For medical emergencies, such as acute ischemic stroke, rapid drug delivery to the target site is essential. For many small molecule drugs, this goal is unachievable due to poor solubility that prevents intravenous administration, and less obviously, by extensive partitioning to plasma proteins and red blood cells (RBCs), which greatly slows delivery to the target. Here we study these effects and how they can be solved by loading into nanoscale drug carriers. We focus on fingolimod, a small molecule drug that is FDA-approved for treatment of multiple sclerosis, which has also shown promise in

BACKGROUND: Preventing the emergence of anti-malarial drug resistance is critical for the success of current malaria elimination efforts. Prevention strategies have focused predominantly on qualitative factors, such as choice of drugs, use of combinations and deployment of multiple first-line treatments. The importance of anti-malarial treatment dosing has been underappreciated. Treatment recommendations are often for the lowest doses that produce "satisfactory" results.
METHODS: The probability of de-novo resistant malaria parasites surviving and transmitting depends on the relationship betw

Complex III of the malaria parasite mitochondrial electron transport chain (mtETC) has been validated as an attractive target for currently used antimalarials. We previously showed that the main function of mtETC in blood stage Plasmodium falciparum is to regenerate ubiquinone, which serves as an obligatory co-substrate of dihydroorotate dehydrogenase (DHOD), an essential mitochondrial enzyme for pyrimidine biosynthesis. P. falciparum can be rendered resistant to all mtETC inhibitors by provision of a bypass mediated by cytosolic yeast DHOD, a fumarate-reducing enzyme. Malaria parasite mitocho

Dermatophyte infections globally account for 20 to 25% of fungal infections. Dermatophytes have begun exhibiting antifungal drug resistance, making it challenging to treat this particular infection. Essential oils could be used as alternative solutions as they have been used for a long period to treat different infections. The research has demonstrated the antifungal efficacy of cinnamon, clove, lemongrass, tea tree, thyme, and garlic essential oils, and the impact of their combinations was assayed against Microsporum canis, Trichophyton tonsurans, T. violaceum, T. verrucosum, and Epidermophyt

E4F1 is essential for early embryonic mouse development and for controlling the balance between proliferation and survival of actively dividing cells. We previously reported that E4F1 is essential for the survival of murine p53-deficient cancer cells by controlling the expression of genes involved in mitochondria functions and metabolism, and in cell-cycle checkpoints, including CHEK1, a major component of the DNA damage and replication stress responses. Here, combining ChIP-Seq and RNA-Seq approaches, we identified the transcriptional program directly controlled by E4F1 in Human Triple-Negati

Chagas disease (CD), caused by Trypanosoma cruzi, remains a serious public health problem. One of the causes of the high morbidity and mortality in patients is the lack of an effective drug therapy. Thus, the aim of this study was to evaluate the efficacy of the essential oil of Syzygium aromaticum alone and in combination with benznidazole (BZ) in mice orally inoculated with strain of T. cruzi IV obtained from oral CD outbreak occurred in Western Brazilian Amazonia. All the animals inoculated with metacyclic trypomastigote forms (AM14 strain, BZ resistant), derived from the insect Rhodnius ro

Experimental studies demonstrated that mineralocorticoid antagonists prevent or reverse myocardial fibrosis. Therefore, we tested the hypothesis that the aldosterone antagonist canrenone can improve left ventricular diastolic function in essential hypertension. Using digitized M-mode echocardiography and 24-hour blood pressure monitoring (ABPM), we realized a prospective, randomized, controlled study on 34 never-treated essential hypertensives with left ventricular diastolic dysfunction. Echocardiogram and ABPM were repeated after 6 months of effective antihypertensive treatment with ACE inhib

Long-term blood pressure stability in humans is not affected by prolonged hypotensive drug therapy. After withdrawal of long-term hypotensive therapy from essential hypertensives, their blood pressures soon return to pre-treatment levels. Since the well-studied stabilizing systems all adapt to prevailing blood pressure levels, they cannot account for long-term stability of blood pressure. There is a parallel between trough blood pressure and the lower limit of cerebral autoregulation. Blood pressure goes down to a trough level once sleep has been established. This level is maintained for 3-4 h

INTRODUCTION: PTK7, as a pseudo-kinase, has its abnormal expression closely related to the progression and prognosis of various malignant tumors. PTK7 is not a typical inhibitory oncogenic target but rather more like an enriched surface recognition on tumor cells, which can be used as drug delivery target and for degradation-based therapeutic strategies.
AREAS COVERED: This review summarizes PTK7's molecular structural traits and transcriptional/post-transcriptional regulatory mechanisms, as well as its critical role in tumorigenesis. It evaluates PTK7's clinical significance across cancers,

Microfluidic systems are increasingly utilized in preclinical research, however, developing platforms that are compatible with large-scale manufacturing remains a critical challenge. In this regard, we show that thermoplastic elastomer (TPE)-based microfluidic chips could be a viable alternative to traditional poly(dimethylsiloxane) (PDMS) devices for drug testing applications. Microfluidic concentration gradient generators are essential components in these systems, enabling local spatial and temporal control of biochemical stimuli for applications such as cellular response studies and drug sc

A comprehensive understanding of tissue mechanics at the microscale is critical for advancing personalised therapies, controlled drug release, and tissue engineering. Characterising the mechanical properties of complex, soft biological materials-particularly multilayered and anisotropic organs such as the skin and eye-remains a significant challenge due to their variable water content and scale-dependent behaviour. Traditional continuum models and linear material responses often fail to capture the dynamic and nonlinear nature of these tissues under physiologically relevant conditions. This re

The complexity of the tumor microenvironment in advanced-stage cancer critically diminishes the activity of chemotherapy, often reducing its efficacy to a point where the inherent risks become clinically unwarranted. This review "unmasks" the tumor microenvironment as a sophisticated barrier system driven primarily by hypoxia, i.e. severe oxygen deprivation. Hypoxia stabilizes master-regulators, including hypoxia-inducible factor-1α, which actively induce multidrug resistance. Specifically, hypoxia-inducible factor-1α orchestrates drug expulsion from the cancer cell via efflux pumps and enhanc

A formidable clinical challenge, colorectal cancer (CRC) has faced a considerable limitation arising in the form of drug resistance, which in turn has affected the optimum outcome of standard chemotherapy, target therapy, and immunotherapies. Peptide scaffolds, in recent research, have turned out to be an effective tool in tackling multidrug resistance. This article reviews recent observations on peptide-mediated therapy against CRC drug resistance that comprises peptide-drug conjugates, which have shown effective implications in intensifying specific antitumor effects, cell-penetrating peptid

PURPOSE: Antiseizure medications (ASMs) are commonly used in the intensive care unit (ICU) and often exhibit pharmacokinetics that differ substantially from those in healthy volunteers or in the outpatient setting. Organ dysfunction, polypharmacy, exogenous devices, and greater severity of illness all influence ASM pharmacokinetics, dosing decisions, and monitoring parameters. It is essential for critical care clinicians to familiarize themselves with the pharmacokinetics, dosing considerations, effects of exogenous devices, and therapeutic drug monitoring (TDM)-all covered in this narrative r

The use of animal models that can reliably predict drug performance in human patients is critical to antibody therapeutic development. Along with assessing toxicity and efficacy, determining the pharmacokinetic (PK) properties of therapeutics in Tg32 and Tg276 mice is essential to preclinical characterization. While Tg32 mice have been well established as indispensable in their ability to model the PK properties of antibody therapeutics, their intact immunity leaves them capable of mounting anti-drug antibody responses that interfere with PK interpretation. Here, we demonstrate the negative im

Accurate monitoring of antitumor drug is essential for therapeutic management and safety evaluation. Herein, an electrochemiluminescence (ECL) sensor was developed for sensitive detection of antitumor drug mitoxantrone (MIT) based on surfactant-modified carbon nanotubes. Cetyltrimethylammonium bromide (CTAB)-modified carboxylated multi-walled carbon nanotube (cMWCNTs) composite (CTAB@cMWCNTs) was prepared through a simple ultrasonic-assisted assembly method and immobilized onto indium tin oxide (ITO) electrode by drop-casting. By forming a stable interfacial modification layer, enhanced ECL re
