Mesenchymal Stromal Cells Improve Islet β-Cell Functional Survival: Analysis of Extracellular Vesicle-Trafficked Proteins and miRNAs.

Type 1 diabetes is caused by autoimmune destruction of insulin-secreting β-cells within islets of Langerhans. Transplantation of donor islets can improve glycaemic control, but current clinical islet transplantation protocols are compromised by extensive loss of β-cell functional mass soon after implantation. Co-incubation in vitro or co-transplantation in vivo of mesenchymal stromal cells (MSCs) with isolated islets improves their functional survival, although the underlying mechanisms remain obscure. Here, we show that MSC-derived extracellular vesicles (MSC-EVs) are alone sufficient to reca

Targeting chitinase-3-like protein-1 in pancolitis by a drug-repurposing approach.

Pancolitis is a severe form of ulcerative colitis. Inflammation affects the entire colon and increases carcinogenesis. Standard therapies often reveal toxicities. We performed transcriptomics of ileum and colon biopsies from pancolitis patients and healthy individuals. We identified 77 differentially expressed genes in the ileum and 89 in the colon. Nine genes were overexpressed in both regions (TCL1A, DUOXA2, CC18, S100A8, CHI3L1, PTGDS, MMP3, CXCL1, and GREM1). We focused on CHI3L1 (chitinase-3-like protein-1) and confirmed its protein expression by immunohistochemistry. Based on gene expres

Regulatory Functions of TDP-43 and FMRP in Non-Neuronal Diseases: Are Co-Targeted mRNAs the Keys?

RNA binding proteins (RBPs) act as the central nodal point in shaping the cellular transcriptome through their involvement in various aspects of RNA metabolism including stability, splicing, polyadenylation, modifications, translation and transport. Dysregulation in the function of various RBPs can be associated with different human pathophysiological conditions. Owing to their ability to regulate various RNA metabolism-associated processes, the same RBPs can functionally be involved in human pathologies with distinct underlying pathophysiological mechanisms. Two such important RBPs, namely TD
