Complexity of prefrontal cortical dysfunction in schizophrenia: more than up or down.

OBJECTIVE: Numerous neuroimaging studies have examined the function of the dorsolateral prefrontal cortex in schizophrenia; although abnormalities usually are identified, it is unclear why some studies find too little activation and others too much. The authors' goal was to explore this phenomenon. METHOD: They used the N-back working memory task and functional magnetic resonance imaging at 3 T to examine a group of 14 patients with schizophrenia and a matched comparison group of 14 healthy subjects. RESULTS: Patients' performance was significantly worse on the two-back working memory task t

Divergent network trajectories in mild cognitive impairment: Converters versus non-converters to Alzheimer's disease.

BackgroundMild cognitive impairment (MCI) is a condition between normal aging and Alzheimer's disease (AD), with around 12% of individuals progressing to AD each year. Understanding how cognitive functions evolve over time in people with MCI, especially those who eventually develop AD, is key to improving early detection and intervention.ObjectiveTo investigate the longitudinal changes in the relationships between cognitive functions in patients with MCI, which will or will not convert to AD.MethodsWe applied cross-lagged panel network (CLPN) to data from the Alzheimer's Disease Neuroimaging I

Multimodal neuroimaging changes and their behavioral, genetic, and neurotransmitter correlates in electroconvulsive therapy for major depressive disorder.

Electroconvulsive therapy (ECT) is an effective treatment for major depressive disorder (MDD), yet its underlying mechanisms remain unclear. This study investigated the antidepressant effects of ECT through a multimodal neuroimage meta-analysis combined with functional, genetic, and neurotransmitter assessments. Resting-state functional magnetic resonance imaging (fMRI) and voxel-based morphometry (VBM) data were analyzed using seed-based d mapping with permutation of subject images (SDM-PSI) to identify changes in spontaneous brain activity and gray matter volume (GMV) before and after ECT. Cognitive and Emotional Predictors of Decision-Making in Adults: Roles of Cognitive Functions and Emotion Regulation Difficulties.

OBJECTIVES: The Iowa Gambling Task (IGT) is a widely used measure of decision-making that mimics real-life situations involving uncertainty and reward. While prior research has separately explored the roles of cognitive functions (e.g., intelligence) and non-cognitive factors (e.g., emotion regulation), few have examined their combined and relative influence. This study aimed to clarify how cognitive abilities and emotion regulation difficulties contribute to decision-making under ambiguity and risk, as assessed by the IGT. METHODS: Fifty healthy adults completed a modified version of the IGT

Ageing and pituitary neuroendocrine tumours (PitNETs): from bench to bedside.

The prevalence of pituitary neuroendocrine tumours (PitNETs) in elderly people (≥65 years old) is raising with life expectancy. Despite an increasing incidental detection on neuroimaging for unrelated conditions, a large majority are macrotumours (≥1 cm) and mass effects are frequent, in particular visual defects. Clinically non-functioning PitNETs are the most prevalent (about 75%), and a majority are gonadotrophs. The clinical presentation of functioning PitNETs differ from younger patients and include somatotroph tumours - often intrasellar with mild acromegaly, prolactinomas - often large

Choroid plexus free-water correlates with glymphatic function in Alzheimer's disease.

INTRODUCTION: Free-water imaging of the choroid plexus (CP) may improve the evaluation of Alzheimer's disease (AD). METHODS: Our study investigated the role of free-water fraction (FWf) of CP in AD among 216 participants (133 Aβ+ participants and 83 Aβ- controls) enrolled in the NeuroBank-Dementia cohort at Ruijin Hospital (RJNB-D). The Alzheimer's Disease Neuroimaging Initiative dataset was used for external validation. RESULTS: At baseline, Aβ+ participants showed higher CP FWf, increased white matter hyperintensity (WMH) volume, and decreased diffusion tensor image analysis along the peri

Neural correlates of personal agency in borderline personality disorder - An fMRI investigation using attachment-informed autobiographical memories.

Individuals with borderline personality disorder (BPD) have identity disturbance and difficulty with self/other distinction which may be linked to difficulties in personal agency. Neuroimaging research has helped inform underlying deficits linked to sociocognitive functioning, however, no research has specifically investigated neural correlates of personal agency in BPD. We used an attachment-informed autobiographical memory recall task to investigate behavioural and neural mechanisms of personal agency in BPD. Individuals with BPD (n = 34) and nonclinical controls (NCC, n = 35) recalled attac

7T magnetic resonance imaging-based investigation of the correlation between mammillary body structure and cognitive impairment in patients with spinocerebellar ataxia type 3.

BACKGROUND: Spinocerebellar ataxia type 3 (SCA3) is a hereditary disease characterized by cerebellar atrophy and motor dysfunction. Patients also exhibit non-ataxic symptoms such as cognitive impairment. While prior neuroimaging studies have identified multiple cognition-associated brain regions in SCA3 patients, research on Papez circuit structural damage (e.g., mammillary bodies (MBs)) remains sparse. Advancements in 7T magnetic resonance imaging (MRI) technology have enabled scanning and quantitative analysis of structures such as the MBs within the Papez circuit. In this study, we investig

Complement factor I deficiency-associated neuroinflammatory disease among Old Order Amish.

BACKGROUND: Complement factor I (CFI) deficiency is an ultrarare inborn disorder of complement regulation that manifests with protean infectious, vasculitic, and neuroinflammatory symptoms. OBJECTIVE: We sought to functionally validate a previously unrecognized, disease-associated CFI variant (Y459S) and determine variant enrichment in the Old Order Amish population. METHODS: Expression and function of the CFI Y459S variant was evaluated via immunoblot, complement factor 3b degradation, and crystal structure analysis. CFI variant frequencies in Old Order Amish populations were determined usi

Individualized functionnectome for the statistical assessment of white matter circuits underlying task-fMRI activations in glioma patients.

The Functionnectome framework enables the projection of task-related fMRI signals onto the underlying white matter pathways, using anatomical priors derived from structural connectivity. However, the existing "standardized" priors are based on averaged tractograms from healthy controls and cannot accurately capture the altered anatomy found in patients with brain lesions. This study extends the Functionnectome framework by generating individualized anatomical priors from subject-specific diffusion MRI tractography, allowing for improved integration of functional and structural data in patients

Brain structural correlates of prior alcohol use disorder and their relation to cognitive impairment in individuals with mood disorders.

BACKGROUND: Alcohol use disorders (AUD) often co-occur with mood disorders (MD), negatively impacting the clinical course of both disorders and exacerbating cognitive impairments. Neuroimaging studies have demonstrated lower gray matter volume (GMV) and hippocampal volume in MD and AUD, and additionally in the orbitofrontal cortex (OFC), nucleus accumbens (NAc), and white matter (WM) volume in AUD. However, whether the pronounced structural alterations in MD persist after remission from AUD, and their relation to cognitive impairments, remains unclear. METHOD: Individuals with MD with (MD + A

Comprehensive examination of resting state fMRI connectomics yields new insights into brain function deficits in Gulf War illness after accounting for heterogeneity in brain impairment across the ill veteran population.

An estimated 200,000 veterans (up to 32% of those deployed) of the 1991 Gulf War (GW) suffer from GW illness (GWI), an incompletely understood chronic medical condition, characterized by multiple symptoms indicative of brain function deficits in various domains. Epidemiologic and animal studies have associated GWI with exposure to neurotoxic chemicals such as nerve agents, organophosphate pesticides and pyridostigmine bromide. One factor that hampers mechanistic investigations into GWI is that there is considerable heterogeneity in brain impairments across the ill GW veteran population. This c

Serotonergic and dopaminergic control of impulsivity in gambling disorder.

Gambling disorder (GD) is major public health issue. The disorder is often characterized by elevated impulsivity with evidence from analogous substance use disorders underlining prominent roles of brain monoamines in addiction susceptibility and outcome. Critically, GD allows the study of addiction mechanisms without the confounder of the effects of chronic substances. Here, we assessed the roles of striatal dopamine transporter binding and extrastriatal serotonin transporter binding in GD as a function of impulsivity using [123 I]FP-CIT SPECT imaging in 20 older adults with GD (DSM-5 criteria

Role of Structural, Metabolic, and Functional MRI in Monitoring Visual System Impairment and Recovery.

The visual system, consisting of the eyes and the visual pathways of the brain, receives and interprets light from the environment so that we can perceive the world around us. A wide variety of disorders can affect human vision, ranging from ocular to neurologic to systemic in nature. While other noninvasive imaging techniques such as optical coherence tomography and ultrasound can image particular sections of the visual system, magnetic resonance imaging (MRI) offers high resolution without depth limitations. MRI also gives superior soft-tissue contrast throughout the entire pathway compared

MRI morphology of the hippocampus in drug-resistant temporal lobe epilepsy: Shape inflation of left hippocampus and correlation of right-sided hippocampal volume and shape with visuospatial function in patients with right-sided TLE.

We sought to quantify the morphology in vivo of hippocampi in patients with drug resistant temporal lobe epilepsy (TLE) via magnetic resonance imaging (MRI), prior to temporal lobe resection, and the correlation of surface-based shape analysis of morphology and clinical cognitive function. Thirty patients with drug-resistant TLE and twenty healthy controls underwent clinical neuropsychological testing, and brain MRI at Lund University Hospital prior to hippocampal resection. A neuroradiologist categorised radiological findings into normal hippocampus, subtle changes or definite hippocampal scl

Impaired instrumental reversal learning is associated with increased medial prefrontal cortex activity in Sapap3 knockout mouse model of compulsive behavior.

Convergent functional neuroimaging findings implicate hyperactivity across the prefrontal cortex (PFC) and striatum in the neuropathology of obsessive compulsive disorder (OCD). The impact of cortico-striatal circuit hyperactivity on executive functions subserved by these circuits is unclear, because impaired recruitment of PFC has also been observed in OCD patients during paradigms assessing cognitive flexibility. To investigate the relationship between cortico-striatal circuit disturbances and cognitive functioning relevant to OCD, Sapap3 knockout mice (KOs) and littermate controls were test…
