Human Bordetella bronchiseptica infection related to contact with infected animals: persistence of bacteria in host.

Within a period of 2 1/2 years, Bordetella bronchiseptica was isolated four times from a 79-year-old woman with bronchopneumonia. We have demonstrated by pulsed-field gel electrophoresis that this infection was related to contact with infected rabbits. The initial human B. bronchiseptica isolate had a phenotype characteristic of usual B. bronchiseptica clinical isolates; it produced toxin and adhesins, such as adenylate cyclase-hemolysin, filamentous hemagglutinin, and pertactin, and was able to induce lethality in a murine respiratory model. By contrast, although the three successive human is

Herpes simplex chronic laryngitis and vocal cord lesions in a patient with acquired immunodeficiency syndrome.

Chronic laryngitis in patients with acquired immunodeficiency syndrome may be due to infections or tumors, such as Kaposi's sarcoma and non-Hodgkin's lymphoma. We present what we believe to be the first proven case of herpes simplex virus chronic laryngitis in a man positive for human immunodeficiency virus. Direct laryngoscopy showed leukoplakic lesions on both vocal cords. Biopsy of the lesions showed squamous epithelial cells with the characteristic features of herpes simplex virus, which was confirmed by immunohistochemical stains. We discuss the differential diagnosis of chronic laryngiti

Morphologic descriptions of Taenia asiatica sp. n.

Among taeniid tapeworms infecting humans through pork or beef, Taenia solium Linnaeus 1758 and Taenia saginata Goeze 1782 have already been known. Based on the morphologic characteristics of adult and metacestodes of Asian Taenia saginata, the third kind of human taeniid tapeworm known to distribute in Asian countries, a new species name of Taenia asiatica is proposed. In addition to the known biology in their intermediate hosts, T. asiatica was different morphologically from Taenia saginata Goeze 1782 in having the unarmed rostellum on the scolex of adult, the large number of 'uterine twigs'

A putative marker for human melanoma. A monoclonal antibody derived from the melanoma gene in the Xiphophorus melanoma model.

A MoAb was raised against a peptide corresponding to an exposed domain of the putative tyrosine kinase receptor protein encoded by Xmrk, a gene involved in melanoma formation and/or progression in the Xiphophorus fish melanoma model. The antibody reacts specifically with cells from human melanocytic lesions, ie, common acquired nevi, primary and metastatic melanoma biopsies. No reactivity with other cells, including normal melanocytes, was observed in the biopsies or with cells in biopsies from normal tissue (skin, liver, lung, spleen) and from other malignancies including those of neuroectode

2',3'-dideoxycytidine permeation of the human erythrocyte membrane.

The mechanism by which 2,3'-dideoxycytidine, an inhibitor of HIV-I infectivity, permeates the cell membrane was investigated. The influx of ddCyd into human erythrocytes was nonconcentrative. The initial velocity of both ddCyd influx and efflux was, in contrast to compounds that permeate the cell membrane via the nucleoside transporter, a linear function of nucleoside concentration in the 1 microM to 10 mM range and relatively insensitive to temperature. Furthermore, potent inhibitors of nucleoside transporter and other nucleosides were found to inhibit ddCyd influx only partially or not at al

Latent herpesviruses of humans.

The herpesviruses that infect humans characteristically establish a latent infection that may be reactivated later. The consequences of reactivation range from asymptomatic shedding to severe disseminated infection. Varicella-zoster and herpes simplex viruses are both highly neurotropic, establishing nonreplicating infections in sensory ganglia. Latent herpes simplex virus is known to reside in neurons, and the virus-cell interactions involved have been defined to an extent. Cytomegalovirus and Epstein-Barr virus interact with peripheral blood leukocytes. Latent cytomegalovirus infection of hu

The human T-cell leukemia/lymphoma virus, lymphoma, lytic bone lesions, and hypercalcemia.

The human T-cell lymphoma (HTL) virus is a type C RNA tumor virus isolated from patients with malignancies of mature T cells. We report three patients with peripheral T-cell lymphoma, hypercalcemia, and antibodies to HTL virus. One patient presented with idiopathic hypercalcemia of 6 months' duration, two with striking lytic bone lesions, and two with circulating malignant lymphocytes. Malignant cells from all patients had surface markers characteristic of thymic-derived lymphocytes (T cells), and all patients had natural serum antibodies to disrupted HTL virus and to one or both viral structu

Legionella infections: a review of five years of research.

In an attempt to place in perspective the role of Legionella pneumophila in human disease, the literature concerning this organism was reviewed. In the five years since the etiology of legionnaires' disease was elucidated, an impressive body of information has been gathered from retrospective investigations of illnesses of previously unidentified etiology and from studies of new cases of nosocomial or community-acquired infection caused by L. pneumophila. Topics on which considerable knowledge has been gained include the characteristics of L. pneumophila itself; the etiology, pathogenesis, and

Epstein-Barr virus: a human pathogen inducing lymphoproliferation in vivo and in vitro.

Epstein-Barr virus (EBV) is a pathogen that is associated with several diseases in humans. It causes most heterophile-positive cases of infectious mononucleosis. The virus is associated with another lymphoproliferative disease, African Burkitt's lymphoma, and with a malignancy of the nasopharynx, nasopharyngeal carcinoma. EBV appears to be a causative agent for some fatal lymphoproliferations in congenitally immunodeficient people. One proliferating cell type in infectious mononucleosis is an EBV-infected B lymphoblast. The tumor cell in African Burkitt's lymphoma is also an EBV-infected B lym

Isolation and culture of human endometrial glands.

A simple method for isolation of glands from human endometrium has been developed. The procedure involves collagenase digestion of the endometrial tissue and filtration through sieves of various pore sizes. Isolated glands retained on the sieves were washed and collected in culture dishes. Tubular organization of the isolated glands was ascertained by examination of the preparations under inverted microscope and light microscopy of stained sections. The appearance of the glands was found to reflect different functional states of the endometrium and, possibly, to reveal abnormalities. Growth of

A quantitative approach to assess influenza A virus fitness and transmission in guinea pigs.

Efforts to estimate the risk posed by potentially pandemic influenza A viruses (IAV), and to understand the mechanisms governing interspecies transmission, have been hampered by a lack of animal models that yield relevant and statistically robust measures of viral fitness. To address this gap, we monitored several quantitative measures of fitness in a guinea pig model: infectivity, magnitude of replication, kinetics of replication, efficiency of transmission, and kinetics of transmission. With the goal of identifying metrics that distinguish human- and non-human-adapted IAV we compared strains

Computational Auditory Periphery Models: the Return of the Rodent.

Animal experiments have provided many insights on auditory function, notably in cases of sensorineural hearing loss (SNHL). However, it is not always clear how these findings translate to the human auditory system, especially in clinically relevant contexts. Cross-species computational models of the auditory periphery can help bridge the gap between non-invasive human diagnostics and experimental evidence from animal studies. In this work we adapted a one-dimensional (1-D) nonlinear cochlear transmission-line (TL) model designed for the human auditory periphery (Verhulst et al., 2018) to mouse

A scoping review of the health effects of fermented foods in specific human populations and their potential role in precision nutrition: current knowledge and gaps.

BACKGROUND: Diets and specific foods have a significant impact on health, and individual responses to nutritional factors vary. This variability among humans can be considered a basis for developing personalized or precision nutrition. Fermented foods (FF) contain a wide range of macro- and micronutrients, bioactive compounds, and live or dead microorganisms. FF represent a diverse range of products and have garnered significant interest due to their potential health benefits. However, consistent evidence remains limited, possibly due to heterogeneity in individual responses. OBJECTIVES: The

A human neuronal model of Niemann Pick C disease developed from stem cells isolated from patient's skin.

BACKGROUND: Niemann Pick C (NPC) disease is a neurovisceral lysosomal storage disorder due to mutations in NPC1 or NPC2 genes, characterized by the accumulation of endocytosed unesterified cholesterol, gangliosides and other lipids within the lysosomes/late endosomes. Even if the neurodegeneration is the main feature of the disease, the analysis of the molecular pathways linking the lipid accumulation and cellular damage in the brain has been challenging due to the limited availability of human neuronal models. OBJECTIVE: The aim of this study was to develop a human neuronal model of NPC dise

A stable atypical H2S-negative, non-motile Salmonella Infantis variant from a human isolate associated with a barA stop-gain mutation.

Salmonella enterica serovar Infantis (S. Infantis) is an increasingly important food-borne pathogen, and detection in broiler flocks via the standardized EN/ISO 6579-1:2017 is of utmost importance. Phenotypic characteristics, such as the production of hydrogen sulfide (H2S) and swarming behavior, are essential for detection. However, the occurrence of atypical variants lacking such phenotypic characteristics bears a risk to public health. In this study, we characterized an Austrian human-derived S. Infantis strain showing, in addition to the normal colony phenotype (NCP), also an atypical colo

Can Large Language Models Preserve Diagnostic Accuracy Despite Patient Self-Diagnosis and Framing Bias in Hypothetical Upper-Extremity Scenarios?

PURPOSE: Online health queries are often addressed by large language models (LLMs) embedded in search engines. It is possible that LLMs, like human clinicians, might be misdirected by vague symptom descriptions or inaccurate self-diagnoses. We examined patient and scenario factors associated with an LLM's ability to identify intended upper-extremity musculoskeletal diagnoses and its tendency to deviate from patient self-diagnoses in structured clinical vignettes. METHODS: ChatGPT (GPT-5) evaluated 180 randomized hypothetical clinical vignettes depicting five common upper-extremity conditions:

The role of thickness inhomogeneities in hierarchical cortical folding.

The mammalian brain cortex is highly folded, with several developmental disorders affecting folding. On the extremes, lissencephaly, a lack of folds in humans, and polymicrogyria, an overly folded brain, can lead to severe mental retardation, short life expectancy, epileptic seizures, and tetraplegia. Not only a specific degree of folding, but also stereotyped patterns, are required for normal brain function. A quantitative model on how and why these folds appear during the development of the brain is the first step in understanding the cause of these conditions. In recent years, there have be

Identifying psychosocial characteristics that predict outcome to the UPLIFT programme for people with persistent back pain: protocol for a prospective cohort study.

INTRODUCTION: Prognostic screening of people with low back pain (LBP) improves utilisation of primary healthcare resources. Whether this also applies to secondary healthcare remains unclear. Therefore, this study aims to develop prognostic models to determine at baseline which patients with persistent LBP are likely to have a good and poor outcome to a 5-week programme of combined education and exercise ('UPLIFT') delivered in a secondary healthcare setting. METHODS AND ANALYSIS: A prospective cohort study of 246 people with persistent LBP will be conducted in a secondary healthcare outpatien
