Renal phenotype of low kallikrein rats.

BACKGROUND: Renal kallikrein has been linked with inheritance of arterial hypertension and with sensitivity to drug nephrotoxicity. Identification of a cause--effect relationship between low kallikrein and intermediate phenotypes has been hampered by the lack of adequate animal models. METHODS: Kallikrein was measured in tissues obtained from rats inbred for low urinary kallikrein excretion (LKR) and wild-type controls. Blood pressure and indices of myocardial contractility were recorded via an intraventricular cannula connected to a transducer. The functional relevance of endogenous angioten

Identification of new Fas mutations in a patient with autoimmune lymphoproliferative syndrome (ALPS) and eosinophilia.

Autoimmune lymphoproliferative syndrome (ALPS) is a rare, newly recognized, chronic lymphoproliferative disorder in children and is characterized by lymphadenopathy, splenomegaly, pancytopenia, autoimmune phenomena and expansion of double-negative (DN) T lymphocytes (TCR alpha beta+, CD4-, CD8-). Defective lymphocyte apoptosis caused by mutations of the Fas (CD95) gene has been linked in the pathogenesis of ALPS, as binding of Fas-ligand to Fas can trigger apoptosis. Of the ALPS cases reported to date, point mutations, frameshifts and silent mutations in Fas all have been identified. We report

Gene encoding a novel murine tissue inhibitor of metalloproteinases (TIMP), TIMP-3, is expressed in developing mouse epithelia, cartilage, and muscle, and is located on mouse chromosome 10.

Remodeling of the extracellular matrix (ECM) is an essential component of normal development and is also involved in the pathogenesis of arthritis and the spread of cancer. The matrix metalloproteinases and their natural inhibitors, the tissue inhibitors of metalloproteinases (TIMPs), play an important role in this context. We have isolated mouse cDNA clones encoding a novel member of the TIMP family, designated TIMP-3. We have assigned the Timp-3 locus to the [C1-D1] region of mouse chromosome 10 using both genetic and cytogenetic methods. The conceptual translation product of the Timp-3 cDNA

Branching Morphogenesis of Mouse Embryonic Submandibular Epithelia Cultured under Three Different Conditions: (mouse submandibular gland/epithelial branching/morphogenesis/collagenase/heparitinase/heparin/Matrigel).

To investigate how the mesenchyme interacts with the epithelium, we employed three different culture systems: System A, in which intact submandibular gland rudiments at the mid 13-day stage were cultured on Millipore filters; System B, in which the 13-day epithelium and mesenchyme were separated once with dispase, recombined again, and cultured on the filter; System C, in which the separated 13-day epithelium was clotted with Matrigel and cultured with the mesenchyme across the filter or in the presence of EGF instead of the mesenchyme. In Systems A and B, 13-day epithelia expanded and produce

Angioimmunoblastic lymphadenopathy with dysproteinemia and dermal T-cell lymphoma.

BACKGROUND: T-cell receptor (TCR)-gamma gene rearrangements provide a specific clonal marker for T-cell malignancies of both the alpha beta and gamma delta varieties. A polymerase chain reaction (PCR)-based method was used in this study for investigation of clonal TCR-gamma gene rearrangements in a patient with a classical presentation of angioimmunoblastic lymphadenopathy with dysproteinemia (AILD) that subsequently progressed into an indolent form of dermal T-cell lymphoma. METHODS: TCR gene rearrangements in patients with cutaneous T-cell lymphoma (CTCL) were examined using conventional So

Superior function of a bicuspid over a monocuspid patch for reconstruction of a hypoplastic pulmonary root in pigs.

In an experimental study in pigs, the function of monocuspid and bicuspid patches was compared to improve surgical reconstruction of a hypoplastic pulmonary root. Segments from glutaraldehyde-treated porcine aortic roots served as monocuspid and bicuspid patches. Their leaflets and commissures were marked with radiopaque metal clips. Marker movements were recorded at 100 frames/sec. Quantification of pulmonary insufficiency, right ventricular stroke volume, as well as pressures in the right ventricle, pulmonary artery, and aorta was performed before and after patch plasty. The cyclic motion of

The internal anal sphincter: mechanisms of control and its role in maintaining anal continence.

The human IAS has particular structural and functional characteristics. This smooth muscle constantly generates rhythmic electrical slow waves, but no action potentials. The slow waves are linked to calcium fluxes and both are essential for mechanical activity, i.e. the ASPW. The IAS is pharmacologically characterized by the presence of alpha excitatory and beta inhibitory adrenergic receptors. Cholinergic drugs have an indirect effect through the release of an inhibitory neurotransmitter, very probably VIP, from NANC nerves. The myogenic activity of the IAS is enhanced by its extrinsic sympat

Clinical pharmacokinetics of local anaesthetics.

The introduction of the new long acting local anaesthetics, bupivacaine and etidocaine, has stimulated an expansion of interest in regional anaesthesia, particularly for obstetrical applications and pain therapy. System toxicity following injection of local anesthetics occurs albeit infrequently, and tentative correlations have been made between the onset of CNS and cardiovascular effects and circulating drug concentrations in both adults and neonates. Amongst other factors, interpretation of these relationships depends upon blood distribution and plasma binding of the agents, sampling sites a

Human adenovirus-induced medulloepitheliomatous neoplasms in Sprague-Dawley rats.

A direct causal relationship between a human virus and malignant transformations in target cells (sensory neuronal precursors) was suggested by the development of a medulloepitheliomatous neoplasm in the central nervous system. Twenty-six newborn Sprague-Dawley rats were given a single intracerebral inoculation of 0.05 ml of adenovirus fluid, 10(3.5) to 10(4.5) TCID(50) HeLa cells/0.1 ml, in the left frontal lobe. Within 37 to 151 days after the virus inoculation, 23 (88.7%) rats autochthonously developed an adenovirus-typical neoplasm in the central nervous system. Nine animals developed a mu

A Probiotic Approach Targeting Hypersialylation Revives CD8+ T-cell Immunity and Boosts Photodynamic Therapy Efficacy in Solid Tumors.

UNLABELLED: Excess terminal sialic acids on tumor glycans engage inhibitory Siglec receptors and mask peptide and glycan epitopes, including major histocompatibility complex class I (MHC-I), thereby reducing antigen visibility and dampening cytotoxic T-cell surveillance. In this study, we showed that a compact, lectin-free Salmonella typhimurium sialidase (Sia) efficiently desialylated melanoma and colorectal carcinoma cells, abolished Siglec-E binding, and spared MHC-I from lysosomal degradation. Functionally, Sia treatment enhanced dendritic cell phagocytosis and tumor antigen presentation,

Blockade of the SP/NK1 receptor axis: multimodal neuroprotection via inflammatory suppression, hypoxic modulation, and lympho-vascular remodeling in global cerebral ischemia.

Transient global cerebral ischemia (tGCI), often precipitated by cardiac arrest, triggers severe neuroinflammation, oxidative stress, and hypoxia, culminating in diffuse delayed neuronal death and debilitating cognitive-affective dysfunctions. Substance P (SP) and its neurokinin-1 (NK1) receptor act as critical upstream mediators in this pathological cascade. To interrogate this biological pathway and rescue acute stroke-induced behavioral deficits, this study investigated the multifaceted neuroprotective efficacy of the NK1 receptor antagonist maropitant, utilizing a rat two-vessel occlusion

Gut microbiota dysbiosis aggravates liver injury in acute-on-chronic liver failure through D-lactate accumulation.

INTRODUCTION: Acute-on-chronic liver failure (ACLF) is a life-threatening condition arising from the abrupt worsening of pre-existing chronic liver disease, typically accompanied by multi-organ failure and markedly elevated short-term mortality. Gut microbiota dysbiosis is frequently observed in patients with ACLF, yet the mechanisms by which microbial alterations contribute to disease progression remain unclear. Here, we investigated whether gut microbiota dysbiosis could exacerbate liver injury in ACLF and explored the underlying mechanisms. METHODS: The correlation between serum lactic aci

A new era for optic pathway glioma: A developmental brain tumor with life-long health consequences.

Optic pathway and hypothalamic glioma (OPHG) are low-grade brain tumors that arise from any part of the visual pathways frequently involving the hypothalamus. The tumors grow slowly and present with features driven by their precise anatomical site, their age at presentation and the stage of growth and development of the host neural and orbital bony tissues. Up to 50% of optic pathway glioma arise in association with Neurofibromatosis type 1 (NF1), which affects 1 in 3,000 births and is a cancer predisposition syndrome. As low-grade tumors, they almost never transform to malignant glioma yet th

Variable alterations in frequencies of CRISPR/Cas9-induced nucleotide deletion and insertion in transgenic maize callus tissues exposed to chromatin modifiers.

Given the growing emphasis on the efficient application of CRISPR/Cas9 in plant genome editing, there is a need for methodologies that enhance targeted mutagenesis efficiency. In the present study, we utilized stable transgenic maize callus tissues harboring active CRISPR/Cas9 vector systems targeting either the abnormal cytokinin response 1 (ARE1) gene or the potassium channel gene (KAT2). The presented DNA sequencing data indicate that the KAT2-targeting vector, which utilizes a 3'-end truncated guide RNA, retained mutagenic activity, albeit at a notably low frequency. We quantified mutageni

Aging as a Potential Modulator of Extramedullary Hematopoiesis: A Hypothesis-Generating Niche-HSPC Competence-Trigger Model.

Extramedullary hematopoiesis (EMH) is a compensatory response to hematopoietic stress that occurs unevenly across disorders with ineffective erythropoiesis (IE). The mechanisms underlying differing EMH patterns despite similarly severe IE remain poorly understood. Thalassemia and congenital dyserythropoietic anemias frequently develop overt EMH, whereas myelodysplastic syndromes (MDS), despite severe IE, rarely show overt EMH. We propose the Niche- HSPC Competence-Trigger Model as a hypothesis-generating framework in which EMH expression reflects the interaction among erythropoietic trigger lo

Restoration of Mucosa-Associated Invariant T-Cell Function in Healthcare-Associated Bacterial Infections Supports Recovery of Carbapenem Efficacy Against Resistant Bacteria Ex Vivo.

BACKGROUND: Antimicrobial resistance (AMR) is a major challenge in healthcare-associated infections (HAIs), especially in immunocompromised individuals and those with comorbidities, who often have an impaired mucosa-associated invariant T (MAIT) cell pool. MAIT cells are innate-like T cells enriched in mucosal tissues with potent antimicrobial activity. Restoring their function may offer a host-directed strategy against drug-resistant pathogens. METHODS: We evaluated how cognate antigen in combination with cytokines modulates MAIT cell cytotoxicity and enhances carbapenem activity. MAIT cell

Quantitative evaluation of T-cell repertoire restoration following hematopoietic stem cell transplantation in patients with and without graft versus host disease.

INTRODUCTION: Acute graft-versus-host disease (aGVHD) is a major cause of mortality following hematopoietic stem cell transplantation (HSCT). Its pathogenesis is primarily driven by alloimmune T cells, which recognize host tissues as foreign. A deeper characterization of the T cell receptor (TCR) repertoires in patients with aGVHD could provide critical insights into the disease's mechanisms and identify potential predictive biomarkers. METHODS: We employed next-generation sequencing to comprehensively profile the T cell receptor alpha (TRA) and beta (TRB) chains in HSCT recipients, comparing
