Association of Milk De Novo Fatty Acid Proportion of the First Week Postpartum With Productivity and Plasma IGF-1 and GLP-1 Concentrations in Dairy Cows.

Immediate postpartum analysis of milk fatty acid (FA) profiles using Fourier transform infra-red spectrometry (FTIR) may be a non-invasive and simple analytical tool for predicting milk production, health problems and reproductive performance. This study evaluated the relationship between milk de novo fatty acid (FA) proportions at the 1st week postpartum and the pre-and postcalving blood metabolic markers, milk yield, health and reproduction performance after calving. Sixty-five Holstein cows were assessed, and blood samples were collected 3 and 1 weeks before the predicted calving date, on t…

Research on the mechanism of cow milk protein dietary intervention in ameliorating systemic chronic inflammation in type 2 diabetes by disrupting the ROS-M1 macrophage axis.

INTRODUCTION: Recent studies suggest that type 2 diabetes mellitus (T2DM) is characterized by a systemic, low-grade chronic inflammatory state. Although cow milk protein (CMP) has been shown to alleviate this inflammation, its underlying mechanisms remain unclear. METHODS: Therefore, we investigated how CMP mitigates systemic chronic inflammation in T2DM using both in vitro digestion and mouse models. RESULTS: The in vitro digestion model demonstrated that CMP, with its low degree of hydrolysis, exhibits significant anti-α-amylase and antioxidant activities. In the in vivo study, CMP markedl…

Effect of alterations in closeup dietary neutral detergent fiber content on colostrum, transition, and mature milk production and composition in primiparous and multiparous Holstein dairy cattle.

There is limited research investigating the impact of closeup nutrition on colostrum and transition milk (TM) production and composition in both primiparous (PP) and multiparous (MP) cattle, despite the potential of alterations in prepartum metabolism to modify the availability of substrates for colostrum and milk synthesis. The objective of this study was to determine the effect of altering closeup dietary NDF content on colostrum, transition and mature milk composition and to characterize potential associations between cow and metabolic factors and colostrum yield and composition. Primiparou…

Feeding Ascophyllum nodosum meal or ethylenediamine dihydroiodide to transition dairy cows: Effects on milk yield and plasma concentrations of thyroid hormones and markers of metabolic status.

Ascophyllum nodosum (ASCO) is a brown seaweed rich in I and bioactive compounds. Previous research demonstrated that feeding incremental amounts of ASCO meal linearly decreased the plasma concentrations of nonesterified fatty acids (NEFA) and cortisol in lactating dairy cows, suggesting that ASCO may improve cows' metabolic status during the transition period. We aimed to investigate the effects of supplementing transition dairy cows with ASCO meal or ethylenediamine dihydroiodide (EDDI) on plasma concentrations of thyroid hormones and markers of metabolic status, DMI, and milk yield and compo…

Feeding biochar to high-yielding dairy cows: effects on feed intake, milk performance, feed efficiency, blood variables and on rumen fermentation in vitro and in vivo.

Biochar, a carbon-rich product from the pyrolysis of biomass, has the potential to enhance feed efficiency and mitigate the environmental impact of milk production by optimising rumen fermentation. However, its effect on dairy cow performance during peak production remains uncertain. We aimed to assess the potential of biochar to improve feed conversion efficiency and reduce ruminal methane production in high-yielding dairy cows. Additionally, through a combination of in vitro and in vivo experiments, we sought to distinguish between intra- and extra-ruminal effects of biochar. In the in vitro…

Impacts of dietary arginine supplementation on performance, nutrient digestion and expression of proteins related to milk fatty acid and casein synthesis in early lactating dairy cows.

Arginine (Arg) can potentially increase milk yields in early lactating dairy cows by stimulating the development of mammary gland and the biosynthesis of milk proteins and fatty acids (FA); however, the underlying mechanism is still unclear. The hypothesis that rumen-protected Arg addition would increase milk, milk fat, and milk protein yields was tested, aiming to dlucidate the possible mechanism. Forty-eight multiparous Chinese Holstein dairy cows (milk yield 34.9 ± 1.27 kg/d, body weight 658 ± 12.7 kg, and days in lactation [DIL] 16.4 ± 2.56) were selected on the principle of proximity and …

Validation and application of an online extraction and liquid chromatography tandem mass spectrometry assay for the analysis of delamanid and its DM-6705 metabolite in human breast milk.

We developed and validated a bioanalytical assay to quantify delamanid and its key metabolite (DM-6705) in breast milk and aimed to quantify the secretion of these compounds in breast milk. Due to the hydrophobic nature of the analytes, special care was taken during sample preparation to prevent the formation of fatty deposits during protein precipitation. This was followed by online solid phase extraction and liquid chromatography with tandem mass spectrometry for detection. A Restek Viva BiPh C18 column (1.0 mm×50 mm, 5 µm) was used for extraction while chromatographic separation was perform…

Ambient 1,2-propanediol exposure accelerates the degradation of lipids and amino acids in milk via allosteric effects and affects the utilization of nutrients containing amide bond.

The scandal of detecting 1, 2-propanediol (PL) in milk brought a crisis to the trust of consumers in the dairy industry, and the potential toxicity of PL has aroused the public concern about dietary exposure. A total of 200 pasteurized milk samples were collected from 15 regions, and the quantity of PL ranged between 0 and 0.31 g kg-1. Pseudo-targeted quantitative metabolomics integrated with proteomics demonstrated that PL enhanced the reduction of κ-casein, β-casein, and 107 substances (41 amines and 66 amides) containing amide bonds. Pathway enrichment and topological analysis indicated tha…

Alterations of milk oligosaccharides in mothers with gestational diabetes mellitus impede colonization of beneficial bacteria and development of RORγt+ Treg cell-mediated immune tolerance in neonates.

Gestational diabetes mellitus (GDM) is an increasing public health concern that significantly increases the risk of early childhood allergic diseases. Altered maternal milk glycobiome may strongly affect gut microbiota and enteric-specific Treg cell-mediated development of immune tolerance in GDM infants. In this study, we found that, compared with healthy Chinese mothers, mothers with GDM had significantly lower levels of total and specific human milk oligosaccharides (HMOs) in their colostrum that subsequently increased with extension of lactation. This alteration in HMO profiles significant…
