Ecological and genetic relationships of the Forest-M form among chromosomal and molecular forms of the malaria vector Anopheles gambiae sensu stricto.

BACKGROUND: Anopheles gambiae sensu stricto, one of the principal vectors of malaria, has been divided into two subspecific groups, known as the M and S molecular forms. Recent studies suggest that the M form found in Cameroon is genetically distinct from the M form found in Mali and elsewhere in West Africa, suggesting further subdivision within that form. METHODS: Chromosomal, microsatellite and geographic/ecological evidence are synthesized to identify sources of genetic polymorphism among chromosomal and molecular forms of the malaria vector Anopheles gambiae s.s. RESULTS: Cytogeneticall

Multilocus microsatellite typing (MLMT) reveals genetic homogeneity of Leishmania donovani strains in the Indian subcontinent.

In this population genetic study of Leishmania donovani parasites in the Indian subcontinent, 132 isolates obtained from patients in Bangladesh, India, Nepal and Sri Lanka suffering from Kala-azar (100), post-Kala-azar dermal leishmaniasis (PKDL) (25) and cutaneous leishmaniasis (CL) (2), and from 5 patients whose clinical patterns were not defined, were analysed by using 15 hyper-variable microsatellite loci. Multilocus microsatellite typing (MLMT) data were analysed by using a Bayesian model-based clustering algorithm and constructing phylogenic tree based on genetic distances. In total, 125

Validating an automated outcomes surveillance application using data from a terminated randomized, controlled trial (OPUS [TIMI-16]).

We sought to validate an automated outcomes surveillance system (DELTA) using OPUS (TIMI-16), a multi-center randomized, controlled trial that was stopped early due to elevated mortality in one of the two intervention arms. Methodologies that were incorporated into the application (Statistical Process Control [SPC] and Bayesian Updating Statistics [BUS]) were compared with standard Data Safety Monitoring Board (DSMB) protocols.

A curious coincidence: mosquito biodiversity and the limits of the Japanese encephalitis virus in Australasia.

BACKGROUND: The mosquito Culex annulirostris Skuse (Diptera: Culicidae) is the major vector of endemic arboviruses in Australia and is also responsible for the establishment of the Japanese encephalitis virus (JEV) in southern Papua New Guinea (PNG) as well as its incursions into northern Australia. Papua New Guinea and mainland Australia are separated by a small stretch of water, the Torres Strait, and its islands. While there has been regular JEV activity on these islands, JEV has not established on mainland Australia despite an abundance of Cx. annulirostris and porcine amplifying hosts. De

Bayesian analysis of multicentre trial outcomes.

Bayesian methods provide flexibility in the analysis of data from multicentre trials that would be difficult to achieve by other means. This paper illustrates some useful applications of Bayesian methods to the analysis of multicentre trials, with emphasis on insights that would be difficult to obtain using conventional frequentist methods. Two trials provide data for illustration: a large multicentre trial comparing two doses of a drug with placebo with respect to an essentially continuous measurement for which the original analysis revealed a significant treatment by centre effect, and a lar

Multicenter study to determine antibody concentrations and assess the safety of administration of INH-A21, a donor-selected human Staphylococcal immune globulin, in low-birth-weight infants.

Nosocomial or late-onset sepsis is a common complication among premature infants, with a frequency inversely correlated with birth weight. Increased susceptibility to infection is due in part to an immature humoral (antibody-mediated) immune response. This study investigated the pharmacokinetics (PKs) and safety of a donor-selected specific intravenous immune globulin (IVIG) preparation, INH-A21 (Veronate), for prevention of sepsis in premature infants. Thirty-six infants weighing between 500 and 1,250 g during the first postnatal week were eligible to begin a series of up to four intravenous

Pharmacokinetic-pharmacodynamic modeling of morphine-6-glucuronide-induced analgesia in healthy volunteers: absence of sex differences.

BACKGROUND: Morphine-6-glucuronide (M6G) is a metabolite of morphine and a micro-opioid agonist. To quantify the potency and speed of onset-offset of M6G and explore putative sex dependency, the authors studied the pharmacokinetics and pharmacodynamics of M6G in volunteers using a placebo-controlled, randomized, double-blind study design. METHODS: Ten men and 10 women received 0.3 mg/kg intravenous M6G and placebo (two thirds of the dose as bolus, one third as a continuous infusion over 1 h) on separate occasions. For 7 h, pain tolerance was measured using gradually increasing transcutaneous

Joint Bayesian estimation of mutation location and age using linkage disequilibrium.

Associations between disease and marker alleles on chromosomes in populations can arise as a consequence of historical forces such as mutation, selection and genetic drift, and is referred to as "linkage disequilibrium" (LD). LD can be used to estimate the map position of a disease mutation relative to a set of linked markers, as well as to estimate other parameters of interest, such as mutation age. Parametric methods for estimating the location of a disease mutation using marker linkage disequilibrium in a sample of normal and affected individuals require a detailed knowledge of population d

Predicting secondary progression in relapsing-remitting multiple sclerosis: a Bayesian analysis.

With the aid of a Bayesian statistical model of the natural course of relapsing remitting Multiple Sclerosis (MS), we identify short-term clinical predictors of long-term evolution of the disease, with particular focus on predicting onset of secondary progressive course (failure event) on the basis of patient information available at an early stage of disease. The model specifies the full joint probability distribution for a set of variables including early indicator variables (observed during the early stage of disease), intermediate indicator variables (observed throughout the course of dise

The effect of age on the apparent clearance of felbamate: a retrospective analysis using nonlinear mixed-effects modeling.

The effects of age on felbamate apparent clearance were examined through a retrospective analysis of plasma concentration data from 700 pediatric and adult epileptic patients (age range, 2-74 years) enrolled in six clinical studies. Patients received felbamate as monotherapy or in combination with either the antiepileptic drugs (AEDs) carbamazepine (CBZ), phenytoin (PHT), or valproate (VPA). Data were analyzed using a nonlinear mixed-effects pharmacostatistical modeling technique (NONMEM). Factors in the model included age, body weight, and concomitant AEDs. Apparent clearance was highest in t

Lifespan normative modeling of brain microstructure.

Normative models of brain metrics based on large populations could be extremely valuable for detecting brain abnormalities in patients with a variety of disorders, including degenerative, psychiatric and neurodevelopmental conditions, but no such models exist for the brain's white matter (WM) microstructure. Here we present a large-scale normative model of brain WM microstructure - based on 19 international diffusion MRI datasets covering almost the entire lifespan (totaling N = 54,583 individuals; age: 4-91 years). We extracted regional diffusion tensor imaging (DTI) metrics using a standardi

Bayesian network models to assess antimicrobial resistance patterns of Streptococcus suis isolated from swine production systems in the United States between 2014-2021.

Multidrug resistance (MDR) is frequently evident in Streptococcus suis, generating distinct antimicrobial resistance (AMR) profiles, which limits the effective antimicrobial drug (AMD) options against S. suis in pigs and humans. Despite its significance, there is a lack of studies and pertinent methodologies that uncover complex interactions among AMDs and associated resistance patterns. This study aimed to identify associations between phenotypic resistance patterns of S. suis isolates from swine production systems in the United States against common AMDs using Bayesian network analysis (BNA)

Refining the Liver Donor Risk Index With Machine Perfusion: A Bayesian Approach.

INTRODUCTION: The Donor Risk Index (DRI) is a widely used liver transplant allograft risk model but does not account for the increasing adoption of machine perfusion (MP). METHODS: Using Bayesian updating, we incorporated MP into the DRI framework (DRI-MP). A Bayesian proportional hazards model with informative priors derived from the original DRI was applied to Organ Procurement and Transplantation Network data from January 2022 to June 2024. Model performance was assessed using Harrell Concordance-statistic, calibration plots, and Brier scores. RESULTS: DRI-MP, defined as DRI × 0.7 for MP

Post-marketing reporting patterns and disproportionality signals of mycophenolate mofetil: a pharmacovigilance study using the FAERS Database.

BACKGROUND: Mycophenolate mofetil (MMF) is widely used in solid-organ transplantation and autoimmune diseases. Spontaneous-reporting databases may assist in identifying adverse-event reporting patterns that warrant further clinical evaluation. This study characterized MMF-associated reports in the U.S. Food and Drug Administration Adverse Event Reporting System (FAERS). METHODS: FAERS reports submitted from the first quarter of 2004 through the fourth quarter of 2024 were analyzed. After deduplication, reports listing MMF as the primary suspect drug were included.
