Effects of thymosin and evidence of monocyte suppression of both T- and B-cell functions in two cases of 'common variable immunodeficiency'.

We have studied two patients with common variable immunodeficiency (CVID), impaired cell-mediated immunity, and high percentages of monocytes in their peripheral blood. Removal of monocytes from cultures of peripheral blood mononuclear cells from both patients increased the in vitro responses to phytohaemagglutinin (PHA) and concanavalin A (Con A) but not to purified protein derivative (PPD), as measured by [3H]thymidine uptake. Similarly, supernatants of monocyte cultures from both patients, unlike supernatants of normal monocytes, suppressed the in vitro responses to PHA and Con A but enhanc

Induction of marginal zinc deficiency in female rhesus monkeys.

Although severe zinc deficiency has been previously induced in rhesus monkeys, marginal zinc deprivation, a state more analogous to that found in human populations has not been characterized. To address this issue, zinc deficiency was induced in mature female rhesus monkeys by restriction of dietary zinc in a semipurified diet based on sprayed egg white as the protein source. Groups of monkeys were fed dietary levels of 0.5, 2, 4, 8, 12, 20, or 100 ppm zinc. Plasma zinc concentration fell from an initial level of 92-148 μg/100 ml to levels between 40 and 90 μg/100 ml in monkeys fed 8 ppm zinc

Staphylococcal pyrogenic exotoxin type C: further characterization.

Staphylococcal pyrogenic exotoxin type C was purified by differential precipitation with ethanol and resolubilization in water followed by thin-layer isoelectric focusing. The purified toxin migrated as a homogeneous protein when re-electrofocused in polyacrylamide (isoelectric point, 7.2), when subjected to sodium dodecyl sulfate polyacrylamide gel electrophoresis (molecular weight, 22,000 daltons), and when reacted against hyperimmune antisera in an Ouchterlony immunodiffusion plate. A 2200-fold purification of the toxin could be obtained. Pyrogenic exotoxin C was elaborated by all of 44 Sta

Cell mediated immunity in idiopathic glomerulonephritis.

Various recent reports have suggested the presence of a functional defect of lymphocyte subpopulations in minimal-change nephropathy during the active phase. A probable role of inhibitory humoral factor(s) has been hypothesized. However, other authors have been unable to detect a significant difference between plasma from patients with nephrotic syndrome due to minimal-change nephropathy and plasma from other glomerulonephritis in the degree of inhibition of mitogen-induced lymphocyte transformation. In our study, T cell function, as measured by the response to PHA in autologous plasma, was de

An in vitro assessment of cellular and humoral immune function in pulmonary tuberculosis: correction of defective neutrophil motility by ascorbate, levamisole, metoprolol and propranolol.

Fifty-six tuberculosis patients and twenty-eight control subjects were evaluated in a comprehensive investigation of cellular and humoral immune function in pulmonary TB. The patient group showed significantly higher levels of secretory IgA and serum IgG, IgA and IgM than did the control group but 7% of patients displayed a selective secretory IgA deficiency. Levels of alpha-1-antitrypsin were also significantly higher in the patient group. There were no significant differences in levels of total haemolytic complement, C'3 and C'4. In moderate to moderately advanced TB patients there were no s

Concanavalin A-mediated in vitro activation of a secondary cytotoxic T-cell response in virus-primed splenocytes.

In a recent report it was shown that what appeared to be secondary cytotoxic T cells could be obtained from lymphocytic choriomeningitis virus (LCMV)-primed splenocytes after stimulation in vitro with the non-specific T cell mitogen concanavalin A (Con A). The present experiments attempt to characterize further these effector cells and, in particular, to establish whether the Con A-activated cytotoxic effectors are qualitatively different from the secondary cytotoxic T cells induced by restimulation with the homologous antigen. It was found that: (1) in vitro activation with Con A could be obt

Revisiting Culture Conditions of Satellite Cells In Vitro for Skeletal Muscle Regeneration and Therapeutic Applications.

Skeletal muscle fibers are largely post-mitotic in adulthood yet retain robust regenerative capacity through satellite cells (SCs), quiescent muscle stem cells located beneath the basal lamina that are activated after injury, proliferate as myoblasts, differentiate, fuse into myotubes, and mature via tightly regulated cues. Given the burden of muscle-wasting and neuromuscular diseases, including Duchenne muscular dystrophy (DMD) (driven by dystrophin defects and impaired SC function), amyotrophic lateral sclerosis (ALS) (with progressive atrophy and evidence of SC perturbation), and myasthenia

Scutellariae Radix and Coptidis Rhizoma Improve Glucose and Lipid Metabolism in T2DM Rats via Regulation of the Metabolic Profiling and MAPK/PI3K/Akt Signaling Pathway.

Aim Scutellariae Radix (SR) and Coptidis Rhizoma (CR) have often been combined to cure type 2 diabetes mellitus (T2DM) in the clinical practice for over thousands of years, but their compatibility mechanism is not clear. Mitogen-activated protein kinase (MAPK) signaling pathway has been suggested to play a critical role during the process of inflammation, insulin resistance, and T2DM. This study was designed to investigate their compatibility effects on T2DM rats and explore the underlying mechanisms by analyzing the metabolic profiling and MAPK/PI3K/Akt signaling pathway. Methods The compatib

Targeting JNK3: An In-silico Approach to Uncover Potential Therapeutics for Alzheimer's Disease.

INTRODUCTION: JNK3 is a specific isoform of c-Jun N-terminal kinase, mainly found in the brain, and is highly sensitive to stress-associated signals in the central nervous system. It has been reported that JNK3 plays a crucial role in neurite formation and cognition. During pathological states such as Alzheimer's disease, cerebral ischemia, Traumatic brain injury (TBI), Parkinson's disease, and epilepsy, it is found to be in a hyperactivated form. Hyperphosphorylation of amyloid precursor protein (APP) and tau leads to toxic Aβ42 and neurofibrillary tangles. Excess Aβ activates JNK3 signaling,

Beyond the scaffold: extracellular matrix uptake in breast cancer.

Breast cancer is associated with a highly fibrotic tumour microenvironment, where cancer-associated fibroblasts (CAFs) secrete excessive amounts of extracellular matrix (ECM). Fibrosis and collagen deposition correlate with poor patient survival, indicating that the ECM plays a role in promoting tumorigenesis. The ECM is constantly remodelled through extracellular and intracellular degradation pathways. While protease-dependent extracellular ECM degradation has been well studied, the intracellular degradation pathway is less understood. Here, I will describe the evidence supporting a role for

The sugar transporter AsSTL is regulated by the kinase Hog1 and is involved in glycerol transport and the response to osmotic stress in the salt-tolerant ascomycete aspergillus sydowii H-1.

Sugar transporters (STs) are critical biological macromolecules that involved in the regulation of fungal development and responses to abiotic stresses. While monosaccharide- and sucrose-specific transporters have been extensively characterized in yeast and plants, knowledge of STs in filamentous fungi remains limited. Here, through genome mining, we identified 173 STs in the salt-tolerant fungus Aspergillus sydowii H-1 and classified them into nine subgroups. Notably, 37 of these STs showed active responses to high-salt stress, with the glycerol transporter AsSTL exhibiting particularly stron

Polyethylene terephthalate (PET) microplastics induced male reproductive toxicity in mice by activating the p38-MAPK pathway.

Microplastics are widespread environmental pollutants that can easily enter the bodies of living organisms through various means, arousing extensive concern. However, research regarding the influence of polyethylene terephthalate microplastics (PET-MPs) on the mammalian reproductive system is limited. This study explored the mechanisms by which PET-MPs induce reproductive toxicity in male mice. The results showed that after 90 days of continuous gavage exposure, PET-MPs with sizes of 50, 200, and 1000 nm accumulated in the testicular tissue. The results of the experiment showed the sperm quali

Targeting the MAPK Pathway in Cancer.

The mitogen-activated protein kinase (MAPK) signaling cascade is fundamental in regulating cellular proliferation and differentiation, cell survival and cell death via apoptosis. Disruption of the MAPK signaling cascade at any point can lead to the evasion of apoptosis and unchecked cell growth and proliferation, leading to oncogenesis. This narrative review describes MAPK pathway dysregulation, its therapeutic targets, and resistance mechanisms. The therapeutic targeting of the MAPK pathway is complex due to the dual context-dependent roles of several kinases in the signaling cascade. Despite

EMP1-driven ferroptosis in liver sinusoidal endothelial cells exacerbates hepatic ischemia-reperfusion injury via P38 MAPK.

Ischemia-reperfusion injury (IRI) remains the leading cause of early graft failure following liver transplantation (LT). Traditional paradigms have centered on hepatocyte death, yet the decisive contribution of liver sinusoidal endothelial cells (LSECs) as the first vascular barrier has long been overlooked. Here, single-cell RNA sequencing of human livers comparing normal controls with those exposed to transplantation-induced IRI, identified EMP1 as the most upregulated gene in LSECs. In LSECs and BRL3A hepatocytes subjected to hypoxia/reoxygenation (H/R), EMP1 silencing with an siRNA, ferrop

The metastasis-associated protein MTA3 promotes cardiac repair by inhibiting the fibroblast to myofibroblast transition during fibrosis.

Cardiac fibrosis is a pathological hallmark of various cardiovascular disorders. Accumulating evidence has demonstrated that fibroblasts transform into myofibroblasts during the occurrence of cardiac fibrosis, but the mechanism remains incompletely understood. This study aims to investigate the relevance of MTA3 as a potential therapeutic target for cardiac fibrosis. The myocardial infarction model was established by ligating the left coronary artery of C57BL6 mice, and myocardial fibrosis was measured by cardiac ultrasound and Sirius red staining of myocardium. MTA3 overexpression plasmid was

Targeting the RAS/MAPK pathway in children with glioma.

PURPOSE: Pediatric gliomas are the most common brain tumor in children, encompassing both low-grade glioma (pLGG) and high-grade glioma (pHGG). Alterations in the RAS/MAPK pathway are the driver event in the majority of pLGG and account for a subset of pHGG. Identification of these alterations has resulted in the transition to targeted therapy as a treatment option. RESULTS: In pLGG, multiple trials have demonstrated superior outcomes using targeted therapy compared to traditional chemotherapy regimens. This has transformed care for these patients over the past decade with targeted therapy mo

Protective effects of functional Nano-Selenium supplementation on spleen injury through regulation of p38 MAPK and NF-κB protein expression.

Selenium (Se) is a trace element necessary for humans to maintain normal physiological activities, and Se deficiency may lead to splenic injury, while Se supplementation can alleviate splenic injury. However, the mechanism is unclear. In this study, we constructed a Se deficiency animal model by feeding Sprague-Dawley (SD) rats with low Se feed. Meanwhile, we observed the repairing effect of Se supplementation on splenic injury with two doses of novel nano-selenium (Nano-Se) supplement by gavage. We measured the Se content in the spleens of the rats by atomic fluorescence spectroscopy (AFS) me

A three-arm randomised phase II study of the MEK inhibitor selumetinib alone or in combination with paclitaxel in metastatic uveal melanoma.

AIMS: The MAPK pathway is constitutively activated in uveal melanoma (UM). Selumetinib (AZD6244, ARRY-142886), a MEK inhibitor, has shown limited activity as monotherapy in metastatic UM. Pre-clinical studies support synergistic cytotoxic activity for MEK inhibitors combined with taxanes, and here we sought to assess the clinical efficacy of combining selumetinib and paclitaxel. PATIENTS AND METHODS: Seventy-seven patients with metastatic UM who had not received prior chemotherapy were randomised to selumetinib alone, or combined with paclitaxel with or without interruption in selumetinib two
