Chikungunya virus at a crossroads: Emergence, interventions, and global health strategies.

Chikungunya virus (CHIKV) has re-emerged as a major arboviral threat across 104 countries, situating nearly 2.8 billion people at risk. This review summarizes evidence on CHIKV epidemiology, molecular evolution, and public health challenges, with an emphasis on the observed expansion. Climate change, international travel, and the spread of Aedes vectors are driving the transmission of CHIKV into temperate regions, exemplified by the first large outbreak in Guangdong, China, in 2025. Adaptive viral mutations, waning population immunity, and urbanization are further accelerating epidemic growth.

Magma as a carrier of non-stoichiometric oxygen linking Earth's interior and atmosphere.

Magma generation and transport govern planetary evolution. Although oxygen is the most abundant anion in Earth's silicate interior, its role in magma is conventionally ignored, with magma oxygen contents assumed to be simply fixed by cation stoichiometry. Here we show, using high-pressure and high-temperature experiments supported by molecular dynamics simulations, that silicate magma can incorporate substantially more oxygen than permitted by traditional valence-state constraints. Excess oxygen acts as a previously unrecognized volatile component, increasing in concentration from near zero at

Mechanisms of formic acid stress response in evolved yeast for microbial-electrocatalytic systems.

Integrated electrocatalytic microbial systems (iEMS) offers a sustainable route for carbon cycling. However, formic acid by-product stress severely hinders microbial conversion. Herein, we reveal the adaptive mechanisms of Saccharomyces cerevisiae under formic acid stress during ethanol cultivation through adaptive laboratory evolution and transcriptomic analysis. Formic acid exposure systematically downregulates central carbon metabolism while upregulating the acetaldehyde branch to replenish carbon skeletons. Concurrently, cell cycle genes are activated via MAPK signaling, and ribosomal biog

The pyrenoid at a glance.

Ribulose-1,5-bisphosphate carboxylase-oxygenase (Rubisco) is the central CO2-fixing enzyme of the biosphere. It is present in virtually all photosynthetic organisms, including cyanobacteria, algae and land plants. Rubisco catalyses the carboxylation step of the Calvin-Benson-Bassham (CBB) cycle that underpins the light-independent 'dark' reactions of oxygenic photosynthesis. However, Rubisco exhibits a low catalytic rate and wasteful oxygenation activity, a constraint rooted in its ancient evolutionary origin. To overcome this specificity limitation, many photosynthetic lineages have evolved C

Solvent Polarity Engineering in Low-DMF ZIF-7 Membrane Growth: Crystallization Behavior, Heterogeneous Intergrowth, and Microstructural Evolution.

Molecular transport membranes are promising alternatives to conventional cryogenic separation processes. Here, solvent polarity effects were investigated by varying the DMF/MeOH ratio during the solvothermal synthesis of supported ZIF-7 membranes. A DMF:MeOH ratio of 1:3 preserved the characteristic sodalite topology while suppressing dense-phase formation. Methanol incorporation modified heterogeneous crystallization behavior, intercrystalline organization, membrane morphology, and film densification on α-alumina supports while reducing DMF consumption by approximately 75%. These effects are

Mapping Meiotic Recombination DNA Double-Strand Breaks (DSBs) Hotspots -Methodological Advances and Challenges.

Programmed DNA double-strand breaks (DSBs) initiate meiotic recombination at discrete genomic hotspots, and their precise mapping is crucial for understanding the molecular regulation of genome stability and evolution. A variety of genome-wide methods have been developed, each leveraging different biochemical principles - from direct capture of Spo11-oligonucleotide covalent intermediates to sequencing protein-bound single-stranded DNA (ssDNA), in situ DNA end labeling, and chromatin immunoprecipitation (ChIP). Direct approaches such as Spo11-oligo mappin, CC-seq and in situ DNA end labeling (

Follicular Lymphoma Transformation is Characterized by Cytokine-associated Remodeling of Stromal and Macrophage Compartments.

Across cancer, one of the most frequent examples of histologic transformation is the evolution of follicular lymphoma (FL) to an aggressive large cell lymphoma. Despite recent progress, understanding of the molecular and cellular underpinnings of transformation remains incomplete. Here, we dissect the interplay of tumor and microenvironment cell populations across transformation through a multimodal investigation of 95 FL and transformed FL (tFL) samples, including single-cell and bulk RNA-sequencing alongside spatial transcriptomics and proteomics, and validate findings across independent FL-

Pleural Effusion Revealing Visceral Leishmaniasis: A Diagnostic Pitfall in an Immunocompetent Patient.

Pleural involvement in visceral leishmaniasis (VL) is rare and may pose a diagnostic challenge, particularly when it mimics pleural tuberculosis. We report the case of a 39-year-old immunocompetent man who presented with a dry cough and night sweats. Initial workup revealed a small lymphocytic exudative pleural effusion, and pleural biopsy showed granulomatous inflammation with a tuberculoid appearance, leading to empirical antituberculosis treatment. Because of clinical deterioration, persistent pleural effusion, and negative molecular testing for Mycobacterium tuberculosis, further investiga

Advances in Five-Membered Heterocyclic Scaffolds as Anti-Tuberculosis Agents: A Decade of Discovery.

Tuberculosis remains a deadly infectious disease with 1.09 million deaths and 10.8 million new cases reported in 2023. Escalating drug resistance and mortality rates worsen the situation. To address these issues, new TB agents with improved activity, better enzyme activation, low toxicity, and acceptable physicochemical and pharmacokinetic profiles are needed. Several reports on five-membered heterocycles with one, two, or more heteroatoms have demonstrated anti-TB properties against Mycobacterium tuberculosis. Clinical candidates such as pretomanid (nitroimidazole), linezolid (oxazolidinone),

Epstein-Barr Virus Infection at Single-Cell Resolution.

Epstein-Barr virus (EBV) infection has been studied at single-cell resolution for six decades and counting. Such investigations can reveal virus-host interactions and their dependence on viral strain, cellular niche, infection program, immune response regulation, and time. Understanding these factors is paramount to treating EBV-associated cancers and autoimmune diseases. This review examines the state of the field in EBV single-cell and spatial-omics spanning experimental models and clinical samples. Topics of primary interest include the growing adoption and emerging biological themes from s

Comparative analysis of rhesus macaque and human placental organoids highlights evolutionary differences in placentation.

Throughout evolution, the placenta has diversified in structure and cellular composition while maintaining its essential role in supporting fetal development. Trophoblasts, key cells responsible for nutrient exchange and immune modulation, are a conserved feature of all placentas. Although primate placentas share broad morphological similarities, species-specific differences in gene expression remain poorly characterized, largely due to the lack of tractable in vitro models. To address this gap, we developed rhesus macaque placental organoids representing both trophoblast and maternal-derived

Organic film evolution and recombination losses in highly efficient perovskite/organic tandem solar cells.

Perovskite/organic tandem solar cells are a promising strategy to surpass the efficiency limits of single-junction devices, yet their performance is restricted by recombination losses in the organic subcells. Here, we investigate these losses by tracking film evolution from the very initial stage of organic film formation. We strategically manipulated donor and acceptor ratios to modulate film growth characteristics, while employing in situ techniques to monitor the real-time crystallization dynamics. Our research findings underscore that the variance in donor content within the organic blend

SELEX-HTCFQ Platform: Developing DNA Enhancers of ADAR1 to Suppress ZBP1-Dependent Immunopathology.

Excessive immune activation drives pathological inflammation through dysregulated ZBP1 signaling, yet this sensor remains crucial for immune surveillance, necessitating targeted therapies that selectively inhibit pathology, while preserving protective functions. Here, we developed an innovative SELEX-HTCFQ platform that combines the Systematic Evolution of Ligands by Exponential Enrichment (SELEX) with high-throughput competitive fluorescence quenching (HTCFQ) to identify ADAR1-specific enhancers. Capitalizing on ADAR1's natural ability to suppress ZBP1 via competitive Z-nucleic acid binding,
