Mechanistic Interplay Between Biomechanical Signaling and Epigenetic Regulation: Targeted Therapy Against Tumor Development and Malignant Progression.

An aberrant mechanical microenvironment is a major driver of malignant tumor progression. Its effects are not limited to the immediate regulation of tumor cell behavior. Through mechanotransduction-mediated epigenetic remodeling, external physical cues can also be converted into persistent malignant programs. Mechanical alterations in the physical tumor microenvironment, including increased matrix stiffness, compressive stress, fluid shear stress, and spatial confinement, can be sensed by integrin adhesion complexes, mechanosensitive ion channels, and the nucleo-cytoskeletal system. These sign

PRL-3 up-regulates exosomal ITGαvβ5 expression to promote liver pre-metastatic niche formation and colon cancer liver metastasis.

Liver pre-metastatic niches (PMN) formation is a pivotal process in colorectal cancer liver metastasis (CLM). Phosphatase of regenerating liver-3 (PRL-3) has been demonstrated as a key factor in promoting CRC progression (e.g., therapeutic resistance and metastasis), but its role in liver PMN formation remains unknown. Using mouse models and CRC patient samples, we herein reveal that high PRL-3 expression in CRC cells could enhance the recruitment of myeloid-derived suppressor cells (MDSCs) into the liver and impair the hepatic infiltration of CD8+ T cells, thereby promoting the liver PMN form

Extracellular Matrix Remodeling as a Mechanobiological Driver of Breast Cancer Aggressiveness: Comparative Oncology, Multi-Omics, and Artificial Intelligence Perspectives.

The extracellular matrix (ECM) is increasingly recognized as an active regulator of breast cancer progression rather than a passive structural scaffold. This narrative review examines how ECM remodeling contributes to tumor aggressiveness through changes in matrix composition, collagen architecture, tissue stiffness, mechanotransduction, stromal permissiveness, immune and metabolic programs, invasion, metastasis and therapeutic response. A structured narrative search of literature published from 1981 to June 2026 was used to support this synthesis. Evidence from breast cancer studies indicates

Biogenic selenium nanoparticles as dual-action antithrombotic agents: Targeting platelet activation cascades and thrombus formation.

Platelet hyperactivation is a central driver of thrombotic disorders, yet current antithrombotic therapies remain constrained by bleeding risk and other limitations. Here, we investigated protein-capped selenium nanoparticles (SeNPs, ~40 nm) as a potential antithrombotic nanoplatform. SeNPs were synthesized and characterized using spectroscopic and microscopic approaches, demonstrating spherical morphology and protein-associated stabilization. Hemocompatibility and metabolic viability studies showed no hemolysis and no significant reduction in viability at concentrations up to 100 μg/ml under

Single-Cell Transcriptomic Profiling Reveals Immunometabolic Reprogramming and Cell-Cell Communication in the Tumor Microenvironment of Human Hepatocellular Carcinoma.

Hepatocellular carcinoma (HCC) is sustained by coordinated interactions among malignant hepatocytes, immune cells, and stromal populations that collectively drive tumor growth, immune evasion, and vascular remodeling. Using integrative single-cell transcriptomics on 93,032 cells from tumor and healthy human liver, we characterized cell-type-specific transcriptional programs underlying immunometabolic reprogramming and reconstructed the intercellular communication circuits that maintain the tumor microenvironment. Malignant hepatocytes displayed upregulation of genes encoding both glycolytic an

Targeting the CD47-SIRPα Axis in Atherosclerosis: From Pathogenesis to Therapeutic Implications.

Atherosclerosis (AS), the pathological basis of cardiovascular diseases, remains a leading global cause of mortality. Its pathogenesis involves chronic inflammation, impaired clearance of apoptotic cells (efferocytosis), and lipid dysregulation within the arterial wall. The CD47-SIRPα signaling axis, a key immune checkpoint, has been identified as a critical regulator integrating these processes in the atherosclerotic plaque microenvironment. This review systematically details the multifaceted role of the aberrantly activated CD47-SIRPα axis in driving AS progression. We elaborate on its molec

Beauvericin Induces Mitochondrial Apoptosis and Attenuates EMT-Associated Phenotypes and Angiogenic Signaling in Colorectal Cancer Cells In Vitro.

Colorectal cancer (CRC) remains a leading cause of cancer-related mortality, largely due to metastasis and therapeutic resistance. Beauvericin, a cyclohexadepsipeptide mycotoxin produced by Beauveria and Cordyceps species, has demonstrated anticancer activity in multiple malignancies; however, its mechanistic effects in CRC have not been fully defined. In this study, we investigated the cellular and molecular effects of beauvericin in HCT116 and SW480 CRC cells using in vitro models. Beauvericin reduced cell viability and clonogenic growth, induced G0/G1 cell-cycle arrest, and activated mitoch

Mapping the antibody-drug-conjugates landscape in non-small cell lung cancer: Where are we and where are we going?

Antibody-drug conjugates (ADCs) represent a rapidly advancing therapeutic class in the treatment of non-small cell lung cancer (NSCLC), offering targeted delivery of cytotoxic agents to tumor cells while minimizing off-target toxicity. In recent years, several ADCs have emerged in both early (phase 1-2) and late (phase 3) clinical development, reflecting a growing recognition of their potential to improve outcomes in defined subsets of patients with advanced NSCLC. Late-phase ADCs include trastuzumab deruxtecan (targeting HER2 [human epidermal growth factor receptor 2]), datopotamab deruxtecan

Biomimetic Macrophage Cell Membrane-Based Nanoparticles for Effective Treatment of Glioblastoma Through Boron Neutron Capture Therapy Combined With Immunotherapy.

Glioblastoma multiforme (GBM) remains largely incurable due to the blood-brain barrier (BBB) and immunosuppressive microenvironment. While boron neutron capture therapy (BNCT) selectively eradicates tumor cells via 10B(n, α)-7Li reactions, its clinical potential in GBM is unrealized because of the suboptimal pharmacokinetics of conventional boron agents and their failure to achieve therapeutic intracranial concentrations. Here, we design macrophage membrane-coated PLGA nanoparticles (M@PLGA-10BN) that exploit innate biological transport mechanisms to overcome dual therapeutic barriers. The mac

In Silico Analyses Suggest That Exercise-Induced Irisin-Mediated Neuroprotection Supports Non-Pharmacological Preventive Strategies for Alzheimer's Disease in Public Health.

Alzheimer's disease (AD) is the leading cause of dementia and imposes a high economic and social burden on healthcare systems. In Brazil, the consistent increase in costs associated with AD hospitalizations, coupled with the absence of curative therapies and population aging, reinforces the need for low-cost, broadly applicable preventive strategies. This study investigated the role of irisin, a myokine induced by physical activity, in the prevention of AD, integrating epidemiological and bioinformatic analyses. Public data on the nutritional status of the Brazilian population in the early 200

The Protein Tyrosine Phosphatase CD45 promotes PMN Transepithelial Migration, Antimicrobial Function and Colonic Mucosal Repair.

Polymorphonuclear neutrophils (PMNs) serve as frontline defenders against injury and infection, eliminating pathogens and initiating mucosal tissue repair. However, excessive PMN transepithelial migration (TEpM) contributes to chronic mucosal inflammatory disorders, including inflammatory bowel disease. PMN pro-inflammatory and pro-repair functions are regulated by incompletely defined signaling cascades involving kinases and phosphatases. Here, we determined how the protein tyrosine phosphatase CD45/PTPRC regulates PMN trafficking and effector functions in the gut. Pharmacologic inhibition of

Defect-Engineered BiO2-X Nanosheets Mediate Sono-Thermomechanical ECM Remodeling for Hepatocellular Carcinoma Immunotherapy.

Immune checkpoint blockade for hepatocellular carcinoma (HCC) is frequently limited by the extracellular matrix (ECM). Through transcriptomic profiling, we identify that acquired anti-PD-1 resistance in HCC models is closely associated with prominent Fibronectin 1 (Fn1) upregulation within the tumor microenvironment. To address these interconnected physical and biological barriers, we developed an ultrasound-responsive nanoplatform utilizing oxygen-defect-abundant 2D BiO2-X nanosheets loaded with Fn1-targeted small interfering RNA (BiO2-X/siFn1). Introducing oxygen vacancies into ultrathin BiO

Adjuvants orchestrate cross-organ induction of mucosal CD8+ T cell immune responses in respiratory tracts.

Mucosal immunity is paramount for combating respiratory infections, but conventional vaccination methods, such as intramuscular injection, often struggle to induce it. Although direct mucosal vaccination is effective, it carries an increased risk of respiratory adverse effects, making it particularly challenging for individuals with chronic respiratory diseases. Consequently, developing a strategy to induce sufficient mucosal immune responses without modifying conventional vaccination methods would be highly desirable. The potential of vaccine adjuvants to facilitate such cross-organ induction

Participation of the purinergic P2X7 receptor in molecular complexes in the nucleus of human chondrocytes.

In addition to the purinergic receptor P2X7R's known activity as a sensor of damage-associated molecular patterns (DAMPs), evidences support its role in maintaining tissue homeostasis. Its presence in cellular compartments other than its usual transmembrane localization suggests its involvement in specific signaling pathways. This study aimed to analyze P2X7R in the nucleus of human chondrocytes and search for potential interacting partners. Through co-immunoprecipitation and proximity ligation assay we discovered that, independent of extracellular ATP levels, P2X7R is abundantly present in bo

Functionalized neutrophil nanovesicles for targeted photodynamic immunotherapy and potentiation of checkpoint blockade in colorectal cancer.

Colorectal cancer (CRC) patients with microsatellite-stable tumors typically resist immune checkpoint blockade (ICB) due to an immunosuppressive "cold" microenvironment. Photodynamic therapy (PDT) generates reactive oxygen species (ROS) to ablate tumor cells and induce immunogenic cell death (ICD), potentiating antitumor immunity. We previously demonstrated that neutrophil-derived nanovesicles (NNVs) retain tumor-suppressive cytotoxic payloads, including granzymes and perforin. Here, we engineered RGD-functionalized neutrophil nanovesicles encapsulating Ce6-loaded liposomes (RNC) via co-extrus

Genomic and epigenetic regulatory mechanisms in exercise-based rehabilitation processes: Cellular and tissue remodeling, microvascular adaptation, and circulating biomarkers.

While exercise-based rehabilitation is known to positively impact functionally related parameters, the role of genomic and epigenomic responses coordinated with cellular, extracellular matrix (ECM), mitochondrial, and microvascular adaptations remains insufficiently investigated. This narrative review summarizes mechanistic evidence linking exercise-associated mechanical, metabolic, hypoxia-redox, inflammatory, and hemodynamic stimuli with tissue remodeling and clinically relevant biomarkers. Current findings indicate that integrin-focal adhesion kinase (FAK) signaling and Hippo YAP/TAZ pathwa
