Herpesvirus-bacteria synergistic interaction in periodontitis.

The etiopathogenesis of severe periodontitis includes herpesvirus-bacteria coinfection. This article evaluates the pathogenicity of herpesviruses (cytomegalovirus and Epstein-Barr virus) and periodontopathic bacteria (Aggregatibacter actinomycetemcomitans and Porphyromonas gingivalis) and coinfection of these infectious agents in the initiation and progression of periodontitis. Cytomegalovirus and A. actinomycetemcomitans/P. gingivalis exercise synergistic pathogenicity in the development of localized ("aggressive") juvenile periodontitis. Cytomegalovirus and Epstein-Barr virus are associated

Interference with the host haemostatic system by schistosomes.

Schistosomes, parasitic flatworms that cause the tropical disease schistosomiasis, are still a threat. They are responsible for 200 million infections worldwide and an estimated 280,000 deaths annually in sub-Saharan Africa alone. The adult parasites reside as pairs in the mesenteric or perivesicular veins of their human host, where they can survive for up to 30 years. The parasite is a potential activator of blood coagulation according to Virchow's triad, because it is expected to alter blood flow and endothelial function, leading to hypercoagulability. In contrast, hepatosplenic schistosomia

Coxiella burnetii alters cyclic AMP-dependent protein kinase signaling during growth in macrophages.

Coxiella burnetii is the bacterial agent of human Q fever, an acute, flu-like illness that can present as chronic endocarditis in immunocompromised individuals. Following aerosol-mediated transmission, C. burnetii replicates in alveolar macrophages in a unique phagolysosome-like parasitophorous vacuole (PV) required for survival. The mechanisms of C. burnetii intracellular survival are poorly defined and a recent Q fever outbreak in the Netherlands emphasizes the need for better understanding this unique host-pathogen interaction. We recently demonstrated that inhibition of host cyclic AMP-dep

Wbm0152, an outer membrane lipoprotein of the Wolbachia endosymbiont of Brugia malayi, inhibits yeast ESCRT complex activity.

Human pathogenic filarial nematodes of the family Onchocercidae, including Brugia malayi and Onchocerca volvulus, cause debilitating filarial diseases such as lymphatic filariasis and river blindness. These arthropod-borne pathogens are obligately colonized by the Gram-negative intracellular alphaproteobacterium, Wolbachia, which is essential for nematode sexual reproduction, long-term survival, and pathogenicity in the mammalian host. Like many intracellular bacteria, Wolbachia likely uses numerous surface-exposed and secreted effector proteins to regulate its ability to persist and replicate

Autophagy in protists and their hosts: When, how and why?

Pathogenic protists are a group of organisms responsible for causing a variety of human diseases including malaria, sleeping sickness, Chagas disease, leishmaniasis, and toxoplasmosis, among others. These diseases, which affect more than one billion people globally, mainly the poorest populations, are characterized by severe chronic stages and the lack of effective antiparasitic treatment. Parasitic protists display complex life-cycles and go through different cellular transformations in order to adapt to the different hosts they live in. Autophagy, a highly conserved cellular degradation proc

Inhibitory receptor paired Ig-like receptor B is exploited by Staphylococcus aureus for virulence.

The innate immune system has developed to acquire a wide variety of pattern-recognition receptors (PRRs) to identify potential pathogens, whereas pathogens have also developed to escape host innate immune responses. ITIM-bearing receptors are attractive targets for pathogens to attenuate immune responses against them; however, the in vivo role of the inhibitory PRRs in host-bacteria interactions remains unknown. We demonstrate in this article that Staphylococcus aureus, a major Gram-positive bacteria, exploits inhibitory PRR paired Ig-like receptor (PIR)-B on macrophages to suppress ERK1/2 and

Spatial pattern formation facilitates eradication of infectious diseases.

Control of animal-born diseases is a major challenge faced by applied ecologists and public health managers. To improve cost-effectiveness, the effort required to control such pathogens needs to be predicted as accurately as possible. In this context, we reviewed the anti-rabies vaccination schemes applied around the world during the past 25 years. We contrasted predictions from classic approaches based on theoretical population ecology (which governs rabies control to date) with a newly developed individual-based model. Our spatially explicit approach allowed for the reproduction of pattern fo

Virulence blockers as alternatives to antibiotics: type III secretion inhibitors against Gram-negative bacteria.

In recent years mounting problems related to antibiotic-resistant bacteria have resulted in the prediction that we are entering the preantibiotic era. A way of preventing such a development would be to introduce novel antibacterial medicines with modes of action distinct from conventional antibiotics. Recent studies of bacterial virulence factors and toxins have resulted in increased understanding of the way in which pathogenic bacteria manipulate host cellular processes. This knowledge may now be used to develop novel antibacterial medicines that disarm pathogenic bacteria. The type III secre

A systems biology perspective on rational design of peptide vaccine against virus infections.

With the recent onset of influenza A (H1N1) pandemic, the need for improved vaccines against virus infections has become an international priority. Strategies for vaccine development have changed over time, from whole-virus to immunogenic proteins and further to antigenic viral peptides. Various algorithms and bioinformatics tools have been developed to predict immunogenic peptide regions in an antigenic protein sequence. Recent advances in next-generation sequencing technologies, as represented by real time DNA sequencing, provide increased throughput and yield of data on viral pathogens and

Quantitative analysis of VirB8-VirB9-VirB10 interactions provides a dynamic model of type IV secretion system core complex assembly.

Type IV secretion systems are multiprotein complexes that translocate macromolecules across the bacterial cell envelope. The type IV secretion system in Brucella species encodes 12 VirB proteins that permit this pathogen to translocate effectors into mammalian cells, where they contribute to its survival inside the host. The "core" complex proteins are conserved in all type IV secretion systems, and they are believed to form the channel for substrate translocation. We have investigated the in vitro interactions between the soluble periplasmic domains of three of these VirB components, VirB8, V

Stochastic SIS Modelling: Coinfection of Two Pathogens in Two-Host Communities.

A pathogen can infect multiple hosts. For example, zoonotic diseases like rabies often colonize both humans and animals. Meanwhile, a single host can sometimes be infected with many pathogens, such as malaria and meningitis. Therefore, we studied two susceptible classes S 1 ( t ) and S 2 ( t ) , each of which can be infected when interacting with two different infectious groups I 1 ( t ) and I 2 ( t ) . The stochastic models were formulated through the continuous time Markov chain (CTMC) along with their deterministic analogues. The statistics for the developed model were studied using the mul

Leptospirosis: aspects of innate immunity, immunopathogenesis and immune evasion from the complement system.

Leptospirosis is a neglected infectious disease caused by spirochetes from the genus Leptospira. It constitutes a major public health problem in developing countries, with outcomes ranging from subclinical infections to fatal pulmonary haemorrhage and Weil's syndrome. To successfully establish an infection, leptospires bind to extracellular matrix compounds and host cells. The interaction of leptospires with pathogen recognition receptors is a fundamental issue in leptospiral immunity as well as in immunophatology. Pathogenic but not saprophytic leptospires are able to evade the host complemen

Exposure of bovine cornea to different strains of Moraxella bovis and to other bacterial species in vitro.

A collection of strains of Moraxella bovis, some pathogenic and some non-pathogenic in cattle, together with other M. bovis preparations, Neisseria ovis, Staphylococcus aureus and Moraxella non-liquefaciens were studied by scanning electron microscopy for their affinity to bovine corneal preparations in vitro. The in vitro procedure provides a convenient method for studies on host-pathogen interactions at the early stage of pathogenesis. The results corresponded well with the pathogenicity of the respective strains and species in cattle. It is considered that the pathogenicity of M. bovis is a…

Triptolide clears Staphylococcus aureus infection by targeting XIAP to induce host apoptosis while maintaining gut microbiota homeostasis.

BACKGROUND: Staphylococcus aureus (SA) remains a global health threat due to its increasing drug resistance and intracellular persistence, which compromise the conventional antibiotic efficacy. Host-directed therapy (HDT) has emerged as a promising alternative by modulating host immunity. With multi-targeting and immunomodulatory properties, traditional Chinese medicine (TCM) monomers represent ideal candidates for HDT. However, their ability to promote host immunity-mediated SA clearance remains largely unexplored. METHODS: Forty-one TCM monomers potentially regulating host apoptosis, a core

Characterization of outer membrane vesicles from a neonatal meningitic strain of Cronobacter sakazakii.

Cronobacter sakazakii is associated with severe and often fatal cases of infant meningitis and necrotizing enterocolitis. The form of meningitis differs from that due to Neisseria meningitidis and Streptococcus spp., in that it is highly invasive and destructive towards human brain cells. However, there is relatively little understanding of the cytopathogenic interaction of C. sakazakii with host cells which results in stimulation of an inflammatory immune response. The production of Cronobacter outer membrane vesicles (OMV) and their potential pathogenic functions have not yet been elucidated

Interactions between bacterial microbiota and protozoan parasites: an intimate relationship.

Interactions between parasites and the gut microbiota play a key role in immune responses and susceptibility to zoonotic diseases; however, many aspects of how protozoan infections alter microbial diversity and how these changes influence parasite pathogenicity and host defense remain poorly understood. This review integrates recenst studies using murine models of the Muroidea superfamily, highlighting patterns in microbial diversity, functional interactions, and potential immunometabolic mechanisms underlying these relationships. Both consistent findings and discrepancies among studies are id

An Antiviral Role for TRIM14 in Ebola Virus Infection.

Ebola virus (EBOV) is a highly pathogenic virus that encodes 7 multifunctional structural proteins. Multiple host factors have been reported to interact with the EBOV proteins. Here, we found that tripartite motif-containing 14 (TRIM14), an interferon-stimulated gene that mediates cellular signaling pathways associated with type I interferon and inflammatory cytokine production, interacts with EBOV nucleoprotein to enhance interferon-β (IFN-β) and nuclear factor-κB (NF-κB) promotor activation. Moreover, TRIM14 overexpression reduced viral replication in an infectious but biologically contained

Biosynthesized selenium nanoparticles increase soybean resistance to root rot by recruiting beneficial microbes and reprogramming host metabolism.

BACKGROUND: Soybean root rot, primarily caused by Fusarium species, is a devastating soil-borne disease that severely threatens global soybean production. Conventional chemical controls are problematic due to their environmental impact and the risk of fostering pathogen resistance. There is therefore an urgent need to develop sustainable and eco-friendly alternative strategies. RESULTS: This study demonstrated that selenium nanoparticles (SeNPs) biosynthesized by Bacillus subtilis ZY56 significantly increased soybean resistance to F. oxysporum. Characterization confirmed that the obtained SeN

Biophysical Elucidation of the Distinct Roles the Nipah Virus Attachment and Fusion Glycoproteins Play in the Entry Process.

Nipah virus (NiV), a World Health Organization priority pathogen, is a lethal enveloped RNA virus within the Paramyxoviridae family. NiV attachment G and fusion F glycoproteins execute viral-cell or cell-cell membrane fusion during viral entry and syncytia formation, respectively. Despite past studies, several aspects of the molecular-level orchestration underlying this process, such as how F, G, and their interactions regulate fusion, remain poorly understood, obscured by challenges in decoupling fusion determinants in complex cellular environments. Here, we report a reconstitution-based appr
