[New data on the transmission of filariae].

This paper summarizes research of the past several years on two phases of filarial transmission: the ingestion of microfilariae by the vector and the regulation of the percentage of successful parasites by the vector. Experiments involved several different models for study: injection of inert particles into the bloodstream of a rodent and their subsequent ingestion by a vector; ingestion of gametocytes of different ages by vectors of a rodent malaria; and a monkey simultaneously infected with four different species of filariae. This has led to the idea that there exist two categories of microf

Transmission of onchocerciasis by local black flies on the Firestone Rubber Plantation, Harbel, Liberia.

A quantitative description of the transmission dynamics of onchocerciasis on the Firestone Rubber Plantation, Harbel, Liberia is presented. The putative vector, Simulium yahense, comprised 98.5% of the flies examined. Diurnal vector-host contact was unimodal, expressing a distinct peak between 0800 and 1100 hours, particularly during the wet season. Perennial breeding occurs in the Firestone biotope, and is manifested by a mean annual daily landing rate of 75 flies per person per day. S. yahense is considered an efficient vector by nature of its anthropophily, moderate infective worm burden (4

Fibronectin-like properties in antigen B from the cysticercus of Taenia solium.

The most prominent antigen of the cysticercus of Taenia solium in the serology of human cysticercosis, antigen B, is capable of agglutinating red blood cells of different species, of aligning in vitro cell growth of transformed mouse fibroblasts, and of strongly binding to pig, rat and human collagen. Scatchard analysis of the adsorption isotherms indicates quite avid heterogeneous binding of antigen B to rat collagen (K's = 10(3), 10(5)). These remarkable biological activities of antigen B, resembling those of fibronectins, allow for some speculation concerning its role in the host-parasite r

[Various factors influencing toxicity and metabolism of metals--metal-metal interactions and host factors (author's transl)].

This paper is a brief overview on metal-metal interactions and various other factors involved in the toxicity and metabolism of metals, including host factors, nutrition, habits, etc. Humans and other organisms are exposed to various metals in many ways, and even in an occupation in which workers are exposed to a metal such as lead, they are at the same time also exposed to other metals, although concentrations of these latter metals are usually not very high. This can result in effects different from those produced by a single metal by itself. Furthermore, individual differences which are cau

Ectoparasites and associated pathogens of free-roaming and captive animals in zoos of South Carolina.

A survey of ectoparasites and their associated pathogens was conducted in two South Carolina zoos, from 2004 to 2007. Dead, wild birds and mammals, as well as captive animals examined during routine veterinary checks constituted the study populations. Ectoparasites were tested for species of Anaplasma, Bartonella, Coxiella burnetii, Ehrlichia, Rickettsia, and Trypanosoma. Forty-six species of ectoparasites were collected from 133 free-roaming and captive hosts and their associated nesting and bedding materials. Six vector-borne pathogens were detected molecularly in the ectoparasites, includin

Modeling Influenza Virus Infection: A Roadmap for Influenza Research.

Influenza A virus (IAV) infection represents a global threat causing seasonal outbreaks and pandemics. Additionally, secondary bacterial infections, caused mainly by Streptococcus pneumoniae, are one of the main complications and responsible for the enhanced morbidity and mortality associated with IAV infections. In spite of the significant advances in our knowledge of IAV infections, holistic comprehension of the interplay between IAV and the host immune response (IR) remains largely fragmented. During the last decade, mathematical modeling has been instrumental to explain and quantify IAV dy

Granzyme B as a potential biological target in toxic encephalopathy: A big data-based exploratory analysis.

Toxic encephalopathy is a central nervous system disorder caused by endogenous or exogenous toxic substances. Granzyme B (GZMB), a key serine protease, plays a crucial role in immune regulation and the progression of various diseases. Abnormal expression of GZMB may contribute to the development and progression of toxic encephalopathy; however, its specific mechanisms remain unclear. This study aims to preliminarily explore the association between granzyme B and toxic encephalopathy through big data. This study retrieved the toxic encephalopathy dataset GSE253309 from the gene expression omnib

Intravacuolar Pathogens Hijack Host Extracellular Vesicle Biogenesis to Secrete Virulence Factors.

Extracellular vesicles (EVs) have garnered significant interest in recent years due to their contributions to cell-to-cell communication and disease processes. EVs are composed of a complex profile of bioactive molecules, which include lipids, nucleic acids, metabolites, and proteins. Although the biogenesis of EVs released by cells under various normal and abnormal conditions has been well-studied, there is incomplete knowledge about how infection influences EV biogenesis. EVs from infected cells contain specific molecules of both host and pathogen origin that may contribute to pathogenesis a

Neuroimmune communication in infection and pain: Friends or foes?

Clinically, a variety of micro-organisms cause painful infections. Before seen as bystanders in the context of infections, recent studies have demonstrated that, as immune cells, nociceptors can sense pathogen-derived products. Nociceptors and immune cells, therefore, have evolved to communicate with each other to control inflammatory and host responses against pathogens in a complementary way. This interaction is named as neuroimmune communication (or axon-axon immune reflex) and initiates after the release of neuropeptides, such as CGRP and VIP by neurons. By this neurogenic response, nocice

Flavivirus replication complex assembly revealed by DNAJC14 functional mapping.

DNAJC14 is an Hsp40 family member that broadly modulates flavivirus replication. The mechanism by which DNAJC14 stoichiometrically participates in flavivirus replication complex (RC) formation is unknown; both reduced and elevated levels result in replication inhibition. Using yellow fever virus (YFV), we demonstrate that DNAJC14 redistributes and clusters with YFV nonstructural proteins via a transmembrane domain and a newly identified membrane-binding domain (MBD), which both mediate targeting to detergent-resistant membranes. Furthermore, the RC and DNAJC14 reside as part of a protein inter

HIV-1-associated central nervous system dysfunction.

Despite more than 15 years of extensive investigative efforts, a complete understanding of the neurological consequences of HIV-1 CNS infection remains elusive. Although the resources of numerous investigators have been focused on studies of HIV-1-associated CNS disease, the complex nature of the disease processes that underlie the clinical, pathological, and cellular manifestations of HIV-1 CNS infection have required a larger volume of studies than was initially envisioned. Several major areas remain as the focus of current research efforts. One of the more pressing issues facing researchers

Coronavirus protein interaction mapping in bat and human cells reveals network rewiring governing immune evasion and zoonotic potential.

Coronaviruses, including severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), can cause severe disease in humans, whereas reservoir hosts such as horseshoe bats remain asymptomatic. To investigate how host-specific protein-protein interactions (PPIs) influence infection, we generated comparative PPI maps for SARS-CoV-2 and its bat progenitor RaTG13, using affinity purification mass spectrometry (AP-MS) in human and greater horseshoe bat cells. We identify both conserved and virus- and host-specific interactions that regulate infection dynamics. Notably, SARS-CoV-2 requires a nonsynony

Pentalinonsterol at the Host-Parasite Interface: Antileishmanial Activity, Host Immunomodulation, and Unresolved Mechanisms.

Leishmaniasis remains difficult to treat because available therapies are constrained by toxicity, administration burden, variable efficacy, cost, and drug resistance. This critical narrative review evaluates the preclinical evidence for pentalinonsterol (PEN), a plant-derived cholestane-type steroidal ketone originally isolated from the Mayan medicinal plant Pentalinon andrieuxii. Experimental studies demonstrate PEN activity against Leishmania promastigotes and intracellular amastigotes, together with marked parasite ultrastructural injury; however, no parasite molecular target has been valid

The multifaceted roles of fibronectin in viral pathogenesis.

Fibronectin is a high molecular weight glycoprotein critical to numerous physiological and pathological processes, including extracellular matrix organization, wound healing, immune responses, and interactions with pathogens. Evidence from various viral systems, including dengue, influenza, hepatitis B, and HIV-1, indicates that targeting fibronectin or disrupting its interactions can notably affect outcomes of viral infections in vitro. Alterations in the expression of fibronectin are associated with immune evasion mechanisms, including the inhibition of interferon signaling, thereby enabling

Pro- and anti-viral functions of lipid droplets in infection at a glance.

Lipid droplets (LDs) were long regarded as passive storage organelles for neutral lipids as this is the most efficient way to store energy. Recently, however, they have emerged as highly dynamic and heterogenous organelles that act as central hubs of lipid metabolic pathways. LDs provide lipids for energy production, supply precursor metabolites for many lipid classes, incorporate lipids under lipotoxic insults and are interconnected with other organelles through membrane contact sites. Viruses and all other obligate intracellular pathogens rely on metabolic networks in the host cell - includi

Bacterial extracellular vesicles as multifaceted regulators of human diseases and emerging therapeutic platforms.

Outer membrane vesicles (OMVs) are nanoscale vesicles actively released by Gram-negative bacteria, and have become key mediators in bacterial physiology, host-pathogen interactions, and disease pathogenesis. The biogenesis of OMVs is a dynamic process, which results from membrane homeostasis imbalance, peptidoglycan remodeling, and stress responses. These mechanisms enable virulence factors, immune-regulatory molecules, and nucleic acids to be selectively or randomly loaded into vesicles. With their ability to penetrate barriers and spread throughout the body, OMVs are widely involved in the o
