Genome-wide association summary statistics-based genetic mapping of brain functional and structural networks in Type 2 diabetes mellitus and its complications.

The disruptions in brain networks have been consistently observed in type 2 diabetes mellitus (T2DM) and complications. However, the causal effects and direction between these factors have yet to be elucidated. We conducted a two-sample bidirectional MR investigation supported by multiple sensitivity analyses. The study incorporated large-scale brain functional/structural network phenotypes from UK Biobank participants, while data for T2DM and related complications were provided by the FinnGen biobank. This dual-source approach ensured a rigorous evaluation of the causal interplay between neur

Major depressive disorder is associated with reduced intrinsic connectivity between the bed nucleus of the stria terminalis and ventral striatum.

BACKGROUND: The bed nucleus of the stria terminalis (BNST) plays a critical role in the neural processing of negative emotions. To date, no studies have investigated whether intrinsic connectivity of the BNST is associated with major depressive disorder (MDD) or with the severity of clinical symptoms in individuals with depression. METHODS: Resting-state functional magnetic resonance imaging (7 Tesla (7T)) with seed-to-voxel-analysis was used to compare the intrinsic connectivity of the BNST between individuals with MDD (N = 17) and healthy volunteers (N = 29). Exploratory analyses were also

Is neurofeedback A reliable therapy for managing Parkinson's Disease?

OBJECTIVE: Standard treatments for Parkinson's disease (PD) often lose effectiveness over time, motivating interest in complementary, non-pharmacological approaches. Neurofeedback, which trains patients to consciously modulate brain activity, has shown theoretical promise in PD; however, its clinical value remains uncertain. We conducted a PRISMA-guided systematic review to evaluate the current evidence. METHODS: We included studies of patients with idiopathic PD undergoing any form of neurofeedback training. Case studies were excluded to reduce intersubject variability. Sixteen studies met i

UniGraphPTMs: a contrastive learning-enhanced universal framework for PTM site prediction via GNN and multimodal fusion.

Protein post-translational modifications (PTMs) represent a core regulatory mechanism governing protein function and cellular fate. Their dynamic alterations profoundly influence critical biological processes. However, Existing research primarily focuses on single-PTM site prediction and remains confined by single-modality analysis. This study introduces UniGraphPTMs, the first universal PTM site prediction framework based on multimodal fusion and graph neural networks. UniGraphPTMs employs a master-slave architecture to break branch independence through multi-stage interactions. We pioneer th

A Study of Unilateral Biportal Endoscopic Discectomy and Transforaminal Endoscopic Discectomy for Single-level Calcified Lumbar Disc Herniation.

Calcified lumbar disc herniation (CLDH) presents unique surgical challenges due to its hard texture and frequent adhesion to neural structures. This study compares the efficacy and safety of unilateral biportal endoscopic discectomy (UBE) and percutaneous endoscopic transforaminal discectomy (PETD) in treating single-level CLDH. An analysis was conducted on 107 patients, with 45 undergoing UBE and 62 PETD. Both techniques significantly improved postoperative visual analogue scale (VAS) and Oswestry Disability Index (ODI) scores. PETD demonstrated advantages in operative time, blood loss, incis

Neurostructural Correlates and Impulsive Mechanisms of Exercise Dependence in Middle-Aged and Elderly Adults: Evidence from Gray Matter and Structural Covariance Networks.

Exercise dependence is characterized by excessive, uncontrollable exercise behavior accompanied by physiological or psychological symptoms. While its existence has been confirmed in young adults, it remains unclear whether the middle-aged and elderly adults exhibit similar neural structural correlates. This study aimed to identify neurostructural changes and the underlying impulsive mechanisms of exercise dependence in middle-aged and elderly adults. A total of 135 participants underwent structural magnetic resonance imaging (MRI) and completed psychological assessments. Whole-brain regression

Compositional Complexity in Text and Images.

Compositionality enables us to derive the meaning of a complex whole from the syntax and semantics of its individual parts. While compositional processing has primarily been studied in language, similar principles apply to visual scenes, in which meaning can be derived from their individual parts and the relationships between them. In this study, we therefore aimed to explore whether there is a shared neural basis for compositional processing in text and images. Based on abstract meaning representation graphs for text, commonly used to capture "who is doing what to whom", and action graph anno

Hyperacusis-inducing drug candidates.

Hyperacusis is a troubling loudness hypersensitivity disorder occasionally involving pain around ear and face. The neural origins of hyperacusis and conditions that evoke it are poorly understood. To gain insights into the neural mechanisms that might give rise to hyperacusis, a pharmacovigilance analysis was conducted on 3833 case reports in which hyperacusis was listed as a drug-induced adverse event in the United States Food and Drug Administration Adverse Event Reporting System (FAERS). The 10 drug classes ranked by the number of reported hyperacusis cases were selective (1) serotonin reup

Strategies to Reduce Rebound Pain and Facilitate Early Recovery After Transforaminal Endoscopic Lumbar Discectomy.

Background: Transforaminal endoscopic lumbar discectomy (TELD) is a minimally invasive and popular surgical method for the treatment of lumbar disc herniation. Although TELD offers favourable outcomes and enables fast recovery, some patients experience rebound pain and transient postoperative pain, which can delay rehabilitation and decrease patient satisfaction. Methods: This narrative review was conducted based on a comprehensive literature search of the MEDLINE database, supplemented by the author's clinical experience. Relevant articles were identified using the keywords "rebound pain" and

Genome-wide consensus transcriptional signatures identify synaptic pruning linking Alzheimer's disease and epilepsy.

Alzheimer's disease (AD) and epilepsy (EP) share a complex bidirectional relationship, yet the molecular mechanisms underlying their comorbidity remain insufficiently explored. To identify potential transcriptional programs across animal models and human patients with AD and EP, we conducted a comprehensive genome-wide transcriptomic analysis. Our investigation included mouse models of temporal lobe epilepsy (pilocarpine- and kainic acid-induced; n = 280), AD transgenic models (7 transgenic models expressing human tau or amyloid pathology; n = 257), and performed cross-species validation in hu

Estimation of 3D Ground Reaction Force and 2D Center of Pressure Using Deep Learning and Load Cells Across Various Gait Conditions.

Traditional force plate-based systems offer high measurement precision but are limited to laboratory settings, restricting their use in real-world environments. To address this, we propose a method for estimating a three-axis ground reaction force (GRF) and two-axis center of pressure (CoP) using a shoe embedded with three uniaxial load cells. The estimation was conducted under five gait conditions: straight walking, turning, uphill, downhill, and running. Data were collected from 40 healthy young adults. Four deep-learning models-Fully Connected Neural Network (FCNN), Convolutional Neural Net

Unveiling Hearts: Deep Learning-Based Electrocardiogram Classification for Congenital Heart Disease Detection.

The electrical activity of the human heart, recorded via an electrocardiogram (ECG), is characterized by distinct waveforms such as the P wave, QRS complex, and T wave. By analyzing the duration, morphology, and intervals between these waveforms, various cardiac disorders can be identified. This study aims to develop a deep learning-based approach for the accurate classification of congenital heart disease (CHD) using ECG data. We employed convolutional neural networks (CNNs) and recurrent neural networks (RNNs) to analyze ECG signals, leveraging their ability to detect multiple features in ti

Pharmacological Therapies for Consequences of Perinatal Hypoxic-Ischemic Brain Injury: Where Are We Now?

Despite significant progress in preclinical research aimed at developing effective therapies for the acute and long-term consequences of perinatal asphyxia, there is still a lack of clinical protocols to regenerate the neonatal brain damaged by hypoxic-ischemic (HI) injury. To date, only therapeutic hypothermia is routinely used in neonates who have experienced perinatal asphyxia. It has been shown to be effective only in limiting the spread of brain damage caused by a cascade of molecular and biochemical events triggered by limited blood supply to the body's organs, including the fragile, dev
