Administration of transforming growth factor-alpha enhances anatomical and behavioral recovery following olfactory nerve transection.

Although replacement of olfactory receptor neurons (ORNs) and subsequent reinnervation of the olfactory bulb occur following ORN injury, the intrinsic and extrinsic factors that contribute to the regulation of this dynamic process have not yet been fully identified. Recent research indicates that several growth factors have neurogenic effects on ORNs in vitro, and that chronic in vivo administration of either basic fibroblast growth factor, epidermal growth factor, or transforming growth factor-alpha (TGF-alpha) following chemical lesion can enhance the normal rate of ORN reinnervation of the

Caregiver Perspectives on Challenges of Caring for Individuals with Intellectual Disability: A Qualitative Study.

This study examined the challenges encountered by caregivers of individuals with intellectual disability in a residential care home and their suggested solutions. A basic qualitative design within the interpretive paradigm was employed, using purposive sampling to recruit 21 caregivers. Thematic analysis revealed three overarching themes: disability-related challenges, care home-related challenges, and strategies to improve quality of life. Disability-related challenges involved behavioral issues, cognitive impairments, and difficulties in managing chronic conditions. Care home-related challen

Exploring thiazole conjugates as cholinesterase inhibitors for Alzheimer's disease treatment.

Alzheimer's Disease (AD), a major global health concern, is marked by memory loss and cognitive decline, especially in the elderly. Cholinergic hypothesis is one of the leading hypotheses toward the treatment of AD, prompting the development of cholinesterase (ChE) inhibitors. Acetylcholinesterase (AChE) remains a vital therapeutic target due to its role in enhancing acetylcholine levels and neural function. The natural products, synthetic analogs, and hybrid molecules have been evaluated as inhibitors of ChEs. Recent studies highlight thiazole-fused heterocyclic scaffolds and their derivative

Andrographolide attenuates microglial senescence in Alzheimer's disease mice by suppressing the STAT3 signaling.

Andrographolide (AP), a diterpenoid extracted from Andrographis paniculata, has emerged as a promising treatment for Alzheimer's disease (AD) in preclinical studies, but the underlying mechanisms remain incompletely defined. Here, we demonstrated that AP treatment improved cognition performance and reduced amyloid-β (Aβ) plaque accumulation in 5×FAD transgenic mice of both sexes, by mitigating microglial senescence. Proteomic analysis revealed that AP markedly decreased cholesterol content in the cerebral cortex. Using an in vitro low-density lipoprotein-induced senescence model, we found that

Managing Opioid Tolerance: Balancing Pain Relief and Tackling Addiction.

This editorial discusses opioid tolerance, a growing challenge in managing pain in chronic patients. With time, overuse of opioids causes less sensitivity and responsiveness of opioid receptors and leads to higher doses of opioids for the same level of pain management. Due to this, it increases the risk of side effects, such as opioid addiction, respiratory issues, and overdose. The editorial highlights the relationship between opioid use disorder (OUD) and opioid tolerance and discusses the difficulty faced by patients and doctors during opioid tolerance. To address these challenges, a multim

Top Ten Tips Palliative Care Clinicians Should Know About Caring for People with Postintensive Care Syndrome.

Intensive care unit (ICU) mortality rates have substantially declined over the past few decades. Accordingly, there has been an increase in the number of ICU survivors, who are often burdened by long-term sequelae and high morbidity following their discharge. The term postintensive care syndrome (PICS) was first coined in 2012 to describe this constellation of physical, psychological, and cognitive sequelae, which may persist long after acute care hospitalization and may also affect family members. In this context, the timely integration and delivery of palliative care has the potential to all

Development of Antisense Gapmers for the Treatment of Huntington's Disease.

The field of neuromuscular and neurodegenerative diseases has been revolutionized by the advent of genetics and molecular biology to evaluate the pathogenicity, thereby providing considerable insight to develop suitable therapies. With the successful translation of antisense oligonucleotides (AOs) from in vitro into animal models and clinical practice, modifications are being continuously made to the AOs to enhance their pharmacokinetics and pharmacodynamics. In order to activate RNase H1-mediated cleavage of the target mRNA, as well as to increase the binding affinity and specificity, gapmer

Sensory encoding and memory retrieval are coordinated with propagating waves in the human brain.

Complex behavior entails a balance between taking in sensory information from the environment and utilizing previously learned internal information. Studies on mice show that the brain continually alternates between outward and inward cognitive modes every few seconds, accompanied by stereotyped cascades of neuronal spiking. Our analysis of large fMRI datasets revealed a similar mechanism in humans. Human brain activity was punctuated every several seconds by coherent, propagating waves emerging in the exteroceptive sensorimotor regions and terminating in the interoceptive default mode network

[Research on auditory neurofeedback technology and its multi-disciplinary applications].

Auditory neurofeedback (ANF) utilizes sound to map brain activity in real-time, guiding individuals to self-regulate neural states via operant conditioning. Compared to visual neurofeedback, ANF offers distinct advantages in millisecond-level response speed and non-visual dependency, significantly enhancing ecological validity and reducing visual fatigue. This paper reviews the physiological basis, signal processing workflows, and three typical experimental paradigms of ANF: threshold regulation, parameter modulation, and state-dependent triggering. Furthermore, it discusses current applicatio

Cognitive Pathways in Atypical Development: Executive Functions in Children With Attention-Deficit/Hyperactivity Disorder and Specific Learning Disorders.

Executive functions encompass a range of higher-level cognitive processes that are crucial for attaining goals and adjusting behaviours. They play a significant role in the early predictions of learning outcomes, academic performance, health, well-being, economic status and social interactions in life. The preschool years are an important period for the development of executive functions, during which an early intervention can have notably beneficial effects. The primary aim of this narrative review is to enhance our understanding of certain executive function impairments, their developmental

A hemochromatosis allele that protects against Alzheimer's disease injury: A role for H-ferritin.

Recent epidemiological evidence showed that mutations to the HFE-63 allele of this hemochromatosis-associated iron-assimilation protein improve chances of avoiding Alzheimer's disease (AD). This is unexpected since increased brain ferroptosis in gray-matter increases the risk for vascular dementia and AD. However, diffusion tensor imaging from a key Alzheimer's Disease Neuroimaging Initiative biomarker study showed that the hemochromatosis H63D allele protected white matter tracts and improved cognitive performance in individuals when APOE4 accelerates AD. H63D-carrying individuals exhibit ele

Active inference and psychodynamics: a novel integration with applications to depression and stress disorders.

This paper introduces Active Intersubjective Inference (AISI), a novel framework that integrates psychodynamic theory with predictive processing to explain self-identity construction and psychopathology. AISI posits that the self emerges from recursive inferences about how others perceive us (second-order self), interacting bidirectionally with interoceptive processes. We map psychodynamic phenomena (e.g., transference, projection) onto neurocomputational mechanisms and apply AISI to Major Depressive Disorder (MDD) and Post-Traumatic Stress Disorder (PTSD), highlighting distorted second-order

Antioxidants as immunomodulators for Tau and Amyloid-β in Alzheimer's disease.

Alzheimer's Disease is a progressive neurodegenerative disorder driven by oxidative stress, neuroinflammation, and pathological protein aggregation, such as Tau tangles and Amyloid-beta plaques. These processes disrupt neuronal integrity, contributing to cognitive decline. Dietary antioxidants, including phenolic compounds, omega-3 fatty acids, carotenoids, and isothiocyanates, have shown significant potential in mitigating these pathological mechanisms. They neutralize free radicals, reduce Tau hyperphosphorylation, inhibit Amyloid-beta aggregation, and alleviate inflammation, offering neurop

Sex Differences in the Neurobiology of Fear and Anxiety.

Although women are diagnosed with anxiety and stress-related disorders at twice the rate of men, there remains a lack of clarity around how to enhance treatment within each sex to reduce disparate rates of anxiety. However, in recent years, a growing literature has identified neural, cognitive, and physiological mechanisms that contribute to sex differences in fear and anxiety, with the promise of informing tailored treatment approaches. Here, we review recent findings, focusing on human studies among healthy populations as well as among patients with generalized anxiety, social anxiety disord

Diagnostic and Prognostic Biomarkers of Idiopathic Normal Pressure Hydrocephalus in Cerebrospinal Fluid and Blood.

Idiopathic normal pressure hydrocephalus (iNPH) is a treatable condition marked by gait disturbances, cognitive decline, and urinary incontinence. Biomarkers play a crucial role in distinguishing iNPH from Alzheimer's disease (AD) and predicting shunt surgery outcomes. Aβ1-42, Aβ1-40, t-tau, and p-tau181 differentiate iNPH from AD, with iNPH showing a higher Aβ1-42/Aβ1-40 ratio and normal tau levels. Neurofilament light chain and soluble amyloid precursor protein derivatives provide prognostic insights with lower preoperative levels linked to better surgical outcomes. Elevated LRG-1 and MCP-1

Neurological impairments in Duchenne muscular dystrophy: A comprehensive review.

Duchenne muscular dystrophy, the most prevalent form of muscular dystrophy, is characterized by neurological complications including cognitive impairment, neuropsychiatric disorders, and epilepsy. Neuroimaging investigations have demonstrated structural brain alterations, hemodynamic disturbances, and metabolic dysregulation in individuals with Duchenne muscular dystrophy. These neurological impairments are primarily attributed to cerebral dystrophin deficiency and subsequent downstream molecular/cellular abnormalities, including altered excitation-inhibition balance, blood-brain barrier disru

Trehalose Ameliorates Zebrafish Emotional and Social Deficits Caused by CLN8 Dysfunction.

CLN8 and other neuronal ceroid lipofuscinoses (NCLs) often lead to cognitive decline, emotional disturbances, and social deficits, worsening with disease progression. Disrupted lysosomal pH, impaired autophagy, and defective dendritic arborization contribute to these symptoms. Using a cln8-/- zebrafish model, we identified significant impairments in locomotion, anxiety, and aggression, along with subtle deficits in social interactions, positioning zebrafish as a useful model for therapeutic studies in NCL. Our findings show that trehalose, an autophagy enhancer, ameliorates anxiety, and modest

"¿Cómo qué, cómo qué? cómo qué?" Single-language echolalia in a bilingual female with progressive supranuclear palsy: a case report.

The case study explores bilingualism and neurodegenerative disorders, specifically progressive supranuclear palsy (PSP) with speech and language disorder (PSP-SL). It features a 78-year-old Mexican American woman who exhibits echolalia only in response to Spanish. This selective impairment suggests unevenly affected language control mechanisms despite her proficiency in both languages. Cognitive function is evaluated with neuropsychological tests; she's diagnosed with PSP-SL, depression, and anxiety. Echolalia in response to one language implies complex phonological retrieval mechanisms. Such

The relationship between sleep quality and depression: Critical roles of reflective and brooding rumination and sleep-related cognition.

There is a complex interplay between sleep problems and depression. This study explored the possible effects of rumination and dysfunctional beliefs about sleep on the relationship between sleep quality and depression. A cross-sectional survey of 1240 Chinese adults was conducted to assess the possible relationships. The results showed a chain mediating effect of reflection rumination and brooding rumination on the relationship between sleep quality and depression, accounting for 38.91% of the total variance. A moderating role of unreasonable attitudes about sleep was also discovered among the
