loci known to be associated with blood pressure. We analyzed data from 5 trials in which genotyping had been performed (n=4696) and in which treatment responses to β-blockers, angiotensin-converting enzyme inhibitors, angiot

Molecular identification and genetic diversity of Lutzomyia gomezi (Diptera: Psychodidae) using DNAbarcodes in Cordoba, Colombia.

Lutzomyia gomezi, a suspected vector of cutaneous leishmaniasis in Colombia is recorded for natural infection of Leishmania parasites, its anthrophilic behaviour and significant abundance supports its vectorial role. The difficulties associated with taxonomic identification due to lack of males require the use of molecular markers (DNA barcodes), which allows us to distinguish the species. In this study, the cytochrome oxidase I fragment was proposed as DNA barcode to identify specimens collected from Cordoba, Colombia (Planeta Rica: Arenoso/ Centro Alegre, Sahagún: Santiago Abajo/San Andresit

Genetic and environmental aspects in the association between attention-deficit hyperactivity disorder symptoms and binge-eating behavior in adults: a twin study.

BACKGROUND: Prior research demonstrated that attention-deficit hyperactivity disorder (ADHD) is associated with binge-eating behavior, binge-eating disorder (BED), and bulimia nervosa (BN). The aim of this study was to investigate these associations in an adult twin population, and to determine the extent to which ADHD symptoms and binge-eating behavior share genetic and environmental factors. METHODS: We used self-reports of current ADHD symptoms and lifetime binge-eating behavior and associated characteristics from a sample of over 18 000 adult twins aged 20-46 years, from the population-ba

Which BRCA genetic testing programs are ready for implementation in health care? A systematic review of economic evaluations.

PURPOSE: There is considerable evidence regarding the efficacy and effectiveness of BRCA genetic testing programs, but whether they represent good use of financial resources is not clear. Therefore, we aimed to identify the main health-care programs for BRCA testing and to evaluate their cost-effectiveness. METHODS: We performed a systematic review of full economic evaluations of health-care programs involving BRCA testing. RESULTS: Nine economic evaluations were included, and four main categories of BRCA testing programs were identified: (i) population-based genetic screening of individuals

'A low risk is still a risk': exploring women's attitudes towards genetic testing for breast cancer susceptibility in order to target disease prevention.

BACKGROUND: Population breast cancer screening programs by mammography are offered to women based on age. It has been suggested that a screening program based on genetic risk profile could be more effective by targeting interventions at those at higher genetic risk. This study explores women's attitudes towards genetic testing for breast cancer susceptibility in order to target breast cancer prevention. METHODS: A qualitative study was conducted using 4 focus groups with 26 women aged 42-73 years. Women were selected irrespective of personal or family history of breast cancer. Discussions wer

Routine assessment of amniotic fluid alpha-Fetoprotein in early second-trimester amniocentesis is no longer justified.

BACKGROUND: Open fetal neural tube defects are often followed by an increase in alpha-Fetoprotein concentration in amniotic fluid. For over 25 years there has been a routine to measure amniotic fluid alpha-Fetoprotein in conjunction with early genetic amniocentesis. The efficacy of such a screening test in a low-risk population has been questioned but never evaluated in a Swedish population. METHODS: Data were reviewed retrospectively from all consecutive early second-trimester genetic amniocenteses from two hospitals during the years 1993-2003. Indications for the genetic amniocenteses were

Long-term dynamics of natural populations of Schistosoma mansoni among Rattus rattus in patchy environment.

Dynamics of natural populations of Schistosoma mansoni were studied during 8 consecutive years among Rattus rattus populations from 8 transmission sites of the marshy forest focus of Guadeloupe (French West Indies). The schistosome population is over-dispersed (k = 0.119) within the murine hosts and ecological factors linked to the patchy environment may be responsible for such aggregated distribution. Analysis of the spatio-temporal variations in prevalences, intensities and abundances showed limited variations of the infection during the 8 years at the level of the whole parasite population

Vertebrate host or tick vector? Which is to blame for Borrelia lusitaniae population structure?

OBJECTIVE: Borrelia lusitaniae is a lizard-associated spirochaete which shows highly genetically structured populations in western Europe. Our objective was to assess whether selective forces acting on the Ixodes tick vector or the reservoir lizard, Psammodromus algirus, shaped its population structure. METHODS: We collected ticks feeding on P. algirus in the Iberian Peninsula and north Africa, and genetically characterized B. lusitaniae, the infected ticks, and the vertebrate host. RESULTS: The goeBURST analysis on seven MLST B. lusitaniae genes identified two main clusters: one from centra

Prevalence of psoriasis in indigenous communities around the world: a scoping review⋆.

Psoriasis is a chronic inflammatory condition with systemic comorbidities and significant impacts on quality of life. Its prevalence in indigenous populations is poorly understood, limiting targeted health interventions. This scoping review aims to review the prevalence of psoriasis in indigenous communities globally, identify genetic, environmental, and socioeconomic influences, and explore barriers to accurate diagnosis and care. A search of PubMed, Ovid MEDLINE, Cochrane Library, and Scopus identified 30 studies, of which 15 met inclusion criteria, encompassing 13 Indigenous populations acr

Skeletal site-specific variations in myeloid cells: insights from single-cell RNA sequencing of the mandible and femur.

To understand differences that exist between the cell populations in different skeletal sites, we performed an unbiased genetic survey via single-cell RNA sequencing of CD11b+ myeloid cells from mandibular- and femur-derived bone marrow of 2-mo-old C57BL/6 mice. Our results reveal transcriptomic evidence that suggests a uniquely inflammatory genetic profile of the mandibular-derived CD11b+ cells. The monocyte cell population found within the CD11b+ cells analyzed by scRNA-seq expressed Csf1r and Tnfrsf11a suggesting that this population contained osteoclast precursors. Osteoclasts of the crani

Genetic Diversity of Sweetpotato (Ipomoea batatas (L.) Lam.) from Portugal, Mozambique and Timor-Leste.

Portugal contributed to the global diffusion of sweetpotato (Ipomoea batatas L. Lam.). Although it is of minor importance on the Portuguese mainland, it is one of the most common crops in the Azores and Madeira archipelagos and is highly relevant in the Portuguese ex-colonies Mozambique and Timor-Leste. We analyzed the genetic diversity and population structure of sweetpotato from these five geographic provenances using twelve nuclear simple sequence repeat (SSR) markers. We studied 45 accessions, 15 of which were collected from farmers' fields in these five regions and 30 of which are held at

The potential causal association of systemic lupus erythematosus with congestive heart failure in the East Asian population: A two-sample mendelian randomization study.

OBJECTIVE: Systemic lupus erythematosus (SLE) is a systemic autoimmune disease that increases the risk of cardiovascular disease, but the causal relationship has remained unknown in the East Asian population. We aim to determine the causal relationship between SLE and congestive heart failure (CHF) in the East Asian population. METHODS: We conducted a two-sample Mendelian randomization MR study to examine the potential causal association of SLE with CHF, using the East Asian genome-wide association study datasets for SLE (4222 cases, 8431 controls) and CHF (9413 cases, 203,040 controls). Lactase deficiency in Russia: multiethnic genetic study.

BACKGROUND: Lactase persistence-the ability to digest lactose through adulthood-is closely related to evolutionary adaptations and has affected many populations since the beginning of cattle breeding. Nevertheless, the contrast initial phenotype, lactase non-persistence or adult lactase deficiency, is still observed in large numbers of people worldwide. METHODS: We performed a multiethnic genetic study of lactase deficiency on 24,439 people, the largest in Russia to date. The percent of each population group was estimated according to the local ancestry inference results. Additionally, we cal

A new approach to estimating the prevalence of hereditary hearing loss: An analysis of the distribution of sign language users based on census data in Russia.

The absence of comparable epidemiological data challenges the correct estimation of the prevalence of congenital hearing loss (HL) around the world. Sign language (SL) is known as the main type of communication of deaf people. We suggest that the distribution of SL can be interpreted as an indirect indicator of the prevalence of congenital HL. Since a significant part of congenital HL is due to genetic causes, an assessment of the distribution of SL users can reveal regions with an extensive accumulation of hereditary HL. For the first time, we analyzed the data on the distribution of SL users

Sexual Selection Does Not Increase the Rate of Compensatory Adaptation to a Mutation Influencing a Secondary Sexual Trait in Drosophila melanogaster.

Theoretical work predicts that sexual selection can enhance natural selection, increasing the rate of adaptation to new environments and helping purge harmful mutations. While some experiments support these predictions, remarkably little work has addressed the role of sexual selection on compensatory adaptation-populations' ability to compensate for the costs of deleterious alleles that are already present. We tested whether sexual selection, as well as the degree of standing genetic variation, affect the rate of compensatory evolution via phenotypic suppression in experimental populations of

Development of polymorphic EST microsatellite markers for the sand fly, Phlebotomus papatasi (Diptera: Psychodidae).

BACKGROUND: Phlebotomus papatasi is a widely distributed sand fly species in different tropical and sub-tropical regions including the Middle East and North Africa. It is considered an important vector that transmits Leishmania major parasites, the causative agents of cutaneous leishmaniasis. The development of microsatellite markers for this sand fly vector is of high interest to understand its population structure and to monitor its geographic dispersal. RESULTS: Fourteen polymorphic microsatellite markers were developed with simple di-, tri- and tetra-nucleotide repeats. The F statistics c
