[Takayasu's arteritis in pregnancy: report seven cases].

Takayasu's arteritis is a chronic and non-specific disease of young women in reproductive age that primarily affects the aorta, its branches and the pulmonary artery. Ramirez Cueto G. and Fernandez Del Castillo C. et al. published a case of pregnancy in Mexico and Takayasu's arteritis in 1968. There are no reports of this disease in pregnancy since. The purpose of this study is to describe the clinical course and perinatal outcome of seven pregnant patients with known diagnosis of Takayasu arteritis. The clinical course, laboratory findings, angiographic findings and perinatal outcomes were as

Study of the evolution of the clinical profile of heart donors.

UNLABELLED: Certain characteristics of the donor have implications for the early and late survival of transplants. The trends show a progressive worsening in the clinical profile of heart donors. Our objective was to analyze changes in the clinical characteristics of donors. MATERIALS AND METHODS: This retrospective study of 396 donors was divided into three periods: 1990 to 1994, 1995 to 1999, and 2000 to 2004. We excluded combination transplantations, retransplantations, and pediatric transplantations. The donor variables included age, cause of death, cardiorespiratory arrest, ICU stay, and

Suicidal high falls.

There are few data on committing suicide by jumping from a height. Information on suicidal high falls in southeast Scotland was prospectively gathered over 7 years (1992-1998). Data sources included ambulance, police, hospital and forensic records. Injuries sustained were scored according to the Abbreviated Injury Scale, generating Injury Severity Scores (ISS). Sixty-three individuals (50 males), appeared to have committed suicide by falling from a height. The backgrounds were diverse, but 44 individuals had known previous psychiatric illness, 18 having attempted suicide before. The most commo

Biomimetic Macrophage Cell Membrane-Based Nanoparticles for Effective Treatment of Glioblastoma Through Boron Neutron Capture Therapy Combined With Immunotherapy.

Glioblastoma multiforme (GBM) remains largely incurable due to the blood-brain barrier (BBB) and immunosuppressive microenvironment. While boron neutron capture therapy (BNCT) selectively eradicates tumor cells via 10B(n, α)-7Li reactions, its clinical potential in GBM is unrealized because of the suboptimal pharmacokinetics of conventional boron agents and their failure to achieve therapeutic intracranial concentrations. Here, we design macrophage membrane-coated PLGA nanoparticles (M@PLGA-10BN) that exploit innate biological transport mechanisms to overcome dual therapeutic barriers. The mac

Microcephaly-like phenotype triggered by novel reassortant and prototypic Oropouche virus strains in brain organoids.

BACKGROUND: Oropouche virus (OROV) is an emerging arbovirus currently spreading across South America, with increasing reports of neurological manifestations, severe systemic disease, and congenital abnormalities. Although traditionally associated with mild febrile illness, the recent geographic expansion and surge in OROV outbreaks have prompted attention to its neurotropic potential. METHODS: We investigated the effects of Oropouche virus (OROV) infection on neural stem cells (NSCs) and brain organoids derived from human induced pluripotent stem cells, using both an emergent reassortant isol

Exerkine-Mediated Regulation of the NLRP3 Inflammasome in Neuroprotection: Mechanistic Insights and the Role of Exercise.

Neurodegeneration is a leading cause of long-term disability and cognitive impairment, and the aberrant activation of the NOD-like receptor protein 3 (NLRP3) inflammasome is closely implicated in its pathogenesis. The NLRP3 inflammasome, as a central mediator of inflammatory cascades, can, when excessively activated, promote neuroinflammation and glial polarization, induce neuronal death, disrupt the blood-brain barrier, suppress angiogenesis and neurogenesis, impair synaptic plasticity, and induce inflammaging, ultimately leading to neurodegeneration. Exerkines, including neurotrophic factors

Pseudo-SAH in a patient with methanol poisoning: a case report.

BACKGROUND: Methanol is a toxic alcohol found in many industrial products. Ingestion can cause severe metabolic acidosis, optic nerve damage, and central nervous system depression. While bilateral putaminal necrosis is a typical radiologic finding, pseudo-subarachnoid hemorrhage (pseudo-SAH) is a rare and often misdiagnosed complication. Prompt recognition is essential to avoid unnecessary interventions. CASE PRESENTATION: A 31-year-old Caucasian Iranian male presented unresponsive after consuming 1-2 L of homemade methanol. He experienced cardiac arrest upon arrival but was successfully resu

Calpastatin, a calpain specific inhibitor, reduce seizures in a mouse model of temporal lobe epilepsy.

Epilepsy is a chronic condition characterized by unpredictable and recurrent spontaneous seizures. In a previous study, we reported that pharmacological inhibition of calpain prevented epileptogenesis in the rat pilocarpine model. In this study, we demonstrate that transgenic overexpression of calpastatin, the endogenous inhibitor of calpain, reduces calpain activation and lessens seizure burden in the mouse intrahippocampal kainate model. Blockade of calpain activation was evidenced by a reduction in the generation of spectrin breakdown products, a hallmark of calpain activation. CAST overexp

Brain-Targeted Near-Infrared Chiral TeSe Nanodrug Against Orthotopic Drug-Resistant Glioma.

Glioblastoma multiforme (GBM) remains a formidable therapeutic challenge due to its high invasiveness, the highly restrictive blood-brain barrier (BBB), and frequent drug resistance, necessitating new therapeutic strategies. Here, we developed tellurium-selenium nanoparticles (TeSe NPs) with exceptional near-infrared (NIR) chiroptical properties and potent antitumor activity against drug-resistant glioma for the first time. Notably, the right-handed nanoparticles (D-TeSe NPs) exhibited higher cellular uptake than left-handed L-TeSe NPs through chirality-selective recognition by drug-resistant

Sex difference in brain functional connectivity of hippocampus in Alzheimer's disease.

Alzheimer's disease (AD), affecting nearly 6.5 million people, is the fifth leading cause of death in individuals 65 years or older in the USA. Prior research has shown that AD disproportionality affects females; females have a greater incidence rate, perform worse on a variety of neuropsychological tasks, and have greater total brain atrophy. Recent research has linked these sex differences to neuroimaging markers of brain pathology, such as hippocampal volumes. Specifically, research from our lab found that functional connectivity from the hippocampus to the precuneus cortex and brain stem w

HMGB1, angel or devil, in ischemic stroke.

INTRODUCTION: High-mobility group box 1 protein (HMGB1) is extensively involved in causing ischemic stroke, pathological damage of ischemic brain injury, and neural tissue repair after ischemic brain injury. However, the precise role of HMGB1 in ischemic stroke remains to be elucidated. METHODS: Comprehensive literature search and narrative review to summarize the current field of HMGB1 in cerebral ischemic based on the basic structure, structural modification, and functional roles of HMGB1 described in the literature. RESULTS: Studies have exhibited the crucial roles of HMGB1 in cell death,

Specific inhibition of NADPH oxidase 2 modifies chronic epilepsy.

Recent work by us and others has implicated NADPH oxidase (NOX) enzymes as main producers of reactive oxygen species (ROS) following a brain insult such as status epilepticus, contributing to neuronal damage and development of epilepsy. Although several NOX isoforms have been examined in the context of epilepsy, most attention has focused on NOX2. In this present study, we demonstrate the effect of gp91ds-tat, a specific competitive inhibitor of NOX2, in in vitro epileptiform activity model as well as in temporal lobe epilepsy (TLE) model in rats. We showed that in in vitro seizure model, gp91

CDR1as regulates α-synuclein-mediated ischemic brain damage by controlling miR-7 availability.

Transient focal ischemia decreased microRNA-7 (miR-7) levels, leading to derepression of its major target α-synuclein (α-Syn) that promotes secondary brain damage. Circular RNA CDR1as is known to regulate miR-7 abundance and function. Hence, we currently evaluated its functional significance after focal ischemia. Transient middle cerebral artery occlusion (MCAO) in adult mice significantly downregulated both CDR1as and miR-7 levels in the peri-infarct cortex between 3 and 72 h of reperfusion. Interestingly, neither pri-miR-7a nor 7b was altered in the ischemic brain. Intracerebral injection of

Application of Nano-based Drug Loading Systems in the Treatment of Neurological Infections: An Updated Review.

Infection of the central nervous system (CNS) is a global healthcare concern with high rates of death and disease. CNS infections mainly include meningitis, encephalitis, and brain abscesses. Bacteria, viruses, fungi, protozoa, and parasites are the most common causes of neuroinfections. There are many types of medications used in the treatment of CNS infections, but drug delivery through the blood-brain barrier (BBB) is a major challenge to overcome. The BBB is a specialized multicellular barrier separating the neural tissue from the peripheral blood circulation. Unique characteristics of the

The Role of DNA Damage in Neural Plasticity in Physiology and Neurodegeneration.

Damage to DNA is generally considered to be a harmful process associated with aging and aging-related disorders such as neurodegenerative diseases that involve the selective death of specific groups of neurons. However, recent studies have provided evidence that DNA damage and its subsequent repair are important processes in the physiology and normal function of neurons. Neurons are unique cells that form new neural connections throughout life by growth and re-organisation in response to various stimuli. This "plasticity" is essential for cognitive processes such as learning and memory as well

Mesenchymal stromal cell-derived extracellular vesicles afford neuroprotection by modulating PI3K/AKT pathway and calcium oscillations.

Mesenchymal stromal cells (MSC) are widely recognized as potential effectors in neuroprotective therapy. The protective properties of MSC were considered to be associated with the secretion of extracellular vesicles (MSC-EV). We explored the effects of MSC-EV in vivo on models of traumatic and hypoxia-ischemia (HI) brain injury. Neuroprotective mechanisms triggered by MSC-EV were also studied in vitro using a primary neuroglial culture. Intranasal administration of MSC-EV reduced the volume of traumatic brain damage, correlating with a recovery of sensorimotor functions. Neonatal HI-induced br

A Novel Anti-Inflammatory d-Peptide Inhibits Disease Phenotype Progression in an ALS Mouse Model.

Amyotrophic lateral sclerosis (ALS) is a progressive neurodegenerative disease characterised by selective neuronal death in the brain stem and spinal cord. The cause is unknown, but an increasing amount of evidence has firmly certified that neuroinflammation plays a key role in ALS pathogenesis. Neuroinflammation is a pathological hallmark of several neurodegenerative disorders and has been implicated as driver of disease progression. Here, we describe a treatment study demonstrating the therapeutic potential of a tandem version of the well-known all-d-peptide RD2 (RD2RD2) in a transgenic mous
