Development of an enzyme-linked immunosorbent assay (ELISA) for identification of venoms from snakes in the Agkistrodon genus.

An enzyme-linked immunosorbent assay (ELISA) using a purified myotoxin from the venom of Agkistrodon contortrix laticintus (broad-banded copperhead) as immunogen was developed for potential use in the identification of envenomation by snakes belonging to the genus Agkistrodon native to North America. The specificity of the assay was tested using a total of 43 venom samples from snakes of diverse geographic locations. Venom samples used for cross-reactivity determination represent eight snake families including 14 species from the genus Crotalus. The assay detected venom from all Agkistrodon sp

A comparative, qualitative and quantitative antimicrobial efficacies of mouthrinses containing chlorhexidine gluconate and essential oils.

A project was launched to evaluate and compare antimicrobial efficacy of Hexidine and Listerine over placebo in 10 day human experimental gingivitis study. A rigid study schedule for subject compliance spanning well over 3 months was sorted out and volunteers recruited for study rinsed all the 3 assigned mouthrinses containing (a) chlorhexidine gluconate, (b) "essential oils" and (c) flavoured distilled water one after the other at certain prefixed intervals. After 10 days of assigned mouthrinse regimen, where the assigned mouthrinse was the only method practiced by the volunteers for oral hyg

Effect of morphine on mesangial immunoglobulin G aggregate kinetics.

Because mesangial expansion is considered a precursor of focal glomerulosclerosis, we studied whether morphine can cause mesangial expansion. We used radiolabeled human immunoglobulin G aggregates (125I-ahIgG) to study mesangial kinetics in control and experimental (morphine-treated) rats. Control and experimental rats were administered 125I-ahIgG by tail vein. Serum levels of 125I-ahIgG and uptake of 125I-ahIgG by liver, spleen, and mesangium were determined at 4, 8, 12, 24, and 36 h after 125I-ahIgG administration. Mesangial 125I-ahIgG levels were higher (P < 0.05) at 4 h and at later period

[Altruism and the donor].

On December 20, 1988, the government of France passed a law to protect people who voluntarily participate in biomedical research. This article makes extensive reference to a major study, titled From Biology to Ethics, by Jean Bernard, a well-respected authority in the field of bioethics. The author looks at models proposed by Bernard, as examples for health volunteers, in particular, the blood donor and the self-experimenter. To set the tone of the article, she recalls the concept of altruism, as first proposed by Auguste Comte, then makes a linkage between his philosophy and Bernard's point o

Viral and bacterial vectors of immunogenes.

A recent development in the production of experimental vaccines has been the use of the smallpox vaccine virus (vaccinia virus) as a carrier (vector) of the genes (immunogenes) which code for the protection-inducing proteins (immunogens) of unrelated viruses. The potential of these vector vaccines lies in the hope that such a vaccine would be cheaper, safer and/or more effective than existing vaccines to some pathogens. Vaccinia virus as a vector has attracted most attention to date because: several immunogenes can be inserted into its genome without destroying its infectivity; the immunogens

Spatial pattern of nerve fiber abnormality indicative of pathologic mechanism.

Estimates of the number, density, and size distribution of myelinated fibers at selected levels of roots, spinal tracts, and sampled levels of peripheral nerves may be used in the detection and characterization of alterations of motor, sensory, and autonomic neurons and their axons with development, aging and disease. Use of imaging techniques, now available, increases the reliability, versatility, and speed of such analysis. In this study, the authors evaluated the spatial pattern of fibers in sampled frames and contour areas of transverse sections of nerve fascicles, utilizing, the coefficie

Nutrition and cancer.

There is increasing epidemiological evidence that nutrition plays a dominant role in the pathogenesis of several types of human cancers. There is considerable epidemiological evidence showing that alcoholism in part because of associated nutritional deficiencies, significantly increases the risk of smokers for cancer of the alimentary tract. There is also some suggestion that nutritional deficiencies may relate to cancers of the stomach, cervix, and thyroid. Of particular importance, and based on relatively new concepts, are data indicating that overnutrition significantly affects the developm

THE EXPERIMENTAL TRANSMISSION OF COLORADO TICK FEVER.

1. The symptoms, history of tick bite, characteristic fever curve, and white blood cell picture should enable the physician to make a diagnosis of Colorado tick fever in nearly every case. 2. The typical white blood cell picture is a depression of the total leucocytes with a shift to the left of the granulocytes. Basophilic cytoplasmic bodies appear occasionally in lymphocytes 3 to 4 days after clinical recovery. 3. The disease can be transmitted serially in human beings by parenteral injection of blood or serum. Such transfers have not resulted in decreased or increased virulence. 4. The natu

The effect of intervertebral disc damage on the mechanical strength of the annulus fibrosus in the adjacent segment.

BACKGROUND CONTEXT: A herniated intervertebral disc (IVD) is a common injury in the human population. Despite the injury being isolated to a singular IVD in the spine, it is important to look at the biomechanical effects that a damaged IVD has on the entire spine, specifically the IVD adjacent to the injury. PURPOSE: This study examined the effects of a damaged IVD on the mechanical properties of the annulus fibrosus (AF) in the adjacent cranial IVD. STUDY DESIGN: Basic science study using an in-vitro porcine model. METHODS: Sixteen porcine cervical spines were used; specifically spinal lev

[Effect of bile on intestinal calcium and vitamin D absorption. Animal experiment studies in swine].

In the human system calcium is the major constituent of bone and the regulator of important bioelectric and biochemical effects. Calcium homeostasis is underlying exact control mechanisms in which vitamin D is a predominant factor. Cholecalciferol (VD3) is metabolized and the active form 1 alpha, 25-dihydroxycholecalciferol (1,25(OH)2D3) is formed by the kidney. 1,25(OH)2D3 acts on the cell nuclei and on the luminal membrane of the intestinal mucosal cell. It enhances intestinal Ca absorption and the Ca transport to the blood system. VD3 metabolism and mechanisms of action are reported in the

Ex Utero Artificial Womb Support: Promising Future for Extremely Preterm Infants.

If they survive, extremely preterm infants (EPIs), born before 28 weeks' gestation, face high risks of chronic lung disease and neurodevelopmental impairment due to organ immaturity and iatrogenic injury. To prevent injury, EPIs should be treated as fetuses rather than small neonates. With the aim of doing so, artificial womb (AW), and artificial placenta (AP) technologies have been investigated for over 60 years. Recent advances have brought these technologies closer to human application, than ever before. This article outlines the rationale for AW and AP, their differences and characteristic

Protocol for characterizing airway epithelial ciliary beating and mucociliary transport using image processing and particle imaging velocimetry.

Lung epithelial cilia play a crucial role in respiratory health by driving mucociliary transport (MCT) and clearing inhaled pathogens and materials. Here, we present a protocol for measuring both ciliary beat frequency (CBF) and MCT in mucociliary human small airway epithelial cells. We describe steps for configuring experimental setups, collecting CBF and MCT data, analyzing datasets, and performing post-acquisition analysis. This protocol is adaptable to mucociliary cells from other lung regions and to mouse tracheal epithelial cells. For complete details on the use and execution of this pro

The role of the endometrial microbiome in embryo implantation and recurrent implantation failure.

There is a suggested pathophysiology associated with endometrial microbiota in cases where repeated implantation failure of high-quality embryos is observed. However, there is a suspected association between endometrial microbiota and the pathogenesis of implantation failure. However, there is still a lack of agreement on the fundamental composition of the physiological microbiome within the uterine cavity. This is primarily due to various limitations in the studies conducted, including small sample sizes and variations in experimental designs. As a result, the impact of bacterial communities

Trypanocidal Essential Oils: A Review.

Trypanosomiases are diseases caused by parasitic protozoan trypanosomes of the genus Trypanosoma. In humans, this includes Chagas disease and African trypanosomiasis. There are few therapeutic options, and there is low efficacy to clinical treatment. Therefore, the search for new drugs for the trypanosomiasis is urgent. This review describes studies of the trypanocidal properties of essential oils, an important group of natural products widely found in several tropical countries. Seventy-seven plants were selected from literature for the trypanocidal activity of their essential oils. The main

Quantification of Leishmania Parasites in Murine Models of Visceral Infection.

Visceral leishmaniasis (VL) is mainly caused by Leishmania donovani (India and East Africa), and Leishmania infantum (Mediterranean Basin and South America) infections. Although murine models of visceral infection lack the clinicopathological aspects of VL in humans, they have been proven useful at advancing our knowledge in the Leishmania field. Indeed, these models have been used not only to better understand the pathophysiology of the infection but also in drug and vaccine development. This chapter focuses on the protocols used to experimentally infect mice and to quantify parasite burdens

Brain Structures Implicated in Inflammation-Associated Depression.

Systemic inflammation rapidly impairs mood, motivation, and cognition inducing a stereotyped cluster of symptoms collectively known as "sickness behaviors." When inflammation is severe or chronic, these behavioral changes can appear indistinguishable from major depressive disorder (MDD). Human and rodent neuroimaging combined with experimental inflammatory challenges has clarified the neural circuitry associated with many of the key features of inflammation-induced-sickness behavior, and in so doing revealed often-remarkable commonalities with circuit abnormalities observed in MDD. This review

The knee joint loose body as a source of viable autologous human chondrocytes.

Loose bodies are fragments of cartilage or bone present in the synovial fluid. In the present study we assessed if loose bodies could be used as a source of autologous human chondrocytes for experimental purposes. Histochemical examination of loose bodies and differential enzymatic digestions were undertaken, the isolated cells were cultured in alginate bead microspheres and immunolocalisations were undertaken for chondrogenic markers such as aggrecan, and type II collagen. Isolated loose body cells had high viability (≥90% viable), expressed chondrogenic markers (aggrecan, type II collagen) b

In vivo imaging methods to assess glaucomatous optic neuropathy.

The goal of this review is to summarize the most common imaging methods currently applied for in vivo assessment of ocular structure in animal models of experimental glaucoma with an emphasis on translational relevance to clinical studies of the human disease. The most common techniques in current use include optical coherence tomography and scanning laser ophthalmoscopy. In reviewing the application of these and other imaging modalities to study glaucomatous optic neuropathy, this article is organized into three major sections: 1) imaging the optic nerve head, 2) imaging the retinal nerve fib

Synthesis and antiproliferative activity of substituted 3[2-(1H-indol-3-yl)- 1,3-thiazol-4-yl]-1H-pyrrolo[3,2-b]pyridines, marine alkaloid nortopsentin analogues.

A large number of indolyl-4-azaindolyl thiazoles, nortopsentin analogues, were conveniently synthesized. The antiproliferative activity of the new derivatives was examined against four human tumor cell lines with different histologic origin. Seven derivatives consistently reduced the growth of the experimental models independently of TP53 gene status and exhibited the highest activity against the malignant peritoneal mesothelioma (STO) cell line. The most active compound of this series acts as a CDK1 inhibitor, and was found to cause cell cycle arrest at G2/M phase, to induce apoptosis by prev
