Placental expression of estrogen and progesterone receptors and vascular endothelial growth factor in buffaloes suffering from uterine torsion.

BACKGROUND: Although several risk factors have been suggested for uterine torsion, the pathogenesis is still unclear. Therefore, the current study aims to investigate the pathogenesis of uterine torsion by assessing the histological, histochemical, and immunohistochemical changes that occur in the placenta obtained from uterine torsion cases. Immunohistochemical changes include investigation of the expression of estrogen receptors (ERs), progesterone receptors (PRs), and the vascular endothelial growth factor (VEGF) in the placental tissue. METHODS: Forty intrapartum dairy cows were included

Advanced multi-modal mass spectrometry imaging reveals functional differences of placental villous compartments at microscale resolution.

The placenta is a complex and heterogeneous organ that links the mother and fetus, playing a crucial role in nourishing and protecting the fetus throughout pregnancy. Integrative spatial multi-omics approaches can provide a systems-level understanding of molecular changes underlying the mechanisms leading to the histological variations of the placenta during healthy pregnancy and pregnancy complications. Herein, we advance our metabolome-informed proteome imaging (MIPI) workflow to include lipidomic imaging, while also expanding the molecular coverage of metabolomic imaging by incorporating on

Subchronic Exposure to Polystyrene Nanoplastics Disrupts Placental Development and Calcium Homeostasis: Insights from In Vivo and In Vitro Models.

Nanoplastics have recently emerged as persistent pollutants of global concern that pose substantial risks to human health. However, the long-term adverse effects of nanoplastics on the female reproductive system remain unclear. Polystyrene nanoplastics (PS-NPs; 50 nm diameter) were selected as representative nanosized plastic particles to investigate the potential effects of subchronic prenatal and gestational exposure via drinking water on placental development in ICR (CD-1) mice. Maternal exposure to 10 mg/L PS-NPs induced an increase in fetal resorption rate and significantly increased feta

Prenatal exposure to environmentally relevant low-dosage dibutyl phthalate reduces placental efficiency in CD-1 mice.

Dibutyl phthalate (DBP), a phthalate congener, is widely utilized in consumer products and medication coatings. Women of reproductive age have a significant burden of DBP exposure. Prenatal DBP exposure is associated with adverse pregnancy/fetal outcomes in the offspring. However, the role of fetal sex and the general mechanisms underlying DBP exposure-associated adverse pregnancy outcomes are unclear. We hypothesize that prenatal DBP exposure at an environmentally relevant low dosage adversely affects fetal-placental development and function during pregnancy in a fetal sex-specific manner. Ad

Placental DNA methylation key topics: sex- and cell-type specificity, mediation, multi-omics, and biomarker discovery.

The placenta is a dynamic organ that serves numerous purposes for fostering a successful pregnancy and the delivery of a healthy infant in humans. It performs critical functions in nutrient and oxygen transport, immune modulation, and hormonal regulation. DNA methylation, a key epigenetic mechanism of transcriptional regulation, plays a key role in the underlying etiologies of placenta-related health complications. Therefore, assessing placental DNA methylation is essential for understanding how adverse prenatal exposures may impact both short-term and long-term health outcomes in women and ch

Dynamic TyG-inflammation-endothelial trajectories and placental histopathology synergistically define a maternal-fetal vascular risk phenotype.

BACKGROUND: Maternal dysmetabolism and low-grade inflammation accelerate fetal aortic wall thickening. Whether concomitant placental vascular or inflammatory lesions amplify this risk is unknown. Participants, Materials and Methods: 88 women with singleton, anatomically normal pregnancies were phenotyped at 24-26 weeks (visit 1, V1) and 32-34 weeks (visit 2, V2). Dynamic maternal markers [(Δ triglyceride-glucose (TyG) index, Δ interleukin-6 (IL-6), Δ high-sensitivity C-reactive protein (hsCRP), Δ flow-mediated dilation (FMD)] again generated three metabolic inflammatory response phenotypes (MI

Serum placental growth factor: Pre-analytical stability, comparison of blood derivates and interference from biotin and hemolysis.

INTRODUCTION: Placental growth factor (PlGF) is an increasingly applied biomarker in the early prediction of preeclampsia. To use PlGF clinically, preanalytical factors and biotin/hemolysis susceptibility must be validated, this forms the aim of this study. MATERIAL AND METHODS: We examined the stability of PlGF including precision, delayed centrifugation, comparability between serum and EDTA-plasma, pneumatic tube transportation, storage at room temperature, 5 °C, -80 °C and undergoing freeze-thaw cycles and further, exposure to biotin and hemolysis. Blood was obtained from pregnant women wi

Prenatal fructose exposure independently impacts placental phenotype and female offspring kidney function and liver composition in rats.

Archaeological and anthropological evidence suggests that human ancestors' diets were rich in fiber, potassium, and complex carbohydrates, while low in sodium, refined sugars, and energy density. Over time, agroindustrialization led to diets poorer in fiber and micronutrients but higher in sodium, simple sugars, and calorie-dense foods. This shift contributed to the rise of noncommunicable diseases (NCDs) such as obesity, type 2 diabetes, and cardiovascular diseases, which now account for 70% of global premature deaths. Maternal nutrition affects fetal development and long-term health. High su

Maternal α-cypermethrin and permethrin exert differential effects on fetal growth, placental morphology, and fetal neurodevelopment in mice.

Pyrethroid insecticides represent a broad class of chemicals used widely in agriculture and household applications. Human studies show mixed effects of maternal pyrethroid exposure on fetal growth and neurodevelopment. Assessment of shared pyrethroid metabolites as a biomarker for exposure obscures effects of specific chemicals within this broader class. To better characterize pyrethroid effects on fetal development, we investigated maternal exposure to permethrin, a type I pyrethroid, and α-cypermethrin, a type II pyrethroid, on fetal development in mice. Pregnant CD1 mice were exposed to per

Casein Kinases 2-dependent phosphorylation of the placental ligand VAR2CSA regulates Plasmodium falciparum-infected erythrocytes cytoadhesion.

Placental malaria is characterized by the massive accumulation and sequestration of infected erythrocytes in the placental intervillous blood spaces, causing severe birth outcomes. The variant surface antigen VAR2CSA is associated with Plasmodium falciparum sequestration in the placenta via its capacity to adhere to chondroitin sulfate A. We have previously shown that the extracellular region of VAR2CSA is phosphorylated on several residues and that the phosphorylation enhances the adhesive properties of CSA-binding infected erythrocytes. Here, we aimed to identify the kinases mediating this p

Bisphenol A alters JUN promoter methylation, impairing steroid metabolism in placental cells and linking to sub-representative phenotypes.

Bisphenol A (BPA) is a widely used industrial compound commonly found in various everyday plastic products. Known for its endocrine-disrupting properties, BPA can enter the human body through multiple pathways. Prenatal exposure to BPA not only disrupts placental structure and function but also interferes with normal steroid metabolism. This study investigates the epigenetic regulatory mechanisms by which BPA influences steroid metabolism in the placenta. Using BPA-treated JEG3 cells, we analyzed hormone levels, gene promoter DNA methylation, and gene expression, further validating our finding

Decreased number of myeloid-derived suppressor cells in the placental trophoblast of gestational diabetes mellitus. Possible role of leptin.

Gestational diabetes mellitus (GDM) is the most common complication of pregnancy and significantly increases both maternal and fetal morbidity and mortality. Inflammation is a hallmark of GDM, and placental inflammation may play a key role in the pathophysiology of the disease. Myeloid-derived suppressor cells (MDSCs), which are innate immunosuppressive, are thought to contribute to feto-maternal tolerance. In normal pregnancies, elevated levels of MDSCs have been observed in both peripheral and umbilical cord blood. Our hypothesis postulates that trophoblasts from placentas belonging to women

Placental Igf1 overexpression sex-specifically impacts mouse placenta structure, altering offspring striatal development and behavior.

Insulin-like growth factor 1 (IGF1) is primarily produced in the placenta and is essential for neurodevelopment. Specifically, how placental IGF1 production persistently influences the brain is unclear, but with rates of complicated pregnancies on the rise, understanding placental contributions to child outcomes is paramount. We hypothesize that placental Igf1 expression alters fetal neurodevelopment relevant to neurodevelopmental disorders. Therefore, this study evaluated the effects of placental Igf1 overexpression on embryonic and postnatal brain development, particularly for striatum, a re

Altered Production and Cellular Levels of Hydrogen Sulfide (H2S) in Placental Trophoblasts from Pregnancies Affected by Pre-Eclampsia.

BACKGROUND/OBJECTIVES: Hydrogen sulfide (H2S) is a vasorelaxant gas and exerts anti-oxidative, anti-inflammatory, and cytoprotective effects. H2S has been implicated in regulating placental vaso-activity and angiogenesis. It is believed that abnormal trophoblast production of vasodilators and angiogenic factors contributes to pre-eclampsia development. However, little is known about whether aberrant H2S production is present in placental trophoblasts from pre-eclamptic pregnancies. METHODS: Trophoblasts were isolated from normal and pre-eclamptic placentas. After incubation, cell production o

Elucidating the role of ABC transporters in the placental efflux of (-)- Δ9-tetrahydrocannabinol (THC) using a cocktail of ABC transport inhibitors.

We previously showed that THC is effluxed in the perfused human placenta cotyledons, but surprisingly this efflux was not inhibited by valspodar, a P-gp and BCRP inhibitor. P-gp and BCRP have multiple binding sites, and therefore THC may be binding to a transport site not blocked by valspodar. To test this hypothesis, we perfused human placenta cotyledons with THC in the absence and presence of a cocktail of P-gp and BCRP inhibitors. The inhibitor cocktail significantly increased the unbound maternal-to-fetal THC clearance index, indicating that P-gp and/or BCRP are likely involved in determin
