Exploiting Bridged Conformations for Precise Molecular Recognition in the Design of KRAS-Selective, Orally Available Macrocyclic Peptides.

Macrocyclic peptides are attracting attention as a promising therapeutic modality for addressing intracellular targets that are traditionally difficult to address with conventional small molecules or antibodies. In this study, we present the chemical optimization process behind the development of AUBE00 (AP7400, compound 30), a KRAS-selective inhibitor based on a scaffold similar to LUNA18, an orally bioavailable macrocyclic peptide RAS inhibitor. Achieving KRAS isoform selectivity is exceptionally challenging due to the nearly identical backbone structures of KRAS, HRAS, and NRAS (Cα RMSD < 1

Lipophilicity and Steric Constraints Govern V-Shaped Inhibition of Human and Rat Placental 3β-Hydroxysteroid Dehydrogenase by Dialkyldimethylammonium Disinfectants: Implications for Progesterone Dysregulation.

Dialkyldimethylammonium chemicals (DDAs) are quaternary ammonium disinfectants. However, their effects on progesterone synthesis in placentas remain unclear. This study aims to study structure-activity relationship (SAR) and mechanism of DDAs (C2-C18) as inhibitors of human (3β-HSD1) and rat (3β-HSD4) placental 3β-hydroxysteroid dehydrogenase/Δ5-Δ4 isomerase (3β-HSD), a key enzyme in progesterone synthesis. DDAs exhibited a distinct V-shaped SAR to inhibit 3β-HSD, which was dependent on alkyl chain length. The inhibitory potency peaked with the C12 analogue, demonstrating the highest potency i

Seminal Plasma Metabolomics Reveals Potential Metabolic Pathways Linking Organophosphate Flame Retardant Exposure to Impaired Semen Quality.

Organophosphate flame retardants (OPFRs) may affect semen quality, but the mechanisms remain unclear. This study included 157 men from the SEARCH cohort and evaluated the associations between urinary OPFR levels and semen quality, and the role of the seminal plasma metabolome. Overall, bis(2-butoxyethyl) phosphate (BBOEP) was negatively associated with progressive motility (β [95% confidence interval (CI)]: -0.090 [-0.175, -0.005]), and some associations between OPFRs and semen quality were more pronounced among men with male-factor infertility. Furthermore, widespread associations were observ

Jellyfish-inspired multilayered responsive hydrogels via micelle-confined photopolymerization for underwater communication and emergency rescue.

Hydrogels offer considerable promise as flexible sensors. However, maintaining their stable performance in complex underwater environments characterized by high pressure, high salinity, and temperature fluctuations remains a significant challenge. Inspired by the dynamic adaptive capabilities of jellyfish, this study reports a biomimetic composite conductive hydrogel with multi-level environmental responsiveness, synthesized via the copolymerization of N-isopropylacrylamide, 2-acrylamido-2-methylpropanesulfonic acid, and octadecyl methacrylate (OMA), incorporating color-changing microcapsules

Persistent chemotherapy-induced alopecia: a scoping review.

PURPOSE OF REVIEW: Persistent chemotherapy-induced alopecia (pCIA) is defined as incomplete hair growth ≥6 months after chemotherapy completion. This scoping review aimed to identify gaps for future research by mapping pCIA's incidence, risk factors, quality-of-life (QoL) impacts, presentation, prevention, and management. RECENT FINDINGS: Thirty-nine articles were included. The most common study type was case reports (12/39, 30.8%). Only 2/39 (5.1%) were randomized controlled trials (RCTs). Most studies (27/39, 69.2%) reported pCIA in patients with breast cancer. Alkylating agents, taxanes, a

Cooperative Fe-LMCT and Ni-SH2 Catalysis for Native Carboxylic Acid Methyl Editing.

Native functional-group conversion represents a powerful complement to C-H methylation, yet the selective installation of methyl groups directly from free carboxylic acids remains an outstanding challenge. Herein, we report a reaction design that addresses these challenges through the integration of Fe-mediated ligand-to-metal charge transfer (LMCT) photocatalysis and Ni-mediated SH2 radical-sorting. Native carboxylic acids are directly converted into alkyl radicals through catalytic Fe-LMCT activation, whereas selective C(sp3)─C(sp3) bond formation is achieved by Ni-mediated SH2 catalysis. Ce

S = 3/2 Iron(I)-Bisphosphine-Ate Complexes: Synthesis, Characterization, and Reactivity for Electrophile Activation.

Iron bisphosphine complexes represent an important class of sustainable catalysts for a variety of important chemical transformations due to the high abundance, low toxicity, and redox-rich chemistry inherent to iron-based systems. In particular, transient, low-coordinate Fe(I) species are postulated to play a pivotal role in multiple catalytic transformations as the principal species for C─X bond activation (X = Cl, Br, I). However, insight into the electronic structure, bonding, and reactivity of Fe(I)-bisphosphine complexes remains poorly developed. In the present study, we report the devel

Redirecting Excited-State Energy Flow Through Programmed Supramolecular Confinement in Twisted Photosensitizers.

Programmed supramolecular confinement is introduced as a physical strategy to investigate aggregation-enhanced reactive oxygen species (ROS) generation in twisted intramolecular charge transfer (TICT)-active twisted photosensitizers without changing the conjugated photosensitizing unit. In this design, TICT functions as a confinement-sensitive regulator of excited-state energy dissipation, while cell membranes (CM) serve as supramolecular platforms that confine photosensitizers within their compact phospholipid bilayer architectures. Through alkyl-chain engineering, the obtained photosensitize

Green Extraction of Phenolic Compounds From Pistacia lentiscus Leaves Coupled With LC-MS Characterization and Structure-Based Enzyme Inhibition Analysis.

Acetylcholinesterase (AChE) and urease are critical enzymes involved in various pathological conditions associated with cellular and tissue damage. Natural products are increasingly explored as safer alternatives to synthetic inhibitors. This study evaluates the phytochemical composition and enzyme inhibitory potential of Pistacia lentiscus leaves using a green extraction approach. Bioactive compounds were extracted via lactic acid-assisted ultrasound, and the crude extract was characterized for total phenolics, flavonoids, LC-MS profile, and in vitro inhibition of acetylcholinesterase and ure

Structural basis of ligand recognition and gating in mosquito IRs.

Ionotropic receptors (IRs) are essential for insect olfaction, enabling the detection of carboxylic acids, amines, moisture, and temperature cues that mediate critical behaviors, including mosquito host-seeking 1,2. However, the structural mechanisms underlying ligand specificity and channel activation in these receptors remain unclear. Here, we report cryo-electron microscopy (cryo-EM) structures and functional validation of the IR8a-IR75k complex from the malaria mosquito Anopheles stephensi, a carboxylic acid detector, captured in four states: the ligand-free full-length closed form (apo st

Non-Negligible Source of PFOS: Biochemical Fate and Metabolic Activation of Precursors.

The widespread detection of PFOS precursors in the environment and human matrices raises a new wave of health concerns. However, their absorption, distribution, metabolism, and excretion (ADME) fates and toxicity profiles in mammals remain largely unexplored. Here, we investigated the toxicokinetic, metabolic transformation, and toxicity of three precursors, N-MeFOSA, N-EtFOSA, and N-EtFOSAA, by integrating toxicokinetic modeling in mice, rat liver microsome metabolism, and hepatocellular toxicity assays. N-MeFOSA and N-EtFOSA were rapidly eliminated in vivo and exhibited high efficiencies of

Adsorption-Engineered Hydrocarbon Ionomers for Durable Proton-Exchange Membrane Fuel Cells.

Reducing reliance on perfluoroalkyl substances (PFAS) in proton-exchange membrane fuel cells requires hydrocarbon ionomers that combine high performance with long-term durability, a persistent challenge in catalyst-layer design. Here, we identify oxidation-driven ionomer-catalyst interfacial degradation as a dominant failure pathway in hydrocarbon ionomer-bonded cathodes and introduce an adsorption-engineering strategy to overcome this limitation. The comparison of a commercial sulfonated poly(phenylene) (Pemion) with structurally engineered sulfonated poly(fluorene)s demonstrated that electro

Development and biological evaluation of new nano copolymer-drug composites designed for selective liver cancer treatment.

This study focuses on the design, synthesis, characterization, and biological evaluation of two novel nano-graft copolymer-drug composites (B3 and B4) for targeted liver cancer therapy. A new nano-graft copolymer was first synthesized via the reaction of phthalic anhydride with glycerol, followed by conversion to its acid chloride derivative (B2). This intermediate was successfully used to graft the therapeutic agent's ampicillin (B3) and 4-aminoantipyrine (B4). Structural confirmation of the grafted composites was achieved using FT-IR, 1H NMR, and 13C NMR spectroscopy, which verified the form

D2O-Labeled Single-Cell Raman Spectroscopy Reveals the Evolutionary Trajectory of Helicobacter pylori During the Phenotypic Silent Window.

Helicobacter pylori is a carcinogenic pathogen with a high gastric cancer risk. The rapid emergence of antibiotic resistance severely compromises treatment efficacy. However current conventional approaches fail to capture early molecular events preceding detectable resistance. Here, we used a gradient intermittent antibiotic exposure model to simulate stepwise resistance evolution and longitudinally acquired single-cell Raman spectra across adaptive cycles. By integrating two-dimensional correlation Raman spectroscopy (2D-COS), we resolved the temporal sequence of molecular alterations during
