Tumor cells often evade immune pressure via metabolic reprogramming, yet the key metabolic regulators orchestrating this process remain inco … tamine uptake and glutathione synthesis. This adaptation limited mitochondrial oxidative stress and cytosolic mitochondrial DNA release, thereby suppressing cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) activation, interferon-β production, and CD8 T cell antitumor responses. These findings define a glutamine-fueled metabolic program …

The cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) pathway has emerged as a critical cytosolic DNA-sensing mechanism that orchestrates innate immune activation in response to cellular stress. In the central nervous system (CNS), this pathway demonstrates highly context-specific and cell-type-dependent functions, ranging from promoting neuroinflammation in neurodegenerative diseases such as Alzheimer's disease (AD) and Parkinson's disease (PD), to modulating immune surveillance and therapeutic responsiveness in glioma. This review systematically delineates the molecular mec…

Macrophages and dendritic cells acquire bacteria and cytosolic content from other cells without killing the donor cell through a trogocytosis-associated process termed merocytophagy. While characteristics of this behavior have been partially identified, the mechanism and potential contribution to the response to infection are unclear. Here, we reveal that a wide range of distinct species of bacteria stimulate enhanced merocytophagy in macrophages through pattern recognition receptor (PRR). Further, we found that cell-to-cell transfer in response to Francisella tularensis infection occurs in a …

Cytosolic aggregation of the nuclear protein TAR DNA-binding protein 43 (TDP-43) is associated with many neurodegenerative diseases, but the triggers for TDP-43 aggregation are still debated. Here, we demonstrate that TDP-43 aggregation requires a double event. One is up-concentration in stress granules beyond a threshold, and the other is oxidative stress. These two events collectively induce intra-condensate demixing, giving rise to a dynamic TDP-43-enriched phase within stress granules, which subsequently transition into pathological aggregates. Intra-condensate demixing of TDP-43 is observ…

The imbalance in cellular ionic homeostasis represents a hallmark of several neurodegenerative diseases, including Amyotrophic Lateral Sclerosis (ALS). Zinc Transporter 1 (ZnT1), the first described member of the ZnT family, stands out as the sole member of the SLC30 family responsible for exporting cytosolic zinc to the extracellular space. While ZnT1 is expressed across all tissues and cell types studied, it exhibits the highest prominence within the central nervous system. In ALS SOD1G93A mice, a reduction in ZnT1 expression consistent with disease progression has been observed, prompting o…

Aldehyde dehydrogenase 1A (ALDH1A), a cytosolic enzyme, plays a critical role in tumorigenesis, cancer progression, and therapy resistance by maintaining cancer stem cell properties across various malignancies. Dysregulation of ALDH1A is implicated in the progression of numerous human diseases, including various cancers, metabolic disorders, and neurodegenerative conditions. Consequently, ALDH1A represents a promising therapeutic target, which underscores the urgent need to develop small-molecule inhibitors targeting ALDH1A. In recent years, research groups have developed a series of highly se…

Stimulator of IFN genes (STING) is an endoplasmic reticulum (ER) signaling receptor involved in the type I interferon response to pathogen- or self-derived cytosolic double-stranded DNA. Excessive activation of STING is associated with many diseases, but the regulatory mechanism of STING activation remains to be further elucidated. Here, we identify VAPB as a negative regulator of STING-mediated innate immune response. VAPB deficiency increases the expression of type I interferons under resting conditions or upon stimulation. Mechanistically, VAPB associates and translocates with STING, thereb…

Calcium ions (Ca2+) are an important secondary messenger in plant signal transduction networks. The cytosolic free Ca2+ concentration ([Ca2+]i) of plants changes rapidly when they are subjected to different abiotic stresses, which drives calcium signaling. Although this process has been extensively studied in spermatophytes, the details of calcium signaling in bryophytes remains largely unknown. In our study, we reconstituted aequorin in the bryophyte Physcomitrella patens, optimized the percentage of ethanol in the Ca2+ discharging solution, and measured the [Ca2+]i changes induced by differe…

Ergothioneine (ET), a dietary thione/thiol, is receiving growing attention for its possible benefits in healthy aging and metabolic resilien … fidation of more than 300 protein targets. Among these, protein-persulfidation-driven activation of cytosolic glycerol-3-phosphate dehydrogenase (cGPDH) primarily contributes to the ET-induced NAD+ increase. ET's effects are abolished in models lacking CSE or cGPDH, highlighting the essential role of H2S signaling and protein persulfidation. These finding …

The necessary energy supply in skeletal muscles is based on either glycolysis or mitochondrial oxidative phosphorylation (OxPhos). These two … g on the muscle type. In the glycolytic muscles, phosphocreatine, produced from creatine and ATP by cytosolic creatine kinase, is mostly considered a cellular energy store for fast ATP delivery, whereas in the oxidative muscles, phosphocreatine and mitochondrial creatine kinase are the main players in the intracellular energy transport.

Neutrophils are the first cells among the innate and adaptive immune cells, which quickly react to infections, injuries, and disease. Neutrophils have a wide range of antimicrobial proteins that are secreted from the cell through the NETosis process. Neutrophil extracellular traps (NETs) are intricate formations composed of chromatin decondensed and coated with granular and cytosolic proteins. The body's defensive mechanism first identified NETs as critical participants due to their capacity to immobilize and even eliminate germs. Follow-up investigations have shown that these substances have …

Studying how single nucleotide polymorphisms (SNPs) crosstalk with non-autologous factors to cause complex autoimmune diseases is challenging. An amino acid replacement in the neutrophil cytosolic factor 1 (NCF1-339/NCF1R90H) leading to lower reactive oxygen species induction has been reported as the major SNP for systemic lupus erythematosus (SLE). Here we show that infection with the murine norovirus (MNV) contributes to the induction of lupus in Ncf190H mice. Mutant NCF190H upregulates the IFN-α/JAK1/STAT1 pathway in macrophages and anti-MNV-antibody production. In parallel, the MNV infecti…

Intracellular infections by Gram-negative bacteria are a significant global health threat. The nuclear receptor Nur77 (also called TR3, NGFI-B, or NR4A1) was recently shown to sense cytosolic bacterial lipopolysaccharide (LPS). However, the potential role for Nur77 in controlling intracellular bacterial infection has not been examined. Here we show that Nur77 protects against intracellular infection in the bladder by uropathogenic Escherichia coli (UPEC), the leading cause of urinary tract infections (UTI). Nur77 deficiency in mice promotes the formation of UPEC intracellular bacterial communi…
