Oxidative stress plays a significant role in regulating the mammalian epigenome, with emerging evidence suggesting imbalances in the cellular redox state trigger stress-responsive epigenetic modifications that drive various human diseases. However, it remains unclear whether, like worms, epigenetic changes caused by redox imbalance or mitochondrial stress can move through the mammalian germline, potentially affecting the health of future generations. Antioxidant therapies are commonly used to reduce oxidative damage and are widely employed in cases of male infertility, where high-dose suppleme

BACKGROUND: Owing to their transcription factor binding sites, endogenous retroviruses (ERVs) can act as cis-regulatory-elements (CREs). By invading genomes in waves, ERVs offer a substrate for lineage-specific adaptations but also, when dysregulated, for lineage-specific disorders. Their role as CREs in rapid placental evolution, and in the human-specific placental disorder preeclampsia, may thus provide a paradigmatic exemplar. Here then we systematically identify ERV-derived CREs controlling human placental gene expression with dysregulation in preeclampsia.
RESULTS: We identify 87 ERV-der

BACKGROUND: Preeclampsia is a hypertensive disorder of pregnancy with major maternal and fetal consequences. While the molecular basis of early-onset preeclampsia is well studied, the mechanisms underlying late-onset disease-and how they differ by fetal sex-remain poorly understood. Placental transcriptomic profiling at term can reveal persistent molecular alterations reflecting cumulative disease processes.
METHODS: We conducted a cross-sectional observational analysis of placental gene expression using RNA sequencing in a subset of 58 term placentas (21 male-bearing and 37 female-bearing pr

INTRODUCTION: Tetrachlorobisphenol A (TCBPA), a BPA alternative, has been identified as a potential toxin, yet its reproductive toxicity remains poorly understood. This study aimed to investigate the effects of gestational TCBPA exposure on fetal development and placental function, focusing on syncytialization and TauT expression.
METHODS: Pregnant ICR mice were divided into three groups: TCBPA-treated (50 mg/kg/day), vehicle-treated (corn oil), and control (water). Treatments were administered until embryonic day 12.5 (E12.5) and 18.5 (E18.5). Embryos and placentas were collected at E12.5 fo

BACKGROUND: Advanced-maternal-age pregnancies carry a heightened risk of pre-eclampsia and fetal growth restriction (FGR), yet current first-trimester screening has limited predictive accuracy. We hypothesized that combining mid-pregnancy placental endocrine biomarkers with a Doppler-based vascular index would improve early identification of women at risk for these complications.
METHODS: In a prospective cohort at Zibo Central Hospital (January 2022-June 2024), 420 singleton pregnancies in women ≥35 years (first antenatal visit <14 weeks) underwent serum assays for human chorionic gonadotrop

Successful placental development and pregnancy rely on effective extravillous trophoblast (EVT) invasion. The mechanisms underlying inadequate EVT invasion in recurrent spontaneous abortion (RSA) remain unclear. WAS/WASL interacting protein family member 1 (WIPF1), the key regulator of cytoskeletal dynamics, is exclusively expressed in first-trimester placental EVTs. Knockdown experiments revealed WIPF1's crucial involvement in successful placental development; reduced levels impaired cell migration, while overexpression induced the opposite effects. Moreover, WIPF1 knockdown in hTSC-derived E

PURPOSE: Placental pathology is directly associated with adverse fetal outcomes congenital heart disease (CHD). However, in vivo placental dysfunction has been poorly characterized. We aimed to compare placental vascular reactivity (PLVR), the fetally-mediated placental response to transient changes in maternal carbon dioxide (CO2) quantified using BOLD MRI, in pregnancies with and without fetal CHD.
METHODS: In a prospective study, placental BOLD MRI was acquired concurrently with maternal end-tidal CO2 (EtCO2) for 7-8 min as pregnant women followed audio-visual breathing cues. Following mot

This study was designed to identify outcome data for non-invasive prenatal testing (NIPT) results suggestive of an atypical sex chromosome abnormality of fetal/placental origin. A single-center descriptive case series was performed between January 1, 2022 and August 1, 2024, which identified 16 cases, 11 of which completed diagnostic testing. Of those 11 cases, only 2 were found to have detectable chromosomal abnormalities of the fetus (monosomy X and mosaic monosomy X). The majority of the 9 cases without detectable fetal chromosome abnormalities cannot be assessed for the presence of confine

The pathogenesis of neuropsychiatric disorders, including autism spectrum disorder and schizophrenia, originates from complex interactions between genetic susceptibility and early environmental exposure. Infectious challenges during pregnancy are well-known environmental risk factors for neurodevelopmental disorders. Our previous research reported the interplay between maternal immune activation (MIA) and nucleotide-binding oligomerization domain-containing protein 2 (Nod2) signaling deficiency as potential genetic and environmental risk factors for schizophrenia pathogenesis. However, the mec

Chlorinated paraffins (CPs) are industrial chemicals detected widely in the environment and human tissues, raising concerns due to their persistence, bioaccumulation, and potential health risks. CPs with lower chlorination or shorter carbon chain lengths can cross the placenta and have been detected in breast milk, indicating fetal and early postnatal exposure. Both in vitro and in vivo studies suggest that CPs exhibit endocrine-disrupting properties, particularly affecting reproductive development. This study investigates the developmental effects and placental transfer of 1,2,9,10-tetrachlor

The proper embryo maintenance depends on tight maternal-fetal communications. C1q/TNF-related protein 6 (C1qtnf6)-encoding a hormone-like CTRP6 protein-exhibits placenta-enriched expression and its deficiency causes partial fetal loss. Here, we demonstrate that abundant C1qtnf6 expression is distinctive for hemochorial placentae, which have deep uterine invasion. C1qtnf6 is most highly expressed in human extravillous trophoblasts and mouse spiral artery-associated trophoblast giant cells, which are functionally analogous cells that invade uterines. The partial embryo loss is mainly due to impa

2-Ethylhexyl-diphenyl phosphate (EHDPP) is an organophosphate ester (OPE) with roles of flame retardant and plasticizer. It is widely used in various applications, detected in environmental matrices and human body, threatening ecological environment and human health. Some OPEs have been reported to disturb the gut microbiota, the gut microbiota mediates placental function. Our previous study showed EHDPP causes placental toxicity and fetal weight loss, it is unknown that whether EHDPP affects fetal development through the gut-placenta axis and whether it is feasible to fight against EHDPP indu

High-altitude pregnancy presents the complex physiological challenge of fulfilling maternal, placental, and fetal metabolic demands under chronic ambient hypoxia. Highland Andeans exhibit signs of adaptation to high-altitude hypoxia, showing relative protection against altitude-associated fetal growth restriction (FGR) and the positive selection of metabolic genes linked to placental mitochondrial capacity. Not all infants are protected, with both FGR and preeclampsia occurring among highland-resident Andeans. In Andeans, placental metabolic dysfunction is evident. By integrating metabolomic s

OBJECTIVE: To evaluate whether plasma levels of placental extracellular vesicles (pcEVs), the EV-scavenging factor lactadherin, and prothrombotic markers predict fetal growth restriction (FGR) and/or fetal distress (FD) in established severe pre-eclampsia (sPE).
METHODS: We recruited 80 sPE patients, 41 normal pregnancies, and 27 non-pregnant women. SPE patients were further dichotomized into event and non-event groups based on the occurrence of FGR/FD during a follow-up period of 77 days. Plasma levels of pcEVs, lactadherin, von Willebrand factor (VWF), antithrombin (AT), and tissue plasmino

Zinc is an essential trace element. The regulatory mechanism of zinc and its transporters in fetal growth in monochorionic diamniotic (MCDA) twins with selective intrauterine growth restriction (MCDA-sIUGR) is unclear. A total of 45 MCDA twins were divided into two groups, MCDA (n=37) and MCDA-sIUGR (n=8), to investigate their possible effects on fetal growth. Maternal and fetal serum zinc levels were measured using a microassay. Immunohistochemistry and western blotting were performed to quantify the expression and localization of zinc transporters. Correlation scatter plots and matrices were

BACKGROUND: Tuberculosis (TB) poses a significant risk during pregnancy and the postpartum period, with evidence linking these stages to an increased likelihood of progression from latent TB infection to active disease. Although TB has been associated with adverse pregnancy outcomes, including congenital transmission, the mechanisms by which Mycobacterium tuberculosis (Mtb) affects placental structure and function remain poorly understood.
OBJECTIVE: This study aimed to investigate the stage-dependent effects of Mtb infection on human placental tissue and assess the potential for vertical tra

BACKGROUND: Placental malaria (PM) is among the leading public health problems associated with adverse outcomes for the mother, the foetus, and the newborn, particularly in sub-Saharan Africa (SSA). Although there are many studies published on PM in SSA, comprehensive evidence for targeted interventions is limited. Thus, this study aimed to synthesize evidence on the prevalence, predictors, and adverse perinatal outcomes of PM in SSA.
METHODS: Pertinent studies online searching, selection, data extraction, and quality assessment were conducted. Data were extracted using Microsoft Excel and an
