Functional characterization of optimized acellular peripheral nerve graft in a rat sciatic nerve injury model.

OBJECTIVES: Acellular grafts are a viable option for use in nerve reconstruction surgeries. Recently, our lab created a novel optimized decellularization procedure that removes immunological material while leaving the majority of the extracellular matrix structure intact. The optimized acellular (OA) graft has been shown to elicit an immune response equal to or less than that elicited by the isograft, the analog of the autograft in the rat model. We investigated the performance of the OA graft to provide functional recovery in a long-term study. METHODS: We performed a long-term functional re

A novel mutation in the nerve-specific 5'UTR of the GJB1 gene causes X-linked Charcot-Marie-Tooth disease.

X-linked Charcot-Marie-Tooth disease (CMT1X) is the second most common cause of CMT, and is usually caused by mutations in the gap junction protein beta 1 (GJB1) gene which codes for connexin 32 (CX32). CX32 has three tissue-specific promoters, P1 which is specific for liver and pancreas, P1a specific for liver, oocytes and embryonic stem cells, and P2 which is nerve-specific. Over 300 mutations have been described in GJB1, spread throughout the coding region. We describe two families with X-linked inheritance and a phenotype consistent with CMT1X who did not have mutations in the GJB1 coding

The use of an ultrasonic bone curette in the surgery of jaw tumors involving the inferior alveolar nerve.

BACKGROUND: Preservation of lip sensation is crucial in ablative surgery of mandibular tumors. When tumor control does not necessitate sacrifice of the inferior alveolar nerve (IAN), as in some cases of benign tumors of the lower jaw, attempts may be made to spare the nerve. The authors present and discuss their experience with an ultrasonic device in the treatment of benign tumors of the jaw in correspondence of the IAN. MATERIALS AND METHODS: Five patients with tumoral lesions involving the IAN underwent surgery with an ultrasonic surgical device (Sonopet Omni Surgical System; Stryker, Kala

Radiculopathy caused by lumbar epidural venous varix: case report.

A 33-year-old man presented with a rare case of lumbar epidural venous varix causing radiculopathy manifesting as sciatica exacerbated during bowel movements. The left straight-leg raising test was positive and patellar tendon reflex was lost. Lumbar magnetic resonance imaging showed a mass lesion in the epidural space at the L4 level just anterior to the dural sac on the left, appearing as hypointense on T(1)- and hyperintense on T(2)-weighted images, with ring-like enhancement with gadopentetate dimeglumine. He underwent surgery under a preoperative diagnosis of cystic nerve sheath tumor. Af

Intraoral approach for the treatment of submandibular salivary gland mixed tumors.

Pleomorphic adenoma represents a most common benign neoplasm of major salivary glands. Most occurrences of this benign tumor in the submandibular gland have been treated surgically without difficulty via transcervical approach. However, a few clinical problems in the transcervical approach have been mentioned, such as nerve injury or aesthetic scaring. In this study, we introduced the intraoral approach for the removal of the submandibular mixed tumor. In total, 12 cases of pleomorphic adenoma of the submandibular gland were treated via intraoral approach. The surgical technique and morbidity

Nerve growth factor expression by PLG-mediated lipofection.

Biomaterials capable of efficient gene delivery provide a fundamental tool for basic and applied research models, such as promoting neural regeneration. We developed a system for the encapsulation and sustained release of plasmid DNA complexed with a cationic lipid and investigated their efficacy using in vitro models of neurite outgrowth. Sustained lipoplex release was obtained for up to 50 days, with rates controlled by the fabrication conditions. Released lipoplexes retained their activity, transfecting 48.2+/-8.3% of NIH3T3 cells with luciferase activity of 3.97x10(7)RLU/mg. Expression of

The 4C5 antigen is associated with Schwann cell migration during development and regeneration of the rat peripheral nervous system.

The monoclonal antibody 4C5 recognizes a cell surface antigen of the developing central nervous system (CNS) and peripheral nervous system (PNS). In vitro antibody perturbation experiments have shown that the 4C5 antigen is involved in horizontal and vertical migration processes of granule cells during development of the rodent cerebellum. Moreover, results concerning the cellular localization and temporal expression of the 4C5 antigen during development and after injury of the rat sciatic nerve suggested that it may participate in Schwann cell migrations that occur during the above processes.

Rapid and sensitive neurotoxicity test based on the morphological changes of PC12 cells with simple computer-assisted image analysis.

In order to develop a rapid and sensitive bioassay for the screening of chemicals with possible neurotoxicity, a computer-assisted simple image-analysis system was developed to quantify small changes in the specific morphology of the cultured pheochromocytoma cell line, PC12. This cell forms a neuron-like microfibril network (neurites) in response to a nerve growth factor (NGF) stimulation in vitro. Dichrolvos (DDVP) and methylmercury chloride (MMC) were employed as model neurotoxicants. In DDVP treatment, there was no large difference in the ED50s (effective dose that reduces the morphologica

Heavy snorer's disease: a progressive local neuropathy.

"Heavy snorer's disease" is defined as progression from heavy snoring to obstructive sleep apnoea syndrome (OSAS). Apart from significant weight gain, the aetiology underlying progression to a collapse of the upper airways during inspiration and sleep remains unclear. Previous studies have shown that nocturnal respiratory disturbances became worse, even in some OSAS patients who did not gain weight. The patency of the upper airways depends on the balance between the negative intrapharyngeal pressure developed during inspiration and its counteraction by dilating muscles. The reflexogenic dilati

Topographical anatomy of the posterior elbow region during fetal development.

We studied the topography of the elbow region with special regard to the ulnar nerve during fetal life. Plastinated 600 microns thick sections through the right and left arms of ten fetuses with a crown-rump length (CRL) of 105 to 360 mm and of a newborn infant were investigated. The ulnar groove does not appear on the dorsal side of the medial humeral epicondyle earlier than at a crown-rump length of 130 mm. It provides the entrance into a formerly described fibro-osseous tunnel called the cubital tunnel. Its floor is formed by the posterior bundle of the ulnar collateral ligament, which bulg

Effects of elevation on nerve function in an acute upper extremity nerve compression model.

External compression was applied to the palmar surface of the wrist over the carpal tunnel in eight healthy volunteer subjects. With the arm in the maximum elevated position, the carpal tunnel pressure was elevated to 50 mm Hg, being continuously monitored by a slit catheter inserted into the carpal canal. Sensory and motor latencies and amplitudes were evaluated at 1-min intervals. When the sensory amplitude decreased by 50% (correlating with subjective sensory changes), the hand was lowered to heart level, still maintaining the carpal tunnel pressure at 50 mm Hg. Nerve conduction velocity an

[Peripheral neuropathies with neural vasculitis. Clinical and electrophysiological aspects of 33 cases].

Lesions of vasculitis of nerve were found in 33 of 140 patients hospitalized for peripheral neuropathy over a little less than 2 years, and in 113 specimens from neuromuscular biopsies. A first group of 7 patients had histological lesions suggestive of systemic necrotizing vasculitis (periarteritis nodosa in 4 cases and Churg and Strauss disease in 2 cases); the 7th patient was treated for myeloproliferative syndrome. The clinical and electromyographic signs were those of an axonal polyneuropathy which was improved by various treatments in 6 cases. A second group of 26 patients had histologica

An Anatomical Cadaveric Demonstration of an Ultrasound-Guided Fascial Plane Injection Pathway in the Deep Gluteal Space.

Background: Deep gluteal syndrome (DGS) has traditionally been attributed to sciatic nerve entrapment within the deep gluteal space. However, increasing evidence suggests that enthesopathy and soft tissue pathology of the short external rotators may also contribute to its pathogenesis. Conventional ultrasound-guided interventions primarily target the sciatic nerve through perineural hydrodissection (HD), which may not address enthesis-related pathology. However, the anatomical feasibility of delivering injectate along the deep gluteal fascial plane has not yet been investigated in cadaveric st

Spinal cord transverse section hinders Wallerian degeneration and neoangiogenesis after peripheral nerve transection: Involvement of Schwann cells' miR-134-5p.

Patients with spinal cord injury (SCI) frequently develop chronic wounds in paralyzed limbs, underscoring the importance of neural regulation and peripheral nerve integrity for tissue repair; however, it remains unclear how concomitant SCI alters the distal responses to peripheral nerve injury (PNI) and the associated microRNA (miRNA) profile. Sprague-Dawley rats underwent sciatic nerve transection alone (PNI group) or complete spinal cord transection at L1-L2 combined with sciatic transection (SCI+PNI group), and distal sciatic nerve stumps were harvested at 0, 3, and 7 days for gross observa

MXene-chitosan photo-responsive conduit for wireless optogenetic stimulation to enhance neural regeneration and functional recovery after optic nerve injury.

Optic nerve injury triggers progressive degeneration of retinal ganglion cells (RGCs) and axonal loss, driven by inhibitory microenvironmental factors such as glial scarring, myelin debris, and growth-inhibitory signaling. Physical stimuli such as photothermal and photoelectric stimulations have gained attention, yet little is known about their potential on normal cells or the optic nerve due to setbacks from over-exposure. Photothermal stimulus presents photoelectric cues and, at the same time, energy conversion for heat generation. Herein, a bio-functional platform was designed by incorporat

Other Nerve Sheath Tumors of Brain and Spinal Cord.

The three main types of nerve sheath tumors are schwannomas, neurofibromas and perineuriomas. Multiple neurofibromas throughout the body are the hallmark of Neurofibromatosis type 1 (NF1). Spinal nerve sheath tumors are classified in the group of intradural extramedullary spinal cord tumors, in which they are the most common type (25-30%). Their incidence is 3-4 per 1 million people. Spinal schwannomas are encountered sporadically or in the context of Neurofibromatosis type 2, while neurofibromas are typical for patients with Neurofibromatosis type 1. Neurofibromas are composed predominantly o

Nerve fibers and endometriotic lesions: partners in crime in inflicting pains in women with endometriosis.

One of major objectives in treating endometriosis is to alleviate pain since dysmenorrhea and other types of pain top the list of complaints from women with endometriosis who seek medical attention. Indeed, endometriosis-associated pain (EAP) is the most debilitating of the disease that negatively impacts on the quality of life in affected women, contributing significantly to the burden of disease and adding to the substantial personal and societal costs. Unfortunately, the mechanisms underlying the EAP are still poorly understood. In the last two decades, one active research field in endometr

Cell relationship in a Wistar rat model of spontaneous prostatitis.

PURPOSE: Prostatitis in men is a painful, noninfectious inflammatory condition. It is similar to interstitial cystitis which is associated with increased bladder mast cell and sensory nerve fiber density as well as suprapubic pain. Certain strains of rats may provide a useful model for studies of the development of spontaneous prostatitis. We evaluated the time course, and involvement of mast cells and sensory nerve fibers in this process using Wistar rats. MATERIALS AND METHODS: The prostates of 4, 6, 8, 10 and 13-week-old male Wistar rats were examined for the degree of inflammation, innerv
